내분비 교란성 DEHP의 항-에스트로젠 활성에 관한 연구 (Study on Anti-estrogenic Activity of DEHP as an Endocrine Disruption Chemical)

  • DEHP는 대표적인 플라스틱 가소제 가운데 하나로서, 광범위하게 사용되고 있으며, 내분비계 장애물질로 분류퇴고 있다. 실제로 하천, 해양, 토양 등 광범위한 환경에서 검출되고 있지만, 이 물질이 수서 생물에 미치는 내분비교란 영향과 기작에 대해서는 거의 알려진 바가 없다. 본 연구에서는 송사리로 불리우는 Oryzias latipes(Japanese medaka)와, 유생 시기에 저니성 무척추 동물로 존재하다가 성충이 되는 Chironomus riparius를 대상으로 DEHP가 내분비 장애물질로서 생식작용에 미치는 영향에 대하여 연구하였다. 먼저, Japanese medaka를 부화 직후부터 3개월간 DEHP 1, 10, 50 $\mu\textrm{g}$/l의 농도로 노출시킨 결과 암놈의 혈중 비텔로제닌의 감소 ,생식소 지수인 GSI(Gonado Somatic Index) 감소. 난자 발달 저해 등이 관찰되었다. 또한, C. riparius를 산란 직후부터 DEHP에 노출시킨 경우에는 성체 출현률, 암수 비율, 산란률에서는 용량-반응 관계를 가진 변화가 발견되지 않았으나, 산란된 알의 부화율은 DEHP에 노출된 경우 유의하게 감소하는 것이 관찰되었다. 이러한 결과를 종합하여 볼 때. DEHP는 O. latpes와 C. riparius 모두 생식 작용에 영향을 미치며, 그 작용 기작은 일반적으로 발견되는 에스트로젠(estrogen) 활성이 아닌 암놈의 생식기관의 발달을 직, 간접적으로 저해함으로서 정상적인 알의 생성을 방해하는 이른바 항-에스트로젠 기작을 보이는 것으로 추정된다. 본 연구에서는 DEHP 위해성 평가를 위한 기본 자료로서, 생식작용 영향에 대한 새로운 자료를 제시하였다.

  • Thousands of new chemicals have been introduced to environment during last decades. Many of them and common consumer products have been shown to be the endocrine disrupting chemicals. One such chemical group is the phthalates, used in soft poly vinyl chloride (PVC) material and in a huge number of consumer products. The prevalence of these modem chemicals have a remarkable increase. Approximately 3.5 million tons of the main phthalate, di-(2-ethylhexyl) phthalate (DEHP), are produced annually worldwide and indeed, DEHP is considered a ubiquitous environmental contaminant. It has been demonstrated that high doses of phthalate can adversely affect adult and developing animals. In this review, we critically discuss the conclusions of recently original research papers and provide an overview of studies on reproductive disrupting effects of phthalates. In addition, we review the reproductive toxicity data of phthalates in some in vitro research and in both male and female reproductive systems in experimental and domestic animals. Finally, we point out some critical issues that should be addressed in order to clarify the implication of phthalates for human reproduction.

제초제 Butachlor의 송사리에 대한 아급성(亞急性) 독성(毒性) (Subchronic Toxicity of Herbicide Butachlor in Fish , the Medaka (Oryzias latipes))

  • 농약의 만성어독성을 평가하기 위한 노력의 일환으로 제초제인 butachlor에 대하여 송사리 (Oryzias latipes)을 대상으로 하여 90일간 아급성 어독성 실험을 실시하였고 또한 만성독성 평가를 위한 생물학적 검정어류로써 송사리의 타당성을 제고한 바 그 결과는 다음과 같다. 1) 송사리는 우리나라에서 고르게 분포하고 있는 소형 민물고기로써 시험기간중 대조군 등에서 산란 및 부화가 이루어져 풍부한 생물학적 자료와 함께 농약에 대한 만성 어독성 평가에 적합하다고 사료된다. 2) Butachlor의 0.16 ppm 처리군은 약간의 길이 성장감소와 산란된 수정란의 부화율이 감소되는 경향을 나타내었고, 0.04ppm과 0.01 ppm의 butachlor 처리군에서는 대조군과 비교할 때 별 다른 영향을 받지 않았음을 관찰할 수 있었다. 고로, butachlor는 송사리에서 0.04${\sim}$0. 16ppm의 MATC 값을 갖는 것으로 사료되며 또한 실험기간중 DO와 pH및 온도를 매일 측정한 결과 송사리에 영향을 주지 않았음이 입증되었다.

마우스 정모세포주에서 스티렌에 대한 삼백초 에탄올 추출물의 보호 효과 (Protective Effect of Saururus chinensis Ethanol Extract against Styrene in Mouse Spermatocyte Cell Line)

  • Background: This study was performed to evaluate the protective effect of Saururus chinensis ethanol extract (SCE) against styrene toxicity in mouse spermatocyte cells [GC-2spd (ts) cell line]. Methods and Results: Cytotoxicity in mouse spermatocyte cells was measured using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Generation of reactive oxygen species (ROS) was determined using 2',7'-dichlorodihydrofluorescein diacetate (DCF-DA) assay. Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) and western blotting were performed to quantify the mRNA and protein expression levels, resepectiviely, of stress or apoptosis-related genes including p21, p53, heat shock protein 70 (Hsp70), heat shock protein 90 (Hsp90), Bax, Bcl-2, and caspase-3. The results of the MTT assay showed that $50 {\mu}g/m{\ell}$ SCE did not affect cell viability. ROS generation in mouse spermatocyte cells increased by treatment with $100{\mu}M$ styrene, and decreased by co-treatment with SCE. SCE repressed the mRNA expression of stress-related genes, which increased by styrene treatment. In addition, SCE inhibited the apoptosis of mouse spermatocyte cells by ameliorating mRNA and protein levels of apoptotic genes that were altered by styrene treatment. Conclusions: These results suggest that SCE may alleviate styrene toxicity in mouse spermatocyte cells by reducing ROS stress and regulating genes related to styrene toxicity.

Estrogenic Activity, and Developmental Toxicity Studies of Pyrethroid Insecticides

  • It is well known that many pesticides possess hormonal activity, and affect the developments of wildlife and mammals including human. Currently, pyrethroid insecticides are in worldwide use to control in and outdoor pests, providing potential far environmental exposure. Hormonal activities of these pyrethroid insecticides, however, have been little studied, and the developmental effects of them were no reported. Therefore, we firstly examined the potential estrogenic activities of some pyrethroid insecticides (permethrin, cypermethrin, tetramethrin, deltamethrin, sumithrin, fenvalerate and bioallethrin) by immature rat uterotrophic assay, luciferase reporter gene assay and Calbindin-D$\sub$9k/ (CaBP-9k) gene expression assay. Uterine wet weights were increased by permethrin and the permethrin-induced weights were inhibited by ICI 182780 in the uterolrophic assay. On the other hand tetramethrin significantly reduced uterine and vaginal wet weights, and also inhibited the E2-induced weight increases at all doses tested. Cypermethrin and sumithrin had a tendency to increase uterine weights, although not statistically significant. Permethrin and cypermethrin dose-dependently increased the luciferase activity in reporter gene assay. Northern blot analysis showed that permethrin induced CaBP-9k mRNA expression whereas tetramethrin inhibted. Subsequent studies were conducted to investigate the possible developmental effects of four pyrethroid insecricides (permethrin, cypermethrin, sumithrin and teramethrin). Either diethlbestrol (DES) or 17${\beta}$ -estradiol (E2) was used as a reference control in this study. Pyrethroid insecticides were administered to Sprague Dawley rats via subcutaneous injection at 6 to 18 days of gestation or 1 to 5 days after birth. In utero treatment of permethrin (10mg/kg/day) in female rat resulted in significant increases in uterine and ovarian weights while significant decreases in serum E2 concentration, uterine and ovarian ER${\alpha}$ mRNA levels. Sumithrin and permethrin led to acceleration in vaginal opening of female rat, while delay in preputial separation of male after neonatal treatment. Anogenital distances of PND 18 were significantly reduced in sumthrin-treated, and permerhrin-treated male rats after neonatal treatment. All the pyrethroid insecticides tested caused significant increases in uterine weights on PND 18, while significant reductions in the first diestrus phase when neonataly treated. In addition, exposure to pyrethroids in neonatal period led to significant reduction in relative brain weight in female rat on PND 18, but its weight was recovered in diestrus phase. In summary, Our experimental data demonstrate the possibilities of developmental effects of pyrethroid insecticides via estrogenic or antiestrogenic activity.

Initial Risk Assessment of Acetanilide in OECD High Production Volume Chemical Program

  • In Korea, 2,320 tonnes of acetanilide were mostly wed as intermediates for synthesis in phar-maceuticals or additives in synthesizing hydrogen peroxide, varnishes, polymers and rubber. Only small amount of 120 kg were wed as a stabilizer for hydrogen peroxide solution for hair colouring agents in 1998. Readily available environmental or human exposure data do not exist in Korea at the present time. However, potential human exposures from drinking water, food, ambient water and in work places are expected to be negligible because this chemical is produced in the closed system in only one company in Korea and the processing factory is equipped with local ventilation and air filtering system. Acetanilide could be distributed mainly to water based on EQC model. This substance is readily biodegradable and its bioaccumulation is low. Acute toxicity of acetanilide is low since the L $D_{50}$ of oral exposure in rats is 1,959 mg/kg bw. The chemical is not irritating to skin, but slightly irritating to the eyes of rabbits. horn repeated dose toxicity, the adverse effects in rats were red pulp hyperplasia of spleen, bone marrow hyperplasia of femur and decreased hemoglobin, hematocrit and mean corpuscular hemoglobin concentration. The LOAEL for repeated dose toxicity in rats was 22 mg/kg/day for both sexes. Acetanilide is not considered to be genotoxic. In a reproductive/developmental toxicity study, no treatment-related changes in precoital time and rate of copulation, impregnation, pregnancy were shown in all treated groups. The NOAELs for reproduction and developmental toxicity (off-spring toxicity) are considered to be 200 mg/kg bw/day and 67 mg/kg bw/day, respectively. Ecotoxicity data has been generated in a limited number of aquatic species of algae (72 hr- $E_{b}$ $C_{50}$; 13.5 mg/l), daphnid (48hr-E $C_{50}$ > 100 mg/l) and fish (Oryzias latipes, 96hr-L $C_{50}$; 100 mg/l). Form the acute toxicity values, the predicted no effect concentration (PNEC) of 0.135 mg/1 was derived win an assessment factor of 100. On the basis of these data, acetanilide was suggested as currently of low priority for further post-SIDS work in OECD.in OECD.D.

Environmental Chemical-Dioxin Impacts on Biological Systems: A Review

  • Worldwide there is concern about the continuing release of a broad range of environmental endocrine disrupting chemicals, including polychlorinated biphenyls, dioxins, phthalates, polybrominated diphenyl ethers (PBDEs), and other halogenated organochlorines persistent organic pollutants (POPs) into the environment. They are condemned for health adverse effects such as cancer, reproductive defects, neurobehavioral abnormalities, endocrine and immunological toxicity. These effects can be elicited via a number of mechanisms among others include disruption of endocrine system, oxidation stress and epigenetic. However, most of the mechanisms are not clear, thus several number of studies are ongoing trying to elucidate them in order to protect the public by reducing these adverse effects. In this review, we briefly limited review the process, the impacts, and the potential mechanisms of dioxin/dioxin like compound, particularly, their possible roles in adverse developmental and reproductive processes, diseases, and gene expression and associated molecular pathways in cells.

  • A new composite antibiotic, SM-101(sulbactam·metampicillin), was at dose levels of 0, 375, 750 and 1500 mg/kg/day administered intravenously to pregnant Sprague-Dawley rats during the organogenetic period. Two-third of dams per group were subjected to caesarean section on day 20 of pregnancy and the remaining 10 dams per group were allowed to deliver. Effects of test substance on dams, embryonal development of F1 fetuses, as well as growth, behaviour and mating performance of F1 offspring were examined. In dams, two deaths occurred at 375 and 1500 mg/kg, respectively. The decrease in the weight of adrenal glands of the 1500 mg/kg group was observed. The prolongation of pregnancy period was found at 1500 mg/kg. F1 fetuses showed no changes related to the treatment of SM-101. In F1 offspring, the increase in spleen weight was seen at all doses treated. No treatment-related abnormalities were observed in each treated group in terms of development, behaviour and reproductive performance. In F2 fetuses, no drug-induced abnormalities occurred at all doses. The results show that the no-effect dose levels (NOELS) for dams and Fl offspring are under 375 mg/kg/day and NOELs for F1/F2 fetuses are over 1500 mg/kg/day.

복합항생제 SM-101(설박탐-메탐피실린)의 생식독성연구: 랫트 주산기 및 수유기시험 (Reproductive Toxicity of SM-101(sulbactam.metampicillin): Peri- and Postnatal Study in Rats)

  • A new composite antibiotic, SM-101 (sulbactam·metampicillin), was at dose levels of 0, 250, 500 and 1000 mg/kg/day administered intravenously to pregnant and subsequently delivered Sprague-Dawley rats from day 17 of gestation to day 21 of lactation. Effects of test agent on dams and growth, behaviour and mating performance of Fl offspring were examined. In dams, one death occurred at 1000 mg/kg. The increase in kidney weight of the 250, 500 and 1000 mg/kg group was found. In F1 offspring, both delayed incisors eruption and decreased body weight were observed in females of the 1000 mg/kg group. The increase in the weights of liver and kidney was found in males of the 1000 mg/kg group. No treatment-related abnormalities were observed in each treated group in terms of behaviour and reproductive performance. In F1/F2 fetuses, no drug-induced abnormalities occurred at all doses tested. The results show that the no effect dose level (NOEL) of SM-101 is under 250 mg/kg/day for dams and 500 mg/kg/day for F1 offspring, and over 1000 mg/kg/day for F1/F2 fetuses.
