Behavioral Characteristics of Bos indicus Cattle after a Superovulatory Treatment Compared to Cows Synchronized for Estrus

  • The intensity and duration of sexual behavior in Bos indicus was assessed through the continuous observation of sexual receptivity. Two groups of cows were formed: only synchronized (n = 50) and other group further superovulated (n = 20). An intravaginal implant that released progesterone over 9 d was used. After removing the implant, 25 mg of $PGF_{2{\alpha}}$ was administered. In the superovulated group, the administration of 280 mg (Follicle stimulant hormone) FSH-P1 per cow with a decreasing dosage over 4 d was utilized. In both groups, behavioral observations began at the moment of implant removal. Sexual behavior was analyzed using a Kruskal-Wallis test to compare the mean of hours in estrus, effective mountings and number of mounts/hour during estrus. A nonparametric survival analysis was performed using the time in two ways: i) when an event happened it was placed in a 24 h timeframe and, ii) the time of observation in continuous form (96 h) assessing the difference between curves by the log rank test Chi-square. The only significant difference was the number of mounts/h during receptivity (p<0.05). In the superovulated group three periods of sexual activity during the day were identified, with these events being of greater frequency and duration than the synchronized group (p = 0.02); besides, the superovulated group began estrus before the synchronized group (p = 0.0035) when using the total period. In a simulation study, when the number of observations went from two (06:00-18:00) to three periods (06:00, 12:00 and 18:00) cows detected accurately (<6 h after the onset) increased more than 20%. The results show that superovulated cows presented greater intensity and duration of sexual activity in contrast with only synchronized animals.

Effects of Osmolality Step during Vitrification on Survival of Vitrified IVP Embryos in Korean Native Cattle (Hanwoo)

  • Solution of glycerol, ethylene glycol, sucrose, dextrose (GESD) and cryotop methods were carried out to investigate the survivability on vitrification of embryos. Embryos cultured in vitro were vitrified by GESD of 10 or 8 step and cryotop methods of 6 step, from cryopreservation step to frozen-thawed and culture step. Survival rate and ICM, TE cells of embryos were investigated after frozen-thawed 24 h. As a results, cryotop method was significantly (p<0.05) higher ($85.76{\pm}5.3$ vs. $66.71{\pm}2.4$, $44.80{\pm}2.1%$) than GESD 10 or 8 step methods on survivability. Also, In ICM cell number, cryotop method was significantly (p<0.05) higher to $45.67{\pm}4.7$ cells than GESD 8 step method. TE cell number was significantly (p<0.05) highest to $111.00{\pm}11.0$ cells in cryotop method. On the other hand, survival rate, TE and total cell number were all the significantly (p<0.05) high, except ICM in GESD 10 step method between GESD 10 step method and GESD 8 step method. In conclusion cryotop method was to be most effective, but it is considered necessary to study vitrification method for step-by-step freezing and thawing process.

Cryopreservation of Embryo by Concentration of Ethylene Glycol and Day 6, 7, 8, 9 Embryo in Korean Native Cattle (Hanwoo)

  • This study was carried out to effects of ethylene glycol concentration, sucrose and culture day of in vitro production embryo on slow-down freezing in Hanwoo. 6, 7, 8 and 9 day embryos produced in vitro were frozen using 1.8M EG+0.1M sucrose, 1.8M EG+0.5% BSA and 1.5M EG+0.1M sucrose media. Survivability was confirmed after frozen-thawed 24 and 48h and ICM, TE cell number were counted by Hoechst 33342 and PI staining after frozen-thawed 24h. As a result, 1.8M EG+0.1M sucrose group was most significantly (p<0.05) higher compared with the other treatment groups on survivability, TE and total cell number after frozen-thawed 24h ($94.2{\pm}2.6%$, $94.67{\pm}3.4$ and $129.67{\pm}5.5$). ICM number did not found significant (p<0.05) differences between the three treatment groups. in 6, 7, 8 and 9 day of embryos using three types of freezing media, frozen-thawed, 1.8M EG+0.1M sucrose groups with embryos cultured 8 day was significantly (p<0.05) highest survivability to $98.3{\pm}1.7%$ after frozen-thawed 24h. 1.5M EG+0.1 sucrose group with embryos cultured 9 day was significantly higher survivability than group of embryos cultured 8 day after frozen-thawed 24 and 48h. In conclusion, 1.8M EG+0.1M sucrose media is considered to be effective to cryopreservation of embryos cultured 8 and 9 day.

Basic reproduction number of African swine fever in wild boars (Sus scrofa) and its spatiotemporal heterogeneity in South Korea

  • Background: African swine fever (ASF) is a hemorrhagic fever occurring in wild boars (Sus scrofa) and domestic pigs. The epidemic situation of ASF in South Korean wild boars has increased the risk of ASF in domestic pig farms. Although basic reproduction number (R0) can be applied for control policies, it is challenging to estimate the R0 for ASF in wild boars due to surveillance bias, lack of wild boar population data, and the effect of ASF-positive wild boar carcass on disease dynamics. Objectives: This study was undertaken to estimate the R0 of ASF in wild boars in South Korea, and subsequently analyze the spatiotemporal heterogeneity. Methods: We detected the local transmission clusters using the spatiotemporal clustering algorithm, which was modified to incorporate the effect of ASF-positive wild boar carcass. With the assumption of exponential growth, R0 was estimated for each cluster. The temporal change of the estimates and its association with the habitat suitability of wild boar were analyzed. Results: Totally, 22 local transmission clusters were detected, showing seasonal patterns occurring in winter and spring. Mean value of R0 of each cluster was 1.54. The estimates showed a temporal increasing trend and positive association with habitat suitability of wild boar. Conclusions: The disease dynamics among wild boars seems to have worsened over time. Thus, in areas with a high elevation and suitable for wild boars, practical methods need to be contrived to ratify the control policies for wild boars.

Studies on the Cycle of the Seminiferous Epithelium in Korean Native Cattle (한우의 세정관상피주기에 관한 연구)

  • The cycle of the seminiferous epthelia in the testis of matured Korean Native Cattle was divided into eight stages. The results were summarized as follows: 1. Type A spermatogonia a, pp.ared twice as many at stage 2 as compared to stage 1, while maximum numbers were the average of 2.8 at stage 2. The intermediate and Type B spermatogonia were found during the stage 3 to 8, stage 6 to 8, respectively. The leptolene primary spermatocytes were not observed during the stage 5 to 7, while the pachytene primary spermatocytes were shown the least in number at stage 4, the secondary supermatocytes could be seen only at stage 4 and the round spermatids were not observed at stage 3, 4. 2. The relative frequencies of the eight stages of the cycle of the seminiferous eptithelia were 24.9, 14.2, 19.0, 6.3, 3.7, 7.9, 10.3 and 13.9%, respectively. 3. Some of the nuclei of Sertoli cells transformed from the "parallel" type to the "perpendicular" type. This evolution took place from stage 1 to 5, when the number of "perpendicular" type nuclei reached a peak and the number was decreased in the rest of the stages.sed in the rest of the stages.

Study on the Sex-Ratio of Fast- and Slow-Developing Mouse Embryo (Mouse 초기배의 발육속도에 따른 성비에 관한 연구)

  • This study was conducted using inbred ICR mice to investigate the sex-ratio of preimplantation mouse embryos. For the investigation of sex-ratio of mouse embryos, the karyotype of embryos collected at 70-72, 74-76, 78-80 and 82-84 hr after HCG injection was analyzed by chromosomal analysis. Eight-cell embryos were cultrued up to blastocyst stage, then divided them into three groups(fast-, intermediate- and slow-) according to the blastocoel formation. The sex-ratio was also investigated by chromosomal analysis. 1. The highest apperance of eight-cell and morula was observed at the embryos collected respectively at 66-68 hr(84.6%) and 82-84 hr(79.3%) compared to any other group. 2. The successful rate of embryos sexing at 4-, 8-cell and morula stage were 23.1% (3/13), 42.1%(138/328) and 32.6%(47/141), respectively. The respective sex ratios (female vs male) of 4-, 8-cell and morula were 66.7:33.3, 49.3:50.7 and 39.5:60.5. 3. Of the 476 eight-cell embryos cultured in vitro, 427(89.7%) embryos were developed to the blastocysts and the number of fast-, intermediate- and show-developing embryos were 139, 144 and 144, respectively. 4. Female to male ratios fo fast-, intermediate- and slow-developing group were 23.0:77.0, 55.2:44.8 and 73.8:26.2, respectively. Significantly higher (P<0.05) number of female (48/65;73.8%) was observed in the group of slow-developing embryo than that out of total number of embryos(82/188;43.6%).

체외배양 기술로 생산된 초기배에 의한 한우 송아지 생사기술 개발. II. 체외성숙, 수정된 소 초기배의 체외발생에 있어서 ITS와 EGF의 효과

  • This study was undertaken to investigate the effects of ITS and EGF on embryonic development of in vitro matured and fertilized bovine oocytes in culture medium su, pp.ementing with or without calf serum. When fertilized oocytes were cultured in TCM-199 containing 0, 0.1, 0.5 and 1.0\ulcorner/ml ITS with 5% calf serum, the rates of development to blastocyst stage and the cell number of blastocysts were not significantly different among all treatments. And also monolayer of cumulus/granulosa cells prepared in containing calf serum and ITS were no beneficial effects of embryonic development. On the other hand, when EGF was su, pp.imented to TCM-199 containing calf serum or calf serum free, embryonic development rates(24.0 2.8% to 29.2 1.7% or 8% to 9%) and cell number of blastocysts(p<0.05) were significantly increased compared with EGF-free(22.1 2.1 or 1.0%, p<0.05). But when fertilized oocytes were cultured with cumulus/granurosa cells in TCM-199 containing EGF and calf serum, the rate of embryos development to the blastocyst stage and cell number of blastocysts were not significantly different compared with EGF-free and any concentrations. These results showed that ITS and EGF was not improved the development of bovine embryo in vitro matured and in vitro fertilized with calf serum and/or monolayer of cumulus/granulosa cells.

Suggestions for Setting on Period of Epidemic Waves in COVID-19 Epidemic of South Korea (한국 코로나19 유행기에 대한 제안)

  • Objectives: In the epidemiology of communicable diseases, the term epidemic period, also referred to as "wave" is often used in the general and academic milieu. A wave refers to a natural pattern of increase in the number of sick individuals, a defined peak, and then a decline in the number of cases. It implies a pattern of peaks and valleys after a particular peak is taken. The idea of epidemic waves is a useful tool for predicting the course as well as helping to accurately describe an epidemic. However, in many domestic and foreign news as well as in various research results in Korea, most of the reports either had no standard, were inaccurate, had a questionable classification of the period of the epidemic, or the basis for classification of a given wave was not presented. Methods: The author reviewed and organized related literature with epidemic wave. The author made several suggestions of an epidemic wave as follows. Results: To start with, it should be based on the number of incident cases in consideration of the size of the outbreak, then the period from the bottom to the peak and then reaching the next bottom; also, the period over a certain scale based on the number of incident cases; and the period according to the change in the major infection type (mutation-dominant species). In addition, according to the period of change in the vaccination rate (formation of herd immunity), as well as the content and duration of the intervention, that is, classification according to the applied quarantine stage. Furthermore, the classification of epidemic periods by the time-dependent reproduction number or time-varying reproduction number (Rt), and lastly the application of mathematical methodology. Conclusions: Therefore, classifying the epidemic period into generally known and accepted time frames is considered to be a very important task for future research analysis and development of intervention strategies.

Effects of Steroid Hormones for Sexual Reproduction of Rotifer, Brachionus rotundiformis (Rotifer Brachionus rotundiformis의 유성생식에 관한 스테로이드 호르몬의 영향)

  • We studied the effect of several sex-related steroid hormones (serotonin, progesterone and ${\beta}$-estradiol) for 6 days on the induction of sexual reproduction for the mass production of resting eggs in the marine rotifer Brachionus rotundiformis. The highest mix rate of 20.6% appeared with the ${\beta}$-estradiol ($E_2$) treatment on the third day. The number of resting eggs was highest with $E_2$ treatment, followed by that of the serotonin treatment group. In addition, we investigated the effect of the hormones on the expression pattern of the genes related to sexual reproduction in the rotifer. NrbP, SRY, Cyclin and MrpmB genes were up-regulated with all the hormone treatments. As a result, ${\beta}$-estradiol was more effective than the other hormone treatments to produce resting eggs in B. rotundiformis. We suggest that the sexual reproduction-related genes in the rotifer are the NrbP, SRY, Cyclin and MrpmB genes. Further study is required to determine the optimum concentration of $E_2$ for the effective production of resting eggs in the rotifer.

Seasonal changes in the reproductive performance in local cows receiving artificial insemination in the Pursat province of Cambodia

  • Objective: The present study aimed to survey seasonal changes in reproductive performance of local cows receiving artificial insemination (AI) in the Pursat province of Cambodia, a tropical country, to investigate if ambient conditions affect the reproductive performance of cows as to better understand the major problems regarding cattle production. Methods: The number of cows receiving AI, resultant number of calving, and calving rate were analyzed for those receiving the first AI from 2016 to 2017. The year was divided into three seasons: cool/dry (from November to February), hot/dry (from March to June), and wet (from July to October), based on the maximal temperature and rainfall in Pursat, to analyze the relationship between ambient conditions and the reproductive performance of cows. Body condition scores (BCS) and feeding schemes were also analyzed in these seasons. Results: The number of cows receiving AI was significantly higher in the cool/dry season than the wet season. The number of calving and calving rate were significantly higher in cows receiving AI in the cool/dry season compared with the hot/dry and wet seasons. The cows showed higher BCSs in the cool/dry season compared to the hot/dry and wet seasons probably due to the seasonal changes in the feeding schemes: these cows grazed on wild grasses in the cool/dry season but fed with a limited amount of grasses and straw in the hot/dry and wet seasons. Conclusion: The present study suggests that the low number of cows receiving AI, low number of calving, and low calving rate could be mainly due to poor body condition as a result of the poor feeding schemes during the hot/dry and wet seasons. The improvement of body condition by the refinement of feeding schemes may contribute to an increase in the reproductive performance in cows during the hot/dry and wet seasons in Cambodia.
