Protein Patterns on a Corpus Luteum during Pregnancy in Korean Native Cows

  • Luteal cells produce progesterone that supports pregnancy. Steroidogenesis requires coordination of the anabolic and catabolic pathways of lipid metabolism. In the present study, the corpus luteum (CL) in early pregnancy established from luteal phase and pregnant phase was analyzed. The first study determined progesterone changes in the bovine CL at day 19 (early maternal recognition period) and day 90 in mid-pregnancy and compared them to the CL from day 12 of the estrous cycle. CL alternation was tested using two-dimensional polyacrylamide gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF). Comparing CL from luteal phase to those from pregnant phase counterparts, significant changes in expression level were found in 23 proteins. Of these proteins 17 were not expressed in pregnant phase CL but expressed in luteal phase counterpart, whereas, the expression of the other 6 proteins was limited only in pregnant phase CL. Among these proteins, vimentin is considered to be involved in regulation of post-implantation development. In particular, vimentin may be used as marker for CL development during pregnancy because the expression level changed considerably in pregnant phase CL tissue compared with its luteal phase counterpart. Data from 2-DE suggest that protein expression was disorientated in mid pregnancy from luteal phase, but these changes was regulated with progression of pregnancy. These findings demonstrate CL development during mid-pregnancy from luteal phase and suggest that alternations of specific CL protein expression may be involved in maintenance of pregnancy.

중규모 해양모형을 이용한 한반도 주변 해역 해양순환 재현 (Reproduction of Ocean Circulation around Korean Peninsula by using a Mesoscale Ocean Circulation Model)

  • 본 연구에서는 중규모 해양모형을 이용하여 주어진 한반도 주변 대기경계조건에 대한 해양모형의 반응을 연구하였다. 연구는 크게 스핀업(spin-up) 실험과 재현(reproduction) 실험으로 나뉘는데, 스핀업 실험은 해양모형을 스핀업시켜 기후학적 월평균 해양상태로 유도하는 실험으로 월평균 대기상태를 경계조건으로 하여 해양모형을 장기적분하였다. 재현 실험은 1980년${\sim}$1998년 동안(19년)의 한반도 주변 해역의 해양변동을 재현하고자 한 실험으로 이를 위하여 스핀업 마지막 해의 해양상태를 모형의 초기상태로 하고 그 기간 동안의 월평균 기상 자료를 경계조건으로 해양모형에 적용하여 19년간 적분하였다. 스핀업 실험에서 모형은 동해지역에서의 동한 해류의 이안, 극전선대(polar front)형성 그리고 소규모 에디들에 의한 냉수대의 형성과 황해지역에서의 연안역과 중앙부 사이에 남북으로 형성되는 수온전선 대 그리고 역풍류(Upwind Current)의 변화에 기인한 순환 구조의 계절적인 변동 등 한반도 주변해역에서 나타나는 특징적인 순환들을 잘 모사하였다. 또한 1982${\sim}$98년까지에 대한 재현 실험에서는 주어진 대기 경계조건에 따른 해면온도의 변동성과 기간동안 약 0.5$^{\circ}$C의 해면온도 아노말리의 상승을 관측과 유사하게 모사하였다. 따라서 실험은 모형이 주어진 경계조건에 대하여 해양의 평균적인 상태 뿐만 아니라 변동성도 잘 모사하고 있음을 보여주었다.

고주석 청동정문경(靑銅精文鏡)의 재현실험을 통한 합금재료의 특성 연구 (A Study on Characteristics of Alloy Materials through Reproduction Experiment of High-tin Bronze Mirror with Geometric Designs)

  • 본 연구는 청동 정문경 제작공정 재현과정에서 생산된 합금 및 부산물을 대상으로 자연과학적 분석을 실시하였다. 재현실험의 합금비율은 국보 제141호 정문경에 대한 분석결과(Cu 61.7%, Sn 32.3%, Pb 5.5%)를 바탕으로 하였다. 사용된 원료에 대한 X-선 형광분석 결과, 구리는 Cu 98.9 wt%, 주석은 Sn 99.0 wt%, 납은 Pb 70.2 wt%, Sn 21.8 wt%로 확인되었다. 합금 용융 시주석과 납의 기화량을 감안하여 Sn, Pb은 5 wt%씩 더 첨가하였다. 1차 재현실험 결과는 Cu 58.8 wt%, Sn 36.9 wt%, Pb 4.4 wt%, 2차 재현실험 결과는 Cu 58.7 wt%, Sn 35.9 wt%, Pb 5.5 wt%로 확인되었다. 성분조성은 Cu와 Sn에서 약 3 wt%씩 차이를 보였으며, 미세조직은 주로 δ상이 관찰되어 선행연구와 유사한 양상을 나타내었다. 본 연구를 통해 향후 고대 청동거울의 재료학적 특성 규명에 있어 기초자료로 활용되었으면 한다.

국내 오프셋 인쇄에서 색재현 특성의 잉크 의존성에 관한 연구 (A Study on the Ink Dependence of Color Reproduction Properties in Domestic Offset Printing)

  • The reality of domestic prints neither possesses an objective standard of standard density nor a spectrophotometric method in regard to process ink. As a result, it creates a difficult communication between a designer and a Prepresser as to estimating the color of reproduction prints. There was an experiment on paper, ink, and printing company to obtain the standard point. The result shows that paper has minimum effect on color reproduction as opposed to ink and different printing companies, which had a great effect. The purpose of this paper is to confirm how different the color reproduction properties of prints are using densitometer and spectrophotometer for three different ink companies. The process color inks of three different companies were used under the same printing environment in this experiment.

고연색 CCFL을 이용한 TFT-LCD 색재현율의 향상 (The Improvement of color reproduction ratio used to CCFL with high color rendering characteristic in TFT-LCD)

  • At present, characteristic of high color reproduction for LCD TV is needed in Display market according to start mass production of LCD TV. Therefore development target for LCD is focused on improvement of color reproduction. The improving methods of high color reproduction are alteration of color Filter or Red, Green, Blue phosphor alteration of CCFL. But decrease of luminance and panel transmittance is caused by alteration of Color Filter. Accordingly, we completed LCD with high color reproduction by the most suitable emission spectrum of CCFL phosphor at panel with conventional color filter. In this experiment, we knew that LCD applied to CCFL with high color rendering characteristic had color reproduction range of 81% compared with NTSC in CIE color coordinate. According to increase of intensity peak and alteration of Red, Green, Blue phosphor spectrum, we made LCD with high color reproduction characteristic. In conclusion we achieved improvement of color reproduction ratio by alteration of CCFL phosphor without changing color filter.

  • At present, Characteristic of high color reproduction for LCD products needed in Display market. Therefore, The improving methods of high color reproduction are alteration of color Filter or Red, Green, Blue phosphor alteration of CCFL. But High color reproduction phosphor is short life time as compared with conventional phosphor. In this experiment, by using split the Red, Green, Blue CCFL with high color reproduction phosphor instead of conventional high color reproduction CCFL. We knew that the high color reproduction RGB split CCFL BLU has same spectrum data and chromaticity, but has long life time as manufacturing RGB split CCFL and reduce chromaticity shift following long time discharge as compared with conventional high color reproduction CCFL.

한우 농장별 번식기록 분석을 통한 번식률 제고 사례 연구 (Case Report on Improvement of Reproduction Rate in Hanwoo Farms)

  • 본 연구는 한우 번식 기록이 잘 유지되고 있는 4개 농장의 2007년 1월부터 2010년 10월까지의 번식 자료 수집하여 분석하였다. 수태 당 평균 수정 횟수와 평균 공태일은 A농장 $1.7{\pm}0.1$회와 $77.4{\pm}4.8$일, B농장 $1.5{\pm}0.1$회와 $150.8{\pm}11.2$일, C농장 $1.5{\pm}0.1$회와 $90.4{\pm}4.5$일, D농장 $1.4{\pm}0.1$회와 $71.4{\pm}2.5$일이었다. 호르몬으로 발정을 유도하는 D농장을 제외한 3개 농장 531두의 번식 기록으로 분만 후 첫 수정 시기에 따른 평균 수정 횟수와 평균 공태일을 분석한 결과, 총 5개의 수정 시기에 따른 수정 횟수는 30일 이전 첫 수정이 $2.1{\pm}0.2$회로 31일 이후 첫 수정보다 유의적으로 높았다. 번식 장애우 58두에 2가지 배란 동기화법을 사용하여 수태율을 확인해 본 결과, Ovsynsh 법은 55.2%의 수태율을, CIDR-based TAI 법은 65.5%의 수태율을 나타냈다. 농장의 번식률을 높이기 위해서는 정확한 번식 기록 작성, 발정 관찰, 수정 후 임신 감정, 번식 기관 검진, 번식률을 고려한 첫 수정 시기 수정 등이 필요하다.

유우종모우 정액의 생상간 상관관계에 관한 연구 (Studies on the Correlation among the Semen Characteristics of Holstein Bulls)

  • This experiment was carried out to examine the correlation among the semen characteristics of Holstein bulls. Seventy-two samples of semen were collected from 6 Holstein bulls, 2 to 10 years old. The characteristics of the semen were estimated by ordinary methods. The results obtained in this experiment were summarized as follows: 1. Semen characteristics on an average were 6.0 ml of volume, 16${\times}$108/ml of concentration, 92.4${\times}$108 of total sperm number(TSN), 72.1% of motility and 4.6% of abnormality. 2. Semen volume was positively high correlated (P<0.01) with total sperm number, but negative correlation was observed with the sperm motility (P<0.05) and semen concentration. 3. In relation to their age, older bulls semen volume and total sperm number were increased (P<0.01), but semen concentration and motility were decreased (P<0.01).

Suggestion of experimental model on the innate kidney essence insufficiency syndrome for the development of humanitas traditional medicine

  • According to the traditional Korean medicine (TKM), kidney has been recognized as the roof of innate endowment because it plays an important role in the birth, growth, sexual reproduction, and aging. Kidney essence insufficiency syndrome (KEIS) is caused when kidney is impaired. KEIS is characterized by retarded development, decreased reproduction, tinnitus, loosening of teeth, and loss of hair and forgetfulness. In traditional Chinese medicine, KEIS mouse model was established by use of threatinjuring the kidney combined with over-fatigue. However, the TKM theory-based KEIS experiment model has not been described correctly. In the present study, we suggest a new KEIS experiment model including following cases; 1) weakness of father essence and mother blood, 2) life nurturing during pregnancy, 3) full-term gestation period.

소 체외수정란의 초자화 동결에 관한 연구 (Studies on Vitrification of Bovine Blastocysts Fertilized In Vitro)

  • Two experiments were conducted to study the production of in vitro fertilized bovine embryos and the viability of blastocysts cryopreserved by vitrification. In experiment 1, production rate of in vitro matured bovine oocytes after fertilization in medium containing bovine oviduct epithelial cells (BOEC), cumulus cells and granulosa cells to blastocysts were 18.4, 14.6 and 13.1%, respectively. Developmental percentages of blastocysts produced at day 6, 7 and 8 were 8.5, 10.6 and 15.2% respectively. Hatching rate of bovine embryos produced was 60.0%. In experiment 2, post-thawed surviving embryos in a vitrification solution consisting of 7.15M ethylene glycol, 2.5 mM ficoll and 0.3 M sucrose were 36.4% (56/154). Also, survival rate of bovine embryos after exposed to vitrification solution at 1, 2, 3, 4 and 5 min were 84.0, 88.0, 71.0, 48.0 and 24.0% respectively.
