Protein Patterns on a Corpus Luteum during Pregnancy in Korean Native Cows

  • Luteal cells produce progesterone that supports pregnancy. Steroidogenesis requires coordination of the anabolic and catabolic pathways of lipid metabolism. In the present study, the corpus luteum (CL) in early pregnancy established from luteal phase and pregnant phase was analyzed. The first study determined progesterone changes in the bovine CL at day 19 (early maternal recognition period) and day 90 in mid-pregnancy and compared them to the CL from day 12 of the estrous cycle. CL alternation was tested using two-dimensional polyacrylamide gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF). Comparing CL from luteal phase to those from pregnant phase counterparts, significant changes in expression level were found in 23 proteins. Of these proteins 17 were not expressed in pregnant phase CL but expressed in luteal phase counterpart, whereas, the expression of the other 6 proteins was limited only in pregnant phase CL. Among these proteins, vimentin is considered to be involved in regulation of post-implantation development. In particular, vimentin may be used as marker for CL development during pregnancy because the expression level changed considerably in pregnant phase CL tissue compared with its luteal phase counterpart. Data from 2-DE suggest that protein expression was disorientated in mid pregnancy from luteal phase, but these changes was regulated with progression of pregnancy. These findings demonstrate CL development during mid-pregnancy from luteal phase and suggest that alternations of specific CL protein expression may be involved in maintenance of pregnancy.

Reproduction of Ocean Circulation around Korean Peninsula by using a Mesoscale Ocean Circulation Model (중규모 해양모형을 이용한 한반도 주변 해역 해양순환 재현)

  • In this study, the oceanic responses to given atmospheric boundary conditions are investigated using a mesoscale ocean circulation model. The numerical experiments are divided into two parts: One is, so called, spin-up experiment and the other is reproduction experiment. The spin-up experiment simulates climatic state of ocean by integrating the ocean model with upper boundary conditions of the monthly mean atmospheric climate data. In the reproduction experiment, for the reproduction of major oceanic changes around Korean Peninsula during the period of 1980-1998 (19 years), the model has been integrated under the boundary condition of the 19year monthly mean atmosphere data. The spined-up state of ocean generated from the spin-up experiment is assigned to the initial boundary condition of the reproduction experiment. In the spin-up experiment, the model properly simulates the major features of circulation structure around Korean Peninsula; such as separation of East Korean Warm Current (EKWC), formation of the polar front, cold water band associated with the small scale eddies in the East Sea, the formation of front along west coast, and the seasonal variation of circulation pattern caused by changing upwind current in the West Sea. In the reproduction experiment, the model has shown the interannual sea surface temperature variations and a warming trend of about 0.5$^{\circ}$C during the period around Korean Peninsula, as in the case of the observation. Therefore, it is concluded that the model is capable of simulating not only the mean states but also the variabilities of ocean under the given atmosphere boundary conditions.

A Study on Characteristics of Alloy Materials through Reproduction Experiment of High-tin Bronze Mirror with Geometric Designs (고주석 청동정문경(靑銅精文鏡)의 재현실험을 통한 합금재료의 특성 연구)

  • This study analyzed on alloys and by-product samples produced through the reproduction experiment of bronze mirror with geometric designs. The alloy ratio used in the first and second reproduction experiments was based on the analysis results of bronze mirror with geometric designs(Cu 61.68%, Sn 32.25%, Pb 5.46%) which is the national treasure No. 141. As a result of portable X-ray fluorescence analysis on the raw materials used in the reproduction experiment, the contents of copper raw materials were 98.85 wt% for Cu, tin raw materials were 99.03 wt% for Sn, and lead raw materials were 70.19 wt% for Pb, and 21.81 wt% for Sn. Sn and Pb were added 5 wt% more considering the evaporation amount of tin and lead during alloy melting. The result produced by the first reproduction experiment were 58.75 wt% for Cu, 36.87 wt% for Sn, 4.39 wt% for Pb, and the other result produced by the second reproduction experiment were 58.66 wt% for Cu, 35.89 wt% for Sn, and 5.50 wt% for Pb. The composition of the components was about 3.00 wt% in Cu and Sn respectively, and the microstructure was similar to the previous studies because the δ phase was observed mainly. The results of this study will be used as basic data for the materialistic characteristics of ancient bronze mirror in the future.

A Study on the Ink Dependence of Color Reproduction Properties in Domestic Offset Printing (국내 오프셋 인쇄에서 색재현 특성의 잉크 의존성에 관한 연구)

  • The reality of domestic prints neither possesses an objective standard of standard density nor a spectrophotometric method in regard to process ink. As a result, it creates a difficult communication between a designer and a Prepresser as to estimating the color of reproduction prints. There was an experiment on paper, ink, and printing company to obtain the standard point. The result shows that paper has minimum effect on color reproduction as opposed to ink and different printing companies, which had a great effect. The purpose of this paper is to confirm how different the color reproduction properties of prints are using densitometer and spectrophotometer for three different ink companies. The process color inks of three different companies were used under the same printing environment in this experiment.

  • At present, characteristic of high color reproduction for LCD TV is needed in Display market according to start mass production of LCD TV. Therefore development target for LCD is focused on improvement of color reproduction. The improving methods of high color reproduction are alteration of color Filter or Red, Green, Blue phosphor alteration of CCFL. But decrease of luminance and panel transmittance is caused by alteration of Color Filter. Accordingly, we completed LCD with high color reproduction by the most suitable emission spectrum of CCFL phosphor at panel with conventional color filter. In this experiment, we knew that LCD applied to CCFL with high color rendering characteristic had color reproduction range of 81% compared with NTSC in CIE color coordinate. According to increase of intensity peak and alteration of Red, Green, Blue phosphor spectrum, we made LCD with high color reproduction characteristic. In conclusion we achieved improvement of color reproduction ratio by alteration of CCFL phosphor without changing color filter.

  • At present, Characteristic of high color reproduction for LCD products needed in Display market. Therefore, The improving methods of high color reproduction are alteration of color Filter or Red, Green, Blue phosphor alteration of CCFL. But High color reproduction phosphor is short life time as compared with conventional phosphor. In this experiment, by using split the Red, Green, Blue CCFL with high color reproduction phosphor instead of conventional high color reproduction CCFL. We knew that the high color reproduction RGB split CCFL BLU has same spectrum data and chromaticity, but has long life time as manufacturing RGB split CCFL and reduce chromaticity shift following long time discharge as compared with conventional high color reproduction CCFL.

Case Report on Improvement of Reproduction Rate in Hanwoo Farms (한우 농장별 번식기록 분석을 통한 번식률 제고 사례 연구)

  • This work was conducted to study the improvement of reproduction rate from the breeding data collected from four farms from January 2007 to October 2010. The average numbers of service per conception were 1) A farm $1.7{\pm}0.1$ times, 2) B farm $1.5{\pm}0.1$ times, 3) C farm $1.5{\pm}0.1$ times, 4) D farm $1.4{\pm}0.1$ times. The average days from calving to conception was $77.4{\pm}4.8$ days in A farm, $150.8{\pm}11.2$ days in B farm, $90.4{\pm}4.5$ days in C farm, and $71.4{\pm}2.5$ days in D farm. Number of artificial insemination (AI) service per conception was higher at the 30 days before first AI ($2.1{\pm}0.2$ times) than at the 31 days after first AI, and the days from calving to conception were shorter at the 90 days before first AI than at the 91 days after first AI. After timed AI (TAI) treatment, the pregnancy rate was 60.3% for the 58 cows with reproductive disorder. In order to improvement of reproduction rates, the farms has to improve the accuracy of estrus detection, pregnancy diagnosis, check-up for reproductive health, and control of day for first AI periods after calving. The result suggests that farmers need the careful management and reproductive examination of farm animals to improve of reproductive efficiency.

Studies on the Correlation among the Semen Characteristics of Holstein Bulls (유우종모우 정액의 생상간 상관관계에 관한 연구)

  • This experiment was carried out to examine the correlation among the semen characteristics of Holstein bulls. Seventy-two samples of semen were collected from 6 Holstein bulls, 2 to 10 years old. The characteristics of the semen were estimated by ordinary methods. The results obtained in this experiment were summarized as follows: 1. Semen characteristics on an average were 6.0 ml of volume, 16${\times}$108/ml of concentration, 92.4${\times}$108 of total sperm number(TSN), 72.1% of motility and 4.6% of abnormality. 2. Semen volume was positively high correlated (P<0.01) with total sperm number, but negative correlation was observed with the sperm motility (P<0.05) and semen concentration. 3. In relation to their age, older bulls semen volume and total sperm number were increased (P<0.01), but semen concentration and motility were decreased (P<0.01).

  • According to the traditional Korean medicine (TKM), kidney has been recognized as the roof of innate endowment because it plays an important role in the birth, growth, sexual reproduction, and aging. Kidney essence insufficiency syndrome (KEIS) is caused when kidney is impaired. KEIS is characterized by retarded development, decreased reproduction, tinnitus, loosening of teeth, and loss of hair and forgetfulness. In traditional Chinese medicine, KEIS mouse model was established by use of threatinjuring the kidney combined with over-fatigue. However, the TKM theory-based KEIS experiment model has not been described correctly. In the present study, we suggest a new KEIS experiment model including following cases; 1) weakness of father essence and mother blood, 2) life nurturing during pregnancy, 3) full-term gestation period.

Studies on Vitrification of Bovine Blastocysts Fertilized In Vitro (소 체외수정란의 초자화 동결에 관한 연구)

  • Two experiments were conducted to study the production of in vitro fertilized bovine embryos and the viability of blastocysts cryopreserved by vitrification. In experiment 1, production rate of in vitro matured bovine oocytes after fertilization in medium containing bovine oviduct epithelial cells (BOEC), cumulus cells and granulosa cells to blastocysts were 18.4, 14.6 and 13.1%, respectively. Developmental percentages of blastocysts produced at day 6, 7 and 8 were 8.5, 10.6 and 15.2% respectively. Hatching rate of bovine embryos produced was 60.0%. In experiment 2, post-thawed surviving embryos in a vitrification solution consisting of 7.15M ethylene glycol, 2.5 mM ficoll and 0.3 M sucrose were 36.4% (56/154). Also, survival rate of bovine embryos after exposed to vitrification solution at 1, 2, 3, 4 and 5 min were 84.0, 88.0, 71.0, 48.0 and 24.0% respectively.
