cDNA Cloning of Farnesoic Acid-Induced Genes in Candida albicans by Differential Display Analysis

  • The yeast Candida albicans has a distinguishing feature, dimorphism, which is the ability to switch between two morphological forms: a budding yeast form and a multicellular invasive filamentous form. This ability has been postulated to contribute to the virulence of this organism. Previously, we reported that the yeast-to-hypha transition in this organism is suppressed by farnesoic acid, a morphogenic autoregulatory substance that accumulates in the medium as the cells proliferate. In this study, using a differential display reverse transcription polymerase chain reaction (DDRT-PCR) technique, we have identified several genes induced in C. albicans by farnesoic acid treatment. These observations indicate that farnesoic acid can alter the expressivity of multiple genes, including the DNA replication machinery and cell-cycle-control proteins.

  • A variety of statistical methods are applied to model and optimize responses, related to product or system's quality, in terms of control and noise factors at design and manufacturing stages. Most of them assume continuous response variables but, assessing the performance of a product or system often involves categorical observations, such as ratings and scores. Although most previous works to deal with the categorical data provide sorhisticated response models and ensure unbiased outcomes, they require heavy computation to estimate the model parameters, as well as enough replications. In this study, we present some practical approaches for optimal parameter design with ordered categorical response when only a few or no replication is available. Two real-life examples are given to illustrate the presented methods.

  • Epigenetics represents a chromatin-mediated transcriptional repression which plays a control role in both animal and plant development. A number of different mechanisms have been described to be involved in the formation of chromatin structure: especially DNA methylation, nucleosomal histone modification, DNA replication timing, and binding of chromatin remodelling proteins. Epigenetic phenomena include genomic imprinting, dosage compensation of X-chromosome linked genes, mutual allelic interactions, paramutation, transvection, silencing of invasive DNA sequences, etc. They are often unstable and inherited in a non-Mendelian way. A number of epigenetic defects has been preferentially described in floral development. Here, epigenetic phenomena in model angiosperm plants and their corresponding mechanisms are reviewed.

Studies on the Effect of Triacontanol on the Growth and Yield of Rice Plants (트리아콘타놀이 벼의 생장과 수량에 미치는 영향에 관한 연구)

  • For the purpose of rice yield, 3 rice varieties S-235 (Sam-Pung), S-287(Tai-Baik), S-294(Nam-Pung), control experiment College of Agriculture, Seoul National University garden in 1982. Method of experiment are 4 treatment, 4 replication and randomized block design. Unhilled rice were increased S-235 14.8%, S-287 of 16.2%, S-294 of 15.7%. Each varieties were each sigmificant of 1%. And the best increased, Triacon 10 and $CaCl_2$ 15mM. It think that problems were the more study. Among factor of yield, increase is panice no. of hill and the same varieties. Only S-287 of panicle of hill is significant of 5% and also 1000 seed weight is not significant. The factor of increase yield is important of no. of flower. No. of flower increase is concerned Triacon, the mechanism will study.

On-off Regulation of 3' Exonuclease Excision to DNA Polymerization by Exo+ Polymerase

  • The role of 3' exonuclease excision in DNA polymerization was evaluated in primer extensions using 3' allele-specific primers that had exonuclease-digestible and exonuclease-resistant 3' termini. With exonuclease-digestible unmodified 3' mismatched primers, the exo+ polymerase yielded template-dependent products. Using exonuclease-resistant 3' mismatched primers, no primer-extended product resulted from exo+ polymerase. As a control, polymerase without proofreading activity yielded primer-dependent products from 3' mismatched primers. These data indicated that a successful removal of the mismatch is required for DNA polymerization from the 3' mismatched primers by exo+ polymerase. In addition to the well-known proofreading from this mismatch removal, the premature termination in DNA polymerization, due to the failure of the efficient removal of the mismatched nucleotides, worked as an off-switch in maintaining the high fidelity in DNA replication from exo+ polymerase.

Influence of Pretreatment with Immunosuppressive Drugs on Viral Proliferation

  • Immunosuppressive drugs are used to make the body less likely to reject transplanted organs or to treat autoimmune diseases. In this study, five immunosuppressive drugs including two glucocorticoids (dexamethasone and prednisolone), one calcineurin inhibitor (cyclosporin A), one non-steroid anti-inflammatory drug (aspirin), and one antimetabolite (methotrexate) were tested for their effects on viral proliferation using feline foamy virus (FFV). The five drugs had different cytotoxic effects on the Crandell-Ress feline kidney (CRFK) cells, the natural host cell of FFV. Dexamethasone-pretreated CRFK cells were susceptible to FFV infection, but pretreatment with prednisolone, cyclosporin A, aspirin, and methotrexate showed obvious inhibitory effects on FFV proliferation, by reducing viral production to 29.8-83.8% of that of an untreated control. These results were supported by western blot, which detected viral Gag structural protein in the infected cell lysate. As our results showed a correlation between immunosuppressive drugs and susceptibility to viral infections, it is proposed that immune-compromised individuals who are using immune-suppressive drugs may be especially vulnerable to viral infection originated from pets.

Replication of High Density Patterned Media (고밀도 패턴드 미디어 성형에 관한 연구)

  • In this paper, we investigated the possibility of replicating patterned media by nano-injection molding process with a metallic nano-stamper. The original nano-master was fabricated by E-beam lithography and ICP etching process. The metallic nano-stamper was fabricated using a nanoimprint lithography and nano-electroforming process. The nano-patterned substrate was replicated using a nano-injection molding process without additional etching process. In nano-injection molding process, since the solidified layer, generated during the polymer filling, deteriorates transcribability of nano patterns by preventing the polymer melt from filling the nano cavities, an injection-mold system was constructed to actively control the stamper surface temperature using MEMS heater and sensors. The replicated polymeric patterns using nano-injection molding process were as small as 50 nm in diameter, 150 nm in pitch, and 50 nm in depth. The replicated polymeric patterns can be applied to high density patterned media.

  • In a naval combat system, the information processing node is a key functional equipment and performs major combat management functions including control sensor and weapon systems. Therefore, a failure of one of the node causes fatal impacts on overall combat system capability. There were many methodologies to enhance system availability by reducing the impact of system failure like a fault tolerant method. This paper proposes a fault tolerant mechanism for information processing node using a replication algorithm with hardware duplication. The mechanism is designed as a generic algorithm and does not require any special hardware. Therefore all applications in combat system can use this functionality. The asynchronous characteristic of this mechanism provides the capability to adapt this algorithm to the module which has low performance hardware.

  • Software copyright are written in text form of documents and stored as files, so it is easy to expose on an illegal copyright. The IOT framework configuration and service environment are also evaluated in software structure and revealed to replication environments. Illegal copyright can be easily created by intelligently modifying the program code in the framework system. This paper deals with similarity comparison to determine the suspicion of illegal copying. In general, original source code should be provided for similarity comparison on both. However, recently, the suspected developer have refused to provide the source code, and comparative evaluation are performed only with executable code. This study dealt with how to analyze the similarity with the execution code and the circuit configuration and interface state of the system without the original source code. In this paper, we propose a method of analyzing the data of the object without source code and verifying the similarity comparison result through evaluation examples.

  • BACKGROUND. Even though excellent impression materials are now available for making accurate replication for hard and soft tissue, the numerous dentists have faced lots of obstacles in making simultaneous impressions of multiple abutments. CASE DESCRIPTION. This article describes a modified method of tray fabrication using auto-polymerizing acrylic resin and impression technique for multiple prepared teeth in cases with limitations and difficulties in taking dental impressions. CLINICAL IMPLICATION. This segmental tray technique has several advantages, including higher impression quality, fewer impressions, and being more comfortable for the patient and less stressful for the clinician.
