Influence of Temperature and Relative Humidity in Infection of Nosema bombycis (Microsporidia: Nosematidae) and Cross-infection of N. mylitta on Growth and Development of Mulberry Silkworm, Bombyx mori

  • The influence of temperature and relative humidity in infection and cross-infection of Nosema bombycis and N. mylitta respectively in mulberry silkworm, Bombyx mori L. on larval mortality, multiplication of pathogens, larval weight and growth rate in three different seasons were studied. Seasons were selected in such condition, when very less fluctuations between minimum and maximum temperature and minimum and maximum relative humidity ($25{\sim}28^{\circ}C$ and $65{\sim}72%$ R.H) was observed i.e., season-1. Fluctuations between minimum and maximum temperature were less ($28.05{\sim}34.50^{\circ}C$) but R.H % was more ($55{\sim}81%$) in season-2. Fluctuations between minimum and maximum temperature and R.H % were more ($20.00{\sim}40.5^{\circ}C$ and $64.00{\sim}90.00%$) in season-3. Growth rate of microsporidian-infected silkworm is directly related to the prevailing temperature and relative humidity in silkworm. Silkworm can tolerate slight variation of temperature but slight variation of relative humidity disfavours the development of silkworm and favours the multiplication of pathogens.

  • The control rod worth is a key safety parameter required to be measured in commercial pressurized water reactors (PWRs). Conventionally, the control rod worth is measured after reaching the critical state, which occupies the considerable time in the zero-power physics test. In this study, an efficient control-rod worth measurement technique has been proposed based on the improved neutron-source multiplication method, which can be implemented with the source-range detector count rates in the subcritical states. Moreover, the noise reduction technique has been adopted to smooth the large fluctuation existing in the original signals. In order to verify the engineering performance of the proposed measurement technique, the measured source-range detector count rates during the rod withdrawal process before reaching critical state in a CNP1000 reactor have been employed. It demonstrated that almost all estimated results of control rod worth satisfy the engineering acceptance criteria, except one control rod with the relative difference over 10 %, which indicates the capability of the proposed method in estimating control rod worth.

  • This study was performed to investigate the biocompatibility of several dental casting alloys (Gold, Dong Myung A-45, Dong Myung AP-35, Verabond, Rexillium, NPG) employing tissue culture. Fibroblast-like cells derived from the subcutaneous tissue of Sprague-Dawley rat were cultivated in DME medium with the addition of those alloys. Results were assessed by calculating the cell multiplication rate and relative growth rate and by observing the morphology of cells in the presence of the specimens. Gold was indicated to be most biocompatible with fibroblast-like cell. Dong Myung A-45 and Dong Myung AP-35 showed very similar effects on the cells as did Gold. Also there was a decrease in cytotoxicity of the alloys as the concentration of gold increased. Verabond and Rexillium showed a decreased in cell mutiplication rate as compared to low gold alloys. NPG exhibited the most severe cell toxicity among the tested alloys.

  • Obtaining the accurate final importance of each customer attribute (CA) is very important in the house of quality(HOQ), because it is deployed to the quality of the final product or service through the quality function deployment(QFD). The final importance is often calculated by the multiplication of the relative importance rate and the competitive priority rate. Traditionally, the sample mean is used for estimating two rates but the dispersion is ignored. This paper proposes a new approach that incorporates statistical significance to consider the dispersion of rates in determining the final importance of CA. The approach is illustrated with a design of car door for each case of crisp and fuzzy numbers.

Effect of Pot Depth on Root Development and Distribution during Seedling Growing Period in Tobacco. (육묘포트 깊이가 담배의 뿌리발달 및 분포에 미치는 영향)

  • The study was carried out to clarify reasonable production of healthy seedling, optimal Pot depth, and appropriate transplanting time, which can be deduced from understanding of seedling quality. Seedling quality results from growth of root and shoot, morphology and distribution of root system under influence of Pot depth during seedling growing period. Stem height, shoot dry weight, leaf area and leaf number were increased in proportion to depth of pots. Growth of shoot and root during seedling growing period showed the most dramatic development between 20th and 25th day after temporary planting. Root number increased as pot depth decrease and total root length and dry weight increased as pot depth increase. In 5cm pot, relative multiplication rate was higher and mean extension rate was lower than other depth of Pot. The limitation of pot volume in which rhizosphere was located enhance the development of roots of second and third order. At 20th days after temporary Planting root distribution was relatively uniform in length and development of adventitious root on stem base was poor as Pot depth decreased.

Effect of Space Limitation of Rhizosphere on Morphology and Development of Root System in Tobacco Seedlings (담배 육묘시 근권의 공간 제한이 근계의 형태와 발달에 미치는 영향)

  • This study was carried out to acquire the basic information of root growth under different pot size, imposing different space limitation on rhizosphere. Different size of pots that had same surface area but different depth, 5cm(Iength)$\times$5cm(width)$\times$30, 15, 5cm(depth), were used during the seedling stage of tobacco plant. Space limitation on rhizosphere affected not only the aerial growth, stem height, leaf area and shoot dry weight, but also root growth and root architecture. Aerial growth was highly related to growth of underground part, so space limitation on rhizosphere decreased aerial growth. Limitation on pot volume by reducing pot depth induced new rooting on crown. Root number and relative multiplication rate were higher in small pot that had 5cm depth than large pot, but total root length and mean extension rate showed reverse patterns. Root numbers of 1st order and 2nd order were increased as pot depth was increased, but the root number of 3rd order was increased in small pot. Root system of seedling grown in large pot distributed more horizontally than that in small pot at 20 days after temporary planting (DAT), but the root architecture of seedling was reversed at 25 DAT.

Effective Multiplication of Somatic Embryos Using Suspension Culture and Regeneration in Soybean

  • The use of liquid-medium-based procedure relative to the solid media led to a 4.5-fold increase in the number of cotyledon-stage embryos. The most efficient system for multiplication and regeneration of somatic embryos was CP6 procedure with the media MSD40/MSD20/MSM6AC/FNL0S3S3GM. However, the rate of regeneration was lower. About 71% of the embryos with dicotyledon were continued to develop the roots after desiccation treatment and 92% of the germinated embryos produced shoots in 10 days. Of the four morphologically different types of embryos, dicotyledonous ones showed a high frequency of conversion, while only a few with fused and horn type cotyledon developed shoots. Mature somatic embryos were desiccated in empty petri dishes for 12-72 h. Embryo survival rate was the highest after 12 h of desiccation, but maximal germination was observed at 24 h. After desiccation, they were placed on MS medium without growth regulators for germination. Germinating embryos were transferred to small pots with vermiculite for plant regeneration. The etiolating the plants during the growth was resolved to add 1% activated charcoal on hormone-free MS medium.

  • The ppage resistance mechanism of Lactococcus lactis subsp. cremoris ATCC 11602-A1 was investigated. When parent and A1 were incubated at 30 and 40$^{\circ}C$, A1 grew well and multiplication of phage(MOI=1)on A1 slightly occurred at 40$^{\circ}C$ in contrast with parent. There was a great difference of proteolytic activity between parent and A1, irrespective of the temperature. As a result of ADS treatment oon culture broth, survival rate of A1 was 27% at the lethal concentration of parent and adsorption rate of phage was increased to 95~97%, which was considered to come from the exposure of phage receptor site masked by an unknown component. These results suggest that acridine orange (AO) treatment leads to the modification of cell wall, conferring resistance to high temperature and lytic phage. No change in plasmid profiles of A1 at 30 and 40$^{\circ}C$ were found, which suggests that plasmid is not relative to temperature-resistance of A1.

AN EXPERIMENTAL STUDY ON THE CYTOTOXICITY OF CAVITY LINERS (이장재의 세포독성에 관한 실험적 연구)

  • The purpose of this study was to evaluate the cytotoxic effects of 6 cavity liners in vitro. Human fibroblasts were cultured in ${\alpha}$-MEM and each liner was manually mixed and filled in glass ring cylinder ($8{\times}8mm$ in diameter, in height). The cylinders filled with the liners were placed in the center of the dish (35mm in diameter) containing 3ml of ${\alpha}$-MEM. Millipore filters (pore size $0.22{\mu}m$) to simulate dentin barrier were also placed between the bottom of cylinder and the dish. Then the culture dishes were stored in 5% $CO_2$ containing incubator for 5 and 10 days at the temperature of $36.6^{\circ}C$. The results of the experiments were analyzed by counting the cells in the period of 5 and 10 days respectively, and were assessed by calculating the cell multiplication rate and the relative growth rate. The experiemntal groups and the control group were compared statistically. The results of the study were summarized as follows: 1. The cell number of Zinc oxide-eugenol was $(4.13{\pm}1.31){\times}10^4$ cells/ml at 5 days and $(4.32{\pm}1.61){\times}10^4$ cells/ml at 10 days. 2. The cell number of Cavitec was ($8.35{\pm}2.87{\times}10^4$ cells/ml and $(10.08{\pm}5.10){\times}10^4$ cells/ml at 5 and 10 days respectively. 3. The cell number of Dycal was $(13.56{\pm}3.89){\times}10^4$ cells/ml at 5 days and $(34.75{\pm}8.85){\times}10^4$ cells/ml at 10 days. 4. The cell number of life was $(11.46{\pm}3.32){\times}10^4$ cells/ml and $(21.92{\pm}6.18){\times}10^4$ cells/ml at 5 and 10 days. 5. The cell number of Base cement was $(13.73{\pm}3.73){\times}10^4$ cells/ml and $(36.68{\pm}5.20){\times}10^4$ cells/ml at 5 and 10 days. 6. The cell number of Dentin cement was $(13.58{\pm}3.90){\times}10$ cells/ml and $(66.95{\pm}24.09){\times}10$ cells/ml at 5 and 10 days. 7. The cell multiplication rate of zinc oxide-eugenol cements was significantly less than that of the calcium hydroxide and glass ionomer cement. (P < 0.05)

Changes in Phytoplasma Densities in Witches' Broom-Infected Jujube Trees over Seasons

  • The relative density of phytoplasmas in witches'broom (WB)-infected jujube trees was investigated using compatitive polymerase chain reaction (PCR). During dormant and defoliating seasons, the densities of phytoplasmas were about the same in roots and twigs. In early growing season, the density showed the highest rates in roots, then in twigs and in petioles. however, the density was highest in petioles and the lowest in roots during actively growing season. Throughout the year, root samples did not show any serious fluctuation compared with that of t2wigs and petioles. Density was lowest during actively growing season in root samples. In contrast, petiole sample densities varied to a great extent depending on the season, very high during actively growing season, but very low during the early growing season, In twig samples, the densities were very high and almost the same in both defoliating and dormant seasons. Among the parts of the trees, phytoplsma density was the most stable in root samples throughout the year. The highest densities of phytoplasmas were about the same in all tree parts. These results suggest that the phytoplasmas may overwinter not only in roots but also in twigs, and that multiplication rate of phytoplsma becomes very high right after the early growing season.
