Effects of three kinds of Radix Rehmanniae Water Extract in Cultured Rat Myocardial Cells (삼종 지황 추출물이 배양 심근세포에 미치는 영향)

  • To test the protective effect of herbal medicine on myocardial damage against oxygen free radical-induced myocardiotoxicity, cytotoxicity was examined using MTT, Beating rate and DNA synthesis assay in the presence of water extract of three kinds of Radix Rehmanniae. Myocardial toxicity was evaluated in neonatal rat myocardiocytes in cultures. The results of these experiments were obtained as follows : Xanthine oxydase/hypoxanthine resulted in a decrease in viability, beating rate and DNA synthesis in cultured myocardial cells. Radix Rehmanniae Recens(生地黃, RRR) water extract shows effects of protection from the cardiocyte toxicity induced by xanthine oxydase/hypoxanthine treatment such as increases in beating rate. Radix Rehmanniae Preparat(熟地黃, RRP) water extract shows effects of protection from the cardiocyte toxicity induced by xanthine oxydase/hypoxanthine treatment such as increases in DNA synthesis. These results show that xanthine oxydase/hypoxanthine elicits toxic effects in cultured myocardial cells derived from neonatal rat, and suggest that water extract of three kinds of Radix Rehmanniae is very effective in the prevention of xanthine oxydase/hypoxanthine-induced cardiotoxicity.

Variation of main components according to the number of steaming and drying of Rehmanniae radix preparata

  • Contents of compounds in Rehmanniae Radix change depending on the number of steaming and drying and the drying method. In this study, as an impregnation method for dried Rehmanniae Radix, takju impregnation and cheongju impregnation were carried out and steaming and drying were repeated for 9 times. The changes of 5-HMF and catalpol contents were analyzed according to the number of repetition times to investigate which stage of steaming and drying is preferable. Also, total nitrogen, crude fat, ash, and crude fiber were measured to analyze changes in general components. 5-HMF was not detected in dried Rehmanniae Radix. As a result of repetitive steaming and drying, the content of 5-HMF increased only slightly from 1 to 4-times steaming and drying but increased significantly from 5-times. The catalpol in dried Rehmanniae Radix was not detected after 5 times of steaming and drying. Sucrose, maltose, and glucose were included in dried Rehmanniae Radix before steaming and drying. However, after the process in both Takju impregnation and Cheongju impregnation, galactose and fructose tended to decrease after production and sucrose and glucose tended to decrease after the increase. In this study condition, 6-times and more steaming and drying were appropriate process which met the content criteria (not less than 0.1%) of the Korean Pharmacopoeia (8th edition) for 5-HMF, an index component for quality control of Rehmanniae Radix Preparata.

  • Rehmanniae Radix Preparata attributes good blood circulation and it has been used for the treatment of dizziness, men's sterility, excessive loss of blood and weakness. On the quality control of commercial Rehmanniae Radix Preparata, quantitative determination of 5-hydroxymethyl (5-HMF) using HPLC method has been conducted. Quantitative analysis of S-hydroxymethyl-2-furaldehyde (5-HMF) in commercial Rehmanniae Radix Preparata showed average of $0.136{\pm}0.057%$ in 14 samples collected throughout the regions of Korea.

Antidiabetic, Antioxidative and Renoprotective Effects of Rehmanniae Radix preparata Extract in Streptozotocin-induced Diabetic Rats

  • This study investigated the effect of Rehmanniae Radix preparata extract on the antioxidant enzymes of kidney and renal function in streptozotocin-induced diabetic rats. Male Sprague-Dawley rats were divided into three groups including normal control (NC), diabetic control (DC), and diabetic treatment with Rehmanniae Radix preparata (DRR). Over a 4-week study period, Rehmanniae Radix preparata aqueous extract was administered orally at 1124 mg/kg BW/day. The serum glucose level in the DRR group was significantly lower (P<0.05) than the DC group. The serum blood urea nitrogen in diabetic groups was significantly higher (P<0.001) than the NC group. The urinary total protein level in the DRR group was significantly lower (P<0.05) than the DC group. The renal xanthine oxidase activity in the DRR group was significantly lower (P<0.01) than the DC group. The renal catalase activity in the DC group was significantly lower (P<0.05) compared to the NC group and that was significantly higher (P<0.05) in the DRR group than the DC group. In conclusion, these results indicated that Rehmanniae Radix preparata can prevent or retard the development of diabetic nephropathy via its beneficial effects for correcting the hyperglycemia and favorable effects on antioxidant enzyme system.

Effects of the Aqueous Extract of Rehmanniae Radix Preparata on Lipopolysaccharide-induced Expressions of Cyclooxygenase-2 and Inducible Nitric Oxide Synthase in Mouse BV2 Microglial Cells

  • Rehmanniae radix preparata is the root of Rehmanniae glutinosa Liboschitz var. purpurea Makino which has been classified into Scrophulariaceae. Rehmanniae radix preparata has been used for the treatment of diabetes, for the relief of the pain, and for the anti-oxidative action. In this study, the effect of the aqueous extract of Rehmanniae radix preparata on lipopolysaccharide-induced inflammation was investigated by using 3-(4,5-dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, reverse transcription-polymerase chain reaction (RT-PCR), Western blot, prostaglandin E2 immunoassay, and nitric oxide (NO) detection in mouse BV2 microglial cells. In the present results, the aqueous extract of Rehmanniae radix preparata suppressed prostaglandin E2 (PGE2) synthesis and nitric oxide production by inhibiting the lipopolysaccharide-stimulated expressions of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) mRNA and protein in mouse BV2 cells. These results show that Rehmanniae radix preparata exerts anti-inflammatory effect probably by suppressing of COX-2 and iNOS expressions.

The Study on the Composition of Rehmanniae Radix Extracts by Supercritical Carbon Dioxide Extraction and by Hydrodistillation Extraction (생지황(生地黃) 증류 추출 약침액과 초임계 유체 추출물의 성분 연구(硏究))

  • Objectives : The purpose of this study was to investigate the composition of Rehmannia glutinosa's essential oils with Rehmanniae Radix herbal acupuncture Methods : I obtained the essential oils of Rehmannia Radix by hydrodistillation extraction method and supercritical fluid extraction(SFE) method, and then I analyzed those by GC/MS(gas chromatography/mass spectrum). Results : 1. With GC(gas chromatography) and GC/MS(gas chromatography/mass spectrum) analysis. I identified 9 compounds in the Rehmanniae Radix's essential oil obtained through the SFE method. The main compounds were as follows : Hexachloroethane(2.24%), N-Butyl-benzenesulfonamide(2.05%), hexadecanoic acid(1.93%), hexadecanoic acid, ethyl ester(3.49%), 9,12-Octadecadienoic acid(z,z)(2.70%), (9E)-9-Octadecenoic acid(6.14%), ethyl linoleate(4.43%), ethyl oleate(5.80%). 2. I failed to get Rehmanniae Radix's essential oil obtained through the hydrodistillation method. 3. With GC(gas chromatography) and GC/MS(gas chromatography/mass spectrum) analysis. I identified 4 compounds in the Rehmanniae Radix's essential oil obtained through the hydrodistillation method. The main compounds were as follows : Ethylbis(trimethylsilyl)amine(1.04%), 2-(Trimethylsiloxy)benzoic methyl ester(2.65%), Hexadecanoic acid trimethylsilyl ester(12.61%), octadecanoic acid, trimethylsilyl ester(6.28%). Conclusions : The substances by hydrodistillation method may not perfectly match with the substances by supercritical fluid extraction(SFE) method in essential oils extracted form Rehmanniae Radix. But, the main substances was assumed Hexadecanoic acid and octadecanoic acid.

The Risk Assessment of Residual Pesticides and Heavy Metals in Rehmanniae Radix Preparata (숙지황 중 잔류농약과 중금속의 안전성 평가)

  • To obtain the risk assessment of hazardous materials in Rehmanniae Radix Preparata, the residual pesticides and heavy metals in samples on the Korea and China are surveyed. Group I ($BHC-{\delta}$, $BHC-{\beta}$, Fenitrothion, Penthoate, Endosulfan-${\alpha}$, Dieldrin, Endosulfan-${\beta}$ and Endosulfan-sulfate), Group II (BHC-${\gamma}$, Aldrin, DDD, DDT-p,p Permethrin and Fenvalerate), Group III(BHC-${\alpha}$, Chlorpyrifos, Tolyfluanid, Captan and DDT-o,p) and Group IV(Quintozene, Vinclozolin, DDE and Chlorfenapyr) could analysed on gas chromatography-ECD for evaluation of residual pesticides. Qualified detection concentration on the GC-ECD are $0.45 ng/g{\sim}2.50 ng/g$. Group I, Group II, Group III and Group IV are not detected in Rehmanniae Radix Preparata on the Korea and China. Concentration of As, Cd and Pb in Rehmanniae Radix Preparata. on the Korea are 3.06%, 7.00% and 5.78% for Korea Food & Drug Administration(KFDA). Concentration of As, Cd and Pb in Rehmanniae Radix Preparata. on the China are 5.16%, 5.33% and 6.50% for Korea Food & Drug Administration(KFDA). The hazardous materials in Rehmanniae Radix Preparata on the Korea and China were verified the safety of the residual heavy metals and pesticides compare with Korea Food & Drug Administration (KFDA) advisory level.

Effects of Ginseng Radix, Zizyphi Spinosae Semen and Rehmanniae Radix Preparat on the Serotonin-immunoreactive Cells of the Mouse Brain (인삼, 산조인, 숙지황이 생쥐 뇌의 serotonin 면역반응성 세포에 미치는 영향)

  • Objectives : The aim of this study was to investigate the effects of Ginseng Radix (Panax ginseng C.A. MEY.), Zizyphi Spinosae Semen (Zizyphus spinosa HU.) and Rehmanniae Radix Preparat (Rehmannia glutinosa LIBOSCH.) on the serotonin- immunoreactivity cells in the Caudal raphe nuclei, Rostal raphe nuclei and hippocampus of the mouse brain. Methods : The mice were stressed by restraining for one hour and treated with herbal medicine by oral feeding. The mice were killed after one hour and observed by electron microscope after immunohistochemical staining. Results : In the caudal raphe nuclei of the medulla oblongata, the highest number of 5-HT immunoreactivity cells were observed at the Zizyphi Spinosae Semen-treated group, while the lowest level among the herbs treatment group was shown at the Ginseng Radix group. In the hippocampus, serotonin-immunoreactive cells were expressed significantly at the CA3 area while the lowest level of it was shown at the control group. In the midbrain, immunoreactive cells were expressed higher than other groups, while observed at the lowest level in the control group. Conclusions : The extracts of Zizyphi Spinosae Semen, Rehmanniae Radix Preparat and Ginseng Radix show a certain degree of effect on the change of serotonin immunoreactive neurons as an index of nerve disorder.

Neuroprotective Effects of Rehmanniae Radix on PC12 Cells and Hippocampal Neural Cells (선지황(鮮地黃)이 PC12 세포 및 뇌해마 신경세포 손상에 미치는 영향)

  • The present study investigated neuroprotective effects Rehmanniae Radix on PC12 cells and hippocampal neural cells. PC12 cells were damage by $H_2O_2$ and nitric oxide and organotypic hippocampal slice cultures were damaged by oxygen-glucose deprivation. Then methanol extract of Rehmanniae Radix was treated with 0.5, 5, and $50\;{\mu}g/ml$ in culture media. Effects of Rehmanniae Radix were evaluated with cell viability assay, PI-staining, and TUNEL-labeling. Treatment of Rehmanniae Radix ($with\;5\;and\;50\;{\mu}g/ml$) produced significant increase of cell viability of PC12 cells damaged by $H_2O_2$ and by SNP-induced nitric oxide. Treatment of Rehmanniae Radix produced significant decrease of PI-uptake % in CA1 ($with\;5\;and\;50\;{\mu}g/ml$) and DG ($with\;50\;{\mu}g/ml$) regions of organotypic hippocampal slice cultures damaged by oxygen-glucose deprivation. Moreover, treatment of Rehmanniae Radix produced significant decrease of TUNEL- positive cells in CA1 ($with\;5\;and\;50\;{\mu}g/ml$) and DG ($with\;50\;{\mu}g/ml$) regions of organotypic hippocampal slice cultures damaged by oxygen-glucose deprivation. These results suggest that methanol extract of Rehmanniae Radix has neuroprotective effects on PC12 cells damaged by oxidative stress and on organotypic hippocampal slice cultures damaged by oxygen-glucose deprivation.

Regulatory Effect of Fresh Rehmanniae Radix Extract on the in Vitro Production of Proinflammatory Cytokines in Pristane-Induced Lupus Mice

  • Fresh Rehmanniae radix is known as a traditional medicine with anti-inflammatory and antioxidant activities. However, whether Rehmanniae radix attenuates autoimmune inflammation in lupus models characterized by T cell-dependent autoimmune disease including overproduction of proinflammatory cytokines, loss of T cell tolerance, and B cell hyperactivity remains unclear. We investigated the effect of fresh Rehmanniae radix methanol extracts (RGMeOH) on the in vitro overproduction of proinflammatory cytokines by immune cells from pristaneinduced lupus BALB/c mice. These results showed that RGMeOH remarkably attenuated Con A-increased overproduction of proinflammatory cytokines, such as IL-2, IFN-${\gamma}$, IL-6 and IL-10 by splenocytes from pristaneinduced lupus mice. RGMeOH greatly reduced LPS-induced production of TNF-${\alpha}$ by splenic macrophages from pristane-induced lupus mice, while significantly enhanced LPS-induced production of IL-10 but did not alter IL-6 by splenic macrophages and splenocytes. These findings suggest that RGMeOH may ameliorate lupus systemic inflammatory autoimmunity via down-regulation of TNF-${\alpha}$ and T cell-dependent cytokine production.
