Bio-antimutagenic effects of water extract from Rehmannia glutinosa Liboschitz in SOS Chromotest (SOS Chromotest에서 숙지황 물 추출물의 세포내 항돌연변이 효과)

  • The antimutagenic activity of the water extract of Rehmannia glutinosa Liboschitz (RG) on the mutagenicity induced by 4-nitroquinoline 1-oxide (4-NQO), N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), mitomycin C (MMC), $aflatoxin\;B_1\;(AFB_1)$ and benzo(a)pyrene [B(a)P] were studied using the SOS Chromotest with Escherichia coli PQ37. The water extract of RG was separated into methanol soluble and methanol insoluble parts. The methanol soluble part exhibited higher inhibition effects than the methanol insoluble part against the mutagenic activities of five mutagens. Step-wise fractionation of methanol soluble part was done using methanol, ethyl acetate and water. Among these fractions, water fraction had the strongest inhibitory effects against the mutagenenicity of five model mutagens, showing $4.5{\sim}29.5%$ inhibition, but the $AFB_1$ mutagenic potency was increased slightly by ethyl acetate fraction. The water fraction was further partitioned by sephadex LH-20 column chromtography, and 9 subfractions were obtained. The fraction III showed the strongest inhibitory effects with dose response against the mutagenic activities induced by all the tested chemical mutagens. The inhibition rates of fraction III at concentration of $400\;{\mu}g/assay$ were 29%, 35%, 38%, 25% and 24% against 4-NQO, MNNG, MMC, AFBl and B(a)P, respectively. The fraction III also exhibited a strong bio-an-timutagenicity against 4-NQO and $AFB_1$ by showing more than 40% inhibition.

Physicochemical Components of Rehmannia glutinosa Fermented with Rhizopus delemar (Rhizopus delemar로 발효된 지황의 이화학적 성분 분석)

  • Background: This study was carried out to determine the physicochemical components of Rehmannia glutinosa (RG) fermented with Rhizopus delemar. Methods and Results: Physicochemical components such as changes in moisture content, pH value, total acidity, amount of reducing sugars as well as quantity of free sugars, free amino acids, and catalpol were investigated. Result showed that, the moisture content ranged from 64.26 to 65.51%. The pH and total acidity of the fermented RG decreased significantly during fermentation. The reducing sugar content ranged from 0.10 to 1.34%. The most abundant main free sugars were identified as raffinose, xylose, glucose, fructose, and sucrose. The sucrose content in 80% ethanol and in water extracts increased during RG fermentation. In total, 26 free amino acids were detected, including seven essential amino acids. In addition, the quantity of free amino acids decreased significantly during fermentation. Finally, the catalpol content of the fermented RG was highest on the $2^{nd}$ day of fermentation at 2,028.67 mg/ 100 g. Conclusions: These results indicated that fermentation of Rhizopus delemar could be used to enhance biological activity, and that fermented RG could be used as a functional material and as an edible resource in food and functional materials industries.

Effect of Planting Method on the Growth and Yield of Rehmannia glutinosa Liboschitz (지황재배시 재식방법이 생육 및 수량에 미치는 영향)

  • This experiment was carried out to investigate the proper planting method of Rehmannia glutinasa Liboschitz. A Chungbuk local cultivar was sown on the seed bed of 200cm with the planting density of 30cm(6 rows) $\times$ 8.5cm(30 rhizome per $\textrm{m}^2$). The experimental design was randomized block design with 3 replications. And the result was summarized as follows. The emergence date of level Planting(control : 0$^{\circ}$) was May 23, but the emergence dates of Oblique planting(45$^{\circ}$) and Upright planting(90$^{\circ}$) treatments were later than the control by 2~4 days. And the leaf length and the ratio of leaf length/leaf width were not significantly different. As compared with the 29.2cm and 10.0mm of the rhizome length and the rhizome diameter of level planting treatment, the rhizome length of Oblique planting treatment was decreased by 3.4cm and 0 mm. The length and diameter of rhizome of the Upright planting treatment was decreased by 2.8cm and 1.0mm. And the order of sugar content was level Planting > Oblique planting > Upright planting. The total yields were decreased by 9% in the Oblique planting and by 15% in the Upright planting compared to 1,203kg/10a in the control, and the commercial yields showed the same tendency. The level planting treatment was found to be most proper for sowing of Rehmannia glutinasa Liboschitz in the middle inland region of Korea.

  • The nucleotide sequence of a Korea isolate of broad bean wilt favavirus from Rehmannia glutinosa Lib., designated BBWV-RE, was determined. Direct amino acid sequencing of the virus coat proteins suggests that a comparison of several favaviruses in terms of nucleotide sequence and amino acid sequences showed that BBWV-2 isolates display high sequence identity. The small coat protein genes of RNA-2 were also determined for three other Japanese isolates(E, L, and 1-2) and two ATCC isolates(PV132 and PV176) of BBWV. the CP sequence suggested distinct evolution lineages. Serotype 2 favaviruses are more prevalent in Asia, Australia and North America, Wheres serotype 1 is more prevalent in europe.(중략)

Changes of Chemical Constituents of Rehmannia Radix during 'Steaming and Drying' Process (증폭(蒸曝)에 의한 지황(地黃)의 성분 변화에 대한 고찰)

  • Objectives : 'Steaming and drying' is a traditional processing method that has been used to produce Suk-ji-hwang (熟地黃; Rehmanniae Radix Preparata) from Ji-hwang (地黃, the fresh root of Rehmannia glutinosa Liboschitz ex Steudel; Rehmanniae Radix). The steaming and drying process, which is proceeded in heating and moisturizing conditions, plays a crucial role in the change of therapeutic effect of Ji-hwang, presumably due to the modification of its chemical constituents. In this article, the chemical influence of the 'Steaming and drying' process was investigated for understanding the underlying mechanism of chemical modification of Ji-hwang. Methods : The articles regarding the modifications of chemical constituents of Ji-hwang during the 'Steaming and drying' process were collected and analyzed to investigate the influence of the processing to Ji-hwang. Results : The results indicated that iridoid glycosides were degraded to their aglycones and sugars, and such degradations occurred faster at a high pressure than at an atmospheric pressure during the process. The contents of catalpol, ajugol, and acteoside were decreased, while those of rehmannioside A and D were slightly increased during the repeated processing. The contents of oligosaccharides, namely sucrose, maltose, raffinose, and stachyose (except for manninotriose), were decreased, while those of monosaccharides, glucose and fructose, were increased by the repeated processing. Conclusions : These results demonstrate that the 'Steaming and drying' process influenced the chemical constituents of Ji-hwang and provide probable basis for the therapeutic modification of Suk-ji-hwang after the processing of Ji-hwang.

Analysis of Index Components in Medicinal Crops of Andong Area Cultivated at Different Altitude (안동지역 약용작물의 표고별 지표성분 분석)

  • The contents of index components in 5 medicinal crops cultivated at different heights in Andong area. The contents of allantoin in Dioscorea batatas Decne and catalpol in Rehmannia glutinosa Liboschitz were not different by cultivation heights (100 ~ 200m). But the contents of index components in Astragalus membranaceus and Angelica acutiloba Kitagawa were higher in samples cultivated at elevated heights. At 700 m of heights, the contents of formononetin and astragaloside I in Astragalus membranaceus were 1.20 and $201.2{\mu}g/g$, respectively. The contents of decursin and nodakenin in Angelica acutiloba Kitagawa were 68.3 and 4.20 mg/g, respectively. Thus Astragalus membranaceus and Angelica acutiloba Kitagawa are recommended to be cultivated in higher altitude.

Plant Regeneration from Adventitious Roots of Rehmannia glutinosa Liboschitz and Bioreactor Culture (지황 부정근을 이용한 식물체 재분화 및 생물반응기 배양)

  • This experiment was carried out to develop rapid mass propagation via shoot organogenesis system from adventitious roots of Rehmannia glutinosa. The induction of adventitious roots from leaf explants was most favorable to MS solid medium supplemented with 2mg/L IBA. However, the growth of adventitious roots was highest when they were cultured on 1/3 strength MS liquid medium supplemented with 2mg/L IBA. When the adventitious roots were grown in 10L bioreactor, 10g roots as initial inoculum was increased to 225g after 6 weeks of culture. The harvested roots were cultured onto solid medium to induce plant regeneration. The optimal adventitious shoot formation was observed on MS medium supplemented with 2mg/L BA. Rooting of individual shoots was induced after transfer to half strength MS medium without growth regulators. Plantlets after acclimatization were successfully transplanted in the field and no phenotypic variation was observed among them.
