Ensuring the Quality of Higher Education in Ukraine

  • The National Agency for Quality Assurance in Higher Education plays a crucial role in education in Ukraine, as an independent entity creates and ensures quality standards of higher education, which allow to properly implement the educational policy of the state, develop the economy and society as a whole.The purpose of the article: to reveal the crucial role of the National Agency for Quality Assurance in Higher Education to create quality management of higher education institutions, to show its mechanism as an independent entity that creates and ensures quality standards of higher education. and society as a whole. The mission of the National Agency for Quality Assurance in Higher Education is to become a catalyst for positive changes in higher education and the formation of a culture of its quality. The strategic goals of the National Agency are implemented in three main areas: the quality of educational services, recognition of the quality of scientific results, ensuring the systemic impact of the National Agency. The National Agency for Quality Assurance in Higher Education exercises various powers, which can be divided into: regulatory, analytical, accreditation, control, communication.The effectiveness of the work of the National Agency for Quality Assurance in Higher Education for 2020 has been proved. The results of a survey conducted by 183 higher education institutions of Ukraine conducted by the National Agency for Quality Assurance in Higher Education are shown. Emphasis was placed on the development of "Recommendations of the National Agency for Quality Assurance in Higher Education regarding the introduction of an internal quality assurance system." The international activity and international recognition of the National Agency for Quality Assurance in Higher Education are shown.

Estimating Minimum Efficient Scale of Korean Mobile Telecom Market: Relation between Cost per Minute and Traffic Volume (국내이동통신서비스 시장에서의 최소효율규모 추정: 분당원가와 통화량 간 관계분석을 중심으로)

  • Competition policy for growth and activation of market should consider structural nature of relevant industry closely. In this viewpoint, whether domestic MNOs(mobile network operators) are competing while securing the effect of scale economies and maximum production efficiency is an important element when looking for competition policy means of domestic mobile communication market. With this, present study analyzed level of MES (Minimum Efficient Scale) and whether achieving them or not through investigating the relation between the cost per minute and call traffic of domestic MNOs. This analysis is differentiated from existing econometric studies entailing several assumptions by enhancing a reality reflection through directly deducing of cost curve using verified data(cost per minute and call traffic). As a result of analysis, the levels of minimum efficient scale were different from each other between domestic MNOs, and an incumbent MNO(SKT) accomplished minimum efficient scale while entrant MNOs(KT and LGU+) could not yet reach minimum efficient scale level. This result demonstrates a gap exists in production efficiency level among domestic MNOs and implies that the policy to alleviate the gap among operators is required when establishing competitive policy in the future.

Detection of a Quorum-Sensing Inhibitor from the Natural Products (천연물로부터 Quorum Sensing 저해제의 탐색)

  • The quorum sensing (QS) regulatory network has been the subject of extensive studies during recent years and has also attracted a lot of attention because it both positively and negatively regulates various putative virulence factors, although initially considered to be a specialized system of Vibrio fischeri and related species. In this study, to identify the novel materials which inhibit QS system of microorganisms, extracts of eighteen natural products were tested by bioassay using N-(3-oxohexanoyl)-$_L$-homoserine lactone and N-(3-oxooctanoyl)-$_L$-homoserine lactone synthesized in this experiment and an Agrobacterium tumefaciens NT1 biosensor strain containing a traI::lacZ fusion. The result indicated that the extracts of cabbage, leek, and onion exhibited the QS inhibition activity. Thus, materials contained in the extracts were isolated via recycling preparative HPLC and were purified via a JAIGEL-LS255 column. The common fraction corresponding to a peak of the 83 min point of them quenched the quorum sensing of A. tumefaciens NT1 biosensor strain in ABMM containing X-gal and was designated quorum sensing inhibitor-83 min (QSI-83). The QSI-83 exhibited the heat stability and did not inhibit the growth of A. tumefaciens NTl. Furthermore, thin layer chromatography (TLC) results suggested that these novel materials may be antagonists of N-acyl homoserine lactone or may inhibit the QS autoinducer synthesis by Pseudomonas syringae pv. tabaci.

Generation of 1E8 Single Chain Fv-Fc Construct Against Human CD59

  • Background: Therapeutic approaches using monoclonal antibodies (mAbs) against complement regulatory proteins (CRPs:i.e.,CD46,CD55 and CD59) have been reported for adjuvant cancer therapy. In this study, we generated a recombinant 1E8 single-chain anti-CD59 antibody (scFv-Fc) and tested anti-cancer effect.by using complement dependent cytotoxicity (CDC). Methods: We isolated mRNA from 1E8 hybridoma cells and amplified the variable regions of the heavy chain (VH) and light chain (VL) genes using reversetranscriptase polymerase chain reaction (RT-PCR). Using a linker, the amplified sequences for the heavy and light chains were each connected to the sequence for a single polypeptide chain that was designed to be expressed. The VL and VH fragments were cloned into the pOptiVEC-TOPO vector that contained the human CH2-CH3 fragment. Then, 293T cells were transfected with the 1E8 single-chain Fv-Fc (scFv-Fc) constructs. CD59 expression was evaluated in the prostate cancer cell lines using flow cytometry. The enhancement of CDC effect by mouse 1E8 and 1E8 scFv-Fc were evaluated using a cytotoxicity assay. Results: The scFv-Fc constructs were expressed by the transfected 293T cells and secreted into the culture medium. The immunoreactivity of the secreted scFv-Fc construct was similar to that of the mouse 1E8 for CCRF-CEM cells. The molecular masses of 1E8 scFv-Fc were about 120 kDa and 55 kDa under reducing and non-reducing conditions, respectively. The DNA sequence of 1E8 scFv-Fc was obtained and presented. CD59 was highly expressed by the prostate cancer cell line. The recombinant 1E8 scFv-Fc mAb revealed significantly enhanced CDC effect similar with mouse 1E8 for prostate cancer cells. Conclusion: A 1E8 scFv-Fc construct for adjuvant cancer therapy was developed.

  • Post-translational modification is an efficient process to rapidly transduce external stimulus into cellular response. Ubiquitination is a typical post-translational modification which is a highly conserved process in eukaryotes. UPS (Ubiquitin/Proteasome System) mediated by the ubiquitination is to target diverse cellular proteins for degradation. Among E3 ubiquitin ligases that function as the key determinant for substrate recognition, CRL (cullin–RING E3 ubiquitin ligase) is the largest family and forms the complex composed of cullin, RBX1, adaptor and substrate receptor. Although CRL1, also known as SCF complex, has been widely researched for its biological role, the functional studies of CRL4 have been relatively elusive. In Arabidopsis, there are 119 substrate receptors named DCAF (DDB1 CUL4 Associated Factor) proteins for CRL4 and a fraction of DCAF proteins have been identified for their potential functions so far. In this paper, current understanding on structure and biological roles of plant CRL4 complexes in a diverse of cellular events is reviewed, especially focusing on CRL4 substrate receptors. Moreover, the regulatory mechanism of CRL4’s activity is also introduced. These studies will be helpful to further understand the signal transduction pathways in which such CRL4 complexes are involved and give a clue to establish the action network of entire CRL4 complexes in plants.

KiSS-1 : A Novel Neuropeptide in Mammalian Reproductive System (KiSS-1 : 포유동물 생식계에서의 새로운 신경펩타이드)

  • The hypothalamo-pituitary-gonadal hormone axis is centrally controlled by a complex regulatory network of excitatory and inhibitory signals, that is dormant during infantile and juvenile periods and activated at puberty. The kisspeptins are the peptide products of the KiSS-1 gene and the endogenous agonists for the G protein-coupled receptor 54(GPR54). Although KiSS-1 was initially discovered as a metastasis suppressor gene, a recent evidence suggests the KiSS-1/GPR54 system is a key regulator of the reproductive system. Yet the actual role of the KiSS-1/GPR54 system in the neuroendocrine control of gonadotropin secretion remains largely unexplored, the system could be the first missing link in the reproductive hormonal axis. Central or peripheral administration of kisspeptin stimulates the hypothalamic-pituitary-gonadal axis, increasing circulating gonadotropin levels in rodents, sheep, monkey and human models. These effects appear likely to be mediated via the hypothalamic GnRH neuron system, although kisspeptins may have direct effects on the anterior pituitary gland. The loss of function mutations of the GPR54(GPR54-/-) have been associated with lack of puberty onset and idiopathic hypogonadotropic hypogonadism(IHH). So kisspeptin infusion may provide a novel mechanism for HPG axis manipulation in disorders of the reproductive system.

Gene Expression Profiling of Oilseed Rape Embryos Using Microarray Analysis (Microarray 분석을 이용한 유채 종자성숙단계별 유전자 발현 양상)

  • We observed that oil began to accumulate at 25 seed days after flowering (DAF) and reached the maximum potential at 35 seed DAF of oilseed rape, and the greatest weight of 100 seeds was obtained at 35 seed DAF. To survey a broad analysis of gene expression in developing embryos of Brassica napus, the Bn 300k microarray have been constructed. The Bn 300k Microarrary was designed from 80,696 unigenes clustered from 543,448 ESTs and 780 cDNA at NCBI. These arrays have been hybridized in a series of experiments with probes derived from seeds and leaf of B. napus. Approximately 8.5% of the 7,000 genes were expressed as ratios 2-fold higher in seed (25 DAF) than leaves and 0.4% at ratios 10. Also we observed that storage and cell differentiation-related genes were highly expressed at 10 DAF, whereas energy-related genes including fatty acid metabolism were increased up depending on seed maturation using Microarray, which was confirmed by reverse transcriptase polymerase chain reaction. These results suggest that B. napus arrays provide a very useful data set of seed-specific expression that can be further analyzed by examination of the promoter regions of these genes and help our understanding of the complex regulatory network in developing seeds.

Feedback Control of Cyclooxygenase-2 Expression by Prostaglandin E2 in Rheumatoid Synoviocytes

  • Objective: The role of prostaglandin $E_2$ (PGE2) in the etiopathogenesis of immune and inflammatory diseases has become the subject of recent debate. To determine the role of PGE2 in rheumatoid arthritis (RA), we tested the effect of exogenous PGE2 on the production of cyclooxygenase-2 (COX-2) by rheumatoid synoviocytes. Methods: Fibroblast-like synoviocytes (FLS) were prepared from the synovial tissues of RA patients, and cultured in the presence of PGE2. The COX-2 mRNA and protein expression levels were determined by RT-PCR and Western blot analysis, respectively. The PGE2 receptor subtypes in the FLS were analyzed by RT-PCR. Electrophoretic mobility shift assay (EMSA) was used to measure the NF-${\kappa}B$ binding activity for COX-2 transcription. The in vivoeffect of PGE2 on the development of arthritis was also tested in collagen induced arthritis (CIA) animals. Results: PGE2 ($10^{-11}$ to $10^{-5}M$) dose-dependently inhibited the expression of COX-2 mRNA and the COX-2 protein stimulated with IL-$1{\beta}$, but not COX-1 mRNA. NS-398, a selective COX-2 inhibitor, displayed an additive effect on PGE2-induced COX-2 downregulation. The FLS predominantly expressed the PGE2 receptor (EP) 2 and EP4, which mediated the COX-2 suppression by PGE2. Treatment with anti-IL-10 monoclonal antibodies partially reversed the PGE2-induced suppression of COX-2 mRNA, suggesting that IL-10 may be involved in modulating COX-2 by PGE2. Experiments using an inducer and an inhibitor of cyclic AMP (cAMP) suggest that cAMP is the major intracellular signal that mediates the regulatory effect of PGE2 on COX-2 expression. EMSA revealed that PGE2 inhibited the binding of NF-${\kappa}B$ in the COX-2 promoter via a cAMP dependent pathway. In addition, a subcutaneous injection of PGE2 twice daily for 2 weeks significantly reduced the incidence and severity of CIA as well as the production of IgG antibodies to type II collagen. Conclusion: Our data suggest that overproduced PGE2 in the RA joints may function as an autocrine regulator of its own synthesis by inhibiting COX-2 production and may, in part, play an anti-inflammatory role in the arthritic joints.

A Case Study on the Application of Security Policy for Outsourcing Personnel in case of Large-Scale Financial IT Projects (금융회사 대형 IT프로젝트 추진 시 외주직원에 대한 보안정책 적용 사례 연구)

  • Financial firms strengthen to protect personal information from the leakage, introducing various security solutions such as print output security, internet network Isolation system, isolationg strorage of customer information, encrypting personal information, personal information detecting system, data loss prevention, personal information monitoring system, and so on. Financial companies are also entering the era of cutthroat competition due to accept of the new channels and the paradigm shift of financial instruments. Accordingly, The needs for security for customer information held by financial firms are keep growing. The large security accidents from the three card companies on January 2014 were happened, the case in which one of the outsourcing personnel seized customer personal information from the system of the thress card companies and sold them illegally to a loan publisher and lender. Three years after the large security accidents had been passed, nevertheless the security threat of the IT outsourcing workforce still exists. The governments including the regulatory agency realted to the financail firms are conducting a review efforts to prevent the leakage of personal information as well as strengthening the extent of the sanction. Through the analysis on the application of security policy for outsourcing personnel in case of large-scale Financial IT projects and the case study of appropriate security policies for security compliance, the theis is proposing a solution for both successfully completing large-scale financial IT Project and so far as possible minizing the risk from the security accidents by the outsouring personnel.
