Production of New Regenerated Plant by Anther Culture of Italian Ryegrass (약(葯)배양 기법 이용한 이탈리안 라이그라스 식물체 생산)

  • We obtained regenerated Italian ryegrass (Lolium multiflorum Lam.) plants by anther culture. When Italian ryegrass anther was incubated for 20 days on callus induction medium, MS medium containing 30 g/$\ell$ of sucrose, 2 mg/$\ell$ of NAA and 1 mg/$\ell$ of kinetin, its callus was induced. The ratio of callus induction was 9.2 %, the mean of callus weight was 8.6 mg/callus/anther. When Italian ryegrass callus was incubated for 50 days on plant regeneration medium, MS medium containing 30 g/$\ell$ of sucrose, 1 mg/$\ell$of NAA and 2 mg/$\ell$of kinetin, Italian ryegrass plant was regenerated. The ratio of plant regeneration was 26%.

  • Purpose: Alveolar bone develops with tooth eruption and is absorbed following tooth extraction. Various ridge preservation techniques have sought to prevent ridge atrophy, with no superior technique evident. Collagen has a long history as a biocompatible material. Its usefulness and safety have been amply verified. The related compound, atelocollagen, is also safe and displays reduced antigenicity since telopeptides are not present. Materials and Methods: The current study evaluated whether the $Rapiderm^{(R)}$ atelocollagen plug (Dalim Tissen, Seoul, Korea) improves tissue healing of extraction sockets and assessed the sequential pattern of bone regeneration using histology and microcomputed tomography in six beagle dogs. To assess the change of extraction socket, hard tissues were examined 2, 4, 6, and 8 weeks after tooth extraction. Result: The experimental groups showed better bone fill with slow remodeling process compared to the control groups although there was no statistical difference between groups. Conclusion: The atelocollagen seems to have a tendency to slow bone remodeling in the early phase of healing period and maintain remodeling capacity until late phase of remodeling. Also, use of atelocollagen increased the bone-to-tissue ratio compared to healing of untreated extraction socket.

  • The effect of medium composition on PLB formation and multiplication and shoot regeneration was studied to establish the micropropagation system of the tropical orchid Phalaenopsis. The highest frequence of PLB formation resulted from the VW medium with 1.2 times ion concentration, 1% sucrose, 1.5g/L PVP or 2.5g/L active charcoal, apple and potato extract and 4g/L gellan gum. The highest ratio of PLB multiplication was obtained from the VW medium with 2% sucrose, apple and potato extract and cotton plate. The shoot regeneration was the most effective with the hyponex medium with 3% apple, 3% potato and 4% banana extract.

Desorption and Regeneration Characteristics for Nickel Ions Loaded onto Sericite Using HNO3 Solution

  • Desorption characteristics for ions adsorbed onto sericite was performed by means of $HNO_3$ solution which was selected as the best desorbing agent in the previous work. Elution of nickel ions adsorbed onto sericite using $HNO_3$ solution was confirmed by means of scanning electron microscopy (SEM) & energy dispersive X-ray spectroscopy (EDX) analysis. Desorption efficiency for nickel ions was 100% at the 20 mM of concentration. Also, nickel ions was completely desorbed within 1.0 of S/L (mg/mL) ratio which is defined as the ratio of adding amount of adsorbent and volume of desorbing agent and desorption process was quickly carried out within 60min. Finally, removal efficiency of reused sericite for nickel ions was constantly maintained until the 4th cycle.

  • With the possibility of hot water being able to be used as a heating source in a liquid desiccant system, an experimental apparatus for regeneration of the liquid desiccant was set up and series of experiments were conducted in a climate-controlled chamber. This study was performed to ascertain the influences of experimental factors on regenerating performance and to suggest the optimal combination of factors affecting regeneration rate. Furthermore. in order to figure out the contribution ratio of the factors on regenerating performance, a multi-way factorial design among the design of experiments was adopted. According to experimental results, the most influential factor on regenerating performance was temperature of the liquid desiccant and its contribution ratio was about 79.4%. In addition. the optimal operating combination was as follows; $60^{\circ}C$ of solution temperature, $14\ell$/min of solution flow rate, and 190m3/h of air volume.

  • In this thesis, improved full wave mode ZVT(Zero-Voltage-Transition) PMW DC-DC Converters are presented to maximize the regeneration ratio of resonant energy by only putting an additional diode In series with the auxiliary switch. The operation of the auxiliary switch in a half wave mode makes it possible soft switching operation of all switches including the auxiliary switch whereas it is turned off with hard switching in conventional converter. The increase of the regeneration ratio to resonant energy results in low commutation losses and minimum voltage and current stresses. The operation principles of the improved ZVT PWM DC-DC Converters are theoretically analyzed using the boost converter topology as an example. Both theoretical analysis and experimental results verify the validity of the PWM boost converter topology with the improved full wave mode ZVT PWM converters.

Optimal Ratio of Wnt3a Expression in Human Mesenchymal Stem Cells Promotes Axonal Regeneration in Spinal Cord Injured Rat Model

  • Objective : Through our previous clinical trials, the demonstrated therapeutic effects of MSC in chronic spinal cord injury (SCI) were found to be not sufficient. Therefore, the need to develop stem cell agent with enhanced efficacy is increased. We transplanted enhanced Wnt3-asecreting human mesenchymal stem cells (hMSC) into injured spines at 6 weeks after SCI to improve axonal regeneration in a rat model of chronic SCI. We hypothesized that enhanced Wnt3a protein expression could augment neuro-regeneration after SCI. Methods : Thirty-six Sprague-Dawley rats were injured using an Infinite Horizon (IH) impactor at the T9-10 vertebrae and separated into five groups : 1) phosphate-buffered saline injection (injury only group, n=7); 2) hMSC transplantation (MSC, n=7); 3) hMSC transfected with pLenti vector (without Wnt3a gene) transplantation (pLenti-MSC, n=7); 4) hMSC transfected with Wnt3a gene transplantation (Wnt3a-MSC, n=7); and 5) hMSC transfected with enhanced Wnt3a gene (1.7 fold Wnt3a mRNA expression) transplantation (1.7 Wnt3a-MSC, n=8). Six weeks after SCI, each 5×105 cells/15 µL at 2 points were injected using stereotactic and microsyringe pump. To evaluate functional recovery from SCI, rats underwent Basso-Beattie-Bresnahan (BBB) locomotor test on the first, second, and third days post-injury and then weekly for 14 weeks. Axonal regeneration was assessed using growth-associated protein 43 (GAP43), microtubule-associated protein 2 (MAP2), and neurofilament (NF) immunostaining. Results : Fourteen weeks after injury (8 weeks after transplantation), BBB score of the 1.7 Wnt3a-MSC group (15.0±0.28) was significantly higher than that of the injury only (10.0±0.48), MSC (12.57±0.48), pLenti-MSC (12.42±0.48), and Wnt3a-MSC (13.71±0.61) groups (p<0.05). Immunostaining revealed increased expression of axonal regeneration markers GAP43, MAP2, and NF in the Wnt3a-MSC and 1.7 Wnt3a-MSC groups. Conclusion : Our results showed that enhanced gene expression of Wnt3a in hMSC can potentiate axonal regeneration and improve functional recovery in a rat model of chronic SCI.

An Experimental Study on the Dehumidification Characteristics and the Effectiveness for Operating Conditions of a Desiccant Rotor (운전조건에 따른 제습로터의 제습특성과 유용도에 관한 실험적 연구)

  • An experimental study has been carried out to investigate the dehumidification characteristics for several operating conditions of a compact desiccant rotor. This problem is of particular interest in the design of a desiccant type of dehumidifier. Room temperature, room humidity, regeneration temperature, revolution speed and frontal air velocity of desiccant rotor are varied as operating conditions. The results obtained indicate that dehumidification rate is increased with an increase in the room humidity while dehumidification effectiveness is not changed much. It is also found that the optimal rotor speed and optimal regeneration temperature exist for maximum dehumidification rate and dehumidification effectiveness.

  • Diesel particulate trap is the most reliable system to reduce the particulate matters from diesel engine. Filter is the core component of DPF and ceramic monolith type is dominantly used, which is expensive and fragile relatively at thermal shock. Porous metal filter, which has superior thermal characteristics and low cost, was tested in order to analyze the regeneration performance by using with ferrocene additive. This filter showed the 72% filtration efficiency, additives itself diminished 48% of PM from engine out emission, and final PM reduction ratio of 89% was achieved by DPF system with D-13 test mode.

  • In tissue engineering, scaffolds play an important role in the growth of cells to 3-D organs or tissues. For the success of tissue engineering, they should be mimicked to meet the requirements of natural extracellular matrix (ECM) in the body, such as mechanical properties, adhesiveness, porosity, biodegradability, and growth factor release, etc. Contrary to other materials, polymeric materials are adequate to engineer scaffolds for tissue engineering because controlling the structure and the ratio of components and designing various shapes and size are possible. In this review, the importance, major characteristics, processes, and recent examples of polymeric scaffolds for tissue engineering applications are discussed.
