Changes in Endogenous Polyamine Levels during Polar Regeneration from Populus Leaf Segments (Populus 잎절편의 극성분화시 내생 Polyamine의 함량과 Polyamine 생합성 효소의 활성도 변화)

  • Polyamine titers and the activities of arginine decarboxylase(ADC) and ornithine decarboxylase (ODC), enzymes which catalyze rate-limiting steps in polyamine biosynthesis, were investigated during polar regeneration of Populus leaf segments. The polar regeneration occurred at the basal cut end of Populus leaf segments through cell division around the vascular bundle. In the process of polar regeneration, the titers of putrescine and spermidine increased rapidly but the content of spermine remained constant. The leaf segments were then divided into three separte part ; the proximal, middle and distal. Spermidine titers showed an increase mainly in the proximal parts where polar regeneration occurred. On the other hand, putrescine titers showed an increase in the other two parts. In the course of polar regeneration, the activities of ADC and ODC increased, the ADC activities being higher than those of ODC. However, ODC activity was higher in the proximal part. Therefore, the spermidine contents and ODC activities are suggested to be related to polar regeneration in Populus leaf segments.

Optimization of Protoplast Preparation and Regeneration of a Medicinal Fungus Antrodia cinnamomea

  • Antrodia cinnamomea is a unique medicinal fungus in Taiwan. It has been found rich in some pharmacologically active compounds for anti-cancer, hangover, and immune regulation etc. With the in-depth study of these components, it would be interesting and important to establish a molecular system for basic studies of A. cinnamomea. Thus, we would like to set up a foundation for this purpose by studying the A. cinnamomea protoplast preparation and regeneration. Firstly, we studied the optimization method of protoplast preparation of A. cinnamomea, and found various factors that may affect the yield during protoplast preparation, such as mycelial ages, pH values, and osmotic stabilizers. Secondly, in the regeneration of protoplasts, we explored the effects of various conditions on the regeneration of protoplasts, including different media and osmotic pressure. In addition, we found that citrate buffer with pH value around 3 dramatically increased the regeneration of protoplasts of A. cinnamomea, and provided a set of regeneration methodology for A. cinnamomea.

Construction of Total Information Portal Service for Korea Urban Regeneration

  • In this study, the total information portal service for urban regeneration was constructed to supply comprehensive information for Korean urban regeneration. The portal service is largely divided into an information analysis service for urban regeneration and an information disclosure service. For total information portal service, the information analysis service constructed a system for making district level decline diagnosis and city and county level potential analysis. Moreover, it can construct and control analyzed information specified at the district level. The information disclosure service consists of functions capable of recycling information, interworking the analysis service and facilitating expert participation. It also supplies data of total information DB in reprocessable format. For revitalization of communities, the information analysis service is constructed to lead experts on urban regeneration to share their opinions.

  • The purpose of this study was to propose the urban regeneration project model that reinforce public functions and contribute effectively to urban development. The results of this study were as follows. First, this study developed objective 22 different types of evaluation indexes. The indexes allows to figure out the necessities, business value and conditions of certain urban regeneration project. Second, the urban is divided to 6 types by characteristics of functionality and location. So, each regeneration project model can be applied to meet the condition of the urban.. Third, this study proposed 5 types of urban regeneration project model to fulfil roles of public and encourage the urban development. The urban regeneration project model is supposed to perform the evaluation of urban condition using the index, typological analysis under the law and scheme and analysis of function and urban location.

Studies on Protoplast Isolation and Regeneration of Lyophyllum ulmarium (느티만가닥 버섯의 원형질체 분리와 재생에 관한 연구)

  • This experiment was undertaken to investigate proper conditions for protoplast isolation and regeneration from the mycelia of Lyophyllum ulmnrium. Protoplast isolation and regeneration are influenced by a variety of factors such as enzyme, osmotic stabilizer, reaction time and age of mycelia. A combination of Novozyme 234 (10mg/ml) and cellulase Onozuka R-10 (10 mg/ml) with 0.6 M $MgSO_4$ was most effective for isolation of the protoplasts. The optimum reaction time of the mycelia with the lytic enzymes was 3.5~4 hours at $28^{\circ}C$ in shaking condition at 120 strokes per min. High yield of the protoplasts were obtained from its 4~5 days old mycelia on complete agar media. Its protoplasts were regenerated to normal hyphae. Regeneration media with 0.6 M sucrose were proper for regeneration of the protoplasts. Their regeneration frequency on complete agar media was 2.3~2.7%.

  • A numerical analysis has been conducted on the dehumidification phenomena of rotary absorptive dehumidifier. Parameters that affect the dehumidification efficiency, such as regeneration temperature, humidity, rotor angular velocity, air flow rate and regeneration section angle are studied and optimum driving conditions are determined from the results, Furthermore three new types of dehumidification method are developed to improve the efficiency They are named MODE 2, 3 and 4, while the present one MODE 1. Cooling zone has been constructed between regeneration and process Bone in MODE 2 and as a result exit temperature of the process air decreases. MODE 3 an improvement of MODE 2, recirculates the cooling air into the regeneration zone and regeneration input as well as exit temperature decreases. In MODE 4, some of tee regeneration air is recirculated and it cuts down the regeneration input. Among them MODE 3, showed the best dehumidification efficiency.

  • This Report intends theoretical background of urban planning, related national law system and instances to be researched and concluded as follows below and also to be shared with planners working in the field of urban planning technique services. Revision of new laws and systems are necessary for the reasonable Urban Regeneration promotion through comprehensive and systematic supporting composition revision. For the reason hereafter urban regeneration shall be various and inclusive in society, economy, culture, environment, resident participation. etc., the development model for regeneration project complying with Korean Urban Regeneration condition is required. Diversity and flexibility through governance system are required to ensure city competitive power.
