Calli obtained from a shoot-tip of garlic, Allium sntivum L., were encapsulated using a calcium alginate gel. Some of the encapsulated calli were cultured on a 1/2 MS medium supplemented with 3% sucrose, 10$\^$-5/ kinetin, and 5 ${\times}$ 10$\^$-6/ M NAA whereas the remainder was stored for 40 days at 4$^{\circ}C$. All the naked calli regenerated on the solid medium, while 95% of the encapsulated calli regenerated, and 88% of the encapsulated calli regenerated after 40 days of storage at 4$^{\circ}C$. The capsule matrix delayed the germination time of the encapsulated calli, yet activated the shoot formation of the artificial garlic seeds. The shoot length of the encapsulated garlic calli was much longer than that of the naked garlic calli. The encapsulated garlic calli were dried in a laminar airflow cabinet and the conversion frequency of the dried artificial garlic seeds on a 1/2 MS medium remained at 93% with a water Loss of Less than 50%.
Shoot proliferation was obtained from shoot tips and nodal explants of Vitex negundo L. on MS medium supplemented with either BAP or KIN (0.1-2.0 mg/L) alone or in combination with NAA (0.1 mg/L). The concentrations of cytokinins combined with NAA produced multiple shoots from shoot tips and nodal explants. The highest mean percentage (84.3$\pm$8.0) of shoot multiplication's were observed on nodal explants in the presence of BAP (1.5 mg/L) and NAA (0.1 mg/L) followed by shoot tips (65.0$\pm$5.0). The regenerated shootlets were rooted on MS basal medium IAA, IBA, NAA (0.1-1.5 mg/L). The maximum number of roots (51.0$\pm$2.6) was achieved on the medium containing IBA (1.0 mg/L) followed by other auxins (NAA, IAA). The regenerated plants were successfully transferred to a mixture of vermiculate and soil. About 95% of the plantlets survived when transferred to the field.
This study was conducted to establish mass propagation system from the axillary bud culture of chrysanthemum zawadskii H. which was used as material of medicinal plants. Shoot egeneration was better on MS medium with NAA and BA. The optimum concentraions of growth regulator for shoot regeneration differed depending on accessionsof C. Zawadskii. Shoot regeneration in Keungucheolcho was better on MS Medium with NAA 0.01mg/1 and BA 0.1mg/1 while Hyangrobonggucheocho was better with NAA 0.1mg/1and BA 0.3mg/1. Addition of NAA into medium was effective for induction of root from shoots regenerated. Shoot multiplcation was more effective when 10mg/1 spermine was added into medium than when other polyamines were treated ino medium . Randomly and specifically amplified polymorphic DAC banding patterns based on polymerase chain reaction (PCR) analysis were used to assess the genetic variation of plants regenerated from in vitro culture.
In vitro shoot regeneration of gladiolus in three different culture systems, viz., semi-solid agar (AS), membrane raft (MR), and duroplast foam liquid (DF) cultures was evaluated following the kinetics of shoot multiplication and hyperhydricity at optimized growth regulator combinations. Compared to the AS system, matrixsupported liquid cultures enhanced shoot multiplication. The peak of shoot multiplication rate was attained at 18 days of incubation in the MR and DF systems, whereas the maximum rate in the AS system was attained at 21 days. An early decline in acceleration trend was observed in liquid cultures than the AS culture. The hyperhydric status of the regenerated shoots in the different culture systems was assessed in terms of stomatal attributes and antioxidative status. Stomatal behavior appeared to be normal in the AS and MR systems. However, structural anomaly of stomata such as large, round shaped guard cells with damage in bordering regions of stomatal pores was pronounced in the DF system along with a relatively higher $K^+$ ion concentration than in the AS and MR systems. Antioxidative status of regenerated shoots was comparable in the AS and MR systems, while a higher incidence of oxidative damages of lipid membrane as evidenced from malondialdehyde and ascorbate content was observed in the DF system. Higher oxidative stress in the DF system was also apparent by elevated activities of superoxide dismutase, ascorbate peroxidase, and catalase. Among the three culture systems, liquid culture with MR resulted in maximum shoot multiplication with little or no symptoms of hyperhydricity. Shoots in the DF system were more prone to hyperhydricity than those in the AS and MR systems. The use of matrix support such as membrane raft as an interface between liquid medium and propagating tissue could be an effective means for rapid and efficient mass propagation with little or no symptoms of hyperhydricity.
In sweet potato cultivars 'Mokpo #29' and 'Sanchunza', shoots from extplants were formed 100% on the MS medium with 0.1 ㎎/L NAA and 2.0 ㎎/L BA after 30 days of culture and roots produced from the base of stem at frequencies of 66.7% ('Mokpo #29') and 69.2% ('Sanchunza'), respectively, The media with 0.5∼4.0 ㎎/L BA were produced the greatest frequency of multiple shoot and the most of shoots developed rapidly into normal plantlets with rooting within 60 days of culture. Whereas the cultivar 'Keumsi' failed to produce normal shoot multiplication on the medium with cytokinins alone because of callusing of adventitious shoots. When single shoots with 1 to 2 nodes were excised from the multiple shoot or shoots covered with callus devoid of root and transferred to MS medium with 4.0 ㎎/L BA or kinetin. Host divided shoots showed the callus induction at the stem base and it was enable to obtain regenerated plantlets with shoot and root normally.

This study was conducted to investigate the optimal plant growth regulator level for the shoot formation of Lycium chinense Mill. In vitro plant propagation was developed for leaf explants of boxthorn. Leaf explants were cultured on MS medium supplemented with cytokinins (BA and 2-iP) alone. Plants were successfully regenerated through in vitro culture by using leaf explants of boxthorn grown in the field. After 4 weeks of culture, 58% of shoot formation had developed from the leaf explants. The shoot formation rate of 'Jangmyeong' was highest followed by 'Myeongan', ;Cheondae', and 'Bullo'. The use of 0.2mg/L BA was critical for enhanced production of shoot formation and resulted in 58% of the culture producing shoot formations. Regenerated plantlets transplanted to pots were developed and successfully acclimatized to greenhouse.
An efficient regeneration protocol for stem disc culture of Populus euramericana, which is important species for bioenergy resource in agroforestry, was established. The number of explants that were obtained and the number of explants that regenerated varied with the genotypes. However, in all the genotypes, stem disc culture produced more regenerated shoots than did in axillary bud culture. A comparison of the effects of cytokinin type and concentration on shoot regeneration in different explants (i.e., petiole, leaf, and root segments of P. euramericana) revealed that a concentration of $0.002mg\;l^{-1}$ thidiazuron (TDZ) used on petiole segments resulted in the greatest shoot regeneration (95.83%). The hormonal requirements for the greatest shoot regeneration in the three explant types varied. Different concentrations of $AgNO_3$ and $CoCl_2$ were added separately to the medium to stop the yellowing and subsequent necrosis of the regenerated shoots. Lower concentrations (3 and $5mg\;l^{-1}$) of these compounds improved shoot regeneration and elongation, compared with the control. The in vitro-regenerated shoots were transferred to rooting medium and subsequently acclimatized. The highly efficient regeneration system of P. euramericana reported here can be used for mass propagation of this recalcitrant for regeneration, economically important tree species.
The effects of explant sources, plant growth regulators on callus induction and plantlet differentiation from leaf blade, petiole, and meristem tissue of Pelalgonium citrosa were investigated under illumination or in dark condition Leaf blade explants cultured on Murashige and Skoog's medium containing 2,4-D and kinetin did not form callus or organ. But those cultured on medium with NAA and BA produced callcus and shoots. Dark condition was more effective than light condition to callus induction and showed that some of shoot were differentiated directly from leaf blade explane. Callus proliferated vigorously on meristem tissue after 7 days of culture, and multiple shoots were obtained Sum callus on medium with 0.5 mg/L NAA and BA. Roots formed readily from about 80% of the shoots cultured on medium with 1.0 mg/L NAA. Regenerated plantlets regenerated had phenotypically normal leaves and roots.

An efficient protocol for shoot regeneration was developed for sesame (Sesamum indicum L.) internodes using the transverse thin cell layer (tTCL) culture method. The frequency of shoot regeneration and the number of adventitious buds produced from regenerated shoots depend significantly on explant age, thickness of the tTCL sections, and the phytohormones supplemented to the culture medium. A combination of 6-benzyladenine (2.0 $mg\;l^{-1}$) and a-naphthaleneacetic acid (0.5 $mg\;l^{-1}$) was found to be the best phytohormone combination for shoot bud induction, with the maximum number of shoots obtained when the tTCL sections were 0.5-1.0 mm thick and derived from 4- to 6-week-old seedlings of sesame. Well-developed shoots were rooted on MS medium without phytohormones, and 80% of the regenerated plantlets were successfully established in soil.
