Optimization of Shoot Induction, Histological Study and Genetic Stability of in vitro Cultured Pisum sativum cv. 'Sparkle'

  • An efficient shoot regeneration condition for pea cv. 'Sparkle' was developed by using optimum explant, plant growth regulator concentrations, and pretreatment of BA onto explant. The average shoot number per explant showed the highest on two kinds of shoot induction media (MSB5 media containing 2 mg/L BA and a combination of 2 mg/L BA and 1 mg/L TDZ) when cotyledonary node explants were cultured. Moreover, the pretreatment of explant in 200 mg/L BA solution was found to be more effective in shoot induction than that of non-pretreatment. By histological study, cell division and proto-meristem were formed near the surface of the sub-epidermal and epidermal cell layers of cotyledonary node in earlier than 3 days after culture. The analysis of genetic stability of regenerants by using thirteen ISSR markers showed that in vitro regenerated plants showed polymorphism with 8.3% compared with their mother plants.

  • Xanthoceras sorbifolium Bunge (yellowhorn) is a woody tree in the soapberry family, Sapindaceae, native to northern China. This species has been identified as a major woody bioenergy plant for bio-diesel production because of high oil content in seed. But the flowers do not bear fruit well while the many flowers blooming. This study was performed to regenerate in vitro plantlet using adventitious shoot formation. To establish the protocol of plant regeneration, adventitious shoots formation rate in the culture of cotyledon of immature zygotic embryos was 68.6% in 1/2 MS medium with 0.5 mg l-1 BA and 3% sucrose (w/v). In the culture of cotyledons of mature zygotic embryos, induction of adventitious shoots was needed to contain high sucrose in pre-culture medium and the frequency of shoot induction was 64.4%. Multiple shoots were induced in 0.5 mg l-1 TDZ, and rooting of shoot was induced 4.0 mg l-1 IBA. Flow cytometry analysis revealed that all the regenerated plantlets were diploid.

Recent advances in seaweed seedling production: a review of eucheumatoids and other valuable seaweeds

  • Modern seaweed farming relies heavily on seedlings from natural beds or vegetative cuttings from previous harvests. However, this farming method has some disadvantages, such as physiological variation in the seed stock and decreased genetic variability, which reduces the growth rate, carrageenan yield, and gel strength of the seaweeds. A new method of seedling production that is sustainable, scalable, and produces a large number of high-quality plantlets is needed to support the seaweed farming industry. Recent use of tissue culture and micropropagation techniques in eucheumatoid seaweed production has yielded promising results in increasing seed supply and growing uniform seedlings in large numbers in a shorter time. Several seaweed species have been successfully cultured and regenerated into new plantlets in laboratories using direct regeneration, callus culture, and protoplast culture. The use of biostimulants and plant growth regulators in culture media increases the seedling quality even further. Seedlings produced by micropropagation grew faster and had better biochemical properties than conventionally cultivated seedlings. Before being transferred to a land-based grow-out system or ocean nets for farming, tissue-cultured seedlings were recommended to undergo an acclimatization process to increase their survival rate. Regular monitoring is needed to prevent disease and pest infestations and grazing by herbivorous fish and turtles during the farming process. The current review discusses recent techniques for producing eucheumatoid and other valuable seaweed farming materials, emphasizing the efficiency of micropropagation and the transition from laboratory culture to cultivation in land-based or open-sea grow-out systems to elucidate optimal conditions for sustainable seaweed production.

땃두릅나무 재분화 유식물체의 순화 체계 및 생육 특성 (Establishment of Acclimatization System and Growth Characteristics for Regenerated Plants of Oplopanax elatus Nakai)

  • Background: Oplopanax elatus is widely distributed at high altitudes (about 1,100 m) in China, Russia and Korea. It is hard to propagate, breed, and difficult to grow. Hence, it has been designated as a rare and endangered medicinal plant. A study was conducted to establish a system for large scale seedling production of Oplopanax elatus in vitro and to find the ideal environment for its seedling growth. Methods and Results: In this study, the explants produced under in vitro conditions during our previous study were grouped into three categories (under 10 mm, 10 mm - 30 mm and above 30 mm) based on plant height and were transferred to the growth-chamber and greenhouse for two weeks in each setting for acclimatization. The plantlet category of above 30 mm showed good performance, and was further evaluated under three acclimatization methods as follows: three different growth media (commercial soil, commercial soil + perlite, commercial soil + sand), four shading levels (0%, 50%, 70%, 90%) and four altitude levels (157 m, 218 m, 601 m, 870 m) in Gangwon province of South Korea. As results, O. elatus seedlings showed better growth characteristics at 870 m of altitude, 70% shading level and in the commercial soil compared to other treatments. Conclusions: The regenerated seedlings of Oplopanax elatus obtained through plant tissue culture would be advantageous for use in large scale seedling production systems paired with a good acclimation method. For obtaining optimal results, it is recommended that seedling be acclimatized in a high altitude environment.

유색칼라(Zantedeschia spp. Southern Light) 미숙배 배양에 의한 다량증식 (Mass Production of Calla Lily(Zantedeschia spp. Southern Light) by the Immature Zygotic Embryo Culture)

  • 유색칼라 미숙배 배양에 의한 급속 증식체계를 확립 하고자 Southern Light 품종을 자가수분 시켜 배발육단계 및 식물생장조절제 종류와 농도가 다량의식물체 재분화에 미치는 효과를 조사하였다. 배 발육단계 별로 MS 기본배지 에 배양한 결과 globular 단계 미숙배는 반응이 없었고 몇 개의 어뢰형 배 및 초기 자업기배는 팽대 및 발아되었는데 어린 자엽기 미숙배가 87.5% 발아율을 보여 가장 효과적이었다. 한편 어린 자엽기 미숙배를 2,4-D, NAA및 BA를 단용 또는 혼용처리한 결과 2,4-D 단용 및 BA와 혼용처리배지에서는 유백색의 점액성 캘러스만이 8개월까지 증식되었고 NAA와 BA혼용처리 배지에서는 담황색의 단단한 배발생적 캘러스가 증식되어 모두 식물체로 재분화되었는데 특히 0.5 mg/L NAA와 1.0 mg/L BA혼용처리 하였을때 한개 자엽기 미숙배로 부터 25-30개의 식물체가 재분화 되었으며 동일배지에 10% coconut water를 첨가하므로 기내 식물의 급속생장이 효과적이었다. 배유래 재분화된 식물체는 vermiculite, perlite 및 모래를 1:1:1로 혼합한 토양에 이식하였으며 100% 정상식물체로 자라고 있다.

  • 양성자와 감마선 조사한 추파형 유채 $M_2$세대의 화뢰의 소포자 배발생에 대해 조사하였다. 세 가지 유채품종 '한라', '내한'과 '탐미' 유채종자를 각각 400 Gy와 600 Gy의 양성자와 감마선으로 전처리 하였다. 일부 종의 양성자와 감마선 조사구에서 배발생이 증가되었고 대조구보다 높았다. 품종별로는 '내한'유채가 배발생 빈도가 가장 높았고 '탐미' 유채가 가장 낮았다. 드물게 배상체로부터 바로 소식물체가 형성되었고 대부분은 비정상적으로 multilobe가 형성되었다. 배상체로부터 식물체가 성공적으로 재분화 되었고 화분에 순화되었다.

Plant Regeneration of B.juncea Through Plant Tissue and Protoplast Culture

  • New types of cytoplasmic male sterility in Brassica species would be very useful for the production of F$_1$, hybrid seeds. Leaves and stems of rapid cycling stock of B.juncea (CrGC4-3) containing Anand CMS were used as experimental materials for plant regeneration from protoplast culture. Very high plant regeneration rate (85%) was found in the Kao & Michayluk medium supplemented with 2 mg/L zeatin, 0.5 mg/L BAP, and 1 mg/L NAA when only leaf, not stem, segments were cultured. Protoplasts were isolated from leaves using mixtures of enzymes (1% Cellulycin, 0.5% Macerozyme) in 0.4 M mannitol and 50 mM $CaCl_2$.$2H_2$O. Mcrocalli induced from protoplasts were transferred to the shoot regeneration medium containing 2 mg/L BAP, 2 mg/L zeatin, and 0.5 mg/L NAA. After 60 days of initial protoplast culture, regenerated plantlets were obtained, acclimatized, transplanted into the pots, and grown up to the flowering stage.

패션에 표현된 환경친화적 디자인의 특성 -1990년대 이후의 국내 디자이너 작품을 중심으로- (A Study on the Environment-Friendly Design Expressed in Fashion -Focused on the Korean Designer′s Work since 1990-)

  • The purpose of this study is to investigate the main characteristics of the environment-friendly design expressed in Koran fashion. Environment-friendly design can be categorized into choice of material, extension of products life cycle, and recycling design. In this study, Korean fashion designers can be found having the conciousness of environment for fashion design since 1990. First, in choice of material, the designers used Natural fibers which are cotton, linen, wool, and etc, and used natural dyes. Some of the designers have moved from using real fur to using fake fur for animal welfare. But fake furs produced from synthetic or regenerated fibers have the environmental problems during textile production processes. Some of the designers used fake leather made from the skins of an edible fish which are otherwise going to waste. Secondly, Design for extension of products life cycle can economize the resources and energy. Design for extension of products life cycle are classified into reversible clothing, many function clothing, modular style, patina clothing, simple style, and layered look. Finally, recycling design are classified into recycling of daily necessaries and expression techniques of recycling design which are designer's works used patchwork, mash techniques, and handmade of knits or buttonhole stitch.

In vitro regeneration from cotyledon explants in figleaf gourd (Cucurbita ficifolia Bouch$\'{e}$), a rootstock for Cucurbitaceae

  • An efficient plant regeneration system has been developed for figleaf gourd (Cucurbita ficifolia Bouch$\'{e}$), which is exclusively used as a rootstock for cucumber. The protocol is based on results obtained from a series of culture experiments involving different parts of the cotyledons and various media. The culture of cotyledon explants was critical for the enhancement of shoot regeneration frequency. The lower parts of the cotyledon excised at the plumule base were found to display a markedly enhanced production of adventitious shoots compared to other cotyledon regions. Culture in silver nitrate-supplemented Murashige and Skoog (MS) medium was not beneficial for shoot regeneration and suppressed root regeneration. Efficient shoot regeneration was obtained on MS medium containing 1.0 $mg\;l^{-1}$ zeatin and 0.1 $mg\;l^{-1}$ indole-3-acetic acid. Regenerated shoots successfully elongated and rooted in medium containing 0.1 $mg\;l^{-1}$ 1-naphthalene-acetic acid after 10-15 days of subculturing. The plantlets were satisfactorily acclimatized in a greenhouse and grew into normal plants without any morphological alterations.

Combination Strategy to Increase Cyclosporin A Productivity by Tolypocladium niveum Using Random Mutagenesis and Protoplast Transformation

  • The cyclic undecapeptide cyclosporin A (CyA), one of the most valuable immunosuppressive drugs, is produced nonribosomally by a multifunctional cyclosporin synthetase enzyme complex by the filamentous fungus Tolypocladium niveum. To increase CyA productivity by wild-type T. niveum (ATCC 34921), random mutagenesis was first performed using an antifungal agar-plug colony assay (APCA) selection approach. This generated a mutant strain producing more than 9-fold greater CyA than the wild-type strain. Additionally, a foreign bacterial gene, Vitreoscilla hemoglobin gene (VHb), was transformed via protoplast regeneration and its transcription was confirmed by RT-PCR in the UV-irradiated mutant cell. This led to an additional 33.5% increase of CyA production. Although most protoplast-regenerated T. niveum transformants tend to lose CyA productivity, the optimized combination of random mutagenesis and protoplast transformation described here should be an efficient strategy to generate a commercially valuable, yet metabolite low-producing, fungal species, such as CyA-producing T. niveum.
