Damping-off is a critical disease on ginseng seedling, which caused by the fungal pathogen Rhizoctonia solani. The disease has been prevented by tolclofos-methyl for the last 20 years. However, the tolclofos-methyl usually detected on the harvested roots of 6-year-old ginseng. Herein, we tried to select an alternative pesticide which not only must be safe but also efficiently inhibits the fungal pathogen. Four fungicides (fludioxonil, flutolanil, pencycuron, and thifluzamide) were applied to their inhibition efficacy against the pathogen. In in vitro test, fludioxonil treatment showed 80% inhibition activity for 25 days. Thifluzamide and flutolanil showed the activity for 10 days. Pencycuron showed for 1 days. In addition, the fludioxonil was more effective to control the pathogen comparing to other three fungicides in field. The incidence of damping-off was reduced to 71% by fludioxonil treatment. The level of the fungicide residue in seedling was 0.44 mg/kg, which value will be a negligible level in final products after 5 years. Consequently, the fludioxonil is a conceivable alternative for tolclofos-methyl to cope with R. solani.
Botrytis cinerea infects stems, leaves and fruits of greenhouse tomato and can cause serious economic losses. This study was conducted to develop organic farming control method against tomato gray mold. Twenty two organic farming materials including mineral and plant extracts were screened for the suppressive activity against Botrytis cinerea, in vitro and in vivo. Among the organic farming materials, sulfur, copper, Chinese twinleaf extract and rhubarb extract decreased by 51.7-90% of the spore germination of Botrytis cinerea. Also, gray mold incidence was reduced more than 90% on tomato stems by treating sulfur, seaweed extracts, rhubarb root extracts and Chinese twinleaf extract. After the selected four organic farming materials were applied on tomato cultivated in greenhouse, their control effects against the tomato gray mold were tested. When the water soluble sulfur was foliar-sprayed on the tomato leaves infected by artificial inoculation with spore suspension of Botrytis cinerea, it showed 87.9% of control value. Also, control activity of the water soluble sulfur was paralleled with chemical fungicide, diethofencarb+carbendazim. The above mentioned results indicate the sulfur formulation can be used as chemical fungicide alternatives for controlling tomato gray mold in the greenhouse.
Resazurin-based microtiter assay was used to evaluate the inhibitory effect of fungicides on the respiration of Colletotrichum acutatum s. lat. 20JDS8 sensitive and 20CDJ6 resistant to strobilurin fungicides. The spores of C. acutatum s. lat. 20JDS8 were inoculated into potato dextrose broth (PDB) at densities of 1x104, 1x105 and 1x106 spores/ml, respectively. The relative fluorescence unit (RFU) of all treatments inoculated at each spore density started to rise after 12 hr of incubation, and were 1,965.5, 5,412.5, and 10,061.0, respectively, after 24 hr of incubation. To evaluate the inhibitory effect of fungicide on the respiration of the pathogen, the spores of the pathogen were inoculated into the PDB and treated with the fungicides 0, 6, 12, and 24 hr after incubation, respectively. After keeping the pathogen culturing for another 24 hr, PrestoBlue reagent was treated into the PDB culturing the pathogen. The RFU of each treatment was examined 1 hr after the reagent was treated. When dithianon, isopyrazam, pyraclostrobin, and fluazinam were treated at high concentrations in the stages of spores (immediately after inoculation [0 hr]), spore germination (after incubation for 6 hr), and hyphal growth (after incubation for 12 hr), the respiration of pathogens was inhibited by 90-100%. When the fungicides were treated after culturing the pathogen for 24 hr, the respiratory inhibitory effects were greatly reduced. With pyraclostrobin-resistant C. acutatum s. lat. 20CDJ6, azxoystrobin, trifloxystrobin and kresoxim-methyl, which have the same mode of action, had very little or no respiratory inhibitory effect in all growth stages of pathogens. Based on the above results, it was thought that the resazurin-based microtiter assay could quickly and accurately evaluate the inhibitory efficacy of the fungicides that inhibited respiration.
Potato late blight caused by Phytophthora infestans Cooke is one of the major diseases in the cultivation of potatoes in Korea. Effect of chitosan preparations (SH-1 and SH-2) was evaluated on the inhibition of mycelial growth of P. infestans, and protective activity using detached potato leaf assay both in vivo and in the field test. SH-1 and SH-2 were showed protective activity of young plant with control values more than 95% potato late blight by inoculation with pathogens under growth chamber conditions. Mycelial growth was inhibited the radial growth over 74% at a concentration of $300{\mu}g/ml$ of both SH-1 and SH-2. Spraying with SH-1 and SH-2 on the leaves for detached leaf assay reduced disease development. The content of total polyphenol in stem was significantly increased by SH-1 and SH-2 application in the field. In field experiments, foliar application with both SH-1 and SH-2 were significantly reduced the development of late blight on potato plants. Control of late blight disease was obtained with control values of 72% and 53% by application of 1% SH-1 and SH-2, respectively, with 4 times at 7 days interval, and reduced with similar disease control values by application with 3 times at 14 days interval compared with untreated control. SH-1 and SH-2 applications increased the fresh weight of potato, and higher grade potatoes were also increased. The results showed that SH-1 and SH-2 applications can be used as eco-friendly natural fungicide for organic farming for the increase of yields and control of late blight.
Bacillus subtilis EBS05, an endophytic bacteria strain isolated from a medicinal plant Cinnamomum camphor, can produce antagonistic compounds that effectively inhibit plant pathogenic fungi. The greenhouse experiments showed that wheat sharp eyespot disease (WSED) was reduced by 91.2%, 88.2% and 43.0% after the treatment with fermentation broth, bacteria-free filter and a fungicide fludioxonil, respectively. The culture broth of strain EBS05 can more effectively control WSED than can fludioxonil. The fermentation broth and bacteria-free filter ability to suppress WSED was not significantly different, suggesting that an active secreted substance played a major role in controlling WSED. Separation and purification of the active compounds was carried out by serial processes, including hydrochloric acid (pH 2.0) treatment, methanol extraction and Sephadex LH-20 column chromatography, silica gel column chromatography and reverse-phase high-pressure liquid chromatography (HPLC), respectively. The purified compounds, one of active peaks in the HPLC spectrum, were obtained from the collection. Analysis of the chemical structures by time-of-flight mass spectrometry (TOF-MS) and electrospray ionization mass spectrometry/mass spectrometry (ESI-MS/MS) showed that the active substances produced by the endophytic bacteria EBS05 are mixture of the ${\beta}$-hydroxy-C12~C15-$Leu^7$ surfactin A isomers with 1035.65 Da, 1021.64 Da, 1007.63 Da and 993.65 Da molecular weights, respectively.
To select antifungal surfactants, control efficacy of various nonionic and anionic surfactants on cucumber powdery mildew was evaluated under greenhouse conditions. Among 14 surfactants, pentaetylene glycol monododecyl ether ($C_{12}E_5$), mixture of heptaethylene glycol monodecyl ether and heptaethylene monododecyl ether, and heptaethylene glycol mono-9-octadecenyl ether effectively reduced the development of powdery mildew on cucumber plants. Among the surfactants, $C_{12}E_5$ gave the best control efficacy on the disease and did not show phytotoxic response in cucumber plants. Whereas, fenarimol at a recommended rate (31.3 mg/L) showed less control activity than the surfactant (1,000 mg/L). In addition, mixtures of the surfactant and DBEDC, a protective fungicide, showed high control efficacies against powdery mildews of cucumber and strawberry by additive effect in greenhouse tests.
This study is conducted to determine the potential of zerovalent iron (ZVI), pyrolusite and birnessite to remediate water contaminated with chlorothalonil. The degradation rate of chlorothalonil by treatment of ZVI, pyrolusite and birnessite was much higher in low condition of pH. Mixing an aqueous solution of chlorothalonil with 1.0% (w/v) ZVI, pyrolusite and birnessite resulted in 4.7, 13.46 and 21.38 hours degradation half-life of chlorothalonil, respectively. Dechlorination number of chlorothalonil by treaonent of ZVI, pyrolusite and birnessite exhibited 2.85, 1.12 and 1.09, respectively. Degradation products of chlorothalonil by teartment of pyrolusite and birnessite were confirmed as trichloro-1,3-dicyanobenzene and dichloro-1,3-dicyanobenzene which were dechlorinated one and two chlorine atoms from parent chlorothalonil by GC-mass. Degradation products of chlorothalonil by ZVI were identified not only as those by pyrolusite and birnessite but as further reduced chloro-1,3-dicyanobenzene and chlorocyanobenzene.

Strawberry Fusarium wilt disease, caused by Fusarium oxysporum f. sp. fragariae, is the most devastating disease in strawberry production. The pathogen produces chlamydospores which tolerate against harsh environment, fungicide and survive for decades in soil. Development of detection and quantification techniques are regarded significantly in many soilborne pathogens to prevent damage from diseases. In this study, we improved specific-quantitative primers for F. oxysporum f. sp. fragariae to reveal correlation between the pathogen density and the disease severity. Standard curve $r^2$ value of the specific-quantitative primers for qRT-PCR and meting curve were over 0.99 and $80.5^{\circ}C$, respectively. Over pathogen $10^5cfu/g$ of soil was required to cause the disease in both lab and field conditions. With the minimum density to develop the wilt disease, the pathogen affected near 60% in nursery plantation. A biological control microbe agent and soil solarization reduced the pathogen population 2-fold and 1.5-fold in soil, respectively. The developed F. oxysporum f. sp. fragariae specific qRT-PCR protocol may contribute to evaluating soil healthiness and appropriate decision making to control the disease.
The biocontrol agent Serratia plymuthica A21-4 readily colonized on the root of pepper plant and the bacterium moves to newly emerging roots continuously. The colonization of A21-4 on the pepper root was influenced by the presence ofPhytophthora capsici in the soil. When P. capsici was introduced in advance, the population density of A21-4 on the root of pepper plant was sustained more than $10^6$ cfu/g root until 3 weeks after transplanting. On the other hand, in the absence of P. capsici, the population density of A21-4 was reduced continuously and less than $10^5$ cfu/g root at 21 days after transplanting. S. plymuthica A21-4 inhibited successfully the P. capsici population in pepper root and rhizosphere soil. In the rhizosphere soil, the population density of P. capsici was not increased more than original inoculum density when A21-4 was treated, but it increased rapidly in non-treated control. Similarly, the population density of P. capsici sharply increased in the non-treated control, however the population of P. capsici in A21-4 treated plant was not increased in pepper roots. The incidence of Phytophthora blight on pepper treated with A21-4 was 12.6%, while that of non-treated pepper was 74.5% in GSNU experimental farm experiment. And in farmer's vinyl house experiment, the incidence of the disease treated with the fungicide was 27.3%, but treatment of A21-4 resulted in only 4.7% of the disease incidence, showing above 80% disease control efficacy.
The microorganisms with the antifungal activity against Phytophthora capsici and Colletotrichum acutatum and the plant growth-promoting activity were screened from a forest and natural fields of Gajang-Dong, Sangju-city. One of the isolates, Streptomyces griseofuscus 200401, was selected as a good plant growth-promoting strain in this study. In greenhouse test, the number of leaf, fresh weight, and dry weight of pepper plants, that were grown with treatment of culture suspension or powder containing S. griseofuscus 200401, were higher than those without the bacterial cells. Cultivation of S. griseofuscus 200401 strain for 7 days in a nutrient rich medium produced ammonium chloride up to 0.13 ${\mu}g/ml$ in the culture solution of S. griseofuscus. Treatment of the selected strain significantly reduced the severity of the late blight of pepper plants to show the equivalent disease control activity to chemical fungicide. This study suggests that S. griseofuscus 200401 strain could be a potential biological agent with the biocontrol activity and the plant growth-promoting activity.
