The yeast Saccharomyces cerevisiae has defense mechanisms identical to higher eukaryotes. It offers the potential for genome-wide experimental approaches owing to its smaller genome size and the availability of the complete sequence. It therefore represents an ideal eukaryotic model for studying cellular redox control and oxidative stress responses. S. cerevisiae Yap1 is a well-known transcription factor that is required for $H_2O_2$-dependent stress responses. Yap1 is involved in various signaling pathways in an oxidative stress response. The Gpx3 (Orp1/PHGpx3) protein is one of the factors related to these signaling pathways. It plays the role of a transducer that transfers the hydroperoxide signal to Yap1. In this study, using extensive proteomic and bioinformatics analyses, the function of the Gpx3 protein in an adaptive response against oxidative stress was investigated in wild-type, gpx3-deletion mutant, and gpx3-deletion mutant overexpressing Gpx3 protein strains. We identified 30 proteins that are related to the Gpx3-dependent oxidative stress responses and 17 proteins that are changed in a Gpx3-dependent manner regardless of oxidative stress. As expected, $H_2O_2$-responsive Gpx3-dependent proteins include a number of antioxidants related with cell rescue and defense. In addition, they contain a variety of proteins related to energy and carbohydrate metabolism, transcription, and protein fate. Based upon the experimental results, it is suggested that Gpx3-dependent stress adaptive response includes the regulation of genes related to the capacity to detoxify oxidants and repair oxidative stress-induced damages affected by Yap1 as well as metabolism and protein fate independent from Yap1.
In this study, we examined the inhibitory effects of propenone derivatives, 1,3-diphenyl-propenone (DPhP), 3-phenyl-1-thiophen-2-yl-propenone (PhT2P), 3-phenyl-1-thiophen-3-yl-propenone (PhT3P) and 1-furan-2-yl-3-phenyl-propenone (FPhP), on $TNF-{\alpha}$-induced nuclear factor (NF)-${\kappa}B$ activity and interleukin (IL)-8-induced monocyte adhesion to colon epithelial cells. 1-Furan-2-yl-3-pyridin-2-yl-propenone (FPP-3) that is previously reported as a $NF-{\kappa}B$ inhibitor suppressed $TNF-{\alpha}$-induced monocyte-epithelial cell adhesion in a concentration-dependent manner. The propenone derivatives, DPhP, PhT2P, PhT3P, FPhP, also inhibited $TNF-{\alpha}$-induced $NF-{\kappa}B$ activation in a similar degree to FPP-3. In a DPPH radical scavenging assay, none of the compounds showed DPPH radical scavenging activity, indicating that the inhibitory actions of the propenone derivatives on redox-sensitive $NF-{\kappa}B$ activity is not due to a simple free radical scavenging activity. In addition, the propenone derivatives also suppressed the IL-8-induced monocyte adhesion to colon epithelial cells. Furthermore, the effective concentrations of the propenone derivatives on both $NF-{\kappa}B$ activation as well as IL-8 induced monocyte-epithelial cell adhesion were 1000 times lower than 5-aminosalicylic acid (5-ASA), a clinically used drug for inflammatory bowel disease. These results suggest that the propenone derivatives may be a potential lead having a strong inhibitory activity against inflammatory cytokine-induced epithelial inflammation.
We previously reported that MSTO-211H cells have a higher capacity to regulate Nrf2 activation in response to changes in the cellular redox environment. To further characterize its biological significance, the response of Nrf2, a transcription factor that regulates ARE-containing genes, on the synergistic cytotoxic effect of clofarabine and resveratrol was investigated in mesothelioma cells. The combination treatment showed a marked growth-inhibitory effect, which was accompanied by suppression of Nrf2 activation and decreased expression of heme oxygenase-1 (HO-1). While transient overexpression of Nrf2 conferred protection against the cytotoxicity caused by their combination, knockdown of Nrf2 expression using siRNA enhanced their cytotoxic effect. Pretreatment with Ly294002, a PI3K inhibitor, augmented the decrease in HO-1 level by their combination, whereas no obvious changes were observed in Nrf2 levels. Altogether, these results suggest that the synergistic cytotoxic effect of clofarabine and resveratrol was mediated, at least in part, through suppression of Nrf2 signaling.
In this paper, an electrochemical sensor for epinephrine (EP), a neurotransmitter was developed by anchoring molecularly imprinted polymeric matrix (MIP) on the surface of gold-coated quartz crystal electrode of electrochemical quartz crystal microbalance (EQCM) using starch nanoparticles (Starch NP) - reduced graphene oxide (RGO) nanocomposite as polymeric format for the first time. Use of EP in therapeutic treatment requires proper dose and route of administration. Proper follow-up of neurological disorders and timely diagnosis of them has been found to depend on EP level. The MIP sensor was developed by electrodeposition of starch NP-RGO composite on EQCM electrode in presence of template EP. As the imprinted sites are located on the surface, high specific surface area enables good accessibility and high binding affinity to template molecule. Differential pulse voltammetry (DPV) and piezoelectrogravimmetry were used for monitoring binding/release, rebinding of template to imprinted cavities. MIP-coated EQCM electrode were characterized by contact angle measurements, AFM images, piezoelectric responses including viscoelasticity of imprinted films, and other voltammetric measurements including direct (DPV) and indirect (using a redox probe) measurements. Selectivity was assessed by imprinting factor (IF) as high as 3.26 (DPV) and 3.88 (EQCM). Sensor was rigorously checked for selectivity in presence of other structurally close analogues, real matrix (blood plasma), reproducibility, repeatability, etc. Under optimized conditions, the EQCM-MIP sensor showed linear dynamic ranges ($1-10{\mu}M$). The limit of detection 40 ppb (DPV) and 290 ppb (EQCM) was achieved without any cross reactivity and matrix effect indicating high sensitivity and selectivity for EP. Hence, an eco-friendly MIP-sensor with high sensitivity and good selectivity was fabricated which could be applied in "real" matrices in a facile manner.
연구배경: 활성산소종(reactive oxygen species)은 발암 기전의 여러 단계 과정에 관여한다. 대부분의 종양 세포주 및 종양 조직내의 종양 세포는 활성산소종을 생성하는 반면 종양 세포의 catalase, Mn- 및 CuZn-SOD등 기존 항산화 단백의 활성도는 대부분 저하되어 있다. 이로 인한 종양 조직내의 지속적인 산화 스트레스는 종양의 국소 침습 및 전이를 촉진한다. 12-kDa thioredoxin은 glutathione 및 glutaredoxin과 함께 세포내 산화-환원 전위를 조절하여 세포 활성, 증식, 분화 및 산화-환원에 의한 아포토시스 조절에 관여하는 것으로 알려져 있다. 한편 histiocytic lymphoma 세포 (U937, human)에서 14-kDa 및 10-kDa의 eosinophilic cytotoxic enhancing factor(ECEF)로 정제되었으며 호산구 자극의 생물학적 기능은 10-kDa에서 20배 이상 높은 것으로 알려져 있다. 성인 T-세포백혈병, 자궁경부상피세포암 및 간세포암에서 thioredoxin 양이 증가 되어 있고 폐암에서는 thioredoxin mRNA가 증가되어 있는 것으로 알려져 있다. 이에 폐암 조직과 주위 정상 조직을 비교하여 catalase, CuZn-SOD 및 glutathione peroxidase 등 기존 항산화 단백과 thioredoxin 발현 변화를 비교 관찰하고 대식세포에서 산화 스트레스 및 내독소에 의한 thioredoxin 발현 변화를 관찰하고자 하였다. 방 법: 동일한 환자의 폐암 조직과 주변의 정상 폐 조직을 immunoblot 분석으로 catalase, CuZn-SOD, glutathione peroxidase 및 thioredoxin 발현을 비교 관찰하였으며 대식세포인 mouse monocyte-macrophage 세포 (RAW 264.7)에 5 ${\mu}M$ menadione 및 1 ${\mu}g/ml$ endotoxin을 처치하여 thioredoxin 발현을 관찰하였다. 결 과: Immunoblot 분석상 12-kDa의 thioredoxin 발현은 폐암 조직에서 정상 폐조직과 비교하여 의미있는 증가를 보였으나 catalase 및 CuZn-SOD의 발현은 폐암 조직에서 정상 폐조직과 비교하여 감소하였고 glutathione peroxidase의 발현은 일정 하지 않은 변화를 보였다. 절단형(truncated) thioredoxin 역시 폐암에서 증가하였다. Mouse monocyte-macrophage cells에 5 ${\mu}M$ menadione 및 1 ${\mu}g/ml$ endotoxin을 처치하였을때 thioredoxin 발현은 12시간에 최고로 증가하여 48 시간까지 지속되었다. 결 론: 폐암에서 기존의 항산화 단백과는 달리 12-kDa 및 절단형 thioredoxin 발현이 증가하며 이는 종양 조직내의 지속적인 산화 스트레스와 밀접한 연관이 있다. 특히 절단형 thioredoxin의 생물학적 기능을 고려할 때 절단형 thioredoxin 발현 증가는 종양 세포 증식을 통한 종양 성장에 더욱 의미있는 역할하리라고 생각된다.
Down-conversion of Eu$^{3+}$ doped LiGdF$_4$ (LGF) for increasing the cell efficiency on dye-sensitized Ti $O_2$ solar cells has been studied. The dye sensitized solar cell (DSC) consisting of mesoporous Ti $O_2$ electrode deposited on transparent substrate, an electrolyte containing I$^{[-10]}$ /I$_3$$^{[-10]}$ redox couple, and Pt counter electrode is a promising alternative to the inorganic solar cell. The structure of DSC is basically a sandwich type, viz., FTO glass/Ru-red dye-absorbed Ti $O_2$/iodine electrolyte/sputtered Pt/FTO glass. The cell without down converter had open circuit potential of approximately 0.66 Volt, the short circuit photocurrent density of 1.632 mA/$\textrm{cm}^2$, and fill factor of about 50 % at the excitation wavelength of 550 nm. In addition, 5.6 mW/$\textrm{cm}^2$ incident light intensity beam was used as a light source. From this result, the calculated monochromatic efficiency at the wavelength of 550 nm of this cell was about 9.62 %. The incident photon to current conversion efficiency (IPCE) of N3 used as a dye in this work is about 80 % at around 590 nm and 610 nm, which is the emission spectrum of Eu$^{3+}$ doped LGF, results in efficiency increasing of DSC.C.
The synergic effect of iron plus blue light on the peroxidation of membrane lipid was investigated, using liposomes made of phospholipid. While strong irradiation did not affect Fe$^{+2}$-promoted lipid peroxidation that turned out to be O$_2$-dependent, ferric iron in bright light exerted a pronounced effect on the initiation of lipid peroxidation: this combined action of light and Fe$^{+3}$ on liposomal membranes was apparently independent of O$_2$. When liposomal samples containing Fe$^{+3}$ were subjected to irradiation, some portions of Fe$^{+3}$ were converted into Fe$^{+2}$. The extent of the Fe$^{+3}$-Fe$^{+2}$ conversion increased with increasing time of irradiation, which resembled the dependence of Fe$^{+3}$-promoted lipid peroxidation on irradiation. Further, it was observed that the effect of irradiation in liposomal samples containing Fe$^{+2}$ was strikingly mimicked by that of Fe$^{+2}$ addition to the same samples. The obligatory requirement of a suitable Fe$^{+3}$/Fe$^{+2}$ ratio for the genesis of iron-dependent lipid peroxidation, a controversial proposition, was also confirmed by the observation that lipid peroxidation was substantially enhanced by the addition of a mixture of Fe$^{+3}$ and Fe$^{+2}$, as compared to the addition of Fe$^{+3}$ or Fe$^{+2}$ alone. The results obtained in this study not only suggest that light acts as an effector for initiating lipid peroxidation, when Fe$^{+3}$ is present in membrane systems, but also imply that any chemical or physical factor that influences the redox states of iron in membranes can play a role in lipid peroxidation reactions.

Reactive oxygen species (ROS) at high concentrations induce oxidative stress, an imbalanced redox state that is a prevalent cause of neurodegenerative disorders. This study aimed to investigate the protective effect of Capsosiphon fulvescens (CF) extract on oxidative stress-induced impairment of cognitive function in models of neurodegenerative diseases. CF was extracted with subcritical water and several solvents and H2O2 (0.25 mM) or aluminum chloride (AlCl3; 25 µM) as an inducer of ROS was treated in PC12 neuronal cells and zebrafish larvae. All statistical analyses were performed using one-way analysis of variance and Dunnett's test using GraphPad Prism. H2O2 and AlCl3 were found to significantly induce ROS production in PC12 neuronal cells and zebrafish larvae. In addition, they strongly affected intracellular Ca2+ levels, antioxidant enzyme activity, brain-derived neurotrophic factor (BDNF) and tropomyosin receptor kinase B (TrkB) signaling, acetylcholinesterase (AChE) activity, and hallmarks of Alzheimer's disease. However, treatment of H2O2-induced PC12 cells or AlCl3-induced zebrafish larvae with CF subcritical water extract at 90℃ and CF water extract effectively regulated excessive ROS production, intracellular Ca2+ levels, and mRNA expression of superoxide dismutase, glutathione peroxide, glycogen synthase kinase-3 beta, β-amyloid, tau, AChE, BDNF, and TrkB. Our study suggested that CF extracts can be a potential source of nutraceuticals that can improve the impairment of cognitive function and synaptic plasticity by regulating ROS generation in neurodegenerative diseases.
Pseudomonas aeruginosa는 기회 감염성 병원균으로, Cystic fibrosis, 미생물 감염성 각막염,화상 부위 2차 감염 등의 다양한 질병을 초래한다. 정족수 인식(쿼럼 센싱)이라고도 알려져 있는 세포간 신호전달 기전이 이러한 감염에서 중요한 역할을 하기 때문에 P. aeruginosa의 정족수 인식 시스템들이 집중적으로 연구되어 왔다. P. aeruginosa의 정족수 인식 시스템들을 소개하는 많은 문헌들이 주로 두 개의 주요 acyl-homoserine lactone (AHL) 계열 정족수 신호물질들인 N-3-oxododecanoyl homoserine lactone (3OC12)과 N-butanoyl homoserine lactone (C4)에 초점을 맞추어 설명하고 있지만, 실제로는 몇 가지 새로운 신호물질들이 발견되어져 왔고, 그들이 P. aeruginosa의 병독성과 신호전달에 중요한 역할을 할 수 있음이 제안되어져 왔다. 그 중 하나가 PQS(Pseudomonas quinolone signal; 2-heptyl-3-hydroxy-4-quinolone)인데, 이 물질은 현재 P. aeruginosa의 잘 규명된 주요 신호물질로 인식되고 있다. 이에 더하여, 최근의 연구들은 또 다른 가능성 있는 P. aeruginosa신호물질들을 제안해 왔는데, diketopiperazines (DKPs)과 pyocyanin이 그들이다. DKPs는 환형 dipeptide로써 이를 구성하는 아미노산의 종류에 따라 다양한 구조를 가진다. P. aeruginosa의 배양액에서 검출된 몇몇 DKPs들이 기존에는 AHL에만 특이적으로 반응한다고 알려졌던 Vibrio 랸�N갸 LuxR biosensor를 활성화 시킬 수 있다는 것이 발견되어 새로운 신호물질로 제안되었다. Pyocyanin (1-hydroxy-5-methyl-phenazine)은 P. aeruginosa가 생산하는 여러 phenazine 화합물들 중의 하나로써, 특징적인 청록색을 띄는 산화-환원 활성물질이다. 이 물질도 정체 성장기 동안 일부 정족수 인식의 조절을 받는 유전자들의 발현을 증가시키는 최종 신호 인자로 최근 제안되었으며, 그 신호는 또 다른 전사 조절 인자인 SoxR에 의해 매개된다고 제안되었다. 본 논문에서는 P. aeruginosa에서 새롭게 발견, 제안되고 있는 이들 신호 전달 물질들에 대해 자세히 다루어 보기로 한다.
Dially disulfide (DADS)는 마늘의 주요한 유기 황화합물 성분으로서 다양한 약리 작용을 나타낸다. 최근 DADS가 항염증과 항동맥경화 작용뿐만 아니라 암세포의 증식을 억제하고 사멸을 유도한다는 보고가 이어지고 있고, 이에 관련된 연구가 활발히 진행되고 있는 실정이다. 한편, DADS가 세포 내 항산화 인자인 glutathione을 증가시킨다는 연구결과와 세포 내 항산화 효소의 일종인 HO-1의 발현을 직접 유도한다는 결과가 보고되었다. 그래서, 본 연구에서는 논란이 되고 있는 DADS의 세포 내 항산화 효소인 HO-1의 발현에서의 효과 및 그 전사인자들의 작용에 관여하는지를 인간 간암세포주 HepG2에서 조사하였다. 배양 중인 HepG2 세포에서 DADS는 독성이 없는 농도에서 세포의 증식을 크게 억제하였고, 전사인자 Nrf2의 발현을 약하게 유도하였으나 HO-1의 발현에는 영향을 미치지 못하는 것으로 나타났다. 또한, DADS는 HO-1 유도제인 CoPP와 hemin에 의해 자극된 HepG2 세포의 HO-1 발현의 증가를 단백질 수준에서 강력하게 억제시키는 것으로 나타났다. 그러나 DADS는 CoPP에 의한 HO-1 유전자의 mRNA 수준의 전사에는 억제 효과를 보이지 않았으며, 또한 Nrf2와 small Maf의 발현을 증가시키고 핵 내에 축적시키는 것으로 나타났다. 이를 종합해 볼 때 DADS는 단독으로 HO-1 발현을 유도하지 못하고, HO-1 유도제에 의한 HO-1 유전자의 발현과정에서는 전사단계가 아닌 번역단계에서 역할을 함으로써 HO-1의 단백질 합성을 억제하는 것으로 보인다. 결론적으로, 항산화 효소인 HO-1의 활성은 외부 자극으로부터 세포를 보호하고 사멸에 저항하게 하는데, DADS는 인간 간암세포주 HepG2에서 이 효소의 발현을 억제함으로써 항암제 및 redox 변화에 따른 암세포주의 성장을 억제하고 세포사멸을 촉진시킬 수 있다고 여겨진다.

③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
