Reduction of Tomato spotted wilt virus on Table Tomatoes in Greenhouses by Soil Fumigation

  • Tomato spotted wilt virus (TSWV) has occurred on fields annually disease occurrence rates were 73.3% in 2005, 53.3% in 2006 and 41.6% in 2007 at Anyang area in Gyeonggi Province. Seasonal occurrence pattern of TSWV showed a dramatic increase from 8.7% in late May to 30.1 % in early June in 2007 at Anyang area, coincided with the high population of a thrip, Frank-liniella occidentalis at that time. The rate of viruliferous thrips with TSWV on lettuce and red pepper was 20.2% and 52.1%, respectively, in greenhouses. Dazomat, soil fumigation pesticide, reduced TSWV disease incidence drastically on table tomato as treatment the chemical into the soil with humidity in early spring in 2006 and 2007. Spraying insecticide periodically after treatment with Dazomat was more effective to control TSWV than spraying if on plants or applying into the soil of the insecticide during growing season. Control efficiency through treatments both of the soil fumigation and of spraying insecticide was significantly high with 85.3% in 2006 and 87.8% in 2007. Removing the potential vector from the soil of TSWV infested area can be an effective strategy for reducing TSWV disease.

Environmental contamination and geochemical behaviour of heavy metals around the abandoned Songcheon Au-Ag mine, Korea

  • The objective of this study is to investigate the contamination levels and dispersion patterns of arsenic and heavy metals and to estimate the bioaccessible fraction of the metals in soil and plant samples in the vicinity of the abandoned Songcheon Au-Ag mine. Tailings, soils, plants (Chinese cabbage, red pepper, soybean, radish, sesame leaves, green onion, lettuce, potato leaves, angelica and groundsel) and waters were collected around the mine site. After appropriate preparation, all samples were analyzed for As, Cd, Cu, Pb and Zn by ICP-AES and ICP-MS. Elevated levels of As and heavy metals were found in tailings. Mean concentrations of As in agricultural soils were higher than the permissible level. Especially, maximum level of As in farmland soil was 513 mg/kg. The highest concentrations of As and Zn were found in Chinese cabbage (6.7 mg/kg and 359 mg/kg, respectively). Concentrations of As, Cd, and Zn in most stream waters which are used for drinking water around this mine area were higher than the permissible levels regulated in Korea. Maximum levels of As, Cd and Zn in stream waters were 0.78 mg/L, 0.19 mg/L and 5.4 mg/L, respectively. These results indicate that mine tailings can be the main contamination sources of As and heavy metals in the soil-water system in the mine area. The average of estimated bioaccessible fraction of As in farmland soils were $3.7\%$ (in simulated stomach) and $10.8\%$ (in simulated small intestine). The highest value of bioaccessible fraction of metal in farmland soils was $46.5\%$ for Cd.

Plant Growth-promoting Activity of Acremonium strictum MJN1 Isolated from Roots of Panax ginseng

  • The plant growth-promoting activity of Acremonium strictum MJN1 isolated from roots of Panox ginseng was explored. The myceliaI extract of A. strictum MJN1 enhanced the rice seedling growth by 14.5 and 9.0% in the dried weight of shoots and roots, and the growth of red pepper by 54 and 85% in the top length and the dried weight in pot experiments, respectively. The plant growth-promoting substances in the myceliaI extract of Acremonium strictum MJN1 were identified as D-adenosine and glycerol. Both commercial D-adenosine and glycerol also promoted significantly the rice seedling growth but, unlike the mycelial extract of A. strictum MJN1, hardly affected the yields of plants grown in pots or field. Therefore, it is possible that other plant growth-promoting substances are produced by A. strictum MJN1. However, this study shows that A. strictum MJN1 has a great potential as a biofertilizer.

Studies on Mild Mutants of Tabacco Mosaic Virus I. Induction of Mild Mutants and Their Characteristics (약독 담배 모자이크 바이러스 I. 약독바이러스의 유기 및 특성)

  • Three mild mutant strains of tobacco mosaic virus (TMV) were isolated from Nicotiana tabacum var. Samsun incubated at $38^{\circ}C$ for 10 days after inoculation with a wild type of TMV-OM strain. They were designated into Tg 5272, Tw 227 and Tw 333. All mild strains could be distinguished from TMV-OM by their reactions on different indicator plants. The mild strains induced the mild mottling without distinct symptoms, whereas the wild strain produced severe mosaic, rugose and stunting on tobacco and red pepper plants. Tw 227 and Tw 333 produced smaller necrotic spots than those of Tg 5272 and TMV-OM on N. glutinosa and Datura stramonium. The former two strains also produced ring spots and mosaic on Gomphrena globosa compared with necrotic spots by the latter strains. Three mild strains were serologically identical to TMV-OM. Their physical properties were thermal inactivation point $80-85^{\circ}C$, dilution end point between $10^{-4}\;and\;10^{-6}$, and longevity in vitro 7days or longer. Ultraviolet absorption spectra of purified preparations of the mild strains and TMV-OM were identical, with a minimum at 247nm, a maximum at 260nm, and a slight shoulder at 290nm. Electrophoresis of the strains in polyacrylamide-agarose gel showed that all the strains formed one major band and two minor bands, except for one minor band of Tw 333. However, when sodium dodecyl sulfate was added to the purified viruses before electrophoresis, each strain formed only one major band.

  • Various types of amendments have been studied for heavy metal stabilization in soil. However, researches on the effect of amendments on alkali soil and their effects on the plants at various edible parts are insufficient. The aim of this study was to evaluate the stabilization efficiency of heavy metals and their transfer into edible parts of food crops. Abandoned mine area was selected and 3 types of amendments (lime stone, LS; steel slag, SS; acid mine drainage sludge, AMDS) was applied with 3% (w/w). in field. After 6 month aging, Chinese cabbage (leafy), bok choy (leafy), garlic (root) and red pepper (fruit) were transplanted and cultivated. For chemical assessment, total concentration and bioavailability using Mehlich-3 solution were determined. For biological assessment, fresh weight and heavy metal uptakes were analyzed. It was revealed that AMDS reduced bioavailability most effectively, resulting in the decrease in heavy metal concentration in edible parts of all crops. When explaining the heavy metal uptake of plants, the bioavailability was more appropriate than the total contents of soil heavy metals. Therefore, bioavailability-based further research and management practices should be carried out continuously for the sustainable environment management, safe crop production, and human health risk reduction.

Identification and Biological Activity of Two New Phytotoxins Isolated from Botrytis cinerea (Botrytis cinerea로부터 분리한 두 개의 새로운 phytotoxin의 구조 결정 및 생물활성)

  • We discovered two novel phytotoxins produced by the pathogenic fungus, Botrytis cinerea. Among the twenty-five B. cinerea isolates, which were obtained from various host plants in 1994 and 1996, twenty-two showed strong or moderate pathogenicity on five plants such as cucumber, tomato, red pepper, tobacco and Chinese cabbage. The culture filtrate of the B. cinerea 2-16 strain showed the most potent phytotoxic activity in a tobacco leaf-wounding assay. Two novel phytotoxins were isolated from the liquid cultures of B. cinerea 2-16 by ethyl acetate extraction, flash silica gel column chromatography, silica gel column chromatography, Sephadex LH-20 column chromatography, preparative TLC and subsequently preparative HPLC. Their chemical structures were determined to be 3-O-acetyl botcinol and 3-O-acetyl botcinolide, respectively, by mass and NMR spectral analyses. These two phytotoxins caused leaf necrosis in a leaf-wounding bioassay, and significant electrolyte leakage from leaf tissues of tobacco. In the two bioassays tested, 3-O-acetyl botcinol exhibited stronger phytotoxic activity than 3-O-acetyl botcinolide. This is the first report on the production of both 3-O-acetyl botcinol and 3-O-acetyl botcinolide from B. cinerea.

Effect of Plants Extracts on Lipid Peroxidation of Rat Brain Tissue Induced by Reactive Oxygen Species (활성산소에 의해 유도된 흰쥐 뇌조직의 지질산화에 대한 식물체 추출물의 효과)

  • Abilities of various edible plants and natural antioxidants to protect brain against oxidative damages were evaluated using brain homogenate of perfused Sprague-Dawley rat. Oxidative damage, expressed as lipid peroxidation (LPO), indicating total quantity of malondialdehyde and 4-hydroxyalkenal, increased from 4.1 to 6.9nmol/mg protein by treatment of $2.5{\mu}M$ ferrous sulfate and 7.5mM hydrogen peroxide as source of reactive oxygen species (ROS) on brain homogenate for 10min at $37^{\circ}C$ Mallow(88%) in leafy vegetables, small potato (93%) in root vegetables, green red pepper (76%) in fruit vegetables, and avocado (96%) in fruits showed highest LPO inhibition capacities. Ability of mushrooms decreased in order of nameko, shiitake, pine mushroom, oyster mushroom, and new type pine mushroom. Among natural antioxidants tested, (+)catechin (91%), (-)epigallocatechin gallate (85%), (-)epicatechin gallate (83%), and kaempferol(83%) showed high LPO inhibition capacities.

Effect of Temperature and Host Plant on Development and Reproduction of the Sweetpotato Whitefly, Bemisia tabaci(Homoptera:Aleyrodidae) (담배가루이의 발육과 생식에 미치는 온도와 기주의 영향)

  • Development and reproduction of the sweetpotato whitefly, Bemisia tabaci(B biotype) were investigated under different temperatures and host plants. Developmental periods from egg to pre-adult of whiteflies measured under four constant temperatures: they were 86.2 days at $15^{\circ}C$ and 17.0 days at $30^{\circ}C$. Lower threshold temperature and total effective temperature for the development of egg and nymph, and for the complete development (egg to emergence) were $10.1,\;11.6,\;11.1^{\circ}C$ and 110.3, 204.7, 317.3 degree days, respectively. The hatching and emergence rates were 87.0% at $25^{\circ}C$ and 76.7% at $20^{\circ}C$, which were higher than the results of other temperatures. The adult longevity was 23.6 and 14.0 days at $20^{\circ}C$ and $30^{\circ}C$, respectively. The highest average fecundity per female was 103.3 at $25^{\circ}C$. But there were no significant differences among the temperatures. The highest intrinsic rate of natural increase($r_{m}$) was 0.196 at $30^{\circ}C$ and the highest net reproduction rate ($R_{o}$) was 97.33 at $25^{\circ}C$. Developmental periods from egg to pre-adult of whiteflies were 21.2 on the tomato, 28.1 on red pepper, 22.2 on eggplant and 25.5 days on poinsetia. The hatching was highest (90.3%) on red paper and emergence rate was highest (89.6%) on eggplant. The longest longevity of adult female was 26.5 days on eggplant, and the average fecundity per female was greater on tomato and eggplant than on other host plants. The intrinsic rate of natural increase($r_{m}$) was the highest on tomato as 0.165 and the net reproduction rate ($R_{o}$) was the highest on eggplant as 106.1. As a result, the optimum range of temperature for the growth of B. tabaci was between $25^{\circ}C$ and $30^{\circ}C$, and the optimum host plant were tomato and eggplant among the plants tested.

  • A wide arry of naturally occurring substances particularly those present in dietary and medicinal plants, have been reported to possess substantial cancer chemopreventive properties. Certain phytochemicals retain strong antioxidative and anti-inflammatory properties which appear to contribute to their chemopreventive or chemoprotective activities. Inducible cyclooxygenase(COX-2) and nitric oxide synthase (iNOS) are important enzymes that mediate inflammatory processes. There is some evidence that expression of both COX-2 and iNOS is co-regulated by the eukaryotic transcription factor NF-$textsc{k}$B. Increased expression of COX-2 and/or iNOS has been associated with pathophysiology of certain types of human cancers as well as inflammatory diseases. Since inflammation is closely linked to tumor promotion, substances with potent anti-inflammatory activies are anticipated to exert chemopreventive effects on carcinogenesis, particularly in the promotion stage. An example is curcumin, a yellow pigment of turmeric (Curcuma longa L., Zingiberaceae), that strongly occurring diaryl heptanoids structurally related to curcumin have substantial anti-tumor promotional activities in two-stage mouse skin carcinogenesis. Thus, yakuchinone A [1-(4'-hydroxy-3'-methoxyphenyl)-7-phenyl-3heptanone] and yakuchinone B [1-(4'-hydroxy-3'methoxyphenyl)-7-phenylhept-1-en-3-one] present in Alpinia oxyphylla Miquel (Zingiberacease) attenuate phorbol ester-induced inflammation and papilloma formation in female ICR mice. These diarylheptanoids also suppressed phorbol ester-induced activation of epdermal ornithine decarboxylase and its mRNA expression when applied onto shaven backs of mice. Yakuchinone A and B as well as curcumin inhibited phorbol ester-induced expression of COX-2 and iNOS and their mRNA in mouse skin via inactivation of NF-$textsc{k}$B. Capsaicin, a major pungent ingredient of red pepper also attenuated phorbol ester-induced NF-$textsc{k}$B activation. Similar suppression of COX-2 and iNOS and down-regulation of NF-$textsc{k}$B activation for its DNA binding were observed with the ginsenosied Rg3 and the ethanol extract of Artemisia asiatica. We have also found that certain anti-inflammatory phytochemicals exert inhibitory effects on phorbol ester-induced COX-2 expression and NF-$textsc{k}$B activation in immortalized human breast epithelial (MCF-10A) cells in culture. One of the plausible mechanisms undelying inhibition by aforementioned phytochemicals of phorbol ester-induced NF-$textsc{k}$B activation involves interference with degragation of the inhibitory unit, I$textsc{k}$Ba, which blocks subsequent nuclear translocation of the functionally active p65 subunit of NF-$textsc{k}$B. the activation of epidermal NF-$textsc{k}$B by phorbol ester and subsequent induction of COX-2 hence appear to play an important role in intracellular signaling pathwasy leading to tumor promotion and targeted inhibition of NF-$textsc{k}$B may provide a new promising cancer chemopreventive strategy.

Environmental Impacts of the Waste Rump in the Dongjin Gold-Silver-Copper Mine (동진 금·은·동 광산 주변에 방치된 폐석의 환경적 영향)

  • Although the Dongjin Au-Ag-Cu mine had been abandoned since about forty years ago, the results of this study on the dispersion patterns and contamination level of heavy metals in the hydrologic system flowing via the waste rump show that the environmental impacts from the mine wastes are still significant. The stream water in the vicinity of the waste rump is severely acidified (pH 3.8 to 4.4) and highly enriched in various dissolved heavy metals. The heavy metal contents of the stream water and stream sediments are systematically attenuated with increasing distance from the mine area. However, it is worth to note that continuous attenuation of heavy metal contents in both media were reenriched in downstream area more than 800 m apart from the mine because it can be acted as a secondary source of heavy metal pollution. The heavy metals, especially Cd, Cu and Zn of polluted downstream sediments mainly occur in Fe-Mn oxides and organic materials, which indicates that these elements are the main pollutants from the waste rump of the Dongjin mine. The heavy metal contents of crops, such as sesame, perilla, red Pepper and brown rice, collected from the polluted farm land in the downstream area are lower than those of land plants from stream sides, but significantly higher in Cd, Cr, Cu and Zn than those from the unpolluted farm land. Especially, almost all of the crops in polluted farm land have been severly contaminated by Cd (>0.4 ppm). On the other hand, the heavy metal contents of the crops collected from refreshed farm land by means of a soil addition method shows significantly lowered level comparing with those of polluted area, which indicates that a soil addition method was effective for the refreshment of polluted farm land by toxic metallic pollutants. Wormwoods from this area showed very high contents in a11 the heavy metals even in unpolluted area (Cd > 1 ppm, Cr > 1 ppm, Cu > 11 ppm, Pb> 4 ppm, Zn > 55 ppm), indicating that a special caution must be payed when one takes ingest them.
