Effect of Dry Powders, Ethanol Extracts and Juices of Radish and Onion on Lipid Metabolism and Antioxidative Capacity in Rats (무, 양파의 시료제조 방법에 따른 흰쥐의 지방대사와 항산화능에 관한 연구)

  • This study was performed to investigate the effects of dry powders, ethanol extracts and juices of radish and onion on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in rats. Forty-nine male Sprague-Dawley rats weighing 157$\pm$6g were blocked into seven groups according to body weight and raised for four weeks with diets containing 5%(w/w) dry powders of two different vegetables consumed frequently by Korean-radish(Raphanus sativus L.) and onion(Allium cepa L.), ethanol extracts and juices from equal amount of each dry powder. All the powders, ethanol extracts and juices of radish and onion decreased total lipids, triglycerides and total cholesterol concentrations in plasma and liver. Above all, onion ethanol extract decreased them most remarkably. It was thought that organosulfur compounds and flavonoids extracted from onion by ethanol inhibited biosynthesis and absorption of lipid and promoted degradation of lipid. Radish powder also decreased them by increasing fecal excretions of total lipids, triglycerides and total cholesterol most effectively. Catalase and glutathine peroxidase(GSH-px) activities in red blood cell(RBC) were most remarkably increased by radish powder and onion powder respectively. Superoxide dismutase(SOD), catalase and GSH-px activities in liver were most remarkably increased by onion ethanol extract, radish powder and onion ethanol extract respectively. Xanthine oxidase(XOD) activities in liver were most effectively decreased by ethanol extracts of radish and onion. Thiobarbituric acid reactive substance (TBARS) levels in plasma and liver of experimental groups were significantly lower than those of controls. Above all, onion powder decreased them most effectively. It was thought that vitamin E and high flavonoids in onion powder inhibited lipid peroxidation, promoting liver and RBC SOD, catalase and GSH-px activities and inhibiting XOD activities effectively. Flavonoids in onion ethanol extract inhibited lipid peroxidation, promoting three antioxident enzyme activities and inhibiting XOD activities most remarkably. Also flavonoids and high vitamin C in radish powder inhibited lipid peroxidation, promoting liver and RBC catalase most remarkably and inhibiting XOD activities. In conclusion, radish and onion were effective in lowering lipid levels and inhibiting of lipid peroxidation in animal tissue. From these data, radish and onion can be recommended in the treatment and prevention of diseases such as cardiovascular disease and cancer and in delaying aging. As ethanol from onion were most effective in lowering lipid level and promoting three antioxident enzymes, and inhibited lipid peroxidation as did we should try to utilize onion skin which is discarded though reported to have abundant flavonoids.   • The growth extent of Leu. mesenteroides and Lac. plantarum in the medium which contain sterilized extract of each of garlic, red pepper powder, and onion was examined. Garlic and onion decreased the growth of Leu. mesenteroides and Lac. Plantarum, and garlic represented more negative effect on the growth of Lac. plantarum than that of onion. Red pepper powder had negative effect on the growth of Lac. plantarum, and positive effect on the growth of Leu. mesenteroides in accordance with incubation processing.

Effects of Onion Extract and Onion-Acanthopanax Senticosus Mixture Extracts on Obese Rats (양파와 가시오가피 혼합 추출액이 비만 흰쥐에 미치는 영향)

  • The aim of this study is to investigate the effects of onion and acanthopanax senticosus on body weight change, serum total cholesterol, HDL cholesterol, LDL cholesterol, triglyceride, free fatty acid, total lipid, phospholipid level, renal and liver function test, and CBC in obese rats and mice. Obese rats induced by high-fat fed are medicated for 7 weeks. Rats are divided into four groups depending on the medication; normal group (general-fat fed and no-medication), control group (high-fat fed and no medication), sample A group (high-fat fed and onion 100% extracted medication), sample B group (high-fat fed and onion 50% & acanthopanax senticosus 50% extracted medication), sample C group (high-fat fed and red onion 50% & acanthopanax senticosus 50% extracted medication). After medication, obesity related index, renal and liver function test, and CBC are analysed. There are significant statistical differences among control group and all experimental groups for the body weight change. There are significant statistical differences among control groups and all experimental groups for the total cholesterol, HDL cholesterol, LDL cholesterol, triglyceride level, free fatty acid, and phospholipid level. These results suggest that medications of onion and acanthopanax senticosus extracted products are effective for the treatment of obesity. Especially, onion 100% extracted product is more effective than the others.

Acid treatment effects on the contents of quercetin glycosides and aglycone in red onion powder (산처리에 의한 적양파 분말의 quercetin 배당체와 aglycone의 농도변화)

  • Flavonoids are bioactive plant metabolites that have a range of beneficial effects on human health. Quercetin 4'-glycoside (Q4'G), quercetin 3,4'-diglycoside (Q3,4'G), and quercetin aglycone (QA) are the main flavonoids found in onions. QA, in particular, is likely to have a greater biological effect than glycosides. To develop an onion extract with high quercetin content, the optimal extraction conditions for red onion powder containing the outer layer of the onion were determined. The effects of acid treatment on the concentration of quercetin glycosides and QA were evaluated. The flavonoids of red onion powder were optimally extracted under 60-70% ethanol at 70℃ for 2 h. The deglycosylation of Q3,4'G and an increase in Q4'G content occurred within 6 h of 0.2% acetic acid treatment. The QA content and deglycosylation of Q4'G eventually peaked at 24 h. In addition, QA content and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity were highly correlated, with a correlation coefficient of 0.90.

  • The Allium tuberosum is a perennial herb. From the ancient times, it has been used for food or medical purpose. Allium tuberosum is widely distributed in Korea, China, Japan and other countries. As herbal medicine, the Allium tuberosum has medical effects on lumbago, sthenia, diuresis, nocturia, hematemesis, anemia, haemorrhoid, hiccups, diarrhea, eczema, sunstroke, pollution and so on. Recently, the Allium tuberosum has been studied on its components, anti-microorganism action, anti-fungal action, anti-arteriosclerosis action, anti-cancer action. However, there has not been any study on hematological effect of allium tuberosum extract on the blood in large dogs whereas the effects of excess garlic and onion on the blood of cattle, horses, dogs, cats and sheep has been reported by many researchers. Thus, this study was performed to observe the hematological effect of excessive Allium tuberosum extract administrated in large dogs. 1. The Red Blood Cell(R3C) count was significantly decreased(p<0.05), Packed Cell Volume (PCV) count were significantly decreased(P<0.05), and Reticulocyte count was significantly increased(p<0.05) 2. The White Blood Cell(WBC) count was significantly increased(p<0.05).3. The Hemoglobin concentration was significantly decreased(p<0.05) and Mean Corpuscular Hemoglobin Concentration (MCHC) was increased. and Mean Corpuscular Volume(MCV) was significantly increased.(p<0.05). 4. The Reduced glutathione(GSH) and Met-Hemoglobin were significantly increased (p<0.05).

Effect of Onion Peel Extracts on Blood Lipid Profile and Blood Coagulation in High Fat Fed SD Rats (고지방 섭취 흰쥐에서 양파 껍질 추출물의 보충 섭취가 혈중 지질농도와 혈행 개선에 미치는 효과)

  • Numerous studies have suggested that dietary flavonoids contribute to prevent cardiovascular disease. Onion contains many functional phytochemicals such as quercetin. The aim of this study was to examine whether onion peel extracts supplementation affect blood lipid profiles and blood coagulation in animal model. Total 48 Sprague-Dawley male rats at 5 weeks old were divided into 6 groups with different diets(C: control, HF: high fat diet, HFOE 0.01%: high fat+onion peel extract 0.01% diet, HFOE 0.02%, HFOE 0.05%, HFOE 0.1%) for 8 weeks. Onion peel extract supplementation significantly decreased serum levels of LDL-cholesterol and increased HDL-cholesterol, while total cholesterol and triglyceride levels were not affected. Hematological parameters(hematocrit, white blood cell, red blood cell, and platelet count) and blood coagulation parameters(prothrombin time, activated partial thromboplastin time, thrombin time, and fibrinogen) were not significantly different among 6 groups. However, activated partial thromboplastin time of HFOE 0.05% group was significantly longer than that of HF group. These results indicate that onion peel extract supplementation displays hypocholestrolemic effects but does not seem to have anti-coagulation effects in high fat fed SD rats.

Antimicrobial effect of Kimchi ingredients of methanol extract on pathogenic microorganisms (김치 재료 methanol 추출물이 식품유해 미생물에 미치는 항균효과)

  • This study was carried out to determine the inhibitory effect of methanol extract from kimchi ingredients against Salmonella typhimurium, Listeria monocytogenes, Staphylococcus aureus, and Esherichia coli which are pathogenic microorganisms and Aspergillus sp. Penicillium sp. Antimicrobial activity of methanol extracts was tested against bacteria by paper disc method. Antifungal activity of methanol extracts was shown by hyphal growth inhibition ratio. The methanol extracts from all materials were effective against E. coli among them. And the antimicrobial activity of the methanol extracts from ginger and onion were lower than the others. The antifungal activity of the methanol extracts from radish, ginger, and garlic were effective against Aspergillus sp. and Penicillium sp. In the result of identifying antimicrobial effect rate, the methanol extracts from red pepper and radish had more than $40\%$ against S. typhimurium and more than $30\%$ against E. coli. Also the methanol extracts from onion had an high inhibitory effect rate of more than $50\%$ against S. typhimurium and that from garlic had more than $60\%$ against S. aureus. The minimum inhibitory concentration(MIC) of red pepper was examined 500 $\mu$g/mL against L. monocytogenes. This value was the lowest among the others.

Effects of Dietary Supplements of processed Onion on the Physiochemical Characteristics by Store Period in Duck Meat (양파함유사료를 급여한 오리 고기의 저장기간중 이화학적 특성 변화)

  • The objectives or this study were to improve duck meat qualify during storage period in onion-red ducks which contained antioxidants. Experimental animals (1 day old, Cherry Valley $F_{1}$, 360 chicks) were randomly assigned in eight treatment group. Treatments were control, 3%, 6% of chopped onion-fed, 5%, 10% of onion extract fed, 6% of onion meal, 3%, 6% of fermented onion-fed. pH was significantly high (P<0.05) in treatment group after 3 day storage in pectoralis muscle. Meat color was low in r* and a* value. TBA value was not different in day 1, was significantly low (P<0.05) 6% chopped and 6% fermented group after day 3 storage. Also, TBA value was significantly high (P<0.05) in control group and was significantly low in 3% chopped, 5%, 10% extract after day 6 storage. VBN value was lower in all treatment groups except 5% ferment group than control group. Meat order was high score in 3% and 6% chopped group and duck specific smell was reduced in all treatment groups. Meat appearance was higher score in treatment groups than in control group during 9 day storage. In result, onion feeding was reduce duck specific smell and improve meat quality during long period storage in ducks.

  • The growth extent of Leuconostoc mesenteroides and Lactobacillus plantarum in the medium which contain sterilized extract of each kimchi minor ingredient (green onion, garlic, ginger, raw red pepper, and red pepper powder) was examined. All minor ingredients decreased the growth of Lac. plantarum, and this effect of garlic is the most distinctive, ginger had the positive effect on the growth of Leu. mesenteroides, and garlic had the negative effect on the growth of Leu. mesenteroides. When the growth extent of two bacteria in the medium which contain sterilized successive extracts of each of garlic, ginger and red pepper powder was examined, the butanol fraction of garlic was reprsented the negative effect on the growth of Leu mesenteroides and Lac. plantarum, and the water fraction of ginger and red pepper powder were represented the positive effect on the growth of Leu. mesenteroides.

Antioxidant Activity According to Each Kind of Natural Plant Pigments (식물유래 천연색소의 항산화 활성)

  • The objective of this study was to determine the effect of antioxidant enzyme activity and radical scavenging activities of thirteen kinds of natural plant pigments. The analytic method of antioxidant activities were measured by estimating DPPH free radical scavenging and nitrite scavenging ability activity. The free radical scavenging activity by method using stable free radical DPPH was the highest in the red cabbaged pigment. Addition of ethanol extract 1mg/ml from onion peel pigment displayed remarkable effect on nitrite scavenging ability about 91.9%. Antioxidative enzyme activity was evaluated in terms of superoxide dismutase(SOD), catalase(CAT), and ascorbate peroxidase(APX) activity. The bitter melon pigment had the highest SOD activity of 87.3%. The activities of CAT and APX were higher in the mulberry leave pigment compared with other natural plant pigments. In contrast, CAT activity of plant pigment samples were unaffected. These results suggest that natural plant pigment had the potent biological activities such as antioxidant enzyme activities, and that their activities exhibited differently depending on each kind of pigments.
