Measurement of red blood cell aggregation by analysis of light transmission in a pressure-driven slit flow system

  • The aggregation characteristics of red blood cells (RBCs) were measured using a newly developed light-transmission slit rheometer. Conventional methods of RBC disaggregation such as the rotational Couette system were replaced with a pressure-driven slit flow system with a vibrational mechanism. Using a vibration generator, one can disaggregate the RBC aggregates stored in the slit. While shear stress decreases exponentially, instantaneous pressure and the transmitted light intensity were measured over time. Applying an abrupt shearing flow after disaggregation caused a rapid elongation of the RBCs followed by loss of elongation with the decreasing shear stress. While the shear stress is further decreasing, the RBCs start to re-aggregate and the corresponding transmitted intensity increases with time, from which the aggregation indices can be obtained using a curve-fitting program.

각종악성종양환자(各種惡性腫瘍患者)의 Ferrokinetics 및 동대사(銅代謝)에 관(關)한 연구(硏究) (Studies on Ferrokinetics and Copper Metabolism in Various Malignant Tumors)

  • Anemia is a usual finding in advanced malignant diseases. Various mechanisms were reported as to be involved in the development of anemia of this kind, and they may differ in individual cases. Tumor anemias may be due, for instance, to chronic blood loss, shortened life span of the red blood cells or a decreased hemopoiesis in the bone marrow. The serum iron and copper levels, total iron binding capacity, apparent half survival of $^{51}Cr$-labelled red blood cells were measured along with the ferrokinetic studies using $^{59}Fe$ in 64 patients with various malignant tumors. Following were the results: 1. The serum iron levels were decreased in all cases. There existed no correlation between the serum iron levels and the severity of the diseases. 2. The serum copper levels were increased, particularly in lung cancer, rectal cancer, hepatoma and various sarcomas. There was also no correlation between the serum copper levels and the severity of the diseases. 3. The serum iron levels appeared to be inversely proportional to the serum copper levels. 4. The total iron binding capacities were within normal limits in all cases. There were also no correlations between the total iron binding capacities, serum iron levels and the severity of the diseases. 5. The patients could be classified according to the ferrokinetic patterns, namely, that of iron deficiency anemia in 10 cases, that of refractory anemia in 6 cases, normal in 1 case and that of atypical abnormal in 9 cases. 6. Apparent half survival time of $^{51}Cr$-labelled red blood cells were definitely shortened in half of the cases.

혈액에 의한 초음파 산란의 이론적 고찰 (A Theoretical Study of Ultrasound Scattering by Blood)

  • In this study, the scattering of ultrasound by blood is theoretically studied. At first, the Chernow equation which represents acoustic propagation inside the region of inhomogenieties is modified to be fitted for blood. Then, scattering amplitude and scattering Cross-section at the far-field region are obtained from this equation. In this case, hemotocrit is assumed to be less than 26 per cent, and the red blood cell is modeled as independent scatterer. This study also develops a practical approximation for the backscattering of periodic sinewave bursts by a volume of randomly distributed scatterers, i.e., whole red blood cells. This expression for the received backscattering ultrasound pressure after the n-th burst of narrowband transducer is obtained from an ideal continuous wave transducer's response.

Solanum Nigrum Polysaccharide (SNL) Extract Effects in Transplanted Tumor-bearing Mice - Erythrocyte Membrane Fluidity and Blocking of Functions

  • Background: Solanum nigrum L. has been used in traditional Chinese medicine because of its diuretic and antipyretic effects. The present research concerned effects of crude polysaccharides isolated from Solanum nigrum L. on erythrocyte membranes of tumor-bearing $S_{180}$ and $H_{22}$ in mice. Materials and Methods: Fluorescence-labeled red blood cell membranes were used with DPH fluorescence spectrophotometry to examine erythrocyte membrane fluidity, and colorimetry to determine degree of erythrocyte surface membrane blocking. Extent of reaction by tumor-bearing mice with the enzyme erythrocyte membrane bubble shadow detection of red cell membrane variation in the degree of closure before and after administration. Results: Solanum nigrum polysaccharide could significantly improve the $S_{180}$ and $H_{22}$ tumor-bearing mice erythrocyte membrane fluidity, compared with the control group, the difference was significant (p<0.01), SNL can significantly improve the red blood cell membrane and then $S_{180}$ tumor-bearing mice sealing ability, compared with the negative control group, the difference was significant(p<0.05, p<0.01). $H_{22}$ tumor-bearing mice can increase red cell membrane and then sealing ability, the difference was significant (p<0.05). Solanum nigrum polysaccharide degree of fluidity and blocking two transplanted tumors in mice restored the ability to raise the red cell membrane has a significant effect. Conclusions: Solanum nigrum L.-type mice transplanted tumor can affect the red blood cell membrane fluidity and re-closed, through the red cell membrane of red blood cells to enhance the immune function of the possibility of erythrocyte immunity against tumor formation garland provide experimental basis.

충전용 농축적혈구의 세척 및 체외순환로의 전순환-초여과법이 개심수술에 의한 사이토카인 형성에 미치는 영향 (The Effects of Packed Red Blood Cell Washing and Circuit Precirculation-Ultrafiltration on the Production of Cytokines by Open Heart Surgery)

  • 배경: 적혈구세척법(체외순환로 충전시 사용되는 농축 적혈구를 헤파린 용액으로 세척하여 충전)은 충전용 농축적혈구액에 포함되어 있는 염증반응 전구 물질 및 각종 손상된 세포 및 미립자들을 제거할 수 있으며, 전순환-초여과법(체외순환전에 체외순환로를 자체 전순환 시킨후 초여과법을 적용)은 체외순환로에 의한 초기 염증반응을 둔화시키고 염증반응 매개체들을 제거할 수 있다 본 연구자는 상기 방법들이 수술시 형성되는 염증반응 매개체중 허혈후 재관류 손상에 관여하는 각종 사이토카인들의 형성을 억제할 수 있을 것이라 가정하여, 실제 임상 환자를 대상으로 상기 방법들을 적용하면서 각종 사이토카인의 체외순환 및 수술 시간에 따른 변화를 측정하여 비교 분석하고자 하였다. 대상 및 방법: 본 연구자는 심실중격결손증으로 개심술을 받는 48명의 환아를 대상으로 하여, 대조군(C군, n=12), 적혈구 세척군(W군, n=12), 전순환-초여과군(F군, n=12), 혼합군(WF군, n=12) 으로 나누어 적혈구 세척법과 전순환-초여과법을 시행한 후에 체외순환을 적용하면서 tomor necrosis factor-$\alpha$(TNF-$\alpha$)와 interleukin-6 (IL-6) 및 interleukin-8 (IL-8) 등의 체외순환 및 수술 시간경과에 따른 변화를 측정 비교 분석 하였다. 결과: TNF-$\alpha$, IL-6, IL-8 등의 변화는 시간에 따라 각군간 유의한 차이를 보였다(p=0.001, p=0.001, p=0.005). TNF-$\alpha$의 변화는 대조군(C군), 세척군(W군), 전순환-초여과군(F군) 사이에서 유의한 차이를 보이지 않았으나(p=0.264), 혼합군(WF군)에서는 다른 군들에 비해 낮게 측정되었다(p<0.05). IL-6의 변화는 세척군(W군), 전순환-초여과군(F군), 혼합군(WF군) 사이에서는 유의한 차이가 없었으나(p=0.053), 각군 모두 대조군(C군)에 비해 낮게 측정되었다(p<0.05). IL-8의 변화는 세척군(W군), 전순환-초여과군(F군), 혼합군(WF군) 사이에서 유의한 차이가 없었으며(p=0.067), 대조군(C군)과 전순환-초여과군(F군) 사이에서도 유의한 차이를 보이지 못하였으나(p=0.086), 세척군(W군)과 혼합군(WF군)에서는 대조군(C군)에 비해 낮게 측정되었다(p<0.05). 결론: 이상의 결과는 적혈구 세척법과 전순환-초여과법이 체외순환을 이용한 개심수술시 사이토카인 형성 억제에 효과가 있음을 구명하였다. 그러나, 상기 방법들을 임상에서 확대적용 하기 위해서는 실질적으로 임상 경과에서 얻을 수 있는 장점에 관한 연구가 추가적으로 시행되어야 하겠다.

AN ELECTRON MICROSCOPIC, RADIOAUTOGRAPHIC STUDY OF ERYTHROPOIESIS IN VITRO

  • Using ³H-proline as a radioactive tracer, the relationship between the ultrastructural differentiation and the site of protein synthesis has been investigated in developing red blood corpuscles. The general ultra-structure of erythropoietic cells in differentiation after 60 minutes of in vitro labeling has confirmed the results from previous investigations by Bessis, M., Thiery, J. and others. In dividing nuclei more than two-thirds of the labeling were present at the interface between heterochromatin and euchromatin. In less differentiated cells most of the grains in interphase cells was localized over the nucleus. As the cells continued to develope beyond a stage where cytoplasmic density was clearly increased over other cell lines in bone marrow, the majority of grains localized over the cytoplasmic area was decreased in more mature cells, as judged by the density of cytoplasm, and the structural changes in mitochondria, Golgi complex and polysomal configurations. These results show; 1) that the cytoplasm of erythroblast series does not change under in vitro conditions employed in the study; 2) that protein synthesis in the nucleus occurs largely at the interface between euchromatin and heterochromatin in active nuclei; and 3) that cytoplasmic synthesis of proteins continues to take place well into the normoblast stage solong as the physically visible polysomes are present in maturing red blood corpuscles.

Aflatoxin-mediated Sperm and Blood Cell Abnormalities in Mice Fed with Contaminated Corn

  • The effect of aflatoxin-contaminated corn on albino mice was investigated using the sperm morphology assay. Blood parameter levels including; total white blood cells (WBC), total red blood cells (RBC), packed cell volume (PCV), serum bilirubin (SB) and fasting blood sugar (FBS) were also determined in the tested mice. Test mice were exposed to aflatoxin-contaminated corn (contamination level of 100 ppb) for $1{\sim}4$ weeks while aflatoxin-free corn and cyclophosphamide were used as negative and positive controls, respectively. Sperm cells showed varieties of morphological abnormality when assessed after 5 weeks. The percentage frequencies of the negative and positive controls were 18.8% and 48.87%, respectively, while the percentage abnormalities for the 1, 2, 3 and 4 weeks exposures were 41.38%, 48.17%, 57.13% and 61.67%, respectively. PCV, WBC, total bilirubin and glucose level values of mice in all concentrations were higher and statistically significant as compared to the negative control values using Dunnett's test. Therefore, abnormal sperm cell induction is concentrationdependent such that continuous consumption of aflatoxin-contaminated corn is capable of negatively affecting spermatogenesis by inducing or increasing the frequency of morphologically abnormal sperm cells produced.

Increased of the Red Blood Cell in Peripheral Plasma of Transgenic Pigs Harboring hEPO Gene

  • The present study were performed to analysis the hematocrit and the red blood cells content into the blood plasma of the transgenic pigs harboring recombinent human erythropoietin gene (rhEPO). Mouse whey acidic protein (mWAP) linked to rhEPO gene was microinjected into pronuclei of porcine one-cell zygotes. After delivered of offspring, PCR analyses identified one mWAP-rhEPO transgenic founder offspring(F$_{0}$). The first generation of transgenic pig (F$_{0}$) harboring mWAP-hEPO appeared to be a male, and the second generation (F$_1$) pigs were made by natural mating of F$_{0}$ with domestic swine, and male and female transgenic pigs (F$_1$) were identified by PCR. The blood samples from transgenic and normal pigs were collected for 50 days during lactation and were counted the red blood cell (RBC) numbers and Hematocrit (HCT) content into the blood. The transgenic pigs expressing rhEPO in their blood gave rise to higher RBC numbers and HCT contents than control animals. rhEPO was secreted both in the blood and milk of genetically engineered pigs harboring rhEPO gene. Therefore, this study provides a model regarding the production of transgenic pig carrying hEPO transgene for biomedical research.earch.

Red Blood Cell Velocity Field in Rat Mesenteric Arterioles Using Micro PIV Technique

  • As endothelial cells are subject to flow shear stress, it is important to determine the detailed velocity distribution in microvessels in the study of mechanical interactions between blood and endothelium. This paper describes a velocity field of the arteriole in the rat mesentery using an intravital microscope and high-speed digital video system obtained by a highly accurate PIV technique. Red blood cells (RBCs) velocity distributions with spatial resolutions of $0.8{\times}0.8{\mu}m$ were obtained even near the wall in the center plane of the arteriole. By making ensemble-averaged time-series of velocity distributions, velocity profiles over different cross-sections were calculated for comparison. The shear rate at the vascular wall also evaluated on the basis of the ensemble-averaged profiles. It was shown that the velocity profiles were blunt in the center region of the vessel cross-section while they were steep in the near wall region. The wall shear rates were significantly small, compared with those estimated from the Poiseuille profiles.

  • The dependence of the rheological properties of blood on shape, aggregation, and deformability of red blood cells (RBCs) has been investigated using hybrid systems by coupling fluid with solid models. We present a simple approach for simulating blood as a multi-component fluid, in which RBCs are modeled as droplets of acquired biconcave shape. We used lattice Boltzmann method (LBM) due to its excellent numerical stability as a simulation tool. The model enables us to control the droplet static shape by imposing non-isotropic surface tension force on the interface between the two components. The use of the proposed non-isotropic surface tension method is justified by the Norris hypothesis. This hypothesis states that the shape of the RBC is due to a non-uniform interfacial surface tension force acting on the RBC periphery. This force is caused by the unbalanced distribution of the lipid molecules on the surface of the RBC. We also used the same concept to investigate the dynamic shape change of the RBC while flowing through the microvasculature, and to explore the physics of the Fahraeus, and the Fahraeus-Lindqvist effects.
