  • The performance of fluidized-bed reactor with Photomedia, immobilized TiO$_2$ onto the porous ceramic ball using a sol-gel method has been studied in this work. A simple model substrate, dilute Rhodamine B (RhB), was decolorized at room temperature. For the purpose of comparison, the slurry reactor and the Inner Wall Coated Reactor (IWCR) were used. The aim of this work was to develop the photocatalytic fluidized bed reactor (PFBR) through contrasting the photodegradability of various reactors such as the TiO$_2$slurry reactor, the inner-wall coated reactor (IWCR). In this study, the RhB was decolorized in three types of reactor. Even though the reaction rate constant of PFBR was lower than that of slurry reactor, PFBR had the advantages of preventing the wash-out of photocatalyst, so it can be operated continuously.

Metabolic Flux Analysis of Beijerinckia indica for PS-7 Production

  • In order to investigate central metabolic changes in Beijerinckia indica, cells were grown on different carbon sources and intracellular flux distributions were studied under varying concentrations of nitrogen. Metabolic fluxes were estimated by combining material balances with extracellular substrate uptake rate, biomass formation rate, and exopolysaccharide (EPS) accumulation rate. Thirty-one metabolic reactions and 30 intracellular metabolites were considered for the flux analysis. The results revealed that most of the carbon source was directed into the Entner-Doudoroff pathway, followed by the recycling of triose-3-phosphate back to Hexose­6-phosphate. The pentose phosphate pathway was operated at a minimal level to supply the precursors for biomass formation. The different metabolic behaviors under varying nitrogen concentrations were observed with flux analysis.

  • Small size privately owned wastewater treatment plants have been difficult to treat their wasted sludge and maintain steady effluent quality compared with publicly owned wastewater treatment plants. Therefore, this study has focused on treatment efficiency enhancement, specially nitrogen removal efficiency by recycling dewatering filtrate as an alkalinity additive from filter press using $CaCO_3$. As the result, it was found that the optimal mixing ratio between the excess sludge and $CaCO_3$ was 1:2. The major operation parameters such as specific substrate utilization rate, specific nitrification rate, and specific denitrification rate were also improved 64% ($0.048-0.079mg\;BOD_5/mg\;MLVSS{\cdot}day$), 35% ($0.020-0.027mg\;NH_3-N/mg\;MLVSS{\cdot}day$) and 68% ($0.051-0.086mg\;NO_3{^-}-N/mg\;MLVSS{\cdot}day$), respectively, after the adoption of new methods. Therefore, both the problem of sludge treatment at small scale plants and the need for efficiency improvement could be solved.

  • Hydrodynamic study of Activated Sludge Process (ASP) is important to optimize the reactor performance and detect anomalies in the system. Residence time distribution (RTD) study has been performed using LiCl as tracer on a pilot scale aeration tank (AT) and ASP, treating the pulp and paper mill effluent. The hydraulic performance and treatment efficiency of the AT and ASP at different operating parameters like residence time, recycle rate was investigated. Flow anomalies were identified and based on the experimental data empirical models was suggested to interpret the hydrodynamics of the reactors using compartment modelling technique. The analysis of the RTD curves and the compartment models indicated increase in back-mixing ratio as the mean hydraulic retention time (MHRT) of the tank was increased. Bypassing stream was observed at lower MHRT. The fraction of dead zone in the tank increased by approximate 20-25% with increase in recycle rate. The fraction of the stagnant zone was found well below 5% for all performed experiments, which was under experimental error. The substrate removal of 91% for Chemical oxygen demand and 96% for Biochemical oxygen demand were observed for the ASP working at a hydraulic mean residence time 39 h MRT with a 20% recycling of activated sludge.

Hydrolysis of Fish Protein Concentration in an Ultrafiltration Membrane Reactor (한외여과막 반응기를 이용한 FPC의 가수분해)

  • In order to improve functional properties, enzymatic hydrolysis of FPC (fish protein concentration) was achieved in ultrafiltration membrane reactor (MWCO 5,000). First, insoluble FPC was hydrolyzed by pepsin in batch reactor to decrease the fouling in ultrafiltration membrane reactor, and second hydrolysis was achieved by pronase E in ultrafiltration membrane reactor The optimum operating conditions in batch reactor using pepsin were at temperature 45$^{\circ}C$, pH 2.0 and the ratio of substrate to pepsin, 150 (w/w) After operating for 5hrs under optimum conditions, 89% of total amount of initial FPC was hydrolyzed. The rate constants, $K_{m}$ and V$_{max}$, were 1.25% and 0.89 mg/$m\ell$/min, respectively, and substrate inhibition was occured above 1.5%. The ultrafiltration membrane reactor was operated with recycling rate of 474 $m\ell$/min and transmembrane pressure of 15 psi. The permeate flux was increased by temperature, transmembrane pressure, but the permeate flux was fixed by pH. The optimum ratio of substrate to pronase E was 200(w/w) and the productivity of ultrafiltration membarane reactor was 702 mg/mg -enzyme, that of batch reactor was 51mg/mg-enzyme. Molecular weight distributions tot first and second hydrolysates were from 2,500 Da to 20,000 Da and from 700 Da to 10,000 Da, respectivelyly.

Effect of Reused Cocopeat Substrate on Growth and Yield of Summer-cultivated Paprika in EC-based Recycling Hydroponic Cultivation (EC기준 순환식 수경재배에서 코크피트배지 재사용이 여름작형 파프리카의 생육 및 수량에 미치는 영향)

  • This experiment was carried out to analyze the effects of substrate reuse on the growth and yield of summer paprika in cyclic hydroponics. The test group was divided into a new coco slab, one year reused coco slab two year reused coco slab based on 30% nutrient solution reuse, and was performed from April 18 to November 31, 2016 for 30 weeks. As a result, plant height of early growth was that the 2 year reused slab was longer than the new slab but the final growth period was 56.58 cm shorter. First group flower position was that reused slab was shorter by 2.92 cm than the new slab and the second group flower position was 0.31 cm long. The relative internode length of early growth, when the reused slab was used, the imbalance in the late growth stage was increased compared with the use of the new slab. The number of growth nodes in the 1 and 2 year reused slab was the smallest with 27.4 nodes. However, the number of harvested nodes did not show the difference in the test group, and the ratio of harvested that the 2 year reused slab was the highest at 26.8%. The ratio of unmarketable fruit tended to increase as the growth progressed. Fresh weight was 227.7g for new slab, 219.2g for 2 year reused slab and 21.2g for 1 year reused slab. The dry weight of the new slab increased with the reuse of the slab. It was 17.13g for new slab, 18.26g for 1 year reused, and 19.28g for 2 year reused. The average water content of the entire growth period was smaller as the slab was reused, and the 1 year reused slab was about 20g less than the 2 year reused slab. This trend was steadily occurring throughout the entire growing season. Especially, the reused slab for 1 year was less than 60g after 3 groups compared to other test groups. In conclusion, If will control seriously occurrence of unmarketable fruits by weakening after medium growth in summer-cultivated paprika in EC-based recycling hydroponic cultivation with reused cocopeat substrate, It is not what I have to worry that decrease of the yield and deterioration of the quality due to the change of physical and chemical properties of the slab and the pathogenic bacteria infection.

  • Food waste is the main source of decay, odors and leachate in collection, transportation and landfill due to the high volatile solids (VS) and moisture content. Acidogenic fermentation of food waste is affected by the fermentation constraints including the biodegradability of substrate and the degrading capability of microorganisms. The biodegradability of food waste is mainly related to cellulosic materials, which are hardly degraded and comprise about 50% of food waste. The efficient and economical method of improving hydrolysis is, therefore, to apply microorganisms with increased cellulose-degrading capability. In this experiment, rumen microorganisms were inoculated to improve the low efficiency of acidogenic fermentation, and then compared with that of mesophilic acidogens. The fermentation of food waste in a leaching bed reactor employing rumen microorganisms resulted in the enhanced acidification (71.2% at $3.0d^{-1}$), which was higher than that (59.8% at $4.5d^{-1}$) employing mesophilic acidogens. This indicated that Rumen microorganisms had an enhanced waste-degrading capability.

  • Chitinases (EC 3.2.1.14) hydrolyze the ${\beta}$-1,4-linkages in chitin, the second most abundant polymer of N-acetyl-${\beta}$-D-glucosamine which is a structural component of protective biological matrices such as fungal cell walls and insect exoskeletons. The glycosyl hydrolases 18 family including chitinases is an ancient gene family widely expressed in archea, prokaryotes and eukaryotes. Since earthworms live in the soil with a lot of microbial activities and fungi are supposed to be a major component of the diet of earthworm, it has been reported that there would be appropriate immune system to protect themselves from microorganisms attacks. In this study, the novel chitinase, EaChi, from the midgut of earthworm, Eisenia andrei, were identified and characterized. To obtain full-length cDNA sequence of chitinase, RT-PCR and RACE-PCR analyses were carried out by using the previously identified EST sequence amongst cDNA library established from the midgut of E. andrei. EaChi, a partial chitinase gene, was composed of 927 nucleotides encoding 309 amino acids. By the multiple sequence alignments of amino acids with other different species, it was revealed that EaCHI is a member of glycosyl hydrolases 18 family, which has two highly conserved domains, substrate binding and catalytic domain.

Effect of Methane Production from Pig Manure Slurry According to The Solids Concentration and The Crushing Solids of Pig Manure Slurry (돼지분뇨 슬러리중의 고형물 농도수준과 분쇄 처리가 메탄 생성에 미치는 효과)

  • Recently, the number of anaerobic digestion facility for livestock manure is on the rise in Korea. All of the livestock manure anaerobic digestion facilities in operation use pig manure slurry as a substrate for anaerobic digestion. Generally, pig manure slurry is composed of 97% water and 3% solids. The particulate matter, such as corn in the form of particles that is undigested by pig is contained in the pig manure slurry. Particulate matter is a factor reducing the effectiveness of biogas production in the anaerobic digestion process. In this study, mechanical grinding treatment was applied to analyze the effect of methane production from pig manure slurry by reducing the particle size of the slurry. On the other hand, the effect of the solid concentration levels on methane production and methane content of the biogas was analyzed. The fine particle concentration in the pig manure slurry was increased by the mechanical grinding treatment. And methane production and methane content of the biogas were higher in grinded pig manure slurry than untreated raw slurry.

The Effect of Fluid Flow on Power Density in a Horizontal-flow Microbial Fuel Cell (수평 흐름형 미생물 연료전지에서 유체의 흐름 형태에 따른 전력수율 평가)

  • This study evaluated the effect of fluid flow on the power density in a horizontal-flow microbial fuel cell (MFC). The maximum power densities in four types of flow induced by different channel types in the anode chamber were investigated. The fluid flow at each channel was analyzed using tracer tests. Results of polarization curves showed that the maximum power densities of case 1, 2, 3 and 4 were 95.7, 129.1, 190.9 and 114.2 mW/m2, respectively. Case 3 with a set of guide walls where flow had an S type-like shape showed the highest power density. Based on the Morrill Dispersion Index (MDI) value of case 4, microbial activity would be enhanced since the reactor allows even distribution of substrate but the overflow occurrence would not guarantee stable performance. Therefore, case 3 could be an effective reactor type for MFC because of high electricity generation and stable performance.
