β-arrestin Promotes c-Jun N-terminal Kinase Mediated Apoptosis via a GABABR·β-arrestin·JNK Signaling Module

  • Evidence is growing that the $GABA_B$ receptor, which belongs to the G protein-coupled receptor (GPCR) superfamily, is involved in tumorigenesis. Recent studies have shown that ${\beta}$-arrestin can serve as a scaffold to recruit signaling protein c-Jun N-terminal knase (JNK) to GPCR. Here we investigated whether ${\beta}$-arrestin recruits JNK to the $GABA_B$ receptor and facilitates its activation to affect the growth of cancer cells. Our results showed that ${\beta}$-arrestin expression is decreased in breast cancer cells in comparison with controls. ${\beta}$-arrestin could enhance interactions of the $GABA_BR{\cdot}{\beta}-arrestin{\cdot}JNK$ signaling module in MCF-7 and T-47D cells. Further studies revealed that increased expression of ${\beta}$-arrestin enhances the phosphorylation of JNK and induces cancer cells apoptosis. Collectively, these results indicate that ${\beta}$-arrestin promotes JNK mediated apoptosis via a $GABA_BR{\cdot}{\beta}-arrestin{\cdot}JNK$ signaling module.

A new function of glucocorticoid receptor: regulation of mRNA stability

  • It has long been thought that glucocorticoid receptor (GR) functions as a DNA-binding transcription factor in response to its ligand (a glucocorticoid) and thus regulates various cellular and physiological processes. It is also known that GR can bind not only to DNA but also to mRNA; this observation points to the possible role of GR in mRNA metabolism. Recent data revealed a molecular mechanism by which binding of GR to target mRNA elicits rapid mRNA degradation. GR binds to specific RNA sequences regardless of the presence of a ligand. In the presence of a ligand, however, the mRNA-associated GR can recruit PNRC2 and UPF1, both of which are specific factors involved in nonsense-mediated mRNA decay (NMD). PNRC2 then recruits the decapping complex, consequently promoting mRNA degradation. This mode of mRNA decay is termed "GR-mediated mRNA decay" (GMD). Further research demonstrated that GMD plays a critical role in chemotaxis of immune cells by targeting CCL2 mRNA. All these observations provide molecular insights into a previously unappreciated function of GR in posttranscriptional regulation of gene expression. [BMB Reports 2015; 48(7): 367-368]

Cloning and characterization of ADP-ribosylation factor 1b from the olive flounder Paralichthys olivaceus

  • Small GTPases are well known as one of the signal transduction factors of immune systems. The ADP-ribosylation factors (ARFs) can be classified into three groups based on the peptide sequence, protein molecular weight, gene structure, and phylogenetic analysis. ARF1 recruits coat proteins to the Golgi membranes when it is bound to GTP. The class I duplicated ARF gene was cloned and characterized from the olive flounder (Paralichthys olivaceus) for this study. PoARF1b contains the GTP-binding motif and the switch 1 and 2 regions. PoARF1b and PoARF1b mutants were transfected into a Hirame natural embryo cell to determine the distribution of its GDP/GTP-bound state; consequently, it was confirmed that PoARF1b associates with the Golgi body when it is in a GTP-binding form. The results of the qPCR-described PoARF1b were expressed for all of the P. olivaceus tissues. The authors plan to study the gene expression patterns of PoARF1b in terms of immunity challenges.

Requirement of Bni5 Phosphorylation for Bud Morphogenesis in Saccharomyces cerevisiae

  • In budding yeast, G2/M transition is tightly correlated with bud morphogenesis regulated by Swel and septin that plays as a scaffold to recruits protein components. BNI5 isolated as a suppressor for septin defect is implicated in septin organization and cytokinesis. The mechanism by which Bni5 regulates normal septin function is not completely understood. Here, we show that Bni5 phosphorylation is required for mitotic entry regulated by Swel pathway. Bni5 modification was evident from late mitosis to G1 phase, and CIP treatment in vitro of affinity-purified Bni5 removed the modification, indicative of phosphorylation on Bni5. The phosphorylation-deficient mutant of BNI5 (bni5-4A) was defective in both growth at semi-restrictive temperature and suppression of septin defect. Loss of Bni5 phosphorylation resulted in abnormal bud morphology and cell cycle delay at G2 phase, as evidenced by the formation of elongated cells with multinuclei. However, deletion of Swel completely eliminated the elongated-bud phenotypes of both bni5 deletion and bni5-4A mutants. These results suggest that the bud morphogenesis and mitotic entry are positively regulated by phosphorylation-dependent function of Bni5 which is under the control of Swel morphogenesis pathway.

Improvement of University Education to Supply Technical Manpower for Plant Engineering Industry in Mechanical Engineering Field (국내 플랜트 엔지니어링 산업의 기계공학 분야 기술인력 수급을 위한 대학교육 개선에 관한 연구)

  • In this study, improvements of university education are suggested so that university graduates can work in plant engineering industry without major retraining which is required in plant engineering companies. Before disposition of manpower to the plant engineering site, new recruits are retrained by the company for about 2~3 years, since university education is not sufficient and appropriate to handle plant engineering tasks. It is urgent to integrate into university education practical plant engineering that can be used effectively after graduation. In case of S2 University located in Seoul, it is enough to supplement interdisciplinary program to plant engineering subjects if proper texts are developed. To replace plant engineering education offered by the company with the university education, following measures should be taken. First, basic plant engineering should be taught for 15 hours. Second, education on design and drawings should be reinforced.

Qualitative Study on the Response of Consumers Experiencing Products' Being Sold Out on the Internet Shopping Malls (인터넷 쇼핑몰 품절상황에서 경험하는 소비자의 반응에 관한 질적 연구)

  • It is common for popular products to sell out, and be out of stock. The purpose of this study is to examine the responses of consumers who have experiences of shopping online, and finding their desired products unavailable from being sold out. In order to carry out the study, in-depth interviews were conducted with ten recruits who were recruited from the Internet community, Soul Dresser, which is part of the Korean web portal, Daum. First, the cognitive reaction of the interviewees showed both positive and negative responses when they found out that their desired product was sold out. However, in the case of emotional reaction, all of them showed negative reactions. Observations showed that consumers researched and found similar products including price when they realized that their desired products were not going to be re-stocked. And in cases of similar products not being available, the consumers looked to buy other types of products such as bags, shoes, and other accessories.

Full-length Fas-associated Death Domain Protein Interacts with Short Form of Cellular FLICE Inhibitory Protein

  • Fas-associated death domain protein (FADD) recruits and activates procaspase-8 through interactions between the death effector domains of these two proteins. Cellular FLICE-inhibitory protein (c-FLIP) was identified as a molecule with sequence homology to caspase-8. It has been postulated that c-FLIP prevents formation of the competent death-inducing signaling complex in a ligand-dependent manner, through its interaction with FADD and/or caspase-8. However, the interaction of FADD and $c-FLIP_s$ (short form) in apoptosis signaling has been controversially discussed. We show the purification and the characterization of human full-length FADD and $c-FLIP_s$ expressed in Escherichia coli. The purified FADD and $c-FLIP_s$ are shown as homogeneity, respectively, in SDS-PAGE analysis and light-scattering measurements. The folding properties of the $\alpha$-helical structure of FADD and the super-secondary structure of $c-FLIP_s$ proteins were characterized by circular dichroism spectroscopy. Furthermore, we report here a series of biochemical and biophysical data for FADD-$c-FLIP_s$ binding in vitro. The binding of both FADD and $c-FLIP_s$ proteins was detected by BIAcore biosensor, fluorescence measurement, and size-exclusion column (SEC).

The Comparison of the Relationship between the Gunfire Shot and Its Resulting Heavy Metal Pollution Rate (소화기 사격장의 사격에 따른 중금속 오염도 비교(I))

  • The following research was initiated in order to compare the relationship between the amount of gunfire shot and its resulting heavy metal pollution rate. The research was conducted at two firing ranges located inside a military unit stationed in the rear strategical area, where one full distance firing range is used by soldiers in active service, and the other is used by recruits and reserves. The heavy metal pollution rate was measured also on water sample collected from the target zone while raining. Based on values such as the real amount of gunshot fired, amount of heavy metal in the soil of the target zone, and the degree of heavy metal pollution for each firing range, the research showed that although pollution rate was higher when more gunshots were fired, there was no close correlation between the two. The water samples showed that this might result from the soils containing heavy metals eroded and transported by rain due to the target zone having no vegetation.

Growth and Maturation of Natural Population of Meristotheca papulosa in Jeju Island (제주도 유용 해조류 갈래곰보(Meristotheca papulosa) 야외개체군의 생장과 성숙)

  • The growth and maturation pattern of a commercial and endemic red algae Meristotheca papulosa were investigated in Jeju Island. Ecological researches were performed in a natural population of 10 m water depth in Sinheung from march 2014 to November 2015. Maximal blade length and weight of M. papulosa were 8.7 cm and 5.49 g in July 2014 and 10.7 cm and 10.17 g in September 2015, respectively. Growth in blade area of the species reached a maximum of $43.84cm^2$ and $67.41cm^2$ in July 2014 and 2015, respectively. Thalli in the field population were rarely observed from October to December but new recruits were found from January, following year. Carposporophytes were observed from June to September when bottom seawater temperatures were around $20^{\circ}C$. At the study site, seawater temperature was in the range of $14.9-25.7^{\circ}C$. Growth parameters of M. papulosa and relative abundance of carposporophytes showed significant correlation with environmental factors (temperature, salinity, DIN). In this result, we clarified the growth and maturation period of M. palpulosa, and provided effective data on the protection and management of natural populations and basic data for mass culture of the alga.

m6A in the Signal Transduction Network

  • In response to environmental changes, signaling pathways rewire gene expression programs through transcription factors. Epigenetic modification of the transcribed RNA can be another layer of gene expression regulation. N6-adenosine methylation (m6A) is one of the most common modifications on mRNA. It is a reversible chemical mark catalyzed by the enzymes that deposit and remove methyl groups. m6A recruits effector proteins that determine the fate of mRNAs through changes in splicing, cellular localization, stability, and translation efficiency. Emerging evidence shows that key signal transduction pathways including TGFβ (transforming growth factor-β), ERK (extracellular signal-regulated kinase), and mTORC1 (mechanistic target of rapamycin complex 1) regulate downstream gene expression through m6A processing. Conversely, m6A can modulate the activity of signal transduction networks via m6A modification of signaling pathway genes or by acting as a ligand for receptors. In this review, we discuss the current understanding of the crosstalk between m6A and signaling pathways and its implication for biological systems.
