Quantitative Source Estimation of PM-10 in Seoul Area (서울시 PM-10 오염원의 정량적 기여도 추정)

  • Recently in Korea, due to the significant drop of lead and bromine levels as a marker of autoemission source in the urban areas, the conventional application of receptor methods has many difficulties to properly apportion mass contribution of some sources. It is then needed to urgently develop alternative source profiles and identify new emission markers. Thus, the study has extensively examined the results obtained from using PAHs and elemental data for receptor modeling and has provided an opportunity to identify alternative source compositions and to determine a proper number of the ambient emission sources in Seoul area. The purpose of the study is to identify the sources of PM-10 and to estimate their mass contributions in Seoul area. Thus, a receptor model, target transformation factor analysis(TTFA) has been massively applied. The TTFA offers the possibility of determining the number of sources and their mass contributions. The input data used in this study are composed of two separate sets: fine (d$_{p}$ < 2.5.mu.m) and coarse (2.5.mu.m < d$_{p}$ < 10.mu.m) mode aerosol samples. Each sample was simultaneously collected by a PM-10 dichotomous sampler during the daytime(8 AM to 8 PM) and the nighttime(8 PM to 8 AM) from February to October 1993 on the Sungdong-Gu, Seoul. All the samples were analyzed to determine the levels of 10 inorganic elements by an XRF system as well as 14 PAHs by a HPLC. However, only 8 inorganic elements and 7 PAHs were used for the various statistical analysis.sis.

Vascular Endothelial Growth Factor Effect on Notch 1 Expression and Proliferation of Fibroblast (혈관내피성장인자의 섬유아세포 증식과 Notch 1 발현에 대한 영향)

  • Purpose: Vascular endothelial growth factor (VEGF) is known as a growth factor of endothelium and fibroblast. The purpose is to know the VEGF effects on fibroblast proliferation and fibroblast's notch receptor expression. Methods: CCD-986sk fibroblast was purchased from the Korean Cell Bank and was used in XTT assay for proliferation and wound healing assay for migration. Immunofluorescent (IF) staining and western blotting were used in testing notch expression of fibroblast. Semiquantitative RT-PCR was used in checking notch 1 mRNA production by fibroblast. Student-t test was used for analyzing results. Results: Cell proliferation assay using XTT showed significant higher proliferation in VEGF treated fibroblast, $2.324{\pm}0.0026$ vs. $2.463{\pm}0.017$ (p=0.002). Wound healing assay showed longer migration in VEGF treated fibroblast (p=0.062). The fluorescence was brighter in VEGF treated cells of notch 1 IF staining. Notch 1 expressions and mRNA productions increased more in VEGF treated cells. Conclusion: VEGF stimulates fibroblast to proliferate, migrate and to express Notch 1 simultaneously. Notch receptor could be related to VEGF mediated wound healing.

AN INTEGRATED APPROACH TO RISK-BASED POST-CLOSURE SAFETY EVALUATION OF COMPLEX RADIATION EXPOSURE SITUATIONS IN RADIOACTIVE WASTE DISPOSAL

  • Embodying the safety of radioactive waste disposal requires the relevant safety criteria and the corresponding stylized methods to demonstrate its compliance with the criteria. This paper proposes a conceptual model of risk-based safety evaluation for integrating complex potential radiation exposure situations in radioactive waste disposal. For demonstrating compliance with a risk constraint, the approach deals with important exposure scenarios from the viewpoint of the receptor to estimate the resulting risk. For respective exposure situations, it considers the occurrence probabilities of the relevant exposure scenarios as their probability of giving rise to doses to estimate the total risk to a representative person by aggregating the respective risks. In this model, an exposure scenario is simply constructed with three components:radionuclide release, radionuclide migration and environment contamination, and interaction between the contaminated media and the receptor. A set of exposure scenarios and the representative person are established from reasonable combinations of the components, based on a balance of their occurrence probabilities and the consequences. In addition, the probability of an exposure scenario is estimated on the assumption that the initiating external factors influence release mechanisms and transport pathways, and its effect on the interaction between the environment and the receptor may be covered in terms of the representative person. This integrated approach enables a systematic risk assessment for complex exposure situations of radioactive waste disposal and facilitates the evaluation of compliance with risk constraints.

  • A neutral ligand is synthesized and studied for the binding properties with anions by electrochemical methods. The binding of 1,8-bis[(N'-phenylureido)ethyloxy]anthraquinone (BPUA) with $H_2PO_4^-$ makes cathodic shift of its electrochemical potentials and red shift of absorption band. This novel neutral anion receptor BPUA binds anions through hydrogen bonding and show high selectivity with $H_2PO_4^-$ over $CH_3CO_2^-,CI^-,{\;}and{\;}HSO_4^-$. The selecivity of H_2PO_4^-$ over $CH_3CO_2^-,CI^-,{\;}and{\;}HSO_4^-$ may be attributed to the stronger hydrogen bonding with urea moiety and also with anthraquinone moiety of BPUA receptor, and also the higher complementarity of the cavity of BPUA for tetrahedral H_2PO_4^-$.

Transient receptor potential melastatin type 7 channels are involved in zinc-induced apoptosis in gastric cancer

  • Transient receptor potential melastatin 7 (TRPM7) channels are novel $Ca^{2+}$-permeable non-selective cation channels that are ubiquitously expressed. Activation of TRPM7 channels has been shown to be involved in the survival of gastric cancer cells. Here we show evidence suggesting that TRPM7 channels play an important role in $Zn^{2+}$- mediated cellular injury. Using a combination of electrophysiology, pharmacological analysis, small interfering RNA (siRNA) methods and cell death assays, we showed that activation of TRPM7 channels augmented $Zn^{2+}$-induced apoptosis of AGS cells, the most common human gastric adenocarcinoma cell line. The $Zn^{2+}$-mediated cytotoxicity was inhibited by the non-specific TRPM7 blockers $Gd^{3+}$ or 2 aminoethoxydiphenyl borate (2-APB) and TRPM7 specific siRNA. In addition, we showed that overexpression of TRPM7 channels in HEK293 cells increased $Zn^{2+}$- induced cell injury. Thus, TRPM7 channels may represent a novel target for physiological disorders where $Zn^{2+}$ toxicity plays an important role.

Analysis of the oxidized low density lipoprotein receptor 1 gene as a potential marker for carcass quality traits in Qinchuan cattle

  • Objective: The oxidized low density lipoprotein receptor 1 (OLR1) gene plays an important role in the degradation of oxidized low-density lipoprotein and adipocyte proliferation in mammals. For this reason, we aimed at investigating the association of OLR1 gene polymorphisms with carcass quality traits in Chinese Qinchuan cattle. Methods: The single nucleotide polymorphism (SNP) was identified in the 3' untranslated region of bovine OLR1 gene by DNA sequencing. In addition, the haplotype frequency and linkage disequilibrium estimates of three SNPs were evaluated in 520 individuals. Results: Results indicated that the studied three SNPs were within the range of moderate genetic diversity (0.25< polymorphism information content<0.5). Haplotype analysis of three SNPs showed that ten different haplotypes were identified, but only five haplotypes were listed as those with a frequency of <0.05 were excluded. The Hap3 ($-G_1T_2C_3-$) had the highest haplotype frequency (42.10%). Linkage disequilibrium analysis showed that the three SNPs had a low linkage ($r^2<0.001$). The T10588C and C10647T were significantly associated with backfat thickness and intramuscular fat content in Qinchuan cattle. Conclusion: Based on our results, we believe that the OLR1 gene could be a strong candidate gene for influencing carcass quality traits in Qinchuan cattle.

Clinical Implications According to Diagnostic Methods of Human Epidermal Growth Factor Receptor 2 Positivity in Breast Cancer: A Retrospective Study

  • Purpose: According to American Society of Clinical Oncology/College of American Pathologists guidelines, breast cancer is human epidermal growth factor receptor 2 (HER2) positive if there is HER2 protein overexpression at a 3+ level on immunohistochemistry (IHC 3+) or gene amplification (more than six copies per nucleus) on fluorescence in situ hybridization (FISH+). However, there have been few reports on whether outcomes differ based on diagnosis by these two techniques. In this study, we compared outcomes based on the two methods in patients with HER2-positive breast cancer. Methods: This study was a retrospective analysis of HER2-positive breast cancer in 18,304 patients, including 14,652 IHC 3+ patients and 3,652 FISH+ patients from the Korean Breast Cancer Society Registry. We compared breast cancer-specific survival and overall survival based on IHC 3+ and FISH+ status with or without trastuzumab. Results: Breast cancer-specific survival was significantly different between the IHC 3+ and FISH+ groups, with 5-year cumulative survival rates of 95.0% for IHC 3+ and 98.5% for FISH+ patients who did not receive trastuzumab (p=0.001) in Kaplan-Meier methods. However, there were no significant differences in breast cancer-specific survival and overall survival between IHC 3+ and FISH+ groups regardless of trastuzumab treatment in Cox proportional hazards models. Conclusion: The survival outcomes were not affected by the different two diagnostic methods of HER2-positive breast cancer. Further research to evaluate differences in prognosis and other characteristics according to the diagnostic methods of HER2 positivity is needed in the future.

Source Identification of PM-10 in Suwon Using the Method of Positive Matrix Factorization (PMF 방법론을 이용한 수원지역 PM-10의 오염원 확인)

  • The receptor modeling is one of the statistical methods to achieve reasonable air pollution strategies. The pur-pose of this study was to survey the concentration variability oi inorganic elements and ionic species in the PM-10 particles, to qualitatively characterize emission sources by an advanced algorithm called positive matrix factoriza-tion(PMF) as a receptor model that can strictly provide results in every loading matrix. A total of 254 samples was collected by a PM-10 high volume air sampler from Mar. 1997 to Feb. 1998 in Kyung Hee University at Suwon Campus. Fourteen chemical species(Zn, Cu, Fe, Pb, Al, Mn, $Na^{+}$, NH$_4$+, $K^{+}$, $Mg^{2+}$, $Ca^{2+}$, $SO_4^{2-}$, $NO_{3}^{-}$, and $Cl^{-}$) were determined by AAS and IC methods. The study results showed that the average monthly concentration of PM-10 particles were 86.3$\mu\textrm{g}$/$\textrm{m}^3$ in March (maximum) and 28.5$\mu\textrm{g}$/$\textrm{m}^3$ in August(minimum), respectively. The concentrations of Na+, NH$_4$+, $K^{+}$ and $Cl^{-}$ in winter, $Mg^{2+}$, $Ca^{2+}$ and $NO_{3}^{-}$, in spring, and $SO_4^{2-}$ in summer showed the largest peak concentration for the respective season. Through and app-lication of a PMF program of Pm-10 concentration data of Suwon, 9 sources were qualitatively identified , such as incineration source, oil burning source, soil related source, open burning source automobile source, coal burning sources, secondary sulfate related source, and secondary nitrate related source.

Overexpression of HER-2/neu in Patients with Prostatic Adenocarcinoma

  • Background: Prostatic adenocarcinoma is one of the main causes of cancer death, and its timely diagnosis and preventing its progression dramatically helps improve life indexes. Given the high disease recurrence rate, today, research is more inclined toward exploring causes of recurrence and development, and innovation of modern treatment methods. Several studies have explored over-expression of human epidermal growth factor receptor 2 (HER-2/neu) in prostatic cancer so far, with different results. Thus, it was decided to investigate HER-2/neu overexpression in patients with prostatic adenocarcinoma in Iran. Materials and Methods: A sample size of 40 patients with prostate cancer entered the study, using a cross-sectional, non-randomized sampling method. Parameters studied included patient age at surgery, Gleason score, serum prostatic specific antigen (PSA) before surgery, and positive sample rate after immunohistochemical staining to investigate HER-2/neu overexpression. Results: In terms of HER-2/neu receptor staining rate, of 40 slides, 16 (40%) scored 0, 13 (32.5%) 1+, 7 (17.5%) 2+, and 4 (10%) 3+. In total 27.5% of slides showed HER-2/neu overexpression. In terms of age, an inverse correlation was found (-0.181), but without significance (p=0.263). In terms of serum PSA, the correlation coefficient was 0.449 (p=0.004). With respect to Gleason score, the coefficient was 0.190 (p=0.240). Conclusions: In this study, HER-2/neu overexpression occurred in 27.5% of prostate cancer cases, which is a relatively high figure, compared to similar studies elsewhere. While, we failed to reveal any relationship between HER-2/neu expression status with progression and prognosis of disease, it was demonstrated that the serum PSA level was significantly higher in cases with increased receptor expression.

A Novel Simple Method to Purify Recombinant Soluble Human Complement Receptor Type 1 (sCR 1) from CHO Cell Culture

  • The human complement receptor type 1 (CR 1, C3 b/C4b receptor) is a polymorphic membrane glycoprotein expressed on human erythrocytes, peripheral leukocytes, plasma and renal glomerular podocytes, which consists of transmembrane and cytoplasmic domains with 30 repeating homologous protein domains known as short consensus repeats (SCR). CR1 has been used as an inhibitor for inflammatory and immune system for the past several years. Recently; it is reported that CRl was found to suppress the hyper-acute rejection in xeno-transplantation and can be used to cure autoimmune diseases. A soluble form of CRl, called sCRl, is a recombinant CRl by cleaving the transmembrane domain at C-terminus and has been expressed in Chinese Hamster Ovary (CHO) cells. Several purification methods for sCR1 from CHO cells have been reported, but most of them require complicated steps at high cost. Moreover, such methods are mostly performed under the pH condition apt to denaturing sCR1 and causes sCRl losing its activity. We here report a rapid and efficient method to purify sCR1 from CHO cell. The new method consists of a two-stage of cell culture by cultivating cells in serum medium followed by serum-free medium, and a two-stage of column purification by means of heparin and gel filtration column chromatography. By using this novel method, sCR1 can be purified in a simple and effective way with high yield and purity, furthermore, the purified sCR1 was confirmed to retain its activity to suppress the complement activation in vivo and ex vivo.
