A Study on the Recognition and Application of the contents of Housign Education among Male/Female Students in the Middle School (가정교과 주생활 단원에 대한 남녀 중학생의 인시과 활용도에 관한 연구)

  • The purpose of this study was to analyze the problems of housing education in Home Economics and then to present some suggestion for improving the housing education in the middle school and to analyze the problems of housing edcation that was investigated on the recognition and application in real life of housing area subject to male/female students in the middle school. The respondents were 600 boys and girls of middle school. Statistical methods adopted for data analysis were Frequency, Chi-square, t-test, F-test. The major findings were follows: 1. Many students(67.2%) responded that housing of home economics was wholly helpful in the actual life. 2. Many students recognized the necessity for the housing education. They particularly recognized the need about the knowlidge of sanitation and facilities of housing. 3. The degree to which housing education applied in the real life was lower than that to which it was needed. 4. There was a tendency that the more students realized the usefulness of housing education, the more positively they applied it in the real life. 5. The contents that students preferred to be additionally included in housing education were improvement of residential environment, solutions to housing environmental deterioration and pollution, future housing, and housing for the elderly.

Development of Life Test Equipment with Real Time Monitoring System for Butterfly Valves

  • Small valves including ball valves, gate valves and butterfly valves have been adopted in the fields of steam power generation, petrochemical industry, carriers, and oil tankers. Butterfly valves have normally been applied to fields where in narrow places installing the existing valves such as gate valves and ball valves have proven difficult due to the surrounding area and the heavier of these valves. Butterfly valves are used to control the mass flow of the piping system under low pressure by rotating the circular disk installed inside. The butterfly valve is benefitted by having simpler structure in which the flow is controlled by rotating the disc circular plate along the center axis, whereas the weight of the valve is light compared to the gate valve and ball valve above-mentioned, as there is no additional bracket supporting the valve body. The manufacturing company needs to acquire the performance and life test equipment, in the case of adopting the improving factors to detect leakage and damage on the seat of the valve disc. However, small companies, which are manufacturing the industrial valves, normally sell their products without the life test, which is the reliability test and environment test, because of financial and manpower problems. Furthermore, the failure mode analysis of the products failed in the field is likewise problematic as there is no system collecting the failure data on sites for analyzing the failures of valves. The analyzing and researching process is not arranged systematically because of the financial problem. Therefore this study firstly tried to obtain information about the failure data from the sites, analyzed the failure mode based on the field data collected from the customers, and then obtained field data using measuring equipment. Secondly, we designed and manufactured the performance and life test equipment which also have the real time monitoring system with the naked eye for the butterfly valves. The concept of this equipment can also be adopted by other valves, such as the ball valve, gate valve, and various others. It can be applied to variously sized valves, ranging from 25 mm to large sized valves exceeding 3000 mm. Finally, this study carries out the life test with square wave pressure, using performance and life test equipment. The performance found out that the failures from the real time monitoring system were good. The results of this study can be expanded to the other valves like ball valves, gate valves, and control valves to find out the failure mode using the real time monitoring system for durability and performance tests.

  • A phosphate-specific and fluorescent probe was prepared for label-free phosphatase assays based on fluorescence polarization. By using the probe, dephosphorylation reactions of DNA and protein substrates by calf intestinal alkaline phosphatase (CIP) could effectively be monitored in real-time. Since this assay method does not require additional materials such as labeled substrates and phosphospecific antibodies to obtain fluorescence polarization signals, it is simple, cost-effective, and expected to be useful not only for measuring activity of phosphatases but also for high-throughput screening of phosphatase inhibitors.

  • RNA interference has been used as a powerful tool in preventing virus proliferation in many species. In this study, we injected the dsRNA in vitro transcripts into Bombyx mori to investigate the resistance to B. mori nuclear polyhedrosis virus (BmNPV). Through vivisectional observation and real-time quantities PCR analysis, we found that these dsRNA can prevent the BmNPV to a certain extent, and delay the viruses' proliferation.

  • Life-Cycle Cost Analysis (LCCA) for highway projects is known as an effective analytical technique that uses economic principles to evaluate long-term alternative investment options, especially for comparing the values of alternative pavement design structures and construction strategies. In the Unites States, the 2012 Moving Ahead for Progress in the 21st Century Act (MAP-21) amended the United States Code to mandate that the United States Government Accountability Office (GOA) conducts a study of the best practices for calculating life-cycle costs and benefits for the federally funded highway projects in 2013. The RealCost 2.5CA program was developed and adapted as an official LCCA tool to comply with regulatory requirements for California state highway projects in 2013. Utilization of this California-customized LCCA software helps Caltrans to achieve substantial economic benefits (agency cost and road user cost savings) for highway projects. Proper implementation of LCCA for roadway construction and rehabilitation would deliver noticeable savings of agency's roadway maintenance cost especially in developing counties where financial difficulties exist.

  • Although Malaysia chooses Islam as national religion, the nation has really shown a variety of belief systems among which we would find Hinduism, Buddhism, and Animism. Most scholars having studied them regarded the aspect of Malay traditional belief systems as syncretism. This trait of the belief systems in Malaysia is revealed well in the sphere specifically as the rite of passage, to which I do rather prefer the rite of "life" from the sense of emphasizing the rituals varied by the cycle of life time. Under the perspective this paper examines the real phases of the rite of passage, or life of Malays. I will argue here the fact that in performing the rite of passage, Malays mobilize various arts on the basis of the syncretic faith accepting the supernatural, or keramat, with almighty power. Therefore, from the study of the rite of passage we can exactly scrutinize how Malays have altered and adjusted their belief systems in real world.

Development of EvaGreen Based Real-time PCR Assay for Detection and Quantification Toxic Dinoflagellate Pfiesteria Piscicida and Field Applications (유독 와편모조류 Pfiesteria Piscicida 탐지 및 정량 분석을 위한 EvaGreen 기반 Real-time PCR기법 개발과 현장 적용)

  • Pfiesteria piscicida is one of heterotrophic dinoflagellate having toxic metaboliges, and it is difficult to detect and quantify this dinoflagellate via light microscope due to small size and morphological similarity with Pfiesteria-like dinoflagellate (PLD) species. Alternatively, we developed quantitative real-time PCR assay based on EvaGreen and determined field accessibility throughout the investigation of distribution in the entire Korean coastal waters and population dynamics in Shihwa Lake. The P. piscicida-specific primers based on internal transcribed spacer 1 (ITS 1) were designed and the specificity of primers was confirmed by PCR with other genomic DNAs which have genetic similarity with target species. Through real-time PCR assay, a standard curve which had a significant linear correlation between log cell number and $C_T$ value ($r^2{\geq}0.999$) and one informative melting peak ($88^{\circ}C$) were obtained. These results implies that developed real-time PCR can accurately detect and quantify P. piscicida. Throughout the field applications of real-time PCR assay, P. piscicida was distributed in western (Mokpo and Kimje) and easthern (Gangneng) Korean coastal water even though light microscopy failed to identify P. piscicida. In the investigation of population dynamics in Shihwa Lake, the density of P. piscicida was peaked in June, July and August 2007 at St. 1 where salinity (${\leq}15psu$) was lower than the other 2 sites. In this study, we successed to develop EvaGreen bassed real-time PCR for detection and quantification of P. piscicida in fields, so this developed assay will be useful for various ecological studies in the future.

Rapid Detection Method of Avian Influenza Subtype H5N1 using Quick Real-Time PCR (Quick Real-time PCR을 이용한 Avian Influenza Virus Subtype H5N1의 신속검출법)

  • The most rapid Real-time PCR based detection method for Avian influenza A virus (AIV) subtype H5N1 was developed. The target DNA sequence in this study was deduced from H5N1 subtype-specific 387 bp partial gene of hemagglutinin, and was synthesized by using PCR-based gene synthesis on the ground of safety. Real-Time PCR was performed by $GenSpector^{TM}$ using microchip-based, total $1{\mu}l$ of reaction mixture with extremely short time in each steps in PCR. The detection including PCR-amplication and analysis of melting temperature was totally completed within 13 min. The H5N1-specific 189 bp PCR product was correctly amplified until 2.4 molecules of hemagglutinin gene as minimum of templates. This kind of PCR was designated as Quick Real-Time PCR in this study and it could be applied to detect not only AIV H5N1, but also other pathogens using PCR-based detection.
