TCE로 오염된 지하수 정화시 부식 광물에 대한 연구 (Investigation of Corrosion Minerals from the Remediation for TCE-Contaminate d Groundwater)

  • 본 연구는 0가 철 (ZVI)의 설치위치와 전극의 배열에 따른 다양한 조합의 전기적 투수성 반응벽을 대상으로 트리클로로에틸렌의 탈염소화 반응에 의한 정화시 ZVI의 부식에 기인하는 광물상침전물에 대해 알아보고, 이에 대한 조절 요소를 알아보고자 한다. 광물학적 연구 결과, 지하수 유입부의 ZVI 시료는 상대적으로 많은 레피도크로사이트, 훼리하이드라이트 혹은 철 수산화물과 (phospho)siderite가 산출되는 반면, 용출부의 ZVI 시료는 아카가나이트, 자철석/마그헤마이트, 그리고 중간 산물인 green rust (CR) I 과 CR II가 산출되었다. 이러한 광물 조합의 변화는 용존 산소 및 pH의 상승에 주로 기인한 것으로 나타났다. 또한 전기적 투수성 반응벽 내에 산출되는 광물상 침전물들의 조절 요소들은 (1) pH, (2) 용존산소, (3) 철의 부식시 중간 산물, (4) 음이온 종류 등으로 밝혀졌다.

Molecular cloning and characterization of peroxiredoxin from Toxoplasma gondii

  • A cDNA of 1.1 kb comprising the gene encoding the peroxiredoxin of Toxo-plasma gondii(TgPrx) has been cloned. The open reading frame of 591 Up was translated into a protein of 196 amino acids with a molecular mass of 25 kDa. Conserved 2 cysteine domains of Phe-Val-Cys-Pro and Glu-Val-Cys-Pro indicated TgPrx belonged to 2-Cys Prx families. TgPrx showed the highest homology with that of Arabidopsis thaliana by 53.9% followed by Entamoeba histolytica with 39.5% by the amino acid sequence alignment. Polyclonal antibody against recombinant TgPrx detected 25 kDa band in T. gondii without binding to host cell proteins TgPrx was located in the cytoplasm of T. gondii extracellularly or intracellularly by immunofluorescence assay. The expression of TgPrx was increased as early as 30 min after the treatment with artemisinin in the intracellular stage, while no changes in those of host Prx I and TgSOD. This result implies that TgPrx may function as an antioxidant protecting the cell from the attack of reactive oxygen intermediates. It is also suggested that TgPrx is a possible target of chemotherapy.

Nuclear Factor-κB Activation: A Question of Life or Death

  • Apoptosis is a mode of cell death that plays an important role in both pathological and physiological processes. Research during the last decade has delineated the entire machinery needed for cell death, and its constituents were found to pre-exist in cells. The apoptotic cascade is triggered when cells are exposed to an apoptotic stimulus. It has been known for several years that inhibitors of protein synthesis can potentiate apoptosis that is induced by cytokines and other inducers. Until 1996, it was not understood why protein synthesis inhibitors potentiate apoptosis. Then three reports appeared that suggested the role of the transcription factor NF-${\kappa}B$ activation in protecting the cells from TNF-induced apoptosis. Since then several proteins have been identified that are regulated by NF-${\kappa}B$ and are involved in cell survival, proliferation, and protection from apoptosis. It now seems that when a cell is attacked by an apoptotic stimulus, the cell responds first by activating anti-apoptotic mechanisms, which mayor may not be followed by apoptosis. Whether or not a cell undergoes proliferation, the survival, or apoptosis, appears to involve a balance between the two mechanisms. Inhibitors of protein synthesis seem to suppress the appearance of protein that are involved in anti-apoptosis. The present review discusses how NF-${\kappa}B$ controls apoptosis.

當歸飮子 水抽出液이 抗ALLERGY 反應과 MOUSE의 免疫細胞機能에 미치는 影響 (Effcts of Dangkiyeumja(當歸飮子) Water Extract of anti-allergic responses and on the Functions of Murine Immunocytes)

  • This study were done to know the effects of Dangkiyeumja on the in vivo and in vitro immune responses of mice. The recipes of Dangkiyeumja used in this study enhanced such, cellular functions of immunocytes as phagocytic capacity of macrophages, rossett-eforming abilities of splenocytes and metabolic activities of lymphocytes, However, the same recipes decreased the formation of such reactive oxygen intermediates(ROI) as superoxide and hydrogenperoxide from the macrophages. The effects of the same recipes on the in vim immune responses was suppressive on the cellular immune response(CIR)measured by delayed-type hypersensitivity against dinitrofluorobenzene and mildly enhancing for the humoral immune response measured by antibody production against sheep red blood cells. The results of this study could be summarized as follow: 1. Administration of Dangkiyeumja enhanced the phagocytic activity of the murine macrophage. 2. Administration of Dangkiyeumja decreased the formation of ROI in the murine macrophage 3. Administration of Dangkiyeumja increased the number of the splenic rotte forming cells in the mouse. 4. Administration of DangKiyeumja did not effect the antibody production against sheep red blood cells. 5. Administration of Dangkiyeumja depressed the delayed-type hypersenitivity against dinitrofluoro benzene in the mouse. The result of this study suggest that Dangkiyeumja could ameliorate the hypersensitivity reactions by reducing the formation of ROI and decreasing the CIR without affecting the other functions of immunocytes.

  • Glutathione(GSH),a tripeptide thiol, found in virtually all cells, functions in metabolism tranasport and cellular protection. It protects cells against the destructive effects of reactive oxygen intermediates and free radicals. Also ${\gamma}-Glutamyl$ transpeptidase(${\gamma}-GTT$), an enzyme of major importance in GSH metabolism, initiates GSH degradation. In order to explore the $GSH-{\gamma}-GTT$ system as periodontal disease activity indicator, we observed the ${\gamma}-GTT$ and arachidonic acid metabolits according to clinical groups(Control, Adult periodontitis, Rapidly progressive Periodontitis). From the experiments, the following results were obtained. 1. When compared with normal, ${\gamma}-GTT$ of A. P. and R. P. P. were increased, and only the change of ${\gamma}-GTT$ of R. P. P. was statistically significant(P<0. 05). 2. The amounts of arachidonic acid metabolites were not different with statistical significance among the clinical groups. 3. ${\gamma}-GTT$ may by useful adjuncts as new cytoprotective indicator and periodontal disease activity indicator in accordance with positive corelation pocket depth, attachment level and ${\gamma}-GTT$.

폐흡충 성충이 분비배설하는 anti-oxidant ensymes의 특성 관찰 및 peroxidase의 정제 (Characterization of a peroxidase in excretory-secretory product of adult Parasonimus westermani)

  • 폐흡충의 분비배설물은 항원성이 높아 폐흡충증의 진단용 항원으로 가치가 있다고 보고되었다. 이 까닭은. 분비배설물에는 숙주내에서 여러 생물학적 반응을 일으키는 물질뿐 아니라 충체의 일부구성 성분 및 여러 효소 등이 포함되어 있기 때문이라 가정할 수 있다. 이 연구는 폐흡충 성충의 분비배설물에서 숙주의 산소라디칼을 분해시키는 효소인 catalase, superoxide dismutasr (SOD), peroxidase 등이 존재하는지를 확인하고 그 활성도를 측정하였으며 그 중 peroxidase를 정제하여 생화학적 특성의 일부 및 항원성을 관찰하였다. 폐흡충 성충 50마리를 $37^{\circ}C$ 부란기에 12시간 배양한 뒤 배양액을 원심분리하고 이를 분비배설 조효소로 사용하였다. 분비배설물에서 catalase, SOD와 peroxidase의 활성도를 측정할 수 있었고 그 비활성도(specific activity)는 각각 11.1, 3.4 및 48.5 이었다. 분비배설물의 peroxidase 비활성도는 충체추출액의 비활성도보다 1.5배 높았다 이 효소를 Sephacryl 5-300 Superfine gel filtration. DEAE-Tnsacryl M anion exchange chromatography로 정제하였다. 정제한 peroxidase의 분자량은 HPLC에서는 19 kDa이었고 SDS-전기영동에서는 16 kDa이었다. 정제한 효소를 전기영동한 후 diaminobenzidine으로 specific staining한 결과 이 효소는 충체추출액의 효소와 같은 영동이동거리를 나타내었다 즉 이 효소는 충체로부터 분비되는 것임을 알 수 있었다. 한편 폐흡충 감염 환자의 혈청과 반응시킨 immunoblot에서 분비배설물의 구성 단백질중 84, 64, 42, 32, 30, 28, 26, 24, 22, 11 및 8 kDa가 항원성을 보인 반면 정제한 peroxidase는 미약한 반응을 보였다 이상의 결과로 폐흡충 peroxidase는 분비배설되어 SOD, catalase와 함께 산소 독성을 제거하는데 작용하고 있으나 패흡충 감염 환자에서 특이 항체 반응을 일으키는데 에는 미약한 작용을 한다는 것을 알 수 있었다.

자유라디칼이 백서의 뇌별아교세포에 미치는 독성작용 (Cytotoxic Effect of Free Radical on Rat Primary Astrocytes)

  • Astrocytes generate free radicals including nitric oxide (NO) and reactive oxygen intermediates(ROI) which in turn play roles in the pathogenesis of degenerative diseases and sclerotic changes of the brain. This study was designed to evaluate the mechanism that free radicals contribute to the cytotoxicty of rat neonatal primary astrocytes. Treatment with NO donors alone including soldium nitroprusside(SNP), S-nitrosoglucathinoe (GSNO), and S-nitroso-n-acetylpenicillamine (SNAP) showed a little effect on the death of rat neonatal primary astrocytes, whereas SNP markedly induced the death of RAW 264.7 cells. ROI inculding H2O2 and O2 donor also slightly induced the death of rat primary astrocytes. However, 3-morpholinosydnonimine(SIN-1), a donor of peroxynitrite (ONOO), which is a reactive compound of NO with superoxide, significantly decreased the viability of rat primary astrocytes in a dose-dependent manner. Cells were retarded in outgrowth of viability of cellular processes with cell shrinkage and detachment from culture dishes. Hoechst staining demonstrated that SIN-1-induced cell death might be due to an apoptosis which was characterized by nuclear condensation and fragmentation. SIN-1-induced apoptosis was prevented by the pretreatment with superoxide dismutase (SOD) and catalase in rat primary astorocytes. Furthermore, prevention of the generation of reduced glutathione (GSH) by DL-buthionine-[S, R]-sulfoximine (BSO) aggravated the cytotoxic effects of SNP, benzene triol, and SIN-1 in rat primary astrocytes. Taken together, it is suggested that peroxynitrite may be a major effector of apoptosis and cellular antioxidant system is important for cell survival in rat prima교 astrocytes.

Molecular and Genomic Approaches on Nickel Toxicity and Carcinogenicity

  • Nickel is the one of potent environmental, the occupational pollutants and the classified human carcinogens. It is a serious hazard to human health, when the metal exposure. To prevent human diseases from the heavy metals, it is seemingly important that understanding of how nickel exerts their toxicity and carcinogenic effect at a molecular and a genomic level. The process of nickel absorption has been demonstrated as phagocytosis, iron channel and diffusion. Uptaked nickel has been suggested to induce carcinogenesis via two pathways, a direct DNA damaging pathway and an indirect DNA damaging pathway. The former was originated from the ability of metal to generate Reactive Oxygen Species (ROS) and the reactive intermediates to interact with DNA directly. Ni-generated ROS or Nickel itself, interacts with DNAs and histones to cause DNA damage and chromosomal abnormality. The latter was originated from an indirect DNA damage via inhibition of DNA repair, or condensation and methylation of DNA. Cells have ability to protect from the genotoxic stresses by changing gene expression. Microarray analysis of the cells treated with nickel or nickel compounds, show the specific altered gene expression profile. For example, HIF-I (Hypoxia-Inducible Factor I) and p53 were well known as transcription factors, which are upregulated in response to stress and activated by both soluble and insoluble nickel compounds. The induction of these important transcription factors exert potent selective pressure and leading to cell transformation. Genes of metallothionein and family of heat shock proteins which have been known to play role in protection and damage control, were also induced by nickel treatment. These gene expressions may give us a clue to understand of the carcinogenesis mechanism of nickel. Further discussions on molecular and genomic, are need in order to understand the specific mechanism of nickel toxicity and carcinogenicity.

Phaleria macrocarpa Suppresses Oxidative Stress in Alloxan-induced Diabetic Rats by Enhancing Hepatic Antioxidant Enzyme Activity

  • Oxidative stress is caused by an imbalance between the production of reactive oxygen and an ability of a biological system, to readily detoxify the reactive intermediates or easily repair the resulting damage. It has been suggested that developmental alloxan-induced liver damage is mediated through increases in oxidative stress. The anti-diabetic effect and antioxidant activity of Phaleria macrocarpa (PM) fractions were investigated in alloxan-induced diabetic rats. After two weeks administration of PM, the liver antioxidant enzyme and hyperglycemic state were evaluated. The results showed that oral administration of PM treatments reduced blood glucose levels in diabetic rats by oral administration (P < 0.05). Serum glutamic-oxaloacetic transaminase (sGOT) and serum glutamic-pyruvate-transaminase (sGPT) were also diminished by PM supplementation. The superoxide dismutase (SOD), catalase (CAT) and glutathione-peroxidase (GPx) activities, and glutathione (GSH) level in the alloxan-induced diabetic rats were significantly decreased (P < 0.05) compared to those in the normal rats but were restored by PM treatments. PM fractions also repressed the level of malondialdehyde (MDA) in the liver. Glutathione reductase (GR), glutathione-S-transferase (GST) and $\gamma$-glutamylcysteine synthase (GCS) were also reduced in alloxan-induced diabetic rats. PM fractions could restore the GR and GST activities, but the GCS activity was not affected in rat livers. From the results of the present study, the diabetic effect of the butanol fraction of PM against alloxan-induced diabetic rats was concluded to be mediated either by preventing the decline of hepatic antioxidant status or due to its indirect radical scavenging capacity.

Rice 7-Hydroxymethyl Chlorophyll a Reductase Is Involved in the Promotion of Chlorophyll Degradation and Modulates Cell Death Signaling

  • The loss of green coloration via chlorophyll (Chl) degradation typically occurs during leaf senescence. To date, many Chl catabolic enzymes have been identified and shown to interact with light harvesting complex II to form a Chl degradation complex in senescing chloroplasts; this complex might metabolically channel phototoxic Chl catabolic intermediates to prevent oxidative damage to cells. The Chl catabolic enzyme 7-hydroxymethyl Chl a reductase (HCAR) converts 7-hydroxymethyl Chl a (7-HMC a) to Chl a. The rice (Oryza sativa) genome contains a single HCAR homolog (OsHCAR), but its exact role remains unknown. Here, we show that an oshcar knockout mutant exhibits persistent green leaves during both dark-induced and natural senescence, and accumulates 7-HMC a and pheophorbide a (Pheo a) in green leaf blades. Interestingly, both rice and Arabidopsis hcar mutants exhibit severe cell death at the vegetative stage; this cell death largely occurs in a light intensity-dependent manner. In addition, 7-HMC a treatment led to the generation of singlet oxygen ($^1O_2$) in Arabidopsis and rice protoplasts in the light. Under herbicide-induced oxidative stress conditions, leaf necrosis was more severe in hcar plants than in wild type, and HCAR-overexpressing plants were more tolerant to reactive oxygen species than wild type. Therefore, in addition to functioning in the conversion of 7-HMC a to Chl a in senescent leaves, HCAR may play a critical role in protecting plants from high light-induced damage by preventing the accumulation of 7-HMC a and Pheo a in developing and mature leaves at the vegetative stage.
