사물탕(四物湯) 가(加) 단참(丹參)의 상처 치료에 대한 효과 (The Effects of Samul-tang-ga-dansam for Wound Healing)

  • Objectives The purpose of this study was to evaluate the antioxidant, anti-inflammatory and wound healing effects of Samul-tang-ga-dansam water extract (SD) in wound-induced mice. Methods The mice were divided into five groups (n=7): the normal group, the control group, the positive control group, the low-dose SD group and the high-dose SD group. The normal group had no wounds and the other groups were wounded on the back with a leather punch. Distilled water was administered to the control group, 200 mg/kg of vitamin E was administered to the positive control group. In the low-dose SD group and the high-dose SD group, 1.23 g/kg and 2.47 g/kg of SD were administered, respectively. Antioxidant and anti-inflammatory protein levels were evaluated using western blot analysis. Skin tissue was analyzed by H&E, Masson's trichrome staining method. Results Oral administration of the SD significantly reduced the visible skin damage and decreased the reactive oxygen species and ONOO- activity of the serum. It significantly increased heme oxygenase-1, superoxide dismutase, catalase, GPx-1/2, Nrf2 and Keap-1 which are antioxidant-related factors in skin tissue and reduced NF-κB p65, inducible nitric oxide synthase, cyclooxygenase-2, tumor necrosis factor α, interleukin (IL)-1β, IL-6 which are inflammation-related factors. Also, SD significantly decreased NOX2, p22phox and p47phox and increased α-smooth muscle actin and COL1A1 protein expression in fibroblasts involved in connective tissue repair. According to histological examination, the thickened epithelial layer was thinned and collagen fibers were increased to accelerate wound healing. Conclusions It is suggested that Samul-tang-ga-dansam has antioxidant and anti-inflammatory effects and promotes wound tissue repair.

사료내 생균제, 일라이트, 활성탄 및 목초액의 첨가가 닭 다리육의 품질 및 저장성에 미치는 영향 (Effect of the Feeding Probiotics, Illite, Activated Carbon, and Hardwood Vinegar on the Meat Quality and Shelf-Life in Chicken Thigh)

  • 본 시험은 육계에게 생균제, 일라이트, 활성탄 및 목초액의 사료적 가치를 평가하기 위하여 시판 생균제 0.2%(T1)와, 일라이트(T2), 활성탄(T3) 및 목초액(T4)을 각각 1.0%를 Arbor Acre Broiler 숫병아리 200수를 35일간 급여하여 도계한 계육의 다리부위 근육을 냉장온도($4{\pm}1^{\circ}C$)에서 10일간 저장하면서 pH, VBN, TBARS, 전단력 및 육색을 분석하였다. pH는 대조구에 비해 T3와 T4에서 높았고, 특히 T4에서 유의적으로 높았으며 저장기간이 경과하면서 모든 처리구에서 증가하였다(p<0.05). VBN은 저장 7일까지 대조구와 T2에 비해 T1, T3및 T4에서 낮았으나(p<0.05), 저장 10일에는 처리구간 유의성이 없었다. TBARS는 대조구, T1 및 T2보다 T3와 T4에서 낮았고, 저장기간이 지남에 따라 모든 처리구에서 급속하게 증가하였다(p<0.05). 전단력은 처리구간에 유의적인 차이가 없었지만, 저장기간이 경과함에 따라 감소하는 경향이었다. 육색중 명도(L)는 대조구보다 처리구에서 높은 경향이었고, 특히 T4에서 전 저장기간 동안 가장 높았다(p<0.05). 황색도는(b) 대조구보다 T3와 T4에서 높았다. 이상의 결과를 종합적으로 고찰해 볼 때 육계사료에 활성탄과 목초액 1%급여는 산화안정성 및 저장성을 향상시키는 효과가 있는 것으로 기대된다.

부추가 Streptozotocin 유발 당뇨쥐의 지질과산화와 항산화방어체계에 미치는 영향 (Effect of buchu (Allium tuberosum) on lipid peroxidation and antioxidative defense system in streptozotocin-induced diabetic rats)

  • STZ(50mg/kg BW)을 대퇴부 근육에 주사하여 당뇨를 유발시킨 당뇨 흰쥐(Sprague Dawley, 수컷)에서 10% 부추 첨가식이가 혈액과 적혈구, 간조직의 지질과산화 정도와 항산화 효소계 활성 및 GSH 수준에 미치는 영향을 조사하였다. 실험군은 정상군, 당뇨대조군, 당뇨부추군의 3군 (n=10)으로 나누었으며 4주간 사육한 후 실험에 이용하였다. STZ로 유발된 당뇨대조군의 적혈구, 간과 LDL의 TBARS와 conjugated dienes 함량은 대조군에 비해 증가하였으나, 당뇨부추군에서는 적혈구의 conjugated dienes 수준이 유의적으로 감소하였고(p<0.05), 간과 LDL의 TBARS 수준은 당뇨대조군에 비해 다소 감소하여 정상대조군과 차이를 보이지 않았다. 간에서의 항산화 효소계 중 SOD 활성은 대조군에 비해 당뇨대조군에서 증가하였고, catalase 활성은 당뇨대조군에 비해 부추를 섭취한 당뇨군에서 유의적으로 증가하였다. GSH-px와 GSH-red 활성은 대조군에 비해 당뇨대조군에서 유의적으로 감소하였으나(p<0.05), 당뇨부추군에서는 다소 증가하였다. 간의 GSH 함량은 대조군과 당뇨대조군에 비해 당뇨부추군에서 유의적으로 증가하였으며(p<0.01), 혈장의 GSH 함량은 대조군에 비해 당뇨군에서 유의적으로 감소하였다(p<0.05). GOT와 GPT 활성은 정상대조군에 비해 당뇨대조군에서 급격히 증가하였으나 당뇨부추군에서는 정상군 수준으로 감소하였다. 이상의 결과로 미루어 볼 때, 부추는 간의 항산화효소계를 활성화시키고 간조직의 GSH수준을 높게 유지하여 고혈당과 STZ로부터 유발된 산화적 스트레스(지질과산화)를 해소함으로써 당뇨로 인한 합병증 예방 및 치료를 위한 식품자원으로 활용이 가능한 것으로 사료된다.도 및 동맥경화지수를 낮추는 효과가 있고, HDL- 콜레스테롤 농도를 증가시키는 효과가 있는 것으로 나타났다.각된다. 7) 발병부위는 대부분 막성유리연의 중간부 및 전방부 1/3에서 발생하였고 수술 결과는 저류낭종 82례 중 25례(30.5%)에서만 낭종의 파열 없이 완전제거가 가능하였으며 유표피낭종 30례 중에서는 21례(70.0%)에서 완전제거가 가능했던 것으로 나타나 저류낭종이 유표피낭종에 비해 낭종을 파열시키지 않고 완전히 제거하는 것이 더욱 어려움을 알 수 있었다. 따라서 저류낭종의 경우 낭종의 절제가 어려운 경우 수술방법으로 조대술(marsupialization)을 시행하는 것도 의미가 있을 것으로 사료된다. 8) 수술 전 후의 음성만족도를 비교한 조사 결과 조사가 가능했던 82례 중 49례(52.0%)에서 만족할 만한 결과를 보였다고 답하여 성대 폴립수술 후의 만족도와 유사한 결과를 보였다.14%, V. mimicus 10%, V. parahaemolyticus 4%, E. coli O157:H7 48%에서 증식되지 않았지만 S. typhimurium은 50%에서도 집락이 검출되었다. 젖산의 경우 V. vulnificus 2%, V. cholerae non-O1 3%, V. mimicus 4%, V. parahaemolyticus 3%, S. typhimurium 14%, E. coli O157:H7 17%에서 증식되지 않았다. 식초와 젖산은 낮은 농도에서 생선회 식중독 유발에 주 원인균이 되는 Vibrio 속의 생육을 억제하였고, S. typhimurium과 E. coli O157:H7의 생육은 비교적 약하게 나타났다.은 고농도로 갈수록 심한 영향을 미쳤으며 저 농도 에서도 폭로시간이 길어짐에 따라 나타나는 SO$_2$의영향이

Effects of a Dietary Chitosan-Alginate-Fe(II) Complex on Meat Quality of Pig Longissimus Muscle during Ageing

  • The current study was conducted to investigate the effects of dietary chitosan-alginate-Fe(II) complex (CAFC) supplementation on carcass and meat qualities of pig m. longissimus during chiller ageing. One hundred and twenty-two LYD (Landrace${\times}$Yorkshire${\times}$Duroc) pigs were sampled from an industrial population. Seventy-four pigs (32 gilts and 42 barrows) were administered 3 ml of dietary supplementation of CAFC per day from 25 to 70 days of age, while the remaining 48 pigs (20 gilts and 28 barrows) were fed the same commercial feeding regime without the supplementation. For assessing the dietary effects on pH, objective meat color, cooking loss, water-holding capacity (WHC), thiobarbituric acid reactive substances (TBARS), volatile basic nitrogen (VBN) and fatty acid composition during ageing, 20 barrows (10 of each treatment) were randomly sampled, and aged for 3, 7, 12, 16, 20 and 25 days in a $1^{\circ}C$ chiller. The results showed that CAFC-fed pigs required approximately 10 fewer feeding days than the control group. Furthermore, the treatment resulted in greatly higher carcass grade whereby the grade A was increased by approximately 35% and 7% for gilts and barrows, respectively. The treatment had no significant effect (p>0.05) on pH, meat color and WHC during ageing. On the other hand, the CAFC-fed pigs showed significantly (p<0.05) lower TBARS values from 20 days of storage. In addition, the sum of unsaturated fatty acids for the treated group was significantly (p<0.05) higher than that for the control group after the storage time. This implied that CAFC supplementation could reduce the formation of free radicals in fatty acids (i.e., lipid oxidation). The treatment also significantly (p<0.05) retarded VBN formation during ageing, indicating a significant reduction in protein degradation. However, as there was no difference in pH between the two groups, the result raised a possibility that antibacterial activity of the CAFC alone could cause reduction in the formation of TBARS and VBN. In this regard, although the treatment effectively slowed down the formation of TBARS and TBA during chiller ageing, it was not resolved whether that was associated with the direct effect of the antioxidant function of chitosan and/or alginate, or a consequence of their antibacterial functions.

키토산의 급여량과 급여 방법에 따른 육계의 성장 및 닭 다리육의 품질에 미치는 영향 (Effects of Dietary Chitosan Supplementation with Chitosan Feeding Levels and Feeding Forms on Growth Performance and Carcass Characteristics of Thigh Muscular in Broiler)

  • 본 연구는 키토산을 육계 사료에 첨가하지 않은 무첨가구를 대조구로 하고, 사료에 1% 첨가구를 T1, 2% 첨가구를 T2, 음수에 1% 첨가구를 T3, 2% 첨가구를 T4로 첨가 급여하면서 육계의 생산성을 조사하였고, 키토산을 급여한 계육의 일반 성분, 혈청콜레스테롤, pH, TBARS, WHC, 전단력, 육색 및 지방산 조성의 변화를 검토하고자 육계 200수(Arbor Acre Broiler, male)를 공시하여 사양한 후 다리 부위 근육을 이용하여 분석하였다. 육계의 증체량, 사료 요구율 등의 생산성은 차이가 없었다. 계육의 일반 성분 중에 조지방 함량은 대조구와 T3에서 유의적으로 높은 함량을 보였고 (P<0.05), 수분, 조단백질 및 조회분은 키토산 급여에 의한 영향은 없었다. pH는 대조구보다 키토산 급여구에서 높게 나타났다(P<0.05). TBARS는 키토산 급여구에서 유의적으로 낮은 결과를 보였고, 음수에 첨가하는 방법보다는 사료에 첨가하는 방법이 효과적이었다. 키토산 급여량이 증가할수록 낮은 TBARS값을 나타내어(P<0.05), 키토산은 계육의 저장성의 향상 가능성이 보였다. 혈청콜레스테롤은 키토산을 급여함으로써 총콜레스테롤, LDL-C은 감소하고, HDL-C은 증가하고, 첨가량이 많을수록 그리고 음수보다는 사료에 첨가하는 방법이 효과적이었다. 육색은 키토산 급여구에서 $L^*$값과 $b^*$값은 높았고, 지방산은 palmitic acid, oleic acid 등은 증가하고, linoleic acid 및 arachidonic acid는 감소하였다(P<0.05). 결론적으로 키토산을 사료에 첨가 급여함으로 총 콜레스테롤, LDL-C은 감소하고, HDL-C은 증가하며, TBARS값을 낮추는 효과가 있다. 이와 같은 결과는 음수보다는 사료에 첨가하는 방법이 유리하며 첨가량이 많을수록 좋았다.

Effects of Gas Composition in the Modified Atmosphere Packaging on the Shelf-life of Longissimus dorsi of Korean Native Black Pigs-Duroc Crossbred during Refrigerated Storage

  • This study was conducted to observe the effects of gas composition in modified atmosphere packaging (MAP) on the shelf-life of Longissimus dorsi of Korean Native Black Pigs-Duroc Crossbred ($KNP{\times}D$) during refrigerated storage. Muscle sample was obtained from the left side of carcass of seven months old of $KNP{\times}D$ barrow. The sample was sliced into 1 cm in thickness, placed on trays (two slices/tray) and filled with different gas composition, i.e. 0:20:80/$O_2:CO_2:N_2$ (MAP1), 30:20:50/$O_2:CO_2:N_2$ (MAP2) and 70:20:10/$O_2:CO_2:N_2$ (MAP3). Other slices of sample were vacuum packed (VP) as a control. All packs were stored at $5{\pm}1^{\circ}C$. At 12 d of storage, pH value of MAP2 and MAP3 were higher (p<0.05) than that of MAP1 and pH value of MAP1 was higher (p<0.05) than that of VP. At 6 d of storage, redness ($a^*$) value of MAP2 and MAP3 were higher (p<0.05) than that of VP and MAP1 and, at 9 and 12 d of storage, redness value of MAP3 was higher (p<0.05) than that of VP, MAP1, and MAP2. At 3, 6, 9, and 12 d of storage, the 2- thiobarbituric acid reactive substances (TBARS) value of MAP3 was higher than that of MAP2 and TBARS value of MAP2 was higher than that of VP and MAP1. At 3, 6, 9, and 12 d of storage, volatile basic nitrogen values of MAP2 and MAP3 were higher (p<0.05) than those of VP and MAP1. At 3 d of storage, total aerobic plate counts of MAP2 and MAP3 were higher (p<0.05) than those of VP and MAP1 and, at 6 d of storage, total aerobic plate counts of MAP3 was higher (p<0.05) than that of MAP1 and MAP2. However, there was no significant different total aerobic plate count among MAP1, MAP2, and MAP3 at 9 and 12 d of storage. There was no significant different total anaerobic plate count among MAP1, MAP2, and MAP3 during storage. It is concluded that the MAP containing 30:20:50/$O_2:CO_2:N_2$ gas composition (MAP2) might be ideal for better meat quality for $KNP{\times}D$ meat.

Antioxidant and hepatoprotective effects of Korean ginseng extract GS-KG9 in a D-galactosamine-induced liver damage animal model

  • BACKGROUND/OBJECTIVES: This study was designed to investigate the improvement effect of white ginseng extract (GS-KG9) on D-galactosamine (Ga1N)-induced oxidative stress and liver injury. SUBJECTS/METHODS: Sixty Sprague-Dawley rats were divided into 6 groups. Rats were orally administrated with GS-KG9 (300, 500, or 700 mg/kg) or silymarin (25 mg/kg) for 2 weeks. The rats of the GS-KG9- and silymarin-treated groups and a control group were then intraperitoneally injected Ga1N at a concentration of 650 mg/kg for 4 days. To investigate the protective effect of GS-KG9 against GalN-induced liver injury, blood liver function indicators, anti-oxidative stress indicators, and histopathological features were analyzed. RESULTS: Serum biochemical analysis indicated that GS-KG9 ameliorated the elevation of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) in GalN-treated rats. The hepatoprotective effects of GS-KG9 involved enhancing components of the hepatic antioxidant defense system, including glutathione, glutathione peroxidase (GPx), superoxide dismutase (SOD), and catalase (CAT). In addition, GS-KG9 treatment inhibited reactive oxygen species (ROS) production induced by GalN treatment in hepatocytes and significantly increased the expression levels of nuclear factor erythroid-2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) proteins, which are antioxidant proteins. In particular, by histological analyses bases on hematoxylin and eosin, Masson's trichrome, α-smooth muscle actin, and transforming growth factor-β1 staining, we determined that the administration of 500 mg/kg GS-KG9 inhibited hepatic inflammation and fibrosis due to the excessive accumulation of collagen. CONCLUSIONS: These findings demonstrate that GS-KG9 improves GalN-induced liver inflammation, necrosis, and fibrosis by attenuating oxidative stress. Therefore, GS-KG9 may be considered a useful candidate in the development of a natural preventive agent against liver injury.

홍삼, 천마, 적하수오 병용투여에 의한 고지혈증 랫드에서의 콜레스테롤 및 발기부전 개선효과 (Beneficial effect of Combination with Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori on Cholesterol and Erectile Dysfunction in Hyperlipidemia rats)

  • Objectives : This study was designed to investigate effects of the combination with Korean Red Ginseng (Panax ginseng C.A. Meyer), Gastrodia Rhizoma (Gastrodia elata Blume) and Polygoni Multiflori Radix (Polygonum multiflorum Thunberg) on metabolic disorders including cholesterol and erectile dysfunction in hyperlipidemia rats.Methods : Animals were divided into six groups; Control with normal diet, high fat/cholesterol-diet (HFCD), fluvastatin, Korean Red Ginseng treated (KRG), and the combination treated (Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori Radix; 1:1:1 for KGP1 and 2:1:1 for KGP2). The experimental groups initially received HFCD for 10 weeks and then treated orally with fluvastatin, KRG, KGP1 and KGP2 during the final 6 weeks. Erectile function was determined by the measurements of intracavernosal pressure (ICP) and maximal arterial pressure (MAP) after electrical stimulation of the cavernosal nerve.Results : KGP2 decreased the level of total cholesterol and LDL cholesterol in the sera of HFCD rats without no changes of body weights. KRG, KGP1 and KGP2 decreased the level of C-reactive protein (CRP) levels except of fluvastatin, synthetic HMG-CoA reductase inhibitor. KRG, KGP1 and KGP2 significantly increased the ICP, ICP/MAP ratio, area under the curve (AUC) compared with those of normal rat. Morphometric analyses showed that KRG, KGP1 and KGP2 increased the volume of smooth muscle and the regular arrangement of collagen fibers in corpus cavernosum of HFCD rats. The penile expression of eNOS was increased by KRG, KGP1 and KGP2.Conclusions : Based on these results, we suggest that the combination with Korean Red Ginseng, Gastrodia Rhizoma and Polygoni Multiflori may improve hyperlipidemia through regulating the lipid profiles and erectile dysfunction in rats.

대황과 실리마린의 병용투여의 간섬유화 보호 효과 (Liver Protective Effect of the Co-treatment of Rhei Radix et Rhizoma and Silymarin on TAA-induced Liver Injury)

  • Objective: Liver fibrosis is a highly conserved wound-healing response and the final common pathway of chronic inflammatory injury. This study aimed to evaluate the potential anti-fibrotic effect of the combination of Rhei Radix et Rhizoma water extract (RW) and silymarin in a thioacetamide (TAA)-induced liver fibrosis model. Methods: The liver fibrosis mouse model was established through the intraperitoneal injection of TAA (1 week 100 mg/kg, 2-3 weeks 200 mg/kg, 4-8 weeks 400 mg/kg) three times per week for eight weeks. Animal experiments were conducted in five groups; Normal, Control (TAA-induced liver fibrosis mice), Sily (silymarin 50 mg/kg), RSL (RW 50 mg/kg+silymarin 50 mg/kg), and RSH (RW 100 mg/kg+silymarin 50 mg/kg). Biochemical analyses were measured in serum, including aspartate aminotransferase (AST), alanine aminotransferase (ALT), malondialdehyde (MDA), and ammonia levels. Liver inflammatory cytokines and fibrous biomarkers were measured by Western blot analysis, and liver histopathology was evaluated through tissue staining. Results: A significant decrease in the liver function markers AST and ALT and a reduction in ammonia and total bilirubin were observed in the group treated with RSL and RSH. Measurement of reactive oxygen species and MDA revealed a significant decrease in the RSL and RSH administration group compared to the TAA induction group. The expression of extracellular matrix-related proteins, such as transforming growth factor β1, α-smooth muscle actin, and collagen type I alpha 1, was likewise significantly decreased. All drug-administered groups had increased matrix metalloproteinase-9 but a decreasing tissue inhibitor of matrix metalloproteinase-1. RSL and RSH exerted a significant upregulation of NADPH oxidase 2, p22phox, and p47phox, which are oxidative stress-related factors. Furthermore, pro-inflammatory proteins such as cyclooxygenase 2 and interleukin-1β were markedly suppressed through the inhibition of nuclear factor kappa B activation. Conclusions: The administration of RW and silymarin suppressed the NADPH oxidase factor protein level and showed a tendency to reduce inflammation-related enzymes. These results suggest that the combined administration of RW and silymarin improves acute liver injury induced by TAA.

Anti-fatigue effect of tormentic acid through alleviating oxidative stress and energy metabolism-modulating property in C2C12 cells and animal models

  • BACKGROUND/OBJECTIVES: Oxidative stress is caused by reactive oxygen species and free radicals that accelerate inflammatory responses and exacerbate fatigue. Tormentic acid (TA) has antioxidant and anti-inflammatory properties. Thus, the aim of present study is to determine the fatigue-regulatory effects of TA in H2O2-stimulated myoblast cell line, C2C12 cells and treadmill stress test (TST) and forced swimming test (FST) animal models. MATERIALS/METHODS: In the in vitro study, C2C12 cells were pretreated with TA before stimulation with H2O2. Then, malondialdehyde (MDA), lactate dehydrogenase (LDH), creatine kinase (CK) activity, tumor necrosis factor (TNF)-α, interleukin (IL)-6, superoxide dismutase (SOD), catalase (CAT), glycogen, and cell viability were analyzed. In the in vivo study, the ICR male mice were administered TA or distilled water orally daily for 28 days. FST and TST were then performed on the last day. In addition, biochemical analysis of the serum, muscle, and liver was performed. RESULTS: TA dose-dependently alleviated the levels of MDA, LDH, CK activity, TNF-α, and IL-6 in H2O2-stimulated C2C12 cells without affecting the cytotoxicity. TA increased the SOD and CAT activities and the glycogen levels in H2O2-stimulated C2C12 cells. In TST and FST animal models, TA decreased the FST immobility time significantly while increasing the TST exhaustion time without weight fluctuations. The in vivo studies showed that the levels of SOD, CAT, citrate synthase, glycogen, and free fatty acid were increased by TA administration, whereas TA significantly reduced the levels of glucose, MDA, LDH, lactate, CK, inflammatory cytokines, alanine transaminase, aspartate transaminase, blood urea nitrogen, and cortisol compared to the control group. CONCLUSIONS: TA improves fatigue by modulating oxidative stress and energy metabolism in C2C12 cells and animal models. Therefore, we suggest that TA can be a powerful substance in healthy functional foods and therapeutics to improve fatigue.
