CDC6 mRNA Expression Is Associated with the Aggressiveness of Prostate Cancer

  • Background: Cell division cycle 6 (CDC6) is an essential regulator of DNA replication and plays important roles in the activation and maintenance of the checkpoint mechanisms in the cell cycle. CDC6 has been associated with oncogenic activities in human cancers; however, the clinical significance of CDC6 in prostate cancer (PCa) remains unclear. Therefore, we investigated whether the CDC6 mRNA expression level is a diagnostic and prognostic marker in PCa. Methods: The study subjects included 121 PCa patients and 66 age-matched benign prostatic hyperplasia (BPH) patients. CDC6 expression was evaluated using real-time polymerase chain reaction and immunohistochemical (IH) staining, and then compared according to the clinicopathological characteristics of PCa. Results: CDC6 mRNA expression was significantly higher in PCa tissues than in BPH control tissues (P = 0.005). In addition, CDC6 expression was significantly higher in patients with elevated prostate-specific antigen (PSA) levels (> 20 ng/mL), a high Gleason score, and advanced stage than in those with low PSA levels, a low Gleason score, and earlier stage, respectively. Multivariate logistic regression analysis showed that high expression of CDC6 was significantly associated with advanced stage (${\geq}T3b$) (odds ratio [OR], 3.005; confidence interval [CI], 1.212-7.450; P = 0.018) and metastasis (OR, 4.192; CI, 1.079-16.286; P = 0.038). Intense IH staining for CDC6 was significantly associated with a high Gleason score and advanced tumor stage including lymph node metastasis stage (linear-by-linear association, P = 0.044 and P = 0.003, respectively). Conclusion: CDC6 expression is associated with aggressive clinicopathological characteristics in PCa. CDC6 may be a potential diagnostic and prognostic marker in PCa patients.

Differential Hrd1 Expression and B-Cell Accumulation in Eosinophilic and Non-eosinophilic Chronic Rhinosinusitis With Nasal Polyps

  • Purpose: Hrd1 has recently emerged as a critical regulator of B-cells in autoimmune diseases. However, its role in the pathogenesis of chronic rhinosinusitis with nasal polyps (CRSwNP) remains largely unexplored. This study aimed to examine Hrd1 expression and B-cell accumulation and their possible roles in CRSwNP. Methods: Quantitative real-time polymerase chain reaction, immunohistochemistry, enzyme-linked immunosorbent assay and Western blotting were used to assess gene and protein expression in nasal tissue extracts. Cells isolated from nasal tissues and peripheral blood mononuclear cells were characterized by flow cytometry. Local antibody production was measured in tissue extracts with a Bio-Plex assay. Additionally, changes in Hrd1 expression in response to specific inflammatory stimuli were measured in cultured dispersed polyp cells. Results: Nasal polyps (NPs) from patients with eosinophilic CRSwNP (ECRS) had increased levels of Hrd1, B-cells and plasma cells compared with NPs from patients with non-eosinophilic CRSwNP (non-ECRS) or other control subjects (P < 0.05). The average Hrd1 levels in B-cells in NPs from ECRS patients were significantly higher than those from non-ECRS patients and control subjects (P < 0.05). NPs also contained significantly increased levels of several antibody isotypes compared with normal controls (P < 0.05). Interestingly, Hrd1 expression in cultured polyp cells from ECRS patients, but not non-ECRS patients, was significantly increased by interleukin-$1{\beta}$, lipopolysaccharide and Poly(I:C) stimulation, and inhibited by dexamethasone treatment (P < 0.05). Conclusions: Differential Hrd1 expression and B-cell accumulation between the ECRS and non-ECRS subsets suggests that they can exhibit distinct pathogenic mechanisms and play important roles in NP.

Ubiquitin D Promotes Progression of Oral Squamous Cell Carcinoma via NF-Kappa B Signaling

  • Ubiquitin D (UBD) is highly upregulated in many cancers, and plays a pivotal role in the pathophysiological processes of cancers. However, its roles and underlying mechanisms in oral squamous cell carcinoma (OSCC) are still unclear. In the present study, we investigated the role of UBD in patients with OSCC. Quantitative real-time polymerase chain reaction and Western blot were used to measure the expression of UBD in OSCC tissues. Immunohistochemistry assay was used to detect the differential expressions of UBD in 244 OSCC patients and 32 cases of normal oral mucosae. In addition, CCK-8, colony formation, wound healing and Transwell assays were performed to evaluate the effect of UBD on the cell proliferation, migration, and invasion in OSCC. Furthermore, a xenograft tumor model was established to verify the role of UBD on tumor formation in vivo. We found that UBD was upregulated in human OSCC tissues and cell lines and was associated with clinical and pathological features of patients. Moreover, the overexpression of UBD promoted the proliferation, migration and invasion of OSCC cells; however, the knockdown of UBD exerted the opposite effects. In this study, our results also suggested that UBD promoted OSCC progression through NF-κB signaling. Our findings indicated that UBD played a critical role in OSCC and may serve as a prognostic biomarker and potential therapeutic target for OSCC treatment.

miR-195/miR-497 Regulate CD274 Expression of Immune Regulatory Ligands in Triple-Negative Breast Cancer

  • Purpose: Immune suppression is common in patients with advanced breast cancer but the mechanisms underlying this phenomenon have not been sufficiently studied. In this study, we aimed to identify B7 family members that were able to predict the immune status of patients, and which may serve as potential targets for the treatment of breast cancer. We also aimed to identify microRNAs that may regulate the expression of B7 family members. Methods: The Cancer Genome Atlas data from 1,092 patients with breast cancer, including gene expression, microRNA expression and survival data, were used for statistical and survival analyses. Polymerase chain reaction and Western blot were used to measure messenger RNA and protein expression, respectively. Luciferase assay was used to investigate direct microRNA target. Results: Bioinformatic analysis predicted that microRNA (miR)-93, miR-195, miR-497, and miR-340 are potential regulators of the immune evasion of breast cancer cells, and that they exert this function by targeting CD274, PDCD1LG2, and NCR3LG1. We chose CD274 for further investigations. We found that miR-195, miR-497, and CD274 expression levels were inversely correlated in MDA-MB-231 cells, and miR-195 and miR-497 expressions mimic inhibited CD274 expression in vitro. Mechanistic investigations demonstrated that miR-195 and miR-497 directly target CD274 3' untranslated region. Conclusion: Our data indicated that the level of B7 family members can predict the prognosis of breast cancer patients, and miR-195/miR-497 regulate CD274 expression in triple negative breast cancer. This regulation may further influence tumor progression and the immune tolerance mechanism in breast cancer and may be able to predict the effect of immunotherapy on patients.

Radio-sensitivity of Human Colorectal Cancer Cell is Regulated by Potassium Cyanate (Potassium cyanate에 의해 조절되는 사람 대장암 세포의 방사선 감수성 변화)

  • Potassium cyanate (KCN) is an inorganic reagent and can induce the post-translation carbamylation of proteins. The carbamylated reaction in the body is involved in cell death in various diseases. According the results in our previous study, KCN enhances the radiosensitivity of human colorectal cancer cell line, HCT 116 cells. However, it was not enough to confirm the mechanism that KCN works in these cells. To determinated the mechanisms of KCN in the cells with increased radiosensitivity, HCT 116 cells were treated KCN with low-dose gamma-radiation. And then, we examined alteration of the cell cycle, cell proliferation, cytokine level and the activation of cell signaling protein. As a result, cell cycle arrest and cell death were induced by the activation of caspase-3 and PARP in the irradiated cells with KCN treatment. These changes of the irradiated cell with KCN treatment were induced by the release of $TNF-{\alpha}$ via $NF-{\kappa}B$ activation. In conclusions, enhanced radio-sensitivity mediated by KCN induced cell death and it occurs by $NF-{\kappa}B$-dependent $TNF-{\alpha}$ production.

Anti-inflammatory Effects of Salvia Miltiorrhizae Radix Water Extract in RAW 264.7 Cells and Mouse Induced by Lipopolysaccharide (단삼 물 추출물의 LPS로 유도된 RAW 264.7 세포와 생쥐 염증모델에서의 항염증 효과)

  • Objectives: This study was performed to identify the anti-inflammatory effects of Salvia miltiorrhizae radix Water extract (SMW) on lipopolysaccharide (LPS) induced inflammation. Methods: RAW 264.7 cells were treated with 500 ng/ml of LPS. SMW (0.1, 0.25, 0.5 mg/ml) was treated 1 h prior to LPS. Cell viability was measured by MTT assay. Levels of nitric oxide (NO) were measured with Griess reagent and pro-inflammatory cytokines were measured by enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (PCR). We also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor-kappa B ($NF-{\kappa}B$) activation by western blot. In addition, we observed mice survival rate after LPS and examined their cytokine levels of serum and liver tissue. Results: SMW itself did not have cytotoxic effects in RAW 264.7 cells less than 0.5 mg/ml. SMW treatment inhibited the production of NO, and interleukin $(IL)-1{\beta}$ which is pro-inflammatory cytokine. And SMW treatment inhibited the LPS-induced activation of MAPKs such as extracellular signal-regulated kinase1/2 (ERK1/2), p38 kinases (p38), c-Jun NH2-terminal kinase (JNK) and $NF-{\kappa}B$. In addition, it also showed reducing the level of $IL-1{\beta}$ on the serum and liver tissue of mice. Also, death of LPS-induced mice was inhibited by SMW. Conclusions: The result suggests that treatment of SMW could reduce the LPS-induced inflammation. Thereby, SMW could be used as a protective agent against inflammation. Also, this study could give a clinical basis that SMW could be a drug or agent to prevent inflammatory diseases.

Review on the mechanism for the reduction of raphide-induced toxicity via processing of Pinelliae Tuber and Arisaematis Rhizoma (포제(炮製)에 의한 반하(半夏)와 천남성(天南星)의 침상결정 유발 독성 감소 기전 고찰)

  • Objectives : The processing of Pinelliae Tuber and Arisaematis Rhizoma is a crucial step to reduce the severe acrid irritation mainly due to the needle-like crystals (raphides). Ginger, alum and bile juice have been used as adjuvant materials for the processing. Methods : Bibliographic research on ancient processing and experimental processing was performed to investigate the toxicity reduction mechanisms of the processing with ginger, alum and bile juice. Results : Ginger has been a major adjuvant for the processing of Pinelliae Tuber, followed by alum and bile juice since Song (宋) and Myeong (明) dynasties, and Arisaematis Rhizoma has been mainly used as Damnamseong (膽南星). The raphides consisting of calcium oxalate, lectin, agglutinin and polysaccharides can induce acrid irritation and the inflammatory reactions. The lipophilic components in the ginger denatured the structure of raphides and 6-gingerol-contained ginger extract attenuated the inflammatory reaction. The calcium ion (Ca2+) of calcium oxalate was substituted to the aluminium ion (Al3+) of the alum, which damaged the calcium oxalate structure. Lectin attached to the surface of raphides was dissolved in alum solution and consequently its structure was denatured. The cholate in the bile juice formed the complex with the oxalate anion or the calcium cation. Moreover, the enzymes activated by Lactobacillus or Bifidobacterium during the fermentation promoted the fragmentation of oxalate. Conclusion : The adjuvant materials damaged the raphides by denaturing or degrading the calcium oxalate, resulting in the reduction of acrid irritation. Further experimental studies would support the toxicity reduction mechanism of the processing.

Numerical Investigation of Aerodynamic Characteristics of a Ducted Fan-Vane Configuration and Improvement of Control Performance in Hover (덕트 팬-베인 형상의 제자리 비행 공력 특성 및 조종 성능 개선에 관한 수치적 연구)

  • In the present study, numerical simulation was performed to investigate aerodynamic characteristics of a ducted fan-upper/lower vanes system in hover. Sensitivity analysis of aerodynamic forces for a system component was conducted with the deflection angle of upper vanes varying but at the constant rotational speed and the collective pitch angle of fan blades. Then, vane control performance and duct airload distributions were analyzed in detail to physically understand operating mechanisms of individual vane and interference effect between duct and vanes. Finally, new control concept of operating upper vanes has been proposed to improve the control performance of the full configuration. It is found that the side force and rolling moment of upper vanes increase linearly with the variation of those deflection angle; however, the total side force is significantly small due to the reaction force acted on the duct. It is also found that upper vanes close to the duct contraction side have a key role in changing vane control forces. It is revealed that the duct suction pressure is induced by the interaction with the suction side of upper vanes, while duct pressure recovery by the interaction with the pressure side, leading to increase in duct asymmetric force. When four upper vanes are kept in situ at 0° deflection angle or removed, the total control performance was improved with duct asymmetric force reduced and the total magnitude of roll remarkably increasing up to 80%.

Recent (2010-2019) foodborne outbreaks caused by viruses in the Republic of Korea along with their detection and inactivation methods (바이러스에 의한 최근(2010-2019) 국내 식중독 사고와 검출법 및 제어법에 대한 동향 조사)

  • In this review, recent foodborne outbreaks caused by viruses in the Republic of Korea (2010-2019) were analyzed. The human norovirus was found to be the major foodborne virus causing an average of 94.9% of the viral outbreaks. Reverse-transcription polymerase chain reaction (PCR) with electrophoresis has been widely used to detect viruses, but several rapid detection methods, including real-time PCR, multiplex PCR, and quantum dot assay, have also been suggested. For norovirus inactivation studies, surrogates such as murine norovirus and feline calicivirus have been widely used to identify the reduction rate owing to the limitations in laboratory cultivation. Conversely, direct cell infection studies have been conducted for other foodborne viruses such as adenovirus, astrovirus, rotavirus, and hepatitis A or E virus. Moreover, virucidal mechanisms using various physical and chemical treatments have been revealed. These recent studies suggest that rapid in situ detection and effective control are valuable for ensuring food safety against viral infections.

Characteristics and Mechanisms of Phosphate Sorption by Calcined Oyster Shell (소성 굴패각에 의한 인산염의 흡착특성 및 메커니즘)

  • BACKGROUND: Although the calcined oyster shell can be used as a calcium-rich adsorbent for phosphate removal, information about it is limited. The purpose of this study was to evaluate the phosphate adsorption characteristics and its mechanism using calcined oyster shells. METHODS AND RESULTS: In this study, calcined oyster shell (C-OS600) was prepared by calcining oyster shells (P-OS) at 600℃ for 20 min. Phosphate adsorption by C-OS600 was performed under various environmental conditions. Phosphate adsorption by C-OS600 occurred rapidly at the beginning of the reaction, and the time to reach equilibrium was less than 1 h. The optimal isotherm and kinetic models for predicting the adsorption of phosphate by C-OS600 were the Langmuir isotherm and pseudo-second order kinetic model, respectively, and the maximum adsorption capacity derived from the Langmuir isotherm was 68.0 mg/g. The adsorption properties of phosphate by C-OS600 were dominantly influenced by the initial pH and C-OS600 dose. In addition, SEM-EDS and FTIR analysis clearly showed a difference in C-OS600 before and after phosphate adsorption, which proved that phosphate was adsorbed on the surface of C-OS600. CONCLUSION: Overall, the calcined oyster shell can be considered as an useful and effective adsorbent to treat wastewater containing phosphate.
