Optimization of Enzymatic Synthesis Condition of Structured Lipids by Response Surface Methodology (반응표면분석에 의한 기능성 유지의 효소적 합성 조건 최적화)

  • Synthesis conditions were optimized using response surface methodology for producing structured lipids (SL) by interesterification of DHA-enriched algae oil derived from microalgae, Schizochytrium sp. and corn oil. Reaction was performed fer 24 hr at $55^{\circ}C$ catalyzed by immobilized lipase from Rhizomucor miehei (RM IM) in shaking water bath. Major fatty acids of SL were palmitic (21.70 mol%), oleic (20.20 mol%), and linoleic (27.34 mol%) acids, and DHA (15.06 mol%). To separate newly synthesized SL-triglycerides (TG) species, HPLC with evaporative light scatting detector (ELSD) was used. Production conditions were optimized using central composite design with reaction temperature $(35-75^{\circ}C,\;X_1)$, reaction time $(2-42\;hr,\;X_2)$, and enzyme concentration $(2-14%,\;X_3)$ as variables. When variables were $70.28^{\circ}C\;(X_1),\;28.74\;hr\;(X_2),\;and\;11.30%\;(X_3)$, maximum content of selected three peaks of synthesized SL-TG species was predicted as 6.97 area%.

Analysis of fatty acid methyl ester in bio-liquid by hollow fiber-liquid phase microextraction

  • Bio-liquid is a liquid by-product of the hydrothermal carbonization (HTC) reaction, converting wet biomass into solid hydrochar, bio-liquid, and bio-gas. Since bio-liquid contains various compounds, it requires efficient sampling method to extract the target compounds from bio-liquid. In this research, fatty acid methyl ester (FAME) in bio-liquid was extracted based on hollow fiber supported liquid phase microextraction (HF-LPME) and determined by Gas Chromatography-Flame Ionization Detector (GC-FID) and Gas Chromatography/Mass Spectrometry (GC/MS). The well-known major components of biodiesel, including methyl myristate, palmitate, methyl palmitoleate, methyl stearate, methyl oleate, and methyl linoleate had been selected as standard materials for FAME analysis using HF-LPME. Physicochemical properties of bio-liquid was measured that the acidity was 3.30 (${\pm}0.01$) and the moisture content was 100.84 (${\pm}3.02$)%. The optimization of HF-LPME method had been investigated by varying the experimental parameters such as extraction solvent, extraction time, stirring speed, and the length of HF at the fixed concentration of NaCl salt. As a result, optimal conditions of HF-LPME for FAMEs were; n-octanol for extraction solvent, 30 min for extraction time, 1200 rpm for stirring speed, 20 mm for the HF length, and 0.5 w/v% for the concentration of NaCl. Validation of HF-LPME was performed with limit of detection (LOD), limit of quantitation (LOQ), dynamic range, reproducibility, and recovery. The results obtained from this study indicated that HF-LPME was suitable for the preconcentration method and the quantitative analysis to characterize FAMEs in bio-liquid generated from food waste via HTC reaction.

  • A liquid-based colorimetric assay using a pH indicator was introduced for high-throughput monitoring of lysine decarboxylase activity. The assay is based on the color change of bromocresol purple, measured at 595 nm in liquid reaction mixture, due to an increase of pH by the production of cadaverine. Bromocresol purple was selected as the indicator because it has higher sensitivity than bromothymol blue and pheonol red within a broad range and shows good linearity within the applied pH. We applied this for simple determination of lysine decarboxylase reusability using 96-well plates, and optimization of conditions for enzyme overexpression with different concentrations of IPTG on lysine decarboxylase. This assay is expected to be applied for monitoring and quantifying the liquid-based enzyme reaction in biotransformation of decarboxylase in a high-throughput way.

Stepwise Addition Technology for the High Yield Production of Chitosanoligosaccharide (키토산올리고당 생산을 위한 다단계 첨가방법)

  • Optimization of the chitosanoligosaccharide production was studied with chitosanase. The optimum conditions for the enzymic reaction have been determined. Enzyme stability was maintained above 90% after 6 days at pH 5.0. The optimum initial reaction rate was appeared in 1.0% of chitosan solution. The production yield of chitosanoligosaccharides was over at 0.4%~2.0% of chitosan. At 4.0% of chitosan solution, however, the production yield was decreased to 64.6%. To increase the yield, stepwise addition of substrate into the reactor was investigated. In this case, the yield was increased from 64.6% to 83.2% and the final concentrations of chitosanoligosaccharide was 12.26 mg/mL. By TLC analysis, most of the chitosanoligosaccharides produced were 3-5 mers.

  • In this study, Al2O3 and H-ZSM-5 were coated on the inner wall of the stainless steel tube for the stable use of liquid hydrocarbon fuel and an endothermic catalyst used as coolant for hypersonic flying vehicles. Coke production is inevitable by the endothermic decomposition reaction of the liquid hydrocarbon fuel, and Fe, Ni metals induce the production of the filamentous coke by using a stainless steel tube reactor as a cooling channel. By coating the stainless steel with H-ZSM-5, Fe and Ni metals are prevented from being directly exposed to the liquid hydrocarbon fuel, and the formation of the filamentous coke is inhibited. In addition, Al2O3 is coated between the stainless steel and H-ZSM-5 to enhance adhesion bond strength.

  • In this study, we investigated the characteristics of biodiesel using the waste coffee oil which was extracted by waste coffee grounds. We tried to deduce the optimum conditions by defining the operating variables, such as mole ratio between methanol and coffee oil (6~18) and the reaction temperature ($45{\sim}60^{\circ}C$) in the biodiesel production processes. The performance was evaluated in terms of yields, contents of fatty acid methyl ester (FAME), viscosities, and heating values. The optimum reaction temperature was $55^{\circ}C$. Also, the best biodiesel was produced at the mole ratio between methanol and coffee oil of 12. The highest heating value of the produced biodiesel made from coffee oil was 39.0~39.4 MJ/kg, which satisfies the general standard for the biodiesel energy density, 39.3~39.8 MJ/kg.

  • Selective amino acid conjugation of bulky antibodies is a valuable asset for real-time diagnosis and therapy. However, selective conjugation incorporating a chelate-bearing radioactive atom into an antibody without affecting its immunoreactivity is a challenging task. A bifunctional chelator (BFC), a selective amino acid-targeting probe, and a linker have been developed to overcome this problem. Here, we report the synthesis of a novel propylene cross-bridged chelator (PCB)-1,8-N,N'-bis-(carboxymethyl)-1,4,8,11-tetraazacyclotetradecane (TE2A)-luminol BFC via a click reaction and radiolabel it with a 64Cu ion for tyrosine-selective conjugation of trastuzumab. In the initial optimization study, we tried different oxidative addition conditions such as electro-oxidation, hemin, horseradish peroxidase, iodogen tube, chloramine-T, and iodo beads. In this study, up to 82% of 64Cu-PCB-TE2A-luminol was conjugated with the antibody in an iodo bead-catalyzed oxidative addition reaction with an isolated yield of 24.4%.

CO2 Conversion by Controlling the Reduction Temperature of Cobalt Catalyst (코발트 촉매의 환원온도 조절을 통한 CO2 전환 공정)

  • This study investigates the impact of reduction temperature on the structure and performance of cobalt-manganese (CM) based catalysts in the direct hydrogenation reaction of carbon dioxide (CO2). It was observed that at a reduction temperature of 350 ℃, these catalysts could successfully facilitate the conversion of CO2 into long-chain hydrocarbons. This efficiency is attributed to the optimal conditions provided by the core-shell structure of the catalysts, which effectively catalyzes both the reverse water-gas shift (RWGS) and Fischer-Tropsch (FT) reactions. However, as the reduction temperature increased to 600 ℃, the effectiveness of the reaction process was hindered, and there was a shift in selectivity towards methane. This shift is due to the excessive reduction of the catalyst's outer shell, which reduces the number of RWGS sites and subsequently suppresses the production of CO. These findings highlight the importance of carefully controlling the reduction temperature in the design and optimization of cobalt-based catalysts. Maintaining a balance between the RWGS and FT reactions is crucial. This emphasizes that the reduction temperature is a key factor in efficiently generating long-chain hydrocarbons from CO2.

Preliminary studies for production of 61Cu using natural nickel target with RFT-30 cyclotron

  • 61Cu is a promising PET radiometal having favorable nuclear decay characteristics with appropriate half-life of 3.3 h. Owing its promising capabilities in radiopharmaceutical chemistry and its chemical similarities with its isotopes 64Cu and 67Cu, in this work we have tried to optimize the production and separation conditions of 61Cu. 61Cu was produced via (p, x) reaction with natural nickel which was electroplated on the high purity silver coated copper backing target holder. The optimization of target electrodeposition, beam energy and current modulation, target dissolution and separation were optimized in this study. Preliminary studies show that 61Cu was successfully produced and separated which can be further extended for the production of 64Cu and 67Cu.

A study on Contact force of Rubber Seal for wheel bearing (휠베어링 고무 실의 접촉력에 관한 연구)

  • Wheel bearing unit has been exclusively applied to car wheel supporting device. The seal for wheel bearing is required to have both high sealing effects and low reaction forces because wheel bearing are operated on adverse environmental conditions such as mud and splash water. High sealing effects are for the protection of bearing ball wear from dust influx. In order to ensure high sealing effects, it is a easiest way to increase contact force which are affected by geometric characteristics, material properties and interferences between seal and inner bearing but induces higher wear phenomena. Interferences in all variables are most important factor to determine the performance of wheel bearing. In this study, optimization of interference amount was performed with finite element analysis with commercial code ABAQUS. For the sake of finite element analysis, tensile tests of rubber material were conducted and governing equation of nonlinear behavior was achieved. Hock-up bearing was manufactured with optimized interference amount. Results of torque and mud spray tests using this bearing unit are performed. Less torque and moisture influx of bearing with optimized interference amount is evidence to validity of this study.
