Influence of Ginsenosides on the Kainic Acid-Induced Seizure Activity in Immature Rats

  • We studied the effects of ginsenosides in immature rats based upon the previous results that ginseng has a suppressive or anticonvulsive activity. To examine the suppressive effect of ginsenosides on kainic acid-induced seizures, the severities and frequencies were observed for 4 h after injection of kainic acid (KA; i.p., 2 mg/kg b.w.) using 10-day-old male Sprague-Dawley rats ($22{\pm}2\;g$). Protopanaxadiol saponins such as ginsenoside-Rb1 (Rb1), ginsenoside-Rb2 (Rb2), ginsenoside-Rc (Rc), and ginsenoside-Rd(Rd) generally reduced the seizure activities while protopanaxatriol saponins such as ginsenoside-Rg1 (Rg1) and ginsenoside-Re (Re) rather increased stereotypic "paddling-like" movements. When vinyl-GABA (v-G) was injected together with Rb1 or Rc, KA-induced seizure severities were additionally reduced only by the injection of Rc, but not by Rb1. The level of gamma isozyme of protein kinase C (PKC-${\gamma}$) in the hippocampus increased about three times as much as that of normal rats at 4 h after KA injection. The increased level of PCK-${\gamma}$ by KA was significantly reduced to about 35% by the coinjection with v-G alone, but it was not changed by v-G together with Rb1 or Rc. The increased level of PKC-${\gamma}$ at 4 h after injection of KA was not consistent with the reduction of seizure severities between Rb1 and Rc. These results suggest that Rc and Rb1 may reduce seizure severity independent of PKC-${\gamma}$ levels, and Rc may additionally act with v-G regarding the GABA metabolism during the stage of KA-induced seizures in the immature rats.

  • Ginseng total saponins (GTS) injected intracerebroventricularly (i.c.v.) at doses from 0.1-1 vs inhibited the i.c.v. injection stress-induced plasma corticosterone levels in mice. The inhibitory action of GTS was blocked by co-administered NG-nitro-L-arginine methyl ester (L-NAME; 1.5 us, i.c.v.), an. inhibitor of nitric oxide synthase (NOS). Of the ginsenosides Rbl, Rba, Rc, Rd, Re, Rf, Rgl,20(S)-Rg3, and 20(R)-Rg3 injected i.c.v. at doses from 0.01 to 0.3ug(or 1 uE),20(5)-Rg3 and Rc significantly inhibited the o.c.v. injection stress-induced Plasma corticosterone levels. The inhibitory actions of 20(S)-Rg3 and Rc were blocked by co-administered L-NAME (1.5 n, i.c.v.). These results suggest that G75, 20(S)-Rg3 and Rc may inhibit the i.c.v. injection stress-induced hypothalamo-pituitary-adrenal response by inducing NO production in the brain.

  • 홍삼 총 사포닌 투여군은 대조군과 비교시 total free radical 및 malondialdchydc 농도는 유의상 있게 감소되었으며, 단백질의 carbonyl 농도는 다소 감소하는 경향을 나타내었다. 그리고 홍삼 총 사포닌 투여군의 경우 Cu, Zn-SOD, catalasc, GSII reductase 등의 항산화 효소와 nonprotein-SH가 대조군 보다 증가되었다. 홍삼 총 사포닌의 구성성분들인 ginsenoside Rb$_1$, Rb$_2$, Rc, Rd, Re, Rg$_1$, Rh$_1$, Rh$_2$, Rf 중 ginsenoside Rh$_2$는 catalase 활성을 대조군보다 유의성있게 증가시켰으며, ginsenoside Rh$_1$ 및 Rc의 경우 GSII peroxidase 활성이 증가하는 경향을 나타내었다. 그리고 Cu, Zn-SOD의 경우 ginsenoside Rc는 대조군보다 유의성있게 감소시켰으며, GSII reductase의 경우 유의성있는 변화는 관찰되지 않았다.

Screening of Antioxidative Components from Red Ginseng Saponin (홍삼 사포닌의 항산화활성 성분 Screening)

  • Aerobic cells are normally protected from the damage of free radicals by antioxidative on , zymes such as superoxide dismutase (SOD), catalase, glutathione (GSH) peroxidase, GSH S- transferase and GSH reductase which scavenge free radicals as well as nonenzymatic antioxidants such as ceruloplasmin, albumin and nonprotein-SH including GSH. The effects of each component (ginsenoside $Rb_1$, $Rb_2$, Rc, Rd, Re, $Rb_1$, Rf, $Rh_1$ and $Rh_2$) of red ginseng on the antioxidative enzyme activities were investigated in the liver in order to screen antioxidative components of red ginseng. Ginsenoside $Rb_1$ and Rc showed a tendency to increase GSH peroxidase activity, while ginsenoside Rc significantly decreased Cu,Zn-SOD activity. Especially, ginsenoside $Rh_2$ significantly increased catalase activity. These results suggest that ginsenoside $Rh_2$ is an important active component among total saponins of red ginseng.

Relationship among Ginsenosides of Panax ginseng Root under the Variation of Mineral Nutrients (무기영양변화에서 인삼근Ginsenoside의 상호관계)

  • Relationships among ginsenosides, panaxadiol(PD), panaxatriol(PT), and total saponin(TS) in Panax ginseng root (2nd Year) grown with culture solotion different in nitrogen, phosphorus and potassium level were analyzed by simple correlation, multiple regression and standard partial regression coefficient. The closeness between ginsenosides by simple correlation was closely related with the similarity of molecular structure. The content of PT was much attributed to Re and Rg1. The contribution order of ginsenosides for PD was Rb1>Rb2$\geq$Rd>Rc. There was significant positive correlation between PT and PD but PD increased more rapidly than PT. Thus total saponin depended much on PD and PT/PD decreased with the increase of total saponin content. All ginsenosides, especially Re showed decreasing tendency with the increase of root weight.

  • The analytical conditions of ginsenosides were optimized using high performance liquid chromatography (HPLC). The optimal analysis conditions were set up from the experiments using different gradient conditions, and the ginsenosides contents of red ginseng products and their raw materials were analysed. Red ginseng contained 0.29% Rg1, 0.82% Rb1, 0.38% Rc, 0.32% Rb2, 0.11% Rd, showing the highest ginsenoside contents. Among the red ginseng products, red ginseng extract showed the highest content. The ginsenoside contents were varied according to the raw material type and product type. Re and Rb1 contents were the highest in most raw materials and products.

An Rapid Extraction of Ginseng Saponin Compounds (인삼사포닌 화합물의 신속한 추출)

  • A new rapid saponin extraction method was developed with using of organic solvent and waring blonder. There was a good correlation between previous distillation method and this method in f major ginsenosides ($Rb_1$, $Rb_2$, Rc, Rd, Re, Rg1) contents. When the ratio of methanol and chloroform was 7:3, this method showed similar saponin contents (total major. ginsenosides contents) comparing with distillation method. Contents of total major ginsenosides were 2.41% in this method and 2.54% in distillation method. However, crude saponin content of this method was higher than that of distillation method.

Effect of Microwave Treatment on Korean Ginseng (고려인삼의 마이크로파 처리 효과)

  • The effect of microwave treatment on Korean ginseng was studied by measuring the changes in moisture, crude lipid, crude ash, crude protein, total dietary fiber and saponin contents, as well as changes in density, color and microstructure. Korean ginseng was treated with 100 or 200 watts of microwaves for 1 or 3 hrs, respectively, followed by drying using an oven at $60^{\circ}C$ for 96 hrs. The moisture contents decreased to 13.12~10.77% from an initial 76.26%. The amounts of lipid and ash were reduced in proportion to the time of microwave treatment and level of microwave power. The amount of protein in ginseng after microwave treatment did not significantly change. The amount of total dietary fiber increased after microwave treatment and the color of dried ginseng became dark. The amounts of ginsenoside-$Rb_1$, $Rb_2+Rb_3$, Rc, Rd, Re, Rf, $Rg_1$, $Rg_2+Rh_1$ and $Rg_3$ were reduced after treatment with 100 watts of microwave radiation for 1 and 3. The amounts of ginsenoside-$Rb_1$, Rd, Re, Rf, $Rg_1$, $Rg_2+Rh_1$ and $Rg_3$ after treatment with 200 watts of microwave radiation for 1 and 3 hr also reduced. On the other hand, the amounts of ginsenoside-$Rb_2+Rb_3$ and Rc after treatment of ginseng with 200 watts of microwave radiation for 1 and 3 hrs were increased.

Effects of Ginsenosides on $GABA_A$ Receptor Channels Expressed in Xenopus Oocytes

  • Ginsenosides, major active ingredients of Panax ginseng, are known to regulate excitatory ligand-gated ion channel activity such as nicotinic acetylcholine and NMDA receptor channel activity. However, it is not known whether ginsenosides affect inhibitory ligand-gated ion channel activity. We investigated the effect of ginsenosides on human recombinant $GABA_A$ receptor (${\alpha}_1{\beta}_1{\gamma}_{2s}$) channel activity expressed in Xenopus oocytes using a two-electrode voltage-clamp technique. Among the eight individual ginsenosides examined, namely, $Rb_1$, $Rb_2$, Rc, Rd, Re, Rf, $Rg_1$ and $Rg_2$, we found that Rc most potently enhanced the GABA-induced inward peak current ($I_{GABA}$). Ginsenoside Rc alone induced an inward membrane current in certain batches of oocytes expressing the $GABA_A$ receptor. The effect of ginsenoside Rc on $I_{GABA}$ was both dose-dependent and reversible. The half-stimulatory concentration ($EC_{50}$) of ginsenoside Rc was 53.2$\pm$12.3 $\mu$M. Both bicuculline, a $GABA_A$ receptor antagonist, and picrotoxin, a $GABA_A$ channel blocker, blocked the stimulatory effect of ginsenoside Rc on $I_{GABA}$. Niflumic acid (NFA) and 4,4'-diisothiocyanostilbene-2,2'-disulfonic acid (DIDS), both $CI^{-1}$ channel blockers, attenuated the effect of ginsenoside Rc on I$I_{GABA}$. This study suggests that ginsenosides regulated $GABA_A$ receptor expressed in Xenopus oocytes and implies that this regulation might be one of the pharmacological actions of Panax ginseng.

The Tentative Plans of Middle-rise Traditional Houses (Han-ok) Located on Seoun-dong in Urban Area of Cheongju, Korea (중층형 생활한옥 모델시안 연구 - 청주 서운동의 사례를 중심으로 -)

  • Focusing on the creation of a new han ok, especially a mid-rise hybrid-structured Han-ok, this study proposes a middle-rise (four-story) Han-ok on one and two lots located in Seoun-dong, the existing Han-ok intensive housing site in downtown Cheongju. 1) In terms of layout and function, according to the existing L-shaped Han-ok corresponding to the road and the direction, the parking lots and shops on the 1st floor, the business facility on the 2nd floor, the Han-ok on the 3rd and 4th floors are placed. There are yards, open roof yards, and semi-open Daecheong(大廳, main hall), which can be shared by residents. 2) In terms of structure and form, one or two floors (some 3 floors) are 5.4m square and 5.4×6.6m modules of the RC(Reinforced Concrete) group, and the upper floor reflects the 2.7m module, size and shape of the existing Han-ok. By extending the outer wall of the RC group in the lower floors (1st to 3rd floors) to the wooden exterior of the upper floors (2nd to 4th floors), it is attempted to avoid the awkward appearance of the RC group being exposed to wooden structures. And it is also attempted to reflect the wooden shape and design elements through the elevation elements such as horizontal windows, corner windows, picture frames, and vertical slits. 3) In environmental control and facilities, it is attempted to smooth the ventilation of the building by forming a vertical upward airflow from the dark space of the low floor to the positive of the upper floor. This doubles the effect through a vertical rise of cold air generated in a narrow alleyway, piloti parking lot, and the various voids. In addition to the Daecheong and Numaru(loft) of Han ok, the rooftop yard, the terrace, and the balcony, horizontal natural ventilation is generated through divided doors and transom windows.
