상수원수의 혼화 및 응집 조건에 따른 DAF pump 장치의 부상분리효율 (Flotation Efficiency of the DAF Pump System for Mixing and Coagulation Conditions in Raw Drinking Water)

  • This study found that flotation efficiencies for removing algae and micro particles in raw water were optimized on mixing intensity and time of the mixing and flocculation conditions with a continuous DAF system. It is more efficient for mixing intensity at 23.1 $s^{-1}$ and time at 660 s(Gt value : 15246) to float flocculated floc with the raw water in M water treatment plant. Flotation efficiency was more than about 0.9 when operated pressure and A/S ratio were sustained at 5 $kg_f{\cdot}cm^{-2}$ and up to 0.056 $mL{\cdot}mg^{-1}$. The continuous DAF system made by the study could be continuously operated for 20 days and sustained not exceeding 4 NTU with raw water with low turbidity(13.4~9.8 NTU).

Ruminal Behavior of Protein and Starch Free Organic Matter of Lupinus Albus and Vicia Faba in Dairy Cows

  • Faba beans (vicia faba) (FB) and lupin seeds (Lupinus Albus) (LS) were dry roasted at three temperatures (110, 130, $150^{\circ}C$) for 15, 30 or 45 min to determine the effects of dry roasting on rumen degradation of crude protein and starch free organic matter ($^{PSF}OM$). Rumen degradation characteristics of $^{PSF}OM$ were determined by the nylon bag incubation technique in dairy cows fed 60% hay and 40% concentrate. Measured characteristics of $^{PSF}OM$ were undegradable fraction (U), degradable fraction (D), soluble fraction (S), lag time (T0), and the rate of degradation (Kd). Based on the measured characteristics, rumen availability ($RA^{PSF}OM$) and bypass $^{PSF}OM$ ($B^{PSF}OM$) were calculated. Dry roasting did not have a greater impact on rumen degradation characteristics of $^{PSF}OM$ (p>0.05). S varied from 32.1 (raw) to 30.0, 27.8, 30.8% (LS) and 15.4 (raw) to 14.4, 20.8, 20.9% (FB); D varied from 65.4 (raw) to 66.3, 66.9, 55.9% (LS) and 54.9 (raw) to 55.0, 51.0, 64.7% (FB); U varied from 2.6 (raw) to 7.3, 7.0, 7.7% (LS) and 29.7 (raw) to 30.6, 28.2, 14.4% (FB); Kd varied from 6.0 (raw) to 7.3, 7.0, 7.7% (LS) and 22.4 (raw) to 24.4, 21.1, 7.9% (FB); $B^{PSF}OM$ varied from 35.5 (raw) to 33.8, 36.6, 38.2% (LS) and 41.3 (raw) to 41.5, 39.7, 47.6% (FB) at 110, 130 and $150^{\circ}C$, respectively. Therefore dry roasting did not significantly affect $RA^{PSF}OM$, which were 353.7, 367.9, 349.6, 336.9 (g/kg DM) (LS) and 12.82, 127.0, 133.7, 117.1 (g/kg DM) (FB) at 110, 130 and $150^{\circ}C$, respectively. These results alone with our previously published reports indicate dry roasting had the differently affected pattern of rumen degradation characteristics of various components in LS and FB. It strongly increased bypass crude protein (BCP) and moderately increased starch (BST) with increasing temperature and time but least affected $^{PSF}OM$. Such desirable degradation patterns in dry roasted LS and FB might be beneficial to the high yielding cows which could use more dry roasted $^{PSF}OM$ as an energy source for microbial protein synthesized in the rumen and absorb more amino acids and glucose in the small intestine.

봉약침액(蜂藥鍼液_이 RAW 264.7 세포의 COX-2, P38, ERK 및 JNK에 미치는 영향(影響) (The Effect of Bee Venom on COX-2, P38, ERK and JNK in RAW 264.7 Cells)

  • The purpose of this study was to investigate the effect of Bee Venom on the lipopolysaccharide(LPS), sodium nitroprusside(SNP), hydrogen peroxide($H_2O_2$)-induced expressions of cyclooxygenase-2(COX-2), p38, jun N-terminal Kinase(JNK) and extra-signal response kinase(ERK) in RAW 264.7 cells, a murine macrophage cell line. Method : The expressions of COX-2, p38, JNK and ERK were determined by western blotting with corresponding antibodies. Results : 1. The 0.5, 1 and $5\;{\mu}g/ml$ of bee venom inhibited significantly LPS and SNP-induced expression of COX-2 compared with control, respectively. The 0.5, 1 and $5\;{\mu}g/ml$ of bee venom inhibited insignificantly $H_2O_2$-induced expression of COX-2 compared with control, respectively. 2. The 0.5, 1 and $5\;{\mu}g/ml$ of bee venom inhibited significantly LPS, SNP and $H_2O_2$-induced expression of p38 compared with control, respectively. 3. The 1 and $5\;{\mu}g/ml$ of bee venom inhibited significantly SNP-induced expression of JNK compared with control, respectively. All of bee venom inhibited insignificantly LPS and $H_2O_2$-induced expression of JNK compared with control, respectively. 4. The $5\;{\mu}g/ml$ of bee venom inhibited significantly SNP-induced expression of ERK, the $0.5\;{\mu}g/ml$ of bee venom increased significantly $H_2O_2$-induced expression of ERK compared with control. The 0.5, 1 and $5\;{\mu}g/ml$ of bee venom inhibited insignificantly LPS-induced expression of ERK compared with control, respectively.

꽃송이버섯 추출물이 RAW 264.7 세포에서 TNF-α, iNOS, IL-1β 유전자 발현에 미치는 영향 (Effect of Water Extract of Sparasis crispa on the Expression of TNF-α, iNOS and IL-1β Genes in RAW 264.7 Cells)

  • Purpose : The purpose of this study was to examine the anti-inflammatory effects of Sparassis crispa (SC). SC is a well-known traditional herbal remedy and its mushroom is used for treatment of inflammation. Many diseases that are increasing recently have characteristics of inflammatory diseases. Researchers are finding bioactive substances from natural products that can promote treatment and prevention of inflammation. We investigated the effect of water extracted from SC on the expression of effector genes involved in the function of RAW 264.7 cells. Methods : Effects of RAW 264.7 cells on cell viability, antioxidation, and mRNA expression were examined using water extracts from SC. A 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay was performed to determine the effect of water extracts from SC on cell viability in RAW 264.7 cells. Inflammation of RAW 264.7 cells induced by lipopolysaccharide (LPS) treatment and expression levels of inflammatory cytokine TNF-α, iNOS and IL-1β gene were analyzed using quantitative reverse transcription PCR (qRT-PCR) analysis. Results : The MTS assay was performed on RAW 264.7 cells after treatment with various concentrations of water extracts of SC. Treatment of RAW 264.7 cells with water extracts from SC and LPS at a concentration of 0.125, 0.5 mg/㎖ for twenty four hours promoted mRNA expression of TNF-α, iNOS and IL-1β. Conclusion : MTS assay was applied to RAW 264.7 cells after various concentrations of water extracts of SC. Through experimental demonstration of anti-oxidant and anti-inflammatory effects of water extracts from SC, we suggest that SC is a valuable material for the prevention and treatment of various inflammatory diseases.

참가자미의 저장 중 오염미생물의 생육에 미치는 아가미의 유무, 건조도, 저장온도 및 키토산-아스코베이트의 처리효과 (Effect of Gill, Drying and Temperature and Chitosan-Ascorbate Treatment on Growth of Contaminated Microorganism in Flounder during Storage)

  • 생 및 반건참가자미의 저장성 향상을 위하여 아가미를 제거하지 않은 것 (WG)과 제거한 것 (OG), 건조도에 따라 생것(0 D), 20%건조한 것 (20 D), 40%건조한 것 (40 D), 0.1% chitosan-ascorbate (CA)의 처리와 무처리로 구분, 진공 포장하여 $5{\sim}10^{\circ}C$에서 10일간 저장하면서 미생물의 증식정도를조사하였다. $10^{\circ}C$에서 저장 중 생 참가자미의 WG와 OG를 비교한 결과, OG가 WG에 비하여 total aerobacter는 $0.3{\sim}0.5$ log cycle이 감소되었으며, coliform과 E. coli의 수도 낮았다. $10^{\circ}C$에서 저장 중 0 D, 20 D 및 40 D를 비교한 결과, total aerobacter, coliform 및 E. coli 모두 20 D에서 가장 낮았으며 total aerobacter의 경우는 생것에 비하여 $0.42{\sim}1.20$ log cycle이 낮았다. 진공포장한 생 참가자미를 $5^{\circ}C$$10^{\circ}C$에서 10일간 저장한 결과, $5^{\circ}C$의 경우는 total aerobacter가 log 4.17 CFU/g로 $10^{\circ}C$보다 $0.6{\sim}1.3$ log cycle이 낮았다. 가자미로부터 분리한 3종의 미생물 [aerobacter (A1), coliform (C12), E-coli (E7)]에 대한 CA의 항균효과를 조사한 결과 0.1%에서 3종 모두 생육이 완전히 억제되었다. 생 참가자미에 0.1% CA를 처리한 후 진공 포장하여 $10^{\circ}C$에서 10일간 저장한 결과 CA 비처리에 비하여 total aerobacter는 1.5 log cycle이 감소되었으며, coliform 및 E. coli도 감소되었으며, CA처리 반건참가자미 (20 D)에서도 유사한 경향을 나타내었다. 0 D 및 20 D를 CA처리한 후 진공 포장하여 $10^{\circ}C$에서 10일간 저장한 생시료 또는 조리하여 관능검사를 행한 결과, 0 D와 20 D 모두 CA 처리군에서 외관, 신선도, 조직감 및 종합적기호도 값이 비처리군의 "보통"에 비하여 우수한 것 (좋다$\sim$매우좋다)으로 평가되었다.

나물 조리시 양념사용에 관한 연구 -염분농도를 중심으로- (A Study on the Use of Seasoning for NAMUL Preparation -in salt concentration-")

  • 조리서를 통하여 염류를 제외한 나물 양념의 분량을 조사하여 사용하고, 염류배합에만 변동을 주어 관능검사로 최적 염분농도 및 최적의 배합조건을 구한 결과를 요약하면 다음과 같다. 1. 재료 300g에 대하여 숙채는 파 15.5g, 마늘 7.9g, 참기름 9.0g, 깨소금 5.0g을, 생채는 파 11.5g, 마늘 7.0g, 참기름 6.0g, 깨소금 3.3g, 설탕 13.9g, 식초 10.6g, 고추가루 5.6g을 사용하였다. 2. 나물의 최적 염분농도는 숙채가 1.412%, 생채가 1.368%였다. 3. 소금과 새우젓으로 간을 한 나물에서는 염분농도 0.84%∼l.89%사이에서 염분농도가 질감에 영향을 주지 않았다. 4. 색의 기호성은 간장의 양을 증가시켜 첨가할수록 낮아졌으며, 색의 기호에 영향을 주지 않고 사용할 수 있는 간장의 양은 오이나물과 도라지나물같이 색이 옅은 경우는 중량의 0.8%∼l.0%였고, 시금치나물같이 색이 진한 경우는 중량의 4.8%였다. 5. 전체적인 맛이 좋은 염분농도는 숙채가 1.223%∼l.513%, 생채가 1.183%∼l.373%로 짠맛의 결과와 일치하는 경향을 보였다.

색소첨가 NIR을 이용한 우유 체세포수 측정법 개발 (Development of Rapid Somatic Cell Counting Method by Using Dye Adding NIR Spectroscopy)

  • 본 연구에서는 근적외선 분광광도계를 이용하여 원유의 체세포수 측정에 필요한 최적의 색소를 선정하기 위하여 eosin-Y, methyl red, methylen blue, resazurin 및 amido black 10B등의 5가지 색소를 0.01%첨가 후 근적외선 분광광도계를 이용하여 400-2,500 nm 영역에서 측청하였으며, PLS(partial least squar)분석 결과는 다음과 같다. 교정부 모델에서의 상관도는 raw milk 0.78, eosin-Y 0.65, methyl red 0.63, methylen blue 0.65, resazurin 0.98 그리고 amido black 10B는 0.99 였다. 또한, 검증부 모델에서의 상관도는 raw milk 0.49, eosin-Y 0.21, methyl red 0.36, methylen blue 0.47, resazurin 0.95 그리고 amido black 10B는 0.98 였다. 위 결과를 종합해 보면, amido black 10B를 첨가한 경우 검증부 모델의 상관도는 0.98, 검증부 오차(SEP)는 0.09로 가장 우수한 결과를 보였다.

Antioxidant Activity of Goldenrod (Solidago virgaurea) Leaf and Stem Powder on Raw Ground Pork during Chilled Storage

  • The effects of adding goldenrod leaf powder (GLP) and goldenrod stem powder (GSP) (0.1% and 0.5%) to raw ground pork on antioxidant activity were examined. The following six treatment groups were used: Control (without antioxidant), GLP1 (with 0.1% GLP), GLP2 (with 0.5% GLP), GSP1 (with 0.1% GSP), GSP2 (with 0.5% GSP) and AS (with 0.05% ascorbic acid). The chemical compositions, pH values, instrumental color, conjugated diene (CD), free fatty acids (FFA) and thiobarbituric acid-reactive substance (TBARS) value were measured during 15 d of storage at chilled temperatures. The addition of GLP and GSP showed no effect on moisture, protein and fat contents of the samples. However, adding 0.5% GSP increased the ash contents of ground pork (p<0.05). The pH values of treated samples decreased until day 7, and then increased thereafter. The addition of GLP and GSP decreased the $L^*$ and $a^*$ values and increased the $b^*$ value (p<0.05). The CD, FFA and TBARS value of the control were higher (p<0.05) than samples containing GLP and GSP. The addition of GLP and GSP resulted in a significant decrease in CD, FFA and TBARS values. Overall, this study demonstrated that GL and GS could be used as an antioxidant of raw ground pork.

Graft 공중합에 의한 생사 Sericin 정착 (Sericin- Fixation of Raw Silk by Graft Copolymerization)

  • 1. 개시제농도에 따른 graft 율의 변화를 보면 CAN농도가 0.003M 일 때 최대를 나타내었다. 2. 단량체농도가 증가함에 따라 graft 율이 상승함을 보였다. 3. HNO$_3$농도 0.lM 부근에 서 graft 율의 최대가 인정되었다. 4. 반응온도가 높아짐에 따라 graft 율이 상승하나 5$0^{\circ}C$ 이상이 되면 homopolymer 의 생성이 많아 비 효율적이다. 5. 중합시간이 1 시간 이상이 되면 graft 율이 급격히 상승함을 보이고 있다. 6. CAN 의 농도가 작을수록 연감율이 작게 나타났다. 7. graft 율이 20% 이상이 되면 sericin 이 대부분 정착되었다.

경남 북부지역 납유원유의 미생물학적 품질에 관한 연구 (A Study on the Microbiological Quality of Raw Milk in the North of Kyeongnam Area)

  • This study was carried out to obtain basic data for the Improvement of microbiological quality of raw milk. Total bacterial, psychrotrophic, thermoduric and spore counts of raw milk samples taken from milk tankers in the nothern part of kyeongnam were investigated for one year from March, 1989. The result obtained were summarized as follow 1. The number of total bacteria in raw milk averaged $4.0{\times}10^6$ CFU / ml and was not affected by seasons 2. The psychrotrophic counts of raw milk averaged $1.5{\times}10^6$ CFU / ml and it was higher in winter than in summer 3. The thermoduric counts of raw milk averaged $5.8{\times}10^4$ CFU / ml and was the lowest in winter 4. The spore counts of raw milk ranging from 3-1, 880/ ml averaged 306/ ml and was the lowest in summer
