국내산 저염분 양식 흰다리새우 유래의 슈와넬라의 병원성 세균으로의 특성 (Shewanellasp., A Potential Pathogen of White Leg Shrimp Cultured in Low Salinity Water in Korea)

  • 2022년 7월, 국내의 저염분 해수 흰다리새우 양식장에서 양식 중이던 흰다리새우가 이상유영과 식욕부진을 보이며 지속적으로 폐사하였다. 일일 평균 폐사율이 0.1%로 완만한 폐사를 보였으나, 2달 넘게 폐사가 지속되었고 성장 지체로 인한 경제적인 피해가 가중되었다. 질병 진단 과정 중, 폐사 새우의 간췌장에서 세균(KNUAF-SHP3)이 분리되었다. 본 세균의 16S rRNA 유전자의 시퀀스 분석 결과, 분리주 KNUAF-SHP3은 Shewanella에 속하는 균으로 동정이 되었고, S. algae MARS 14, S. chilikensis JC5T와 근연 관계를 보이며 같은 그룹으로 분류되었다. 실험 동물을 활용한 인위 감염 실험에서는 상대적으로 고농도(2.1×108CFU/ml와 2.1×109CFU/ml)를 적용한 군에서는 모든 새우들이 감염 증상을 보이며 감염 후 7일 내에 100% 폐사율을 나타내었다. 본 연구는 Shewanella 세균이 저염분 해수 양식 흰다리새우에서의 잠재적인 기회 감염균이 될 수 있음을 기술하고 있다.

식이내 Calcium 수준이 흰쥐의 카드뮴과 납중독에 미치는 영향 (Effect of dietary Calcium Level on Cadmium and Lead Toxicity in Rats)

  • This study was performed to investigate the effect of dietary calcium level on cadmium and lead toxicity in rats. Fifty-four male rats of Sprague-Dawely strain weighing 152$\pm$12g were blocked into 9 groups according to body weight, and were raised for 30 days. Nine experimental diets different with cadmium(0%, 0.04%), lead (0%, 0.071%) and calcijm(0.5%, 1.0%, 1.5%) levels were prepared. The results are summarized as follow. Weight gain, F.E.R.(food efficiency ratio), and weights of liver, kidney and femur were lower in cadmium exposed groups than those of heavy metal free groups. Weight gain F.E.R. and ash weight of lead groups were lower than those of heavy metal free groups. But, these were increased with increasing dietary calcium level. Cadmium and lead concentrations in blood, liver, kidney and femur were lower in rats fed 1.5% calcium than 0.5% calcium diet. Fecal cadmium and lead excretions were remarkably increased in 1.5% calcium groups, and cadmium and lead retention rates were decreased in 1.5% calcium groups. Metallothionein concentrations in liver, kidney and small intestine were higher in rats exposed to cadmium and lead. Calcium content in blood, femur and daily urinary and fecal calcium excretion were decreased by cadmium and lead additions, and increased in 1.5% calcium groups. Creatinine clearance were decreased with cadmium administratino and calcium addition. In conclusion, weight gain and organ weights were decreased with cadmium or lead administration. But, cadmium administration was more toxic than lead adminstration. Cadmium or lead toxicity was alleviated by increasing dietary calcium level. Especially, lead toxicity was alleviated in proportion to dietary calcium level.

Dislocation-oxide interaction in Y2O3 embedded Fe: A molecular dynamics simulation study

  • Oxide dispersed strengthened (ODS) steel is an important candidate for Gen-IV reactors. Oxide embedded in Fe can help to trap irradiation defects and enhances the strength of steel. It was observed in this study that the size of oxide has a profound impact on the depinning mechanism. For smaller sizes, the oxide acts as a void; thus, letting the dislocation bypass without any shear. On the other hand, oxides larger than 2 nm generate new dislocation segments around themselves. The depinning is similar to that of Orowan mechanism and the strengthening effect is likely to be greater for larger oxides. It was found that higher shear deformation rates produce more fine-tuned stress-strain curve. Both molecular dynamics (MD) simulations and BKS (Bacon-Knocks-Scattergood) model display similar characteristics whereby establishing an inverse relation between the depinning stress and the obstacle distance. It was found that (110)oxide || (111)Fe (oriented oxide) also had similar characteristics as that of (100)oxide || (111)Fe but resulted in an increased depinning stress thereby providing greater resistance to dislocation bypass. Our simulation results concluded that critical depinning stress depends significantly on the size and orientation of the oxide.

젓갈로부터 분리된 젖산균 및 효모의 프로바이오틱 특성 (Probiotic Properties of Lactic Acid Bacteria and Yeasts Isolated from Korean Traditional Food, Jeot-gal)

  • 젓갈은 전남 목포지역의 대형마트 및 가정에서 수집하였으며 오징어젓, 갈치속젓, 꼴뚜기젓, 멸치젓 등 10여종의 젓갈로부터 균을 분리하였다. 이들 분리 유산균 중 3균주가 인공위액과 인공담즙액에서 내성을 나타내었다. 이들 분리된 균주 중 생존력이 왕성한 3균주는 모두 인공위액에서 2시간, 인공담즙액에서 24시간 생존 가능하였다. 특히 유산균 ML 36, ML 128, ML 178 외 2종의 효모는 초기 생균수와 인공위액에서 2시간 배양 후 생균수의 변화가 거의 없어 가장 강한 내성을 나타내었다. 인공위액에서 분리된 균주의 생존율은 낮은 pH로 인하여 낮은 경향을 나타내었으나, 인공담즙액에 대해서는 높은 생존율을 나타내었다. 인공위액 및 인공담즙에 내성을 나타낸 3종의 유산균이 항생물질인 nicin, rifamycin, streptomycin, 그리고 tetracycline에 대하여 내성을 나타내었다. 또한 이들 분리 유산균은 병원성 미생물인 Listeria monocytogenes 대해 뚜렷한 항균 효과가 있는 것으로 나타났다.

비구속 삽입된 직물 섬유를 이용한 샌드위치 쉴드의 초고속 충격 해석 (Computational analysis of sandwich shield with free boundary inserted fabric at hypervelocity impact)

  • 본 연구에서는 우주 파편들과의 초고속 충돌로부터 우주 구조물을 보호하기 위한 새로운 하이브리드 복합재료 쉴드가 제안되었다. 제안된 쉴드의 유한요소 모델을 구성하고, 에너지 흡수율을 예측하기 위해서 유한 요소 해석을 수행하였다. 최종모델의 해석에 앞서 각 구성 요소인 알루미늄 판, PMMA 판 그리고 중간층인 직물 섬유의 해석이 먼저 수행되었으며, 각 요소의 유한요소 모델의 타당성이 검증되었다. 해석에 사용된 재료 물성은 고 변형률 속도에서의 재료 물성들을 예측하여 사용하였으며, 해석 결과 개별 요소의 에너지 흡수율이 직물섬유를 제외하고는 잘 맞음을 확인하였다. 이후 하이브리드 복합재료 쉴드의 유한 요소 모델을 구성하였고, 직물섬유의 구속 조건을 고정과 비 구속의 두 가지로 나누어 해석을 수행하여 비교하였다. 이를 통해서 비구속 삽입된 섬유를 이용한 하이브리드 쉴드가 섬유 풀아웃 현상이 잘 구현되었고, 이로 인해 에너지 흡수율이 향상 될 수 있음을 최종 확인하였다.

Biohydrogen production from engineered microalgae Chlamydomonas reinhardtii

  • The green microalgae Chlamydomonas reinhardtti is well-known specie in the terms of $H_2$ production by photo fermentation and has been studying for a long time. Although the $H_2$ production yield is promising; there are some bottlenecks to enhance the yield and efficiency to focus on a well-designed, sustainable production and also scaling up for further studies. D1 protein of photosystem II (PSII) plays an important role in photosystem damage repair and related to $H_2$ production. Because Chlamydomonas is the model algae and the genetic basis is well-studied; metabolic engineering tools are intended to use for enhanced production. Mutations are focused on D1 protein which aims long-lasting hydrogen production by blocking the PSII repair system thus $O_2$ sensitive hydrogenases catalysis hydrogen production for a longer period of time under anaerobic and sulfur deprived conditions. Chlamydomonas CC124 as control strain and D1 mutant strains(D240, D239-40 and D240-41)are cultured photomixotrophically at $80{\mu}mol\;photons\;m^{-2}s^{-1}$, by two sides. Cells are grown in TAP medium as aerobic stage for culture growth; in logarithmic phase cells are transferred from aerobic to an anaerobic and sulfur deprived TAP- S medium and 12 mg/L initial chlorophyll content for $H_2$ production which is monitored by the water columns and later detected by Gas Chromatography. Total produced hydrogen was $82{\pm}10$, $180{\pm}20$, $196{\pm}20$, $290{\pm}30mL$ for CC124, D240, D239-40, D240-41, respectively. $H_2$ production rates for mutant strains was $1.3{\pm}0.5mL/L.h$ meanwhile CC124 showed 2-3 fold lower rate as $0.57{\pm}0.2mL/L.h$. Hydrogen production period was $5{\pm}2days$ for CC124 and mutants showed a longer production time for $9{\pm}2days$. It is seen from the results that $H_2$ productions for mutant strains have a significant effect in terms of productivity, yield and production time.

창난젓에서 분리한 Rhodococcus sp. 3T6-5Mj가 생산하는 Cholesterol Oxidase의 정제 및 특성 (Pruification and Characterization of Cholesterol Oxidase Produced by Rhodococcus sp. 3T6-5Mj isolated from Changran-jeot)

  • Rhodococcus sp. 3T6-5Mj 균주의 배양액으로부터 0.2${\mu}m$ membrane filter을 통과시키고, 아세톤 침전, DEAE-Sephadex A-50 column chromatography의 약음이온 교환수지,cholesterol affinity column chromatography를 통과시켜서 콜레스테롤 산화 효소를 25.6units/mg의 비활성도, 15%의 회수율,88배로 정제할 수 있었다. 본 균주가 생산하는 콜레스테롤산화효소의 분자량은 SDS-PAGE로 측정한 결과 약,52,000 daltons 정도로 추정되었다. 그리고 이 효소의 Km값은 콜레스테롤을 기질로 측정하였을 때 $3.2{\times}10^{-4}M$ 이었다. 본 효소의 최적온도는 $50^{\circ}C$최적 pH는 7.0으로 나타났으며, 열에 대한 안정성은 30~$45^{\circ}C$,pH에 대한 안정성은 6.5~11.0으로 나타났다. 본 효소는 cholesterol, campesterol, stigmasterol, hecogenin, $\beta$m-sistosterol의 기질에 특이성을 나타내었다. 본 효소의 아미노산 440개의 잔기로 이루어져 있으며,그 아미노산 조성은 aspararine/aspartate > glutamine/glutamate > proline > alaline > histidine 의 순으로 나타났다. 본 효소는 효소적 혈중 콜레스테롤의 측정방법에 필수적인 콜레스테롤 산화 효소의 이용 가능성을 보여 주었다.용 가능성을 보여 주었다.

표고 등록균주의 초저온 보존에 관한 연구 (Studies on Cryo-preservation of Registered Strains of Lentinula edodes)

  • 새로운 균주가 육종되었을 때 그 균주가 장기간 고유특성을 유지하기란 쉽지 않으며, 시간이 경과 할수록 균주는 퇴화를 거듭한다. 이에 따라 현재 많이 이용되고 있는 초저온에서의 균주보존 후 균사의 활력과 온도가 생존에 미치는 영향과 표고균주가 동결하는 동안 나타나는 상태변화를 이해하고자 하였다. 저온 처리별 균주보존결과 표고균주의 생존은 $-20^{\circ}C$에서 50일간 보존하면 사멸했으며, $-80^{\circ}C$$-196^{\circ}C$에서 보존한 균주들은 온도에 영향을 받지 않고 생존하였다. 저온 처리별 균사생장속도는 $-80^{\circ}C$에서 보존한 균주들의 재생속도가 가장 빨랐다. 균사의 정단부분과 말단부분에 대한 저온 처리별 균주보존 결과 부분별 위치는 균사의 활력과 생존에 영향을 끼치지 못했다. 프로그램이 가능한 동결장치를 이용하여 표고균주의 동결과정에서 나타나는 현상을 조사한 결과 균주별 각기 다른 어는점을 가지고 있었다. 또한 표고의 자실체 발생 온도형과 균주별 어는점과의 관계는 관련이 없었다. 프로그램이 가능한 동결장치를 이용하여 표고균주의 동결과정에서 나타나는 상태변화를 최소화하기 위해 일시적으로 낮은 온도로 동결을 시도하였지만 균주들은 온도에 영향을 받지 않았다.

  • In analyzing the operational characteristics of a rice whitening machine, the internal radial pressure of the machine was measured using strain gage equipment. Changes in cylinder and feed screw configurations, screen type, cylinder speed and counter-pressure levels were examined to determine their impact on the quality and quantity of milled rice and the performance of the machine. The results are summarized as follows: 1. The internal radial pressure in the whitening chamber varied with the surface condition of the grain being processed. During the first or second pass through the machine, pressure was relatively low, reached a maximum after two to three passes with combinations I and II, three to six with combination III and then began to fall. 2. The pitch of the feed screw and the size of the feed gate opening which determine the rate of entry of grain into the whitening chamber, appeared to be the most important factor aff-::cting the degree of radial pressure, quality and quantity of milled rice and the efficiency of the machine. Using a feed screw with a wide pitch (4.8cm), radial pressure was relatively high and head rice recovery ratio \vere quite low. In this case capacity and machine effic?iency were much higher than obtained when using a feed screw with a narrow pitch (2.3cm). Very significant responses in radial pressure, head rice recovery rates and machine capacity were observed with changes in cylinder speed and counter-pressure levels when using the wide pitch feed screw. 3. The characteristics of the screen which surrounds the whitening chamber had an important effect on whitening efficiency. The existence of small protuberances on the original screen resulted in significant increases in both machine capacity and efficiency but without a significant decrease in head rice recovery or development of excessive radial pressure. Further work is required to determine the effects of screen surface conditions and the shape of the cylinderical steel roller on the rate of bran removal, machine efficiency and recovery rates. The size of the slotted perforations 0:1 the screen affects total milled rice recovery. The opening size on the original screen was fabricated to accommodate the round shape of Japonica rice varieties but was not suitable for the more slender Indica type. Milling Indica varieties with this screen resulted in a reduction in total milled rice recovery. 4. An increase in cylinder speed from 380 to 820 rpm produced a positive effect on head rice recovery for all machine combinations at every level of counter-pressure used in the tests. Head rice recovery was considerably lower at 380rpm using a wide screw pitch when compared to the results obtained at speeds from 600 to 820 r.p.m. The effects of cylinder speed On radial pressure, capacity and machine efficiency showed contrasting results, depending on the width of the feed screw pitch. With a narrow feed screw pitch (2.3cm), a direct proportional relationship was observed bet?ween cylinder speed and both radial pressure and machine efficiency. In contrast, using a 4.8 centimeter pitch feed roller produced a series of inverse relationships between the above variables. Based on the results of this study it is recommended when milling Indica type long grain rice varieties that the cylinder speed of the original machine be increased from 500-600 rmp up to a minimum of 800 rpm to obtain a greater abrasive effect between the grain and the screen. The pitch of the feed screw should be also reduced to decr?ease the level of internal radial pressure and to obtain higher machine efficiency and increased quality of milled rice with increased cylinder speeds. Further study on the interaction between cylinder speed and feed screw pitch is recommended. 5. An increase in the counter pressure level produced a negative effect On the head rice recovery with an increase in radial pressure, capacity, and machine efficiency over all combinations and at every level of cylinder speed. 6. Head rice recovery rates were conditioned primarily by the pressure inside the whitening chamber. According to the empirical cha racteristics curve developed in this study, the relationships of head rice recovery ($Y_h$) and machine capacity ($Y_c$/TEX>) to internal radial pressure ($X_p$) followed an inverse quadratic function and a linear function respectively: $$Y_h^\Delta=\frac{1}{{1.4383-0.2951X_p^\ast+0.1425X_p^{\ast\ast}}^2} , (R^2=0.98)$$$$Y_c^\Delta=-305.83+374.37X_p^{\ast\ast}, (R^2=0.88)$$The correlation between capacity and power consumption per unit of brown rice expressed in the following exponential function: $$Y_c^\Delta=1.63Y_c^{-0.7786^\{\ast\ast}, (R^2=0.94)$$These relationships indicate that when radial pressure increases above a certain range (1. 6 to 2.0 kg/$cm^2$ based On the results of the experiment) head ricerecovery decrea?ses in a quadratic relation with a inear increase in capacity but without any decrease in power consump tion per unit of brown rice. On the other hand, if radial pressure is below the range shown above, power consumption increases dramatically with a lin?ear decrease in capacity but without significant increases in head rice recovery. During the operation of a given whitening machine, the optimum radial pressure range or the correct capacity range should be selected by controlling the feed rate and/or counter-pressure keeping in mind the condition of the grain, particulary the hardness. It was observed that the total number of passes is related to radial pessure level, feed rate and counter-pressure level. The higher theradial pressure the fewer num?ber of pass required but with decreased head rice recovery. In particular, when using high feed rates, the total number of passes should be increased to more than three by reducing the counter-pressure level to avoid decreaseases in head rice recovery (less than 65 percent head rice recovery on the basis of brown rice) at every cylinder speed. 7. A rapid rise in grain temperature seemed to have a close relationship with the pressure generated inside the whitening chamber and, subsequently with head rice reco?very rates. The higher the rate of increase, the lower were the resulting head rice recoveries.
