Objectives : This study was designed to evaluate the effects of Desmodii herb pharmacopuncture (DH-HA) at $KI_{10}$(Umgok) on nephritis induced by lipopolysaccharide(LPS) in rat. Methods : Rats were injected LPS to induce nephritis. DH-HA group was treated with DH-HA at $KI_{10}$ three times for a week, NP group with 26 gauge needle, saline group with normal saline. To evaluate the effects of DH-HA at $KI_{10}$ on nephritis in rats, RBC, WBC, neutrophil in blood, TNF-a, CINC-1 in serum, creatinine and total protein in urine, renal TNF-a, renal MPO were measured and renal tissue was analyzed. Results : WBC in blood, CINC-1 in serum, renal MPO significantly reduced in DH-HA group. Conclusions : DH-HA at $KI_{10}$ is effective for nephritis in LPS-induced rats. Therefore, DH-HA at $KI_{10}$ may be useful for nephritis in clinical field.
Objective&Methods: The purpose of this study is to observe the effects of HK-1000 Herbal-acupuncture solution at GB34(Yangleungchean) on hyperlipidemia and arteriosclerosis in rats. The author performed several experimental items to analyze the levels of various components and enzymes in serum, urine and liver, as well as the histological change of liver and aorta. Results: 1. HK-1000 herbal-acupuncture infusion increased the cell viability rate, DPPH radical scavenging activity and HMG-CoA reductase inhibition rate in rat liver cells. 2. The levels of total cholesterol, free cholesterol, LDL-cholesterol, phospholipid in serum and AI(atherogenic index) were decreased, and the ratio of HDL to TCL(HDL/TCL) and the level of TG in serum were increased as compared with those of the control group. 3. In the HK-1000 group, serum AST and ALT were significantly lower than those of the HG group and saline group. 4. Hepatic GSH and catalase activities were significantly increased as compared with those of the saline group. Conclusions: From the above results, it is suggested that HK-1000 herbal-acupuncture at GB34 has a therapeutic effect on hyperlipidemia and arteriosclerosis.
Benzene and toluene, which are widely used aromatic hydrocarbons in workplace, are recently proved to cause health hazards due to their toxic effects. This study investigated the influence of toluene on the urinary excretion of benzene metabolite by administering short term exposure limit(STEL) of these compounds(i.e., 13.8mg/kg of benzene and 108.8 mg/kg of toluene) intraperitoneally into Sprague-Dawley rats. After administration, urinary phenol concentration of rat was measured by gas chromatography for every three hours. Data were analyzed by non-parametric statistical methods using Kruskal-Wallis multi-sample test and Mann-Whitney U test. The following results were obtained : 1. Administration of STEL-benzene increased urinary phenol concentration in lats. 2. Urinary phenol concentration was increased logarithmically according to the dosage of benzene. 3. Excretion of phenol in urine was decreased when benzene and toluene were administered simultaneously compared with administering benzene alone. In summary, these results reveal that administration of STEL of toluene has antagonistic effect of urinary excretion of benzene metabolite in rats.
Objectives : We investigated the repeated-dose toxicity of Wenpitang-Hab-Wulingsan(WHW), a Korean traditional medicine prescribed with twelve herbs, which has been used for the treatment of renal disease. Methods : WHW extract prepared by GLP company. WHW was supplemented by gavage at 0, 100, 500 and 1000 mg/kg/day for 13-week consecutive days. We recorded the clinical signs of toxicity, body weight, organ weights, hematology, gross and histological changes in target organs rats and clinical chemistry analysis for all rats. Results : WHW extract at all doses was shown no mortality or abnormal clinical signs in rats during at the observation period. Furthermore, there was no difference in body weight and food-take consumption, organ weight, gross pathological findings, and urine analysis among the groups of rats treated with different doses of WHW extract. The hematological analysis and clinical blood chemistry data were revealed no toxic effects from WHW-treated rats. Conclusions : The results suggest that WHW extract in rats is a wide margin of safety on a acute toxicity.
The purpose of this study was to examine the impact of soy protein and soy isoflavones on bone and mineral density in young female Sprague-Dawley rats. Fifty eight rats (body Weight 75 $\pm$ 5 g) were randomly assigned to one of four groups, consuming casein, soy protein concentrate, soy protein isolate (57 mg isoflavones/100 g diet) or casein added isoflavones (57 mg isoflavones /100 g diet). All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXImus (GE Lunar Co, Wisconsin, USA) in spine and femur on 3, 6, 9 weeks after feeding. The serum and urine concentrations of Ca and P were determined. Diet did not affect weight gain and mean food intake. Food efficiency ratio was lower In soy protein groups. The serum concentration of Ca and P were not changed by soy protein and isoflavones. Urinary Ca and P excretion were not significantly different. Spine BMD was significantly increased by soy protein isolate on 3 and 6 weeks after feeding. Femur BMD was significantly increased in the groups of soy protein isolate and isoflavones adding on after 9 weeks. Therefore, soy protein with rich isoflavones may be beneficial on spine and femur BMD increasement in growing female rats.

The total contents of polyphenolics in potatoes measured by Folin Denis method were 42~76mg/100g in fresh weight. A major phenolic component contained in potato polyphenolics was identified as chlorogenic acid(3.6~15mg/100g in fresh weight). The antioxidative effects of potato polyphenolics and chlorogenic acid on the lipid peroxidation of liver microsome were studied in vivo and in vitro systems by measuring the formation of thiobarbituric acid reactive substances(TBARS) and the content of urine 8 hydroxy deoxyguanosine(OHdG). The TBARS contents of liver showed an increase in the cholesterol diet compared to those in the normal diet. This trend, however, was minimized when potato polyphenolics and chlorogenic acid were supplemented in the cholesterol diet. On the other hands, urinary 8 OHdG contents showed a marked increase with the supplementation of potato polyphenolics in the cholesterol diet. However, there was a trend of marked decrease by the supplementation of chlorogenic acid. In vitro study, potato poly phenolics and chlorogenic acid effectively inhibited the formation of TBARS in liver microsomal system in a dose dependent manners. These results suggest that potato polyphenolics exerts an antioxidative activity in cholesterol fed rat liver.

Oryeong-san which was first recorded in Shanghanrun describing the treatments of acute febrile disease is one of the frequently used oriental medicines. Oryeong-san has been prescribed for the treatment of symptoms accompanied by edema. The purpose of this study was to examine the diuretic effects of Oryeong-san by different routes of administration. Oryeong-san (100 mg/kg body weight) was administrated by three different routes in Sprague-Dawley rats: intravenous infusion, intraperitoneal injection and oral intake. Oral intake of Oryeong-san significantly increased urinary volume and excretion of $Na^+$, $Cl^-$, and $K^+$ compared to vehicle-treated control group. The effects were concentration-dependent. Intravenously administrated Oryeong-san increased urinary volume and electrolyte excretion but without significance in hydrated (0.02 ml/min/rat for 90 min) anesthetized rats. Similarly, intraperitoneally injected Oryeong-san had no effects on water and urine electrolyte excretion compared with saline control group. These findings suggest that Oryeong-san has different effects on water and electrolyte balance by routes of administration.
A procedure for the determination of KR-30075 and its metabolites in plasma and urine by high performance liquid chromatography is described. For the study of pharmacokinetic properties of KR-30075, a new PDE III inhibitor, the plasma concentration and urinary excretion after an oral administration of KR-30075 (4 mg/kg) in the male rat (Sprague Dawley) were determined by high performance liquid chromatography. The best extraction efficiency of KR-30075 and KR-30072 is obtained with ethyl ether adjusted to pH 4.0. Retention times of both KR-30072 and KR-30075 were within 5 min and resolution was complete at the flow rate of 1.0 ml/min. The sensitivity and specificity of this HPLC assay appears to be satisfactory for the pharmacokinetic study of KR-30075 and its metabolites. One-compartment open model with first-order absorption was applied to evaluate the pharmacokinetic parameters of KR-30075 according to Minimum AIC Estimation. $T_{max}$ was 1 hr, $C_{max}$ was $0.789{\pm}0.31\;{\mu}g/ml$ and elimination half $T_{1/2}$ was 6.31 min after oral administration of 4 mg/kg KR-30075 to male rats.

The influence of arginine supplements on bone mineral density and content were studied in young female Sprague-Dawley rats fed either an arginine supplemented diet or control diet. Twenty four rats (body weight $83{\pm}5g$) were randomly assigned to one of two groups, consuming casein or casein with supplemented arginine diet. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXImus (GE Lunar Co, Wisconsin, USA) in spine and femur 3, 6, and 9 weeks after feeding. The serum and urine concentrations of Ca and P were determined. Diet did not affect weight gain and mean food intake. The serum concentration of Ca and P were not changed by arginine supplementation. Urinary Ca excretion was significantly decreased by arginine supplementation. Spine BMD was significantly increased by arginine supplementation on 3 and 6 weeks after feeding. Femur BMD was significantly increased in the group of arginine supplementation on 3, 6, and 9 weeks. Rats fed the arginine-supplemented diet had better bone mineral content than did control diet rats in the experimental period. Therefore, arginine supplementation may be beneficial on spine and femur BMD increment in growing female rats. These are thought to be associated with an arginine-induced growth hormone release. The exact mechanism of this effect remains to be elucidated.
