  • These studies were carried out to investigate the effect of Onpaetang water extract on the inhibitory contractile action of acetylcholine in sensitized rat. The results of these studies were as follows; 1. Contractile force of acetylcholine from trachea smooth muscle in sensitized rat was significantly inhibited by Onpaetang. 2. Dose-response of acetylcholine pretreated Onpaetang in sensitized rat was not significantly changed. 3. Inhibitory contractile action of acetylcholine pretreated propranolol in sensitized rat was decreased. 4. Inhibitory contractile action of acetylcholine pretreated indomethacin was not significantly changed by Onpaetang. 5. Inhibitory contractile action of acetylcholine pretreated methylene blue was not significantly changed by Onpaetang. 6. Inhibitory contractile action of acetylcholine pretreated tetrodotoxin was not significantly changed by Onpaetang.

Effects of Biological Active Plants on the Isolated Rat and Guinea Pig Trachea Smooth muscle (수종(數種) 한약재(韓藥材)가 기관지평골근(氣管支平滑筋)에 미치는 영향(影響)(I))

  • Several medicinal Plants in Jeonbuk are screened for the contraction or relaxation to the acetylcholine and histamine induced contraction in the isolated rat and guinea pig's trachea smooth muscle. Contractions evoked by acetylcholine and histamine were inhibited by Flos Farfarae, Poria and Rhizoma alismatis, but contractions were increased Tuber Pinelliae, Herba Chelidonii, Fructus Qusqualis, Radix Asari, Semen Perillae, Folium Artemisiae and Fructus Schizamdrae increased the contractions evoked by acetylcholine and histamine. Radix Puerariae, Radix Osterici Koreani, Rhizoma Zingiberis siccatum, Radix Sanguisorbae, Rhizoma Atractylodis, Fructus gardeniae and Cortex Magnoliae did not effect the contractions evoked by acetylcholine and histamine.

Study on the Expectorant Activity of Ethanol Extracts from Some Crude Drugs Containing Saponins (Saponin을 함유한 생약추출물의 거담작용에 관한 연구)

  • The effect of ethanol extracts of Gilkyung(Platicodi Radix, Platicodongrandiflorum A, DC), Onji(Polygalae Radix, Polygala tennuifolia Willdenow) and Deoduk(Codonopsis lanceolate Radix, Codonopsis lanceolata) on expectorant activity of rat trachea was investigated. Following treatment of 50% ethanol extract of these medicinal plants (25 mg/rat), the content of neurotransmitters such as acetylcholine and histamine in tissue was significantly increased. The secretions of acid glycoproteins and the artificially injected phenol red were also increased. However, there was no significant difference except Onji From the histological study through periodic acid Schiff and alcian blue stain, the thickness of inner membrane of acinar glands and the stained glycoproteins on surface of epithelium and on the glands were observed in all the rats trachea treated with extract of medicinal plants. In vitro, the viscosity of mucin solution was slightly decreased with an addition of the extracts. Onji showed the most effective expectorant activity among them at the identical conditions. The mechanism of expectorant activity of these medicinal plants seems to be due to stimulation of secretion and changes of rheological properties of mucus.

An Immuno-Electron Microscopic Study of Expression of $\alpha$-tubulin on Ciliogenesis in Newborn Rat Trachea (신생 횐쥐의 기관섬모에서 $\alpha$-tubulin의 발현과 섬모형성 양상에 대한 면역전자현미경적 연구)

  • Background: This study was undertaken to detect the patterns of ciliogenesis in newborn rats trachea. Materials and Methods: The experimental animals(Sprague-Dawley strain) were divided five groups, one day, two day, three day, five day and seven day newborns as experimental groups. To obtain differential distribution of $\alpha$-tubulins in the ciliated cells and patterns of ciliogenesis, we used immunohistochemical stain method with mouse anti $\alpha$-tubulin monoclonal antibody as the primary antibody and gold particles conjugated goat anti mouse IgG as the secondary antibody. And we observed the specimens by electron microscope (Hitach-600 Model). Results : 1) The distribution of the $\alpha$ -tubulin reactions in apical zone was slightly decreased from three day after birth. 2) From 5th day after birth, the decreasing number of gold particles in intermediate zone was remarkable. 3) On the comparison with the other zones, the number of gold particles in the Golgi zone for seven days showed no statistical difference. Conclusion : The ciliogenesis of the tracheal ciliated cells in early newborn rat were made via centriolar and acentriolar pathways, in late groups, from five day after birth, the major ciliogenesis pattern might be centriolar pathway.

Mucolytic Effects of Various Parts of FRUCTUS BENINCASAE Extracts in the Rat Trachea (백동과(白冬瓜)와 청동과(靑冬瓜)의 부위별(部位別) 추출물(抽出物)에 의(依)한 거담효과(祛痰效果)의 비교연구(比較硏究))

  • The in vivo effects of Fructus Benincasae (FB-Baekdongkwa(B) and FB- on the expectoration (decrease in sputum viscoelasticity) by their sorts and using rats (Sp. D. male, $150{\sim}160g$). FB was divided by seed, flesh, and bark, extracted by 95% ethanol for 3 hr. The extracts were given to rats administration and the following results were obtained: 1. When FB Recens-C extract was administered at the concentration of 300 mg/kg b.w., mucus secretion effect in the trachea was desirably stimulated. 2. The secretion of phenol red was increased in the FB-treated tracheas in the order of Semen Benincasae(SB)-B $(153{\pm}8\;%)$, FB-B $(149{\pm}10\;%)$, and FBR-C $(117{\pm}26\;%)$. In general, the effect of FB-B extract on phenol red secretion was stronger than that of FB-C. 3. When tracheobronchial lavage fluid was analyzed, the mucus secretion was relatively high $(111{\pm}14\;%)$ in FB-B compared with other extracts. 4. Microscopic analysis after direct treatment of the FB extracts to the rat tracheal tissue showed that all the FB extracts possessed no effects for the activity of the ciliary movement. 5. Glycoprotein content secreted by the seed extract of FB-B was increased compared with the control group, which represents the highest secretion effect of mucus. From the above results. we could conclude that the seed of SB-B possesses better activity for mucus secretion from trachea than the extracts of any other parts. Therefore, it is expected that the seed of SB-B may be available for the purpose of expectorant activity in the prescription of traditional medicine.

  • Objective: Diseases of the respiratory system are one of the main causes of death and include situation such as chronic obstructive pulmonary disease, pneumonia or asthma. Medicinal plants have beneficial effects on multiple diseases include respiratory disorders like asthma and bronchitis. The aim of this study was to evaluate the effects of U. dioica and L. Album on tracheal smooth muscle contraction. Material and methods: Hydroalcoholic extracts of L. Album and U. Dioica aerial parts were prepared by maceration method and standardized based on their total phenol content. The effect of the extracts on the KCl-induced contraction of rat trachea was examined in an organ bath chamber. Data was analyzed with spss software 22. Results: The extract of L. Album (5 mg/ml), similar to theophylline (20 mM), significantly reduced the KCl-induced tracheal contraction. On the other hand, U. Dioica (1 mg/ml and 5 mg/ml) augmented the KCl-induced contraction. Conclusion: The relaxant effect of L. Album on the trachea makes it as a candidate for the managing patients with asthma and obstructive pulmonary diseases. But because of U. Dioica potential constrictive effect on the trachea it is suggested that patients avoid consuming it.

Calcium Channel Blocking and Phosphodiesterase Inhibitory Action of GS386, a Dihydroisoquinoline Derivative, in Isolated Rat Trachea (흰쥐 기관평활근에 대한 GS 386의 칼슘억제 및 포스포디에스테라제 억제 작용)

  • Recently we reported that GS 386, 1-(4'-methoxybenzyl)-6,7-dimethoxy-3,4-dihydroisoquinoline, inhibited amplitude of the $Ca^{2+}$ current by reducing the probability of $Ca^{2+}$ channel opening without changing channel kinetics in isolated rabbit atrial myocyte. In the present study, further investigation of the mechanism of action of GS 386 was performed using isolated rat trachealis. GS 386 concentration-dependently relaxed rat trachealis contracted by carbachol $(0.3{\mu}M)$ and high $K^+$(65.4 mM) with $IC_{50}$ 5.24 and 5.67 ${\mu}M$, respectively. Verapamil inhibited more effectively the high $K^+-contracted$ tissues than those with carbachol in the rat trachealis muscle. In $Ca^{2+}-free$ media, $Ca^{2+}-induced$ contraction was inhibited by GS 386. Furthermore, high concentration of GS 386 $(100{\mu}M)$ but not verapamil, attenuated a phasic contraction induced not only by carbachol but also caffeine, indicating that GS386 can enter into the cytoplasm where it may exert secondary actions on internal sites of the muscle, such as sarcoplasmic reticulum. Moreover, GS 386 showed verapamil-resistant component of relaxation and increased cAMP levels in rat trachal smooth muscle. These results suggest that the mechanism of action of GS 386 attributes to not only $Ca^{2+}$ antagonistic action but also weak phosphodiesterase inhibitory action.

The Morphological Changes of Cryopreserved Rat Trachea After Heterotopic Transplantation (쥐의 초냉동기관 이소 이식 후 형태학적 변화)

  • The best treatment of congenital or acquired tracheal stenosis is resection and end to end anastomosis. Various prosthetic material and tissue graft replacement can be considered when the stenotic segment is too long, but their uses are still limited due to many serious complications. The present study examined the effect of immunosuppression and cryopreserved allograft trachea after intraperitoneal omental implantation for evaluation of the possibility of tracheal transplantation. Thirty tracheal segments were harvested from fifteen donor Wistar rats. Among them eighteen segments were implanted immediately(group I, II, III) and twelve segments were used for cryopreservation(group IV, V). Heterotopical intraperitoneal implantation was performed in five groups of rats(n=6); Group I was Wistar syngeneic controls and received no immunosuppression. Group II and III were those of Sprague-Dawley recipients, the former receiving no immunosuppression and the latter receiving immunosuppression(Cyclosporin A 15mg/kg/day, Methylprednisolone 2mg/kg/day). Group IV and V were groups of Sprague-Dawley recipients, the former receiving immunosuppression and the latter receiving no Immunosuppression. After 28 days, rats were sacrificed and the tracheal segments were histologically evaluated. Epithelial thickness was significantly decreased in group II, IV. Epithelial regeneration score was also significantly decreased in II. All rats maintained well their round tracheal contour. In conclusion; I) trachea could be preserved for a long time with cryo method, 2) epithelium could regenerate fully with omentopexy in cryopreserved trachea, 3) immunosuppresion was not necessary with cryopreserved trachea.

Effect of Benincasa hispida Extract on Mucus Secretion in Rat Tracheobronchial Lumen (동과추출물이 흰쥐의 기관·기관지 내 점액분비에 미치는 영향)

  • Mucus in the respiratory system is a factor to cause not only respiratory diseases but also chronic diseases. In this regard, this study is to investigate whether Benincasa hispida extracts (BHE) would stimulate secreting mucus in trachea and tracheobronchial lumen of rats. As a result, effect of stimulating the mucus secretion into trachea and trachebronchial lumen of rats is significantly increased in accordance with the administration concentration of Benincasa hispida seed extracts (BHSE). The secretion amounts of phenol red (PR) and sodium fluoreseceine (NaFI) are significantly increased by 25% and 34%, respectively, when 70% EtOH extracts of Benincasae pericarpium (BPE) are administrated to rats at the concentration of 200 mg/kg. In addition, the secretion amounts of PR and NaFI are significantly increased when BuOH fraction and water fraction are intraperitoneally administrated to rats at the concentration of 200 mg/kg. It seems the result implies the physiologically active substance that stimulates the secretion of mucus is contained in Benincasa hispida. Thus, Benincasa hispida is considered to be effective for Korean medicine treatment as a medicinal plant having effect of antitussive and apophlegmatic, and it also can be useful in developing functional food and beverage.

The Effect of Cyclosporin A and Methylprednisolone on the Epithelial Regeneration of the Allograft Rat Tracheal Segments (Cyclosporin A와 methylprednisolone이 이인자형이식된 쥐기관의 상피조직재생에 미치는 영향에 대한 연구)

  • This study examined the effects of cyclosporin A [CsA] and methylprednisolone[MP] on the viability of the devascularized trachea after heterotopic transplantation. Fourty-two tracheal segments were harvested from 21 donor Wistar rats. Those tracheal segments were heterotopically implanted into the abdomen of recipient rats after wrapping in omentum. Heterotopical implantation was performed in six groups of rats:Group I was Wistar syngeneic controls, and five groups of Sprague Dawley recipients, receiving no immunosuppression[Group II], CsA alone[Group III, V], and CsA in combination with MP[Group IV, VI]. After 14 days, the tracheal segments were histologically evaluated.Epithelial thickness and the degree of epithelial regeneration were significantly different between group I and group II, III, VI, VI [p< 0.05]. There were significant differences in the epithelial thickness between group II, III, IV and group V, VI. In the degree of epithelial regeneration, there were significant differences in group II, group III-IV, and group V-VI. Without immunosuppression there was virtually no epithelium, whereas low-dose immunosuppression yielded intermediated viability, and with high dose CsA and MP we observed improved tracheal viability. Our results suggest that optimal combination of CsA and MP may improve the viability in heterotopic tracheal allografts.
