Skeletal muscle differentiation or myogenesis is important to maintain muscle mass and metabolic homeostasis. Muscle-specific microRNAs (miRNAs) are known to play a critical role in skeletal myogenic differentiation. In this study, we examined the expression profiling of miRNAs during myogenic differentiation in rat L6 myoblasts using rat miRNA microarrays. We identified the upregulated expression of miR-128 as well as several well-known myogenic miRNAs, including miR-1, miR-133b, and miR-206. We additionally confirmed the increased expression of miR-128 observed on microarray through quantitative real-time PCR (qRT-PCR), which showed similarly upregulated expression of both primary miR-128 and mature miR-128, consistent with the microarray findings. Furthermore, transfection of miR-128 into rat L6 myoblasts induced gene expression of myogenic markers such as muscle creatine kinase (MCK), myogenin, and myosin heavy chain (MHC). Protein expression of MHC was increased as well. Inhibition of miR-128 by inhibitory peptide nucleic acids (PNAs) blocked the expression of those myogenic markers. In addition, the transfection of miR-128 into rat L6 myoblasts enhanced the phosphorylation of Erk and Akt proteins stimulated by insulin, while simultaneously reversing the inhibited phosphorylation of Erk and Akt due to insulin resistance. These findings suggest that miR-128 may play important roles in myogenic differentiation and insulin signaling.
Sulfamethazine sodium was orally administrated to sprague-dawley strain male rats(body weight, 200-300g) with using sonde at the rate of 20 mg/100g body weight(recommended therapeutic dose) on once a day for 3 days. There were investigated the depletion rate of the sulfamethazine in serum, liver and skeletal muscle of rat at the time 8 hours, 1st, 2nd, 3rd, 4th, 5th and 6th day after administration sulfamethazine sodium. 1. The mean concentrations of sulfamethazine in serum according to the time lapsed after oral administration of the sulfamethazine soudijm were showed 215.53$\pm$42.99 ppm at the 8 hours after withdrawal of medicated sulfamethazine. And gradually according to the time lapsed, the concentrations of sulfamethazine residues in serum were significantly (p<.05) decreased 25.87$\pm$5.18 ppm at 1st day, 2.30$\pm$0.61 ppm at 3rd day and 0.11$\pm$0.02 ppm at 6th day after withdrawal of medicated sulfamethazine. 2. The mean concentrations of sulfamethazine in liver were significantly (p<.05) decreased 81.77 $\pm$ 12.88 ppm to 0.11$\pm$0.03 ppm between 8 hours and 6th day according to the time lapsed after oral administration of the sulfamethazine sodium for 3 days. 3. After oral administration of the sulfamethazine sodium, the mean concentrations of sulfamethazine in skeletal muscle were significantly (p<.05) decreased 35.96$\pm$1.39 ppm to 0.009$\pm$0.001 ppm between 8 hours and 6th day after withdrawal of medicated sulfamethazine. At the 4th day, the concentrations of sulfamethazine residues were showed 0.10 $\pm$ 0.04 ppm below 0.1 ppm at the permitted limit concentration of muscle in Korea. 4. After oral administration of the sulfamethazine sodium once a day for 3 days, there were showed the highest concentration in serum (215.53$\pm$42.99 ppm) than in liver(81.77$\pm$12.88 ppm) and skeletal muscle (35.96$\pm$1.39 ppm) at the 8 hours after withdrawal of medicated sulfamethazine. The mean concentration of sulfamethazine residues in serum, liver and skeletal muscle were gradually decreased according to the time lapsed.

The purpose of this study is to investigate the effects of eight weeks high fat intake and regular exercise in skeletal muscle and adipose tissue for Endoplasmic Reticulum (ER) stress in rats. This experiment involved 32 subjects (sprague-dawley rats) divided into four groups as follows: chow group (Chow, n = 8), chow and exercise group (Chow + EX, n = 8), high fat diet-induced hyperlipidemia group (HF, n = 8), and HF and exercise group (HF + EX, n = 8). As a result, there were significant decrease in body weight and abdominal fat, and blood lipid level was significantly improved by exercise for eight weeks (p < .05). There were variables changed about the skeletal muscle and ER stress in GRP78, XBP-1, ATF4, CHOP and JNK mRNA. There increased in mRNA factor by exercise, especially GRP78, and ATF4 mRNA were significantly increased in exercise (p < .05). However, there were increased in adipose tissue by exercise and there were significantly decreased in mRNA factor by high fat diet (p < .05). Consequently, this study suggests that the consistent exercise was more improved of obesity factor, such as dyslipidemia, hyperlipidemia, hyperglycemia, as well as body weight or abdominal fat. The response of ER stress in adipose tissue and skeletal muscle were more sensitive in exercise than high fat diet feed.
To evaluate the adrenergic receptors for glycogenolytic responses to catecholamine, the blood glucose level, liver glycogen content and muscle glycogen level in rats were studied with treatment of epinephrine, norepinephrine and isoproterenol. In addition, to study the possibility of interconversion of adrenergic receptors, the effects of catecholamines in feeding animal were compared with those in fasting animal. The results are summarized as follow; 1) Epinephrine and norepinephine showed dose dependent increase of blood glucose level but the effect of isproterenol was not significant. 2) The prandial states of animal did not influence on effects of catecholamines on blood glucose level. 3) Liver glycogen contents were lowered by epinephrine or by norepinephrins but effect of isoproterenol was not significant. 4) Glycogen content of skeletal muscle was significantly lowered by isoproterenol and. epinephrine shifted the dose-response curve to right, but the effect of norepinephrine was not significant. 5) The effects of epinephrine and norepinephrine on blood glucose were significantly blocked by ergotamine but not by propranolol. These results indicate that the glycogenolytic response to adrenergic agents in rat is mediated by an alpha-receptor in liver and by a beta-receptor in skeletal muscle.
The purpose of this study was to determined the effect of low-frequency electrical stimulation on the denervated gastrocnemius muscles of the albino rats, Sprague-Dawley. Fifteen Sprague-Dawley adult male albino rats were divided into non-treated (normal) group, denervated (control) group, denervated and electrical stimulated (experiments). The gastrocnemius muscles of the right leg were submaximally stimulated with 30 Hz electrical stimulation. After 4-week period, the animals were sacrificed, and muscle were removed, fixed by immersion, and processed for light and electron microscopy. The numbers of Ag-NOR increased significantly (p<0.001), but significant reductions of girth(p<0.01), wet muscle weight (p<0.001), high glycogen content fiber (p<0.01), and mitochondrial number (p<0.05) were found in denervated control group. In comparison with control group, significant increase of right leg girth (p<0.05), wet muscle weight (p<0.001), high glycogen content fiber (p<0.05), numbers of Ag-NOR(p<0.001), number of mitochondria (p<0.01), mitochondrial volume found in electrical stimulated experimental group. The results suggest that the electrical stimulation of the muscle partially prevented the denervated atrophy in the rat gastrocnemius muscles.
The effect of orally administered ginseng ethanol extract on potassium deficient and normal rat skeletal muscles was investigated in terms of Ca uptake by sarcoplastic reticulum fragments. The ginseng ethanol extract (100mg/kg/day) was administered orally to Sprague-Dawley rats for 21 days and their changes of body weights, $K^+$ content in skeletal muscles, and calcium uptake capacity of sarcoplamic reticulurn of each groups were measured. The growth rate of rats fed with the potassium deficient diet was significantly decreased compared to that of normal rats. Ginseng components did not show any effect on the decreased growth rate of the postassium deficient rats. Potassium content in skeletal muscle from potassium deficient rats was significantly reduced compared to that of normal rats. Ginseng components showed the tendency to prevent the reduction in potassium content of potassium deficient rats, but differences were not statistically significant. Calcium uptake of SR prepared from skeletal muscles of potassium deficient rats was increased significantly compared to that of normal rats. Ginseng components prevented such increase of calcium uptake by 30%. In summary, it can be concluded that ginseng may prevent the increase in Cauptake of SR obtained from potassium deficient rats.

This study conducts electrical stimulation to male white rat of Spargue-Dawley which is 7 weeks, has the weight of 240 g and is seemingly healthy for one or two weeks by means of neuromuscular electrical stimulator in order to examine the effects of neuromuscular electrical stimulation on its gastrocnemius, measures change of weight of gastrocnemius, serum and enzyme activity and then obtains the following conclusions. There is little difference in AST and CPK of weight and serum of gastrocnemius after one or two weeks of conducting neuromuscular electrical stimulation in all experimental groups. On the one hand, as a result of histochemical observation, NMES I group showed hypertrophy of perimysium and increase of sectional diameter of muscle fiber compared to comparison group, but NMES II group showed a similar result to comparison group. When ultrasubstructure was observed under electron microscope, I-type muscle fiber of NMES I group showed well-arranged mitochondria and it was similar to comparison group. II-type muscle fiber showed a large quantity of glycogen granules within sarcoplasmatic and the extension of luminal of T-tubule. I-type muscle fiber of NMES II group had small mitochondria and showed the vacuolar degeneration of mitochondria and extended T-tubule. II-type muscle fiber showed the extension of agranule cytoplasma reticulum with T-tubule and the reduction of amount of glycogen granule within partial sarcoplasmatic.
This study was carried out to determine effects of electrical stimulation on the soleus, target muscle of the sciatic newt, of white rat normal muscles. The biometric, histochemical, ultrastructural observations were made. The following results were obtained. A daily electrical stimulation of the skeletal muscle of the normally-functioning rat caused an increase of girth and weight of the muscle fibers for 2 weeks. No noticeable change was observed afterwards. More specifically, the density of volume of the red muscle fiber increased. whereas the density of the white muscle fiber decreased. The electrical stimulation group(experimental group) showed hypertrophy of the muscle fibers and narrowing of the space between perimysium and endomysium. Normally, glycogen granules are accumulated regardless of classification of muscle fibers. In addition, the NADH-TR reaction results were in agreement with the biometric findings, in that the red muscle fibers significantly increased. The ultrastructural observations revealed that mitochondria was formed in the red muscle fiber parallel to the muscle fibers of normal muscle, while mitochondria was observed in the sarcomere region of the white muscle fiber. However, activation of mitochondria took place in the sarcolemma region of the muscle fiber, and generation of mitochondria was observed in the sarcomere region of the white muscle fiber.
In this study we investigated the effects of physical therapeutic intervention through electromyography, ultrasonographic imaging and changes of the IL-$1{\beta}$, IGF-1 and IGF-2 in skeletal muscle of rats injured experimentally. The twenty Sprague-Dawley male rats were randomly divided into the 4 groups: a normal, a control, a low power laser and a neuromuscular electrical stimulation group. Abnormal spontaneous activities had not been shown, both in normal and skeletal muscle injured rats. The maximum diameter of the calf muscle was significantly increased in the low power laser and neuromuscular electrical stimulation groups compared with control group. The level of the serum IL-$1{\beta}$ was more decreased in the low power laser and neuromuscular electrical stimulation groups than that of control group. The activation level of the IGF-1 and the IGF-2 were significantly higher in the control, low power laser and neuromuscular electrical stimulation groups than that of normal group. However, there was no statistically significant difference among the control, low power laser and neuromuscular electrical stimulation groups.
The aim of this study was to identify the effect of suspension unloading (SU) and electrical stimulation upon the development of neonatal muscular system. For this study, the neonatal rats were randomly divided into three groups: a control group, an experimental group I, and an experimental group II. The SU for experimental group I and II was applied from postnatal day (PD) 5 to PD 30. The electrical stimulation for soleus muscle of experimental group IIwas applied from PD 16 to PD 30 using neuromuscular electrical stimulation (NMES), which gave isometric contraction with 10 pps for 30 minutes twice a day. In order to observe the effect of SU and ES, this study observed myocyte enhancer factor 2C (MEF2C) and vascular endothelial growth factor (VEGF) immunoreactivity in the soleus muscles at PD 15 and PD 30. In addition, the motor behavior test was performed through footprint analysis at PD 30. The following is the result. At PD 15, the soleus muscles of experimental group Iand II had significantly lower MEF2C, VEGF immunoreactivity than the control group. It proved that microgravity conditions restricted the development of the skeletal muscle cells at PD 15. At PD 30, soleus muscles of the control group and experimental group II had significantly higher MEF2C, VEGF, immunoreactivity than experimental group I. It proved that the NMES facilitated the development of the skeletal muscle cells. At PD 30, it showed that SU caused the decrease in stride length of parameter of gait analysis and an increase in toe-out angle, and that the NMES decreased these variations. These results suggest that weight bearing during neonatal developmental period is essential for muscular development. They also reveal that NMES can encourage the development of muscular systems by fully supplementing the effect of weight bearing, which is an essential factor in the neonatal developmental process.
