Studies on the Rapid Plasma Reagin(RPR) Card Test for the Diagnosis of Syphilis (매독진단(梅毒診斷)을 위(爲)한 신속(迅速)한 혈장항체검사법(血漿抗體檢査法)에 관(關)한 연구(硏究))

  • For the effective control of Syphilis, many investigators have developed a more rapid, simple and economical screening serological test which is adequately sensitive and specific. To fulfill the requirements of a more rapid serologic test for syphlis, a substitute for the conventional serum specimen was needed since considerable time and labor are involved in the processing of blood to serum. Burdon suggested the use of plasma in the serologic tests for syphilis as a substitute for serum. He noticed that plasma was more sensitive than serum in the Kline and Kahn tests, and attributed this to the presence of more antibody-like substance, "reagin" in plasma than in serum. However, to make plasma sufficiently sensitive, it was necessary to inactivate plasma by heating at a temperature of $56^{\circ}C$ for about 30 minutes. Heating of plasma resulted in the precipitation of fibrinogen which made centrifugation necessary to obtain dear plasma. Since the chief disadvantage to the use of unheated plasma(or serum) was a reduction in sensitivity of results-which probably was due to a labile factor such as complement-Portnoy et al began to consider rapid chemical methods of inactivation of plasma(or serum). They experienced that choline chloirde was shown to be anticomplementary which suggested its use as an inactivating agent for unheated plasma(or serum). In 1959 Portnoy et al reported the Rapid Plasma Reagin(RPR) Test for syphilis which is a more rapid, economical and simple. But still this test has many disadvantages as a rapid performing, field and office procedure, because it requires the usual laboratory equipments such as centrifuge, rotating machine, microscope etc. To substitute these disadvantages of the RPR test, in 1962, Portnoy et al developed the Rapid Plasma Reagin(RPR) card test for syphilis, which has the following advantages: a) Simplicity and rapidity of performance, b) Requires no laboratory equipments, c) Stable antigen suspension, d) Adequate sensitivity and specificity. This RPR card test can be used as a rapidly performing and screening test in field investigation, outpatient clinics, small laboratories and hospitals doing limited syphilis serology, and predonor in blood bank. Private clinic which has limited laboratory equipment and technic for syphilis serology can also use this RPR card test as a tool in the rapid diagnosis of syphilis. It was thought that this RPR card test is a useful tool in Korea for private physician and mass survey for syphilis diagnosis. But Portnoy patented the reagents needed for the performing the RPR card test. Therefore authors developed newly the reagents and according to Portnoy's method evaluated the newly developed. RPR card test compared with the VDRL, Kolmer CF, and RPCF tests. The RPR card and VDRL tests were performed plasma and serum from the total 1,132 cases. Among these 1,131 cases, 521 were syphilis suspected laboratory specimens, and 611 were syphilis unsuspected healthy young men. After screening with these two tests, the RPR card and VDRL tests, reactive specimens to the above one or both tests were retested by the Kolmer CF and RPCF tests.

The effecal of irradiance during leaf development on photoinhibition in Panag ginseng C. A. Meyer

  • This experiment used chlorophyll fluorescence techniques to assess the effect of irradiant during leaf development on photoinhibition of photosynthesis in Panax ginseng. Seedlings of p. ginseng were grown in the 91asshouse at four shade levels. The maximum mid-day irradiant in each treatment between emergence (January 4) and completion of the experiment (February 25) was 1220, 485, 235, 125 $\mu$mol/$\textrm{m}^2$/s. To assess the rapidity of photosynthetic readaptation to changes in light levels, fluorescence parameters (Fo, F, Fm, Fm', AF/Fm;, Fv/Fm) were measured for three days before and after transfer of plants (on February 21) from each light treatment into each of the other light treatments. Before transfer, dark adapted values of Fv/Fm in the 1220 (0.699) and 485 (0.739) treatments were different from each other and lower than values in the 235 (0.764) and 125 (0.768) treatments, indicating mild photoinhibition. Patterns of change in F during the day also differed between treatments, with low light treatments tracking irradiant levels, but F in the high light treatment (1220) declined in the morning, presumably due to fluorescence quenching. Although plants grown at high irradiant had relatively low photosynthetic efficiency, relative electron transport rate was greater than in lower irradiant treatments. After transfer, plants adopted the daily pattern of change in F of the treatment to which they were moved with little change in absolute levels of F, except in plants transferred from the highest (1220) to the lowest light level (125), where F increased over the course of the three days following transfer. After plants were transferred, Fm' converged on values similar to those in plants raised in the treatments to which they were moved. Values of Fv/Fm in plants moved from low to high light declined dramatically, but there was no decline in plants from 485 moved to 1220. Values of Pv/Fm in plants that were moved from high light to lower light increased to values above those recorded in plants raised in the lower light treatments. Reductions in quantum efficiency caused by photoinhibition at high irradiant may be more than compensated for by higher electron transport rates, although evidence suggests that under high irradiant this tends to be balanced by reduced leaf area and earlier senescence. Chlorophyll fluorescence techniques appear capable of indicating effects of irradiant induced stress in ginseng, yielding results comparable to those obtained with gas exchange techniques but in less time and with greater replication.

  • Social commerce is a new internet business model which are types of joint purchase, social link, promotion, and on-off association etc. The recent public organization has the dual target to increase sound finance, and to improve customer satisfaction and the quality of public service. The purpose of this study is to propose the strategy of public social commerce which makes it's customers become positive purchasers. We analyzed 31 public organizations, and found that the type of social link interlocking with SNS site in the various purpose of public, and the offline association type using the service of location base would be utilized very highly. We also found that the joint purchase type and the promotion type would be used in the area of public and private selective services intending to make a profit. The anticipated performance are as follows: rapidity and reliance, customer made thing and goodwill, convenience for the service environment quality, publicity and diffusion, and sales promotion, productivity increase and new finding of revenue model for the profit increase outcome.

Preparation of the Proteus vulgaris Bacterial Electrodes for the Determination of Urea and Their Application (요소 정량을 위한 Proteus vulgaris 박테리아 전극의 개발과 그 응용)

  • The bacteria containing urease convert each molecule of urea into two molecules of ammonia and one molecule of carbon dioxide gas. Bacterial electrodes have been constructed by immobilizing the Proteus vulgaris on an ammonia and a carbon dioxide gas-sensors, and were investigated for the effects of pH, temperature, buffer solution, bacterial amounts and interferences, and life time. NH3-bacterial electrode based on ammonia gas-sensor had linearity in the range of $7.0{\times}10^{-4}\;-\;3.0{\times}10^{-2}$M urea in pH 7.4, 0.05M phosphate buffer at $25^{\circ}C$ with a slope of 116.7 mV/decade. While $CO_{2-}$bacterial electrode based on carbon dioxide gas-sensor bad linearity in the range of $7.0{\times}10^{-4}\;-\;5. 0{\times}10^{-2}$M urea in pH 7.0, 0.1M phosphate buffer at $30^{\circ}C$with a slope of $45.4{\times}45.7mV/decade$. As the clinical application, urea in urine was determined by these devices and this result was compared with spectrophotometric method. Consequently, these electrodes could be used for the analysis of many samples because of simplicity, rapidity and convenience of the experimental procedure.

Simultaneous Determination of Heavy Metals in Cosmetic Products by Ion Chromatography (이온 크로마토그래피를 이용한 화장품 중 중금속 동시분석)

  • No matter how small amount of heavy metals it may be cause skin allergies through percutaneous adsorption when existing in cosmetic products as impurities. In order to develop a highly sensitive method for simultaneous determination of $Pb^{2+},\;Fe^{2+},\;Cu^{2+},\;Ni^{2+},\;Zn^{2+},\;Co^{2+},\;Cd^{2+},\;and\;Mn^{2+}$ in coloring agents and cosmetic products with rapidity and accuracy, we carried out the determination on ion chromatography. All of these metals are well separated through a bifunctional ion-exchange column(IonPac CS5A) and detected by post-column reaction and spectrophotometric detection. The calibration graphs are linear($r^2>0.999$) in the range $0.1{\sim}1000{\mu}g/mL$. Detection limits for 200 ${\mu}L$ of sample solution are at the level of ${\mu}g/L$, which is sufficient for judging whether the product is safe or not. The relative standard deviations(RSDs) of the retention time and the peak area are less than 0.21 and 1.24%, respectively. The recovery rates are $97{\sim}104%$. The new method was applied to analyze the amount of heavy metals which were contained in 22 cosmetic products and 11 coloring agents.

Quality characteristics of cut kimchi cabbage during short-term storage depending on the packaging materials (절단배추의 포장재질에 따른 단기 저장 중 품질특성)

  • Fresh cut vegetables provide convenience and rapidity to consumers. However, they have a weakness with respect to their short shelf-life due to browning and quality degradation via increased respiration. To overcome this problem, the effect of packaging film on the short-term storage of cut kimchi cabbages was investigated. Polypropylene (PP), oriented polypropylene (OPP), and low-density polyethylene (LDPE) films were used as packaging film, and cut kimchi cabbages were stored in the packaging films at $5^{\circ}C$ for 4 weeks. PP film packaging showed the least weight loss and soluble solids loss after 4 weeks. Titratable acidity increased during storage for all samples, however, the increase rate of titratable acidity in PP and OPP film packaging decreased, which was lower than that of LDPE film packaging. Color values decreased over time during storage. In appearance, PP film packaging was better than other films due to their high transparency. In a sensory test, there was no statistical difference among samples. Taken together, the transparent PP film packaging was more effective for short-term storage of cut kimchi cabbages. Thus, this study provides useful information for the selection of packaging materials for cut kimchi cabbage marketing.

Website Falsification Detection System Based on Image and Code Analysis for Enhanced Security Monitoring and Response (이미지 및 코드분석을 활용한 보안관제 지향적 웹사이트 위·변조 탐지 시스템)

  • New types of attacks that mainly compromise the public, portal and financial websites for the purpose of economic profit or national confusion are being emerged and evolved. In addition, in case of 'drive by download' attack, if a host just visits the compromised websites, then the host is infected by a malware. Website falsification detection system is one of the most powerful solutions to cope with such cyber threats that try to attack the websites. Many domestic CERTs including NCSC (National Cyber Security Center) that carry out security monitoring and response service deploy it into the target organizations. However, the existing techniques for the website falsification detection system have practical problems in that their time complexity is high and the detection accuracy is not high. In this paper, we propose website falsification detection system based on image and code analysis for improving the performance of the security monitoring and response service in CERTs. The proposed system focuses on improvement of the accuracy as well as the rapidity in detecting falsification of the target websites.

Development of antigen for the microplate latex agglutination test on toxoplasmosis in animals (Latex 응집반응을 이용한 동물의 톡소플라즈마병 진단액 개발에 관한 연구)

  • This study was conducted to develop a sensitized latex-antigen for serodiagnosis of toxoplasmosis in animals. Tachyzoites of T gondii(RH-strain) harvested from mouse peritoneal cavity were purified through the filtraton of polycarbonate membrane(pore size, $3.0{{\mu}m}$, Costar Co.) and disrupted by ultrasonicator. The tachyzoite suspension was ultracentrifuged for 30 min at $60,000{\times}g(4{^{\circ}C})$ and the supernatant was used as a water-lysate antigen. Polystyrene latex particles of $0.8{{\mu}m}$ in diameter(Sigma) were used for the preparation of sensitized latex-antigen suspension. The several parameters including the preparation conditions, incubation buffer. serum dilution buffer and stability of agglutination reactions were evaluated and the results obtained were summarized as follows : 1. The antigen consisting of a water-lysate of T gondii tachyzoites was adsorbed onto polystyrene latex particles of $0.8{{\mu}m}$ in diameter by adding a latex suspension to an equal volume of diluted antigen solution and by incubating the mixture at $37{^{\circ}C}$ under different conditions. 2. The optimum incubation buffer used for the antigen sensitization was 0.1M Tris-HCl buffer(pH 8.0). 3. The optimum serum dilution buffer used for the latex agglutination test was 0.1M Tris-HCl-NaCl buffer(pH 7.4) containing 300 mM NaCl. But 0.1M Tris-HCl-NaCl buffer(pH 7.4) containing 300-600 mM NaCl, 0.5% BSA and 0.01% Tween-20 improved the agglutination pattems and cleared the background of microplate well without the effects on L.A titer. 4. The time required for antigen sensitization was 40 and 60 min in incubation buffer(pH 8.0) at $37{^{\circ}C}$. But the optimun time for antigen sensitization was min at $37{^{\circ}C}$. 5. The optimun quantity of antigen absorbed on latex particles for proper agglutination was the range of 20 to $32{\mu}g$ of latex particles. 6. The optimun concentration of the latex-antigen suspension for the proper agglutination reaction was determined as 0.2%(w/v). 7. The specificity, rapidity and simplicity of the latex-particle agglutination test suggested that it might be adaptable to large scale serum screening.

  • The technique, which reproduces the figures of objects from measured data of the objects using 3-D laser scanner, is called reverse engineering. Recently, research studies into applications of reverse engineering to rock engineering are increasing in number, in the discontinuity surveys for rock slopes out of man's reach, or rapid discontinuity surveys for wide range areas. For analysis of discontinuity using reverse engineering, a program for processing point clouds data from the 3-D laser scanner, for sampling from these point clouds data, and finally analyzing the discontinuity is needed. However, existing programs rarely have sufficient functions to properly analyze the discontinuities. In this study, a program was developed, which can automatically sample discontinuities from the point clouds data which measured in a rock slope using a 3-D laser scanner, and which can also undertake statistical analysis of the discontinuities. This developed program was verified by the application of discontinuity surveys in a rock slope and a tunnel. By undertaking the discontinuity survey using a 3-D laser scanner and the developed program, the feasibility and rapidity of such surveys is expected to improve in areas out of man's reach in geotechnical surveys. Taking into consideration the fact that the international level of related techniques is at a rudimentary stage, the possibility of prior occupation of a broad market is also expected.

Analysis on Displacement Characteristics of Slow-Moving Landslide on a slope near road Using the Topographic Map and Airborne LiDAR (수치지형도와 항공 LiDAR를 이용한 도로인접 사면 땅밀림 발생지 변위 특성 분석)

  • The purpose of this study is to analyze the displacement characteristics in slow-moving landslide area using digital elevation model and airborne LiDAR when unpredictable disaster such as slow-moving landslide occurred. We also aimed to provide basic data for establishing a rapid, reasonable and effective restoration plan. In this study, slow-moving landslide occurrence cracks were selected through the airborne LiDAR data, and the topographic changes and the scale of occurrence were quantitatively analyzed. As a result of the analysis, the study area showed horseshoe shape similar to the general form of slow-moving landslide occurrence in Korea, and the direction of movement was in the north direction. The total area of slow-moving landslide damage was estimated to about 2.5ha, length of landsldie scrap 327.3m, average width 19.3m, and average depth 8.6m. The slow-moving landslides did not occur on a large scale but occurred on the adjacent slope where roads were located, caused damage to retaining walls and roads. The field survey of slow-moving landslides was limited by accessibility and safety issues, but there was an advantage that accurate analysis was possible through the airborne LiDAR. However, because airborne LiDAR has costly disadvantages, it has proposed a technique to mount LiDAR on UAV for rapidity, long-term monitoring. In a slow-moving landslide damage area, information such as direction of movement of cracks and change of scale should be acquired continuously to be used in restoration planning and prevention of damage.
