Chemical Genomics with Natural Products

  • Natural products are a rich source of biologically active small molecules and a fertile area for lead discovery of new drugs [10, 52]. For instance, 5% of the 1,031 new chemical entities approved as drugs by the US Food and Drug Administration (FDA) were natural products between 1981 and 2002, and another 23% were natural product-derived molecules [53]. These molecules have evolved through millions of years of natural selection to interact with biomolecules in the cells or organisms and offer unrivaled chemical and structural diversity [14, 37]. Nonetheless, a large percentage of nature remains unexplored, in particular, in the marine and microbial environments. Therefore, natural products are still major valuable sources of innovative therapeutic agents for human diseases. However, even when a natural product is found to exhibit biological activity, the cellular target and mode of action of the compound are mostly mysterious. This is also true of many natural products that are currently under clinical trials or have already been approved as clinical drugs [11]. The lack of information on a definitive cellular target for a biologically active natural product prevents the rational design and development of more potent therapeutics. Therefore, there is a great need for new techniques to expedite the rapid identification and validation of cellular targets for biologically active natural products. Chemical genomics is a new integrated research engine toward functional studies of genome and drug discovery [40, 69]. The identification and validation of cellular receptors of biologically active small molecules is one of the key goals of the discipline. This eventually facilitates subsequent rational drug design, and provides valuable information on the receptors in cellular processes. Indeed, several biologically crucial proteins have already been identified as targets for natural products using chemical genomics approach (Table 1). Herein, the representative case studies of chemical genomics using natural products derived from microbes, marine sources, and plants will be introduced.

Development for Reliability Quality and Performance Evaluate Model of Fingerprint Recognition System (지문인식시스템의 신뢰성 품질 성능 평가모델 개발)

  • Although products with the fingerprint recognition system currently show a rapid growth in quantity, it is also true that efforts to consider the product quality have been lacking until now. Accordingly, this paper analyzed technological elements with domestic and foreign market situations for products with the fingerprint recognition system to develop an evaluation model to support the quality increase by evaluating aspects of product quality for the knowledge information security, identifying the level of quality and deriving directions for improvements. A model for the reliability quality evaluation was constructed that can be applied comprehensively to non-functional elements that have not been done in the existing evaluations central to the security functions by analyzing requirements for the security, performance and reliability in consideration of features on products. It is considered that this paper can make contributions to the overall quality increase for products with the knowledge information security by reflecting features and trends for the fingerprint recognition products and building a model for the reliability and evaluation to perform evaluations by product.

  • The collaboration hub has been developed since 2004 as an online collaboration space, which supports the various collaborative works amongst small and medium enterprises using information sharing, collaboration project management, and project history management. Because of the change of manufacturing environment and rapid development of information technologies, it should be evolved from the existing version called Collaboration Hub 1.0. Recently, a lot of manufacturing enterprises know the importance of business process management(BPM) and start to introduce BPM systems. Our research group has developed the new version of Collaboration Hub 1.0 called Collaboration Hub 2.0 which contains the BPM concept, the consistent product data management, and the specialized functions overcoming the various variation of manufacturing. This study scrutinizes the meaning and role of the Collaboration Hub 2.0 and introduces an application study of it to the value chain of automobile module development consisted of a leading company and subcontractors. The case study covers the definition, execution and monitoring of collaboration process, the specialized functions overcoming the manufacturing variation and the key performance index of collaboration business.

  • According to the rapid growth of Internet service, electronic commerce usage scale is growing every year. It requires the method of vitalizations of electronic commerce through reducing the software development cost and making easy of data construction. Web service, which is adapting distributed technology based on internet supports the infrastructure of reducing the software development cost through software sharing. As the shared software modules access the same data, they can reference the same data on the other site. In this paper, we designed and implemented database Processing module of the electronic commerce program using web service technology, which can make easy of site construction and sharing database. Implemented database processing module supports data selection modification insertion deletion and searching. Using this functions we showed product item information could be shared among the other electronic commerce sit.

  • As manufacturing system have become complex and globalized, rapid development and production of products are essentially requisites for competitiveness. The importance of agility in manufacturing is being emphasized and a new paradigm is necessary for reduction of the time and expenses related to planning, product development and production. To meet such requirements, virtual manufacturing (VM) environment was suggested. In this paper, Multi-agent system is adopted into VM operating system. Because our system is flexible due to agent technology, agents can be added or deleted with ease. VM unit modules which were defined as DEVS models execute independent simulation of other modules in unit level and compose one VM system with other modules. They also execute simulation in system level. This research can contribute to usefulness of VM environment due to flexibility and extensibility of this system.

MULTI STAGE SHAPE OPTIMIZATION OF CENTRIFUGAL FAN FOR HOME APPLIANCE USING CFD (전산유체역학을 활용한 가전 제품용 원심팬 블레이드의 단계별 형상 최적화)

  • We conducted a multi-stage optimization to secure the desired performance of a centrifugal fan for home appliance in an early stage of product development. In optimization, the static pressure at the outlet of the fan is chosen as an objective function that is to be maximized, providing the required flow rate at the operating point of the fan. The optimization procedure begins with parameters for an initial baseline fan design. The baseline design is optimized by using a commercial optimization package. Accordingly, the corresponding blade models with a set of geometrical parameters are generated. Flow through a fan is simulated by solving the Reynolds-averaged Navier-Stokes equations. A multi-stage optimization scheme is employed to determine the family of optimum values for the parameters, leading to the pressure increase at the outlet of the fan. To validate the numerically obtained optimal design parameters, we fabricated the three types of fans using rapid prototyping and assessed the performance using a fan tester. Experimental results show that the design parameters at each stage satisfy the goal of optimization. The multi-stage optimization process turned out to be a useful tool in the development of a centrifugal fan.

Characteristics and Industrial Linkage of Science Parks : A Case Study of the United Kingdom (영국 과학단지의 특성과 기업연계)

  • A rapid growth of science parks worldwide since 1980s is caused by both the widespread perception that technological innovation is the essential element for the economic development and the notified cases of successful high technology cluster like Silicon Valley. Analyzed in terms of R & D employment and expenditure, new product launches, patent registration, qualifications of the founder, self-rated technological level and the like, the technological level of the science parks in the U.K. reveals very high. While nealy half of the firms on the science parks are represented to be formally linked with universities, the use of the library is referred most frequently among the types of the linkages. R & D links such as contract research, sponsored research, test and analysis were rarely mentioned. Summing up, science parks play an important role for the development of high-tech industries with superior technological characteristics. But in reatlity, there remains so much difference among the individual parks and their location. In addition the individual parks and their location. In addition, institutionalization of the technology transfer with univeristies that only when the science parks are based on the innovative environment they can be successful in the capability of the technological innovation.

  • In recent years, environments of our globe has been getting worse as a result of rapid growth of socioeconomic activities. The global environmental issues of acid rain, green house effect and ozone depletion are caused by various chemical pollutants, emitted from industries, automobiles and home. Most of these pollutants are produced by combustion processes. CO2 as a chief criminal of the greenhouse effect is a main combustion product of fossil fuels. Development of solid-state electrochemical devices for detecting CO2 is demonstrated based on various combination of solid electrolytes and auxiliary sensing materials. The object of this research is to develop various sensor performance for solid electrolyte gas sensor, and to test gas sensor performance manufactured. So we try to present a guidance for developing potential type gas sensor. We concentrated on development of manufacturing process and performance test.

  • This study was concerned about mobile web-service of a VMI in the SCM system. The VMI would be a main module part of the SCM and regarded as very important thing on supply chain. Therefore, it was essential to react in real time of materials production and situation of used auto-part in procurement company. We could find an affordable method to realize the VMI in a web service. The web service has made inter relevant enterprise easy to collaborate because of supporting most operating system and frame-work. Moreover, the use of a mobile tools was guaranteed a product recording and requirement of materials to real time in a rapid changing of production environments. The result of this research was very useful to apply the VMI system in medium and small sized auto parts company. This web service package has programmed the Visual Studio Dot Net 2003 and the MS SQL 2000 as database.

Facile and Rapid Glycosylation Monitoring of Therapeutic Antibodies Through Intact Protein Analysis

  • The therapeutic antibody drug market has experienced explosive growth as mAbs become the main therapeutic modality for a variety of diseases. Characterization of glycosylation that directly affects the efficacy and safety of therapeutic monoclonal antibodies (mAbs) is critical for therapeutics development, bioprocess system optimization, lot release, and comparability evaluation. The LC/MS approach has been widely used to structurally characterize mAbs, and recently attempts have been made to obtain comprehensive information on the primary structure and post-translational modifications (PTMs) of mAbs through intact protein analysis. In this study, we performed state-of-the-art LC/MS based intact protein analysis to readily identify and characterize glycoforms of various mAbs. Different glycoforms of mAbs produced in different expression cell lines including CHO, SP2/0 and HEK cells were monitored and compared. In addition, the comparability of protein molecular weight, glycoform pattern, and relative abundances of glycoforms between the commercialized trastuzumab biosimilar and the original product was determined in detail using the given platform. Intact mAb analysis allowed us to gain insight into the overall mAb structure, including the complexity and diversity of glycosylation. Furthermore, our analytical platform with high reproducibility is expected to be widely used for biopharmaceutical characterization required at all stages of drug development and manufacturing.
