Anthrax is an infectious disease caused by the gram-positive bacterium, Bacillus anthracis. Anthrax toxin is a tripartite toxin comprising of protective antigen (PA), lethal factor (LF) and edema factor (EF). PA is the receptor-binding component, which facilitates the entry of LF or EF onto the cytosol. LF is a zinc-dependent metalloprotease, which is a critical virulence factor in cytotoxicity of infected animals. Therefore, it is of interest to develop its potent inhibitors for the neutralization of anthrax toxin. The first step to identify the inhibitors is the development of a rapid, sensitive, and simple assay method with a high-throughput ability. Much efforts have been concentrated on the preparation of powerful assays and on the screening of inhibitors using these system. In the present study, we have tried to construct anthrax lethal factor in yeast expression system to prepare cell-based high-throughput assay system. Here, we have shown the results covering the construction of a new vector system, subcloning of LF gene, and the expression of target gene. Our results are first trial to express LF gene in eukaryote and provide the basic steps in design of cell-based assay system.

Our domestic economy has been developed very rapidly after 1960's. Also, it is dramatically increasing traffic on road and surround environmental issues. Especially, rapid economic growth has been induced large construction of pavement, and bigger and higher traffic for transportation. These are making air pollution, traffic noise and vibration. The social requirement against the revealed road environment and traffic sound reduction is being demanded. Traffic noise of city zone is showed over the environmental specification more than 57%. In order to overcome these situations, the social attention is being increased. The quiet pavement is the same format of permeable pavement, but is not same for functional performance. In this research, it has been carried out to evaluate the fundamental-mechanical properties of hot mix asphalt for quiet pavement. Especially, couple of laboratory tests are conducted like marshall stability, resilient modulus, indirect tensile test, and compaction energy analysis with gyratory compaction curve. Also, two-layer pavement system has been adopted for developing of quiet pavement. The basic performance of hot mix asphalt of quiet pavement show a satisfaction of specification of hot mix asphalt.

Due to the recent shortage of well-graded river sand resulting from a rapid growth of concrete construction, sea sand, crushed sand, and etc. are increasingly used instead. It is, however, well noted that non-washed sea sand leads to corrosion of the reinforcing steel in concrete, and thus eventually results in damage to concrete. Also, the crushed sand is not being widely used, since it is difficult to maintain the allowable amount of passing 0.08mm sieve and to adjust grading. On the other hand, because the fine sand of Nakdong-River has a poor grading but good quality as a fine aggregate for concrete, it is strongly needed to investigate the fine sand as an alternative fine aggregate. Thus, the purpose of this research is to evaluate the physical properties of the fine sand of Nakdong-River to utilize it actively as a fine aggregate. For this purpose, after the sand samples were collected according to typical location of main stream of Nakdong-River, the physical properties such as density in oven-dry condition, grading, unit volume mass, and etc. of them were estimated. It was observed from the test results that physical properties of the fine sand of Nakdong-River except grading were found to be excellent.
Purpose - The ICT market in the EU is lagging behind that of the US; however, algorithm and software development within the EU have grown steadily, and they involve focusing on the creative cultural convergence conceptualized as part of Horizon 2020 and connecting neighboring markets in the EE and the Mediterranean region. It is essential to study the requirements to market the EU's creative ICT development in emerging industrial countries after examining its applicability in these countries. Research design, data, and methodology - This study deals with data pertaining to the EU's creative industry and competitive edge. The global cultural expansion of the EU facilitates a new concept involving not only low-cost IT products to enhance local cultural artifacts through R&D and the construction of efficient infrastructure services, but also information exchange with a realistic commercialization of the technology that can be applied for creative cultural localization. In the European industry, research on algorithms has been applied for the benefit of consumers. We investigated how the process is conducted in the EU. Results - Europe needs to adjust its economic structure to the local culture as part of IT distribution convergence. The convergence has been converted into a production algorithm with IT in the form of low-cost production. This is because there is an attempt to improve the quality of transport infrastructure, workforce availability, and the distribution of the distance to the local industries and consumers, using IT algorithms. Integrated into the manufacturing industry, based on the ICT infrastructure and solutions, smart localized regional clusters are formed with the help of grafting. Europe has own strategy to increase the number of hub-and-spoke cities. Europe is now becoming integrated, with an EPC system for regional cooperation rather than national competition in ICT technology. Europe has also been recognized in this study as changing the step-by-step paradigm for global competitiveness through new creative culture industries. Conclusions - As a result, there are several ways of converging with others through EU R&D intensity; therefore, the EU can be seen as successfully increasing marginal value, which is useful in developing a special industrial cluster or local cultural cities that create converged development by connecting people and objects with IT. In fact, when compared to the US, Europe has a strong culture and the car industries have a tendency to overshadow the IT industries with integration of services in IT distribution. Considering the rapid environmental changes, the convergence of IT services is likely to take place in Europe, similar to the pharmaceutical industry and the automotive industry. This requires a focus on human resources and automated systems management. The trend is to move away from low-wage industries, switched to key personnel centers of the local university-industry. EU emphasizes the creation of IT market demand in Europe involving local cultural convergence for marketing as the second step to strengthen the economic hub-and-spoke areas.

The act of design in landscape architecture consists of a concept within a designer's mind, technical representations, and finally, a process of construction. In the 4th Industrial Revolution, the design process is facing many changes due to the rapid development of computer technology and the IT ecosystem. Computer technology was initially developed for simple functions, such as mathematical calculation and graphic representation. However, after the spread of Personal Computers, starting with IBM and Macintosh, programming languages and hardware rapidly developed, algorithms and applications became specialized, and the purpose of using computers became very diverse. This study diagnoses issues concerning the functions and roles that new design methods, such as computational design, parametric design, and algorithmic design, can play in landscape architecture based on changes in the digital society. The study focused on the design methodology using parametric technology, which has recently received the most attention. First, the basis for discussion was developed by examining the main concepts and characteristics of parametric design in modern landscape architecture. Prior research on the use of parametric design in landscape architecture was analyzed, as were the case studies conducted by landscape design firms. As a result, it was confirmed that parametric design has not been sufficiently discussed in terms of the number and diversity of studies compared to other techniques investigated by landscape design firms. Finally, based on the discussion, the study examined specific cases and future possibilities of the parametric design in landscape architecture.

The number of deaths for railway traffic accidents is mainly caused by working close to the track, or when unauthorized passage pass through the track. The safety fences are being used to ensure safety for workers close to the track, and to improve the efficiency of the work, without interfering with the passage of trains. However, a safety fence for railway tracks needs to be examined to see if it will interfere with the passage of trains. The purpose of this study is to analyze the safe distance between train and safety fence developed in Korea. In addition, the lateral load condition of wind pressure by trains is estimated and numerical analysis is carried out according to the installation intervals of railway safety fences. It has been confirmed that the proper spacing between the train and the railway safety fence should be at least 200 mm from the vehicle limit, and that the proper spacing of railway safety fence must be calculated in consideration of the wind pressure by trains.
The flood prevention capacity of drainage facilities in urban areas has weakened because of the increase in impervious surface areas downtown owing to rapid urbanization as well as localized heavy rains caused by climate change. Detention can be installed in trunk sewers and linked to existing drainage facilities for the efficient drainage of runoff in various urban areas with increasing stormwater discharge and changing runoff patterns. In this study, the concept of detention in trunk sewers, which are storage facilities linked to existing sewer pipes, was applied. By selecting a virtual watershed with a different watershed shape, the relationship between the characteristic factors of detention in the trunk sewer and the design parameters was analyzed. The effect of reducing stormwater runoff according to the installation location and capacity of the reservoir was examined. The relationship between the installation location and the capacity of the detention trunk sewer in the Dowon district of the city of Yeosu, South Korea was verified. The effects of the existing water runoff reduction facility and the detention trunk sewer were also compared and analyzed. As a result of analyzing the effects of reducing internal inundation, it was found that the inundation area decreased by approximately 66.5% depending on the installation location of the detention trunk sewer. The detention trunk sewer proposed in this paper could effectively reduce internal inundation in urban areas.

Recently, the rapid proliferation, introduction, and application of the fourth industrial revolution technology has emerged as a trend in the construction market. Building Information Model (BIM) technology is a multidimensional information system that forms the basis of the fourth industrial revolution technology. The river sector utilizing this information-based system is also being actively reviewed, for example, the current measures for maintenance. In recent years, active research and current work should be done to reflect the need for river experts to introduce BIM into the river field. In addition, the development of tools and support software for establishing various information systems is essential for the activation of facility maintenance information systems reflecting advanced technology and to establish and operate management plans. A study on the maintenance of river facilities involves using existing drawings to build a three-dimensional (3D) information model, check the damage utilizing it, and inform it, and utilize it as the data for maintenance reinforcement. This study involved determining a method to build a river facility without the existing information system and using the property maintenance information with 3D modeling to provide a more effective and highly utilized management plan to check maintenance operations and manage damages.

Rock sheds are a type of rockfall protection facility that is installed on the road near steep slopes, where large amount of rockfall is expected. Rock sheds are generally designed to resist approximately 200 kJ to 3,000 kJ of rockfall energy. In a previous study, a new type rock shed structure having a concrete-filled tube (CFT) main frame was proposed. By using CFT as the main frame in a rock shed, rapid construction is possible. Additionally, high load carrying capacity and ductility can be achieved. The behavior of the proposed rock shed structure was studied via elastic analysis with the equivalent static load of rockfall energy as in a previous study. However, it is necessary to investigate the behavior of the proposed rock shed in more detail with a full 3D finite element (FE) model considering realistic rockfall load. The FE model for the CFT rock shed main frame was developed first in this study. Then, the resistance of the CFT rock shed main frame Under ultimate rockfall energy was investigated.
The recent spread of novel and highly variant pathogenic viruses, including the coronavirus (SARS-CoV-2), has increased the demand for diagnostic testing for rapid confirmation. This has resulted in investigating the functional capability of each space, and preparing facility guidelines to secure the safety of medical technologists. During viral evaluations, there is a requirement of negative pressure facilities along with thread separation, during pre-treatment of samples and before nucleic acid amplification. Space composition therefore needs to be planned by considering unidirectional air flow. This classification of safety management facilities is designated as biosafety level 2, and personal protective equipment is placed accordingly. In case of handling dangerous materials, they need to be carried out of the biosafety cabinet, and sterilizers are required for suitable disposal of infectious agents. A common feature of domestic laboratories is maintenance of the sample pre-treatment space at a negative pressure of -2.5 Pa or less, and arranging separate pre-treatment and reagent preparation spaces during the test process. We believe that the data generated in this study is meaningful, and offers an efficient direction and detailed flow for separation of the inspection process and space functions. Moreover, this study introduces construction of the laboratory by applying the safety management standards.
