Study Analysis of Isocycloseram and Its Metabolites in Agricultural Food Commodities

  • An accurate and easy-to-use analytical method for determining isocycloseram and its metabolites (SYN549431 and SYN548569) residue is necessary in various food matrixes. Additionally, this method should satisfy domestic and international guidelines (Ministry of Food and Drug Safety and Codex Alimentarius Commission CAC/GL 40). Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS) was used to determine the isocycloseram and its metabolites residue in foods. To determine the residue and its metabolites, a sample was extracted with 20 mL of 0.1% formic acid in acetonitrile, 4 g magnesium sulfate anhydrous and 1 g sodium chloride and centrifuged (4,700 G, 10 min, 4℃). To remove the interferences and moisture, d-SPE cartridge was performed before LC-MS/MS analysis with C18 column. To verify the method, a total of five agricultural commodities (hulled rice, potato, soybean, mandarin, and red pepper) were used as a representative group. The matrix-matched calibration curves were confirmed with coefficients of determination (R2) ≥ 0.99 at a calibration range of 0.001-0.05 mg/kg. The limits of detection and quantification were 0.003 and 0.01 mg/kg, respectively. Mean average recoveries were 71.5-109.8% and precision was less than 10% for all five samples. In addition, inter-laboratory validation testing revealed that average recovery was 75.4-107.0% and the coefficient of variation (CV) was below 19.4%. The method is suitable for MFDS, CODEX, and EU guideline for residue analysis. Thus, this method can be useful for determining the residue in various food matrixes in routine analysis.

Mineralogical Studies on Luster of Seawater Cultured Pearls, Tongyeong, Korea (경남 통영 해수양식진주의 광택에 대한 광물학적 연구)

  • A mineralogical analysis on the factors affecting the luster of pearls was carried out using gravity measurement, optical microscope observation, X-ray diffraction analysis, and scanning electron microscopy. We divided the seawater cultured pearls from Tongyeong into the following four types based on luster and shape; good luster and round (LR), lackluster and round (LLR), lackluster and baroque (LLB), and lackluster and two nucleus (LTN) pearls. Pearls with high-quality luster had slightly lower specific gravity as compared to pearls with low-quality luster, but both these types of pearls are within the specific gravity range of commercial pearls. Regarding the cross-sectional thickness of the mother-of-pearl layer, LR pearls showed a uniform thickness of about 0.3 mm in average. On the other hand, LLR pearls were characterized by relatively thinner, but uniform thickness. LTN and LLB pearls showed a tendency of significantly large variation in thickness even within a single pearl. For the surface of pearls, pearls with high-quality luster showed narrower and clearer growth lines of aragonite crystals as compared to pearls with low-quality luster. Pearls with high-quality luster were characterized by fewer aragonite crystal lattice defects as compared to pearls with low-quality luster, and the former showed parallel arrangement, thinner thickness, and less difference in thickness on the surface and inside. If a pearl has a prismatic layer, it is composed of aragonite with calcite in the prismatic and nacreous layer, and calcite content is very high in the lackluster pearl. Pearls without a prismatic layer were devoid of calcite irrespective of their quality of luster, and were composed of aragonite.

Determination of the water content in citrus leaves by portable near infrared (NIR) system (근적외분광분석법을 이용한 감귤잎의 수분 측정)

  • The amount of water for the cultivation of citrus is different based on the growing period. The effect of water stress induces to enhance of sugar accumulation in citrus. The water content in the leaves of citrus can be a index for watering during cultivation. The purpose of this study is to determine the water content of citrus leaves non-destructively by using near infrared spectroscopy (NIRS). Citrus leaves were prepared from 'Okitsu' Satusuma mandarin leaves (Citrus unshiu Marc.) ranging from 20.80 to 69.98% of water content by loss on drying method, and NIR reflectance spectra of citrus leaves were acquired by using a fiber optic probe. It was found that the variation of absorbance band 1450 nm from OH vibration of water depending on the water content change. Partial least squares regression (PLSR) was applied to develop a calibration model over the spectral range 1100-1700 nm. The calibration model predicted the water content for the validation set with a standard errors of prediction (SEP) of 0.97%. In order to validate the developed calibration model, routine analyses were performed using independently prepared citrus leaves. The NIR routine analyses showed good results with those of loss on drying method with a SEP of 0.81%. The rapid and non-destructive determination of the water content in citrus leaves was successfully performed by portable NIR system.

Determination of human breast cancer cells viability by near infrared spectroscopy

  • Near infrared spectroscopy (NIRS) was employed to qualify and quantify on survival, the injury rate and apoptosis of the human breast cancer cell line MCF-7 cells. MCF-7 cells were cultured in RPMI medium supplemented with 10% FCS in a 95% air and 5% CO2 atmosphere at 37$^{\circ}C$. For the viable cells preparation, cells were de-touched by 0.1% of trypsin treatment and washed with RPMI supplemented with 10% FCS medium by centrifugation at 1000 rpm for 3min. For the dead cells preparation, cells were de-touched by a cell scraper. The cells were counted by a hemacytometer, and the viability was estimated by the exclusion method with frypan blue dye. Each viable and dead cells were suspended in PBS (phosphate bufferred saline) or milk at the cell density desired. For the quantitative determination of cell death by measuring the LDH (lactate dehydrogenase) activity liberated from cells with cell membrane injuries, LDH-Cytotoxic Test Wako (Wako, Pure Pharmaceutical Co. Ltd., Japan) was used. We found that NIRS measurement of MCF-7 cells at the density range could evaluate and monitor the different characteristics of living cells and dead cells. The spectral analysis was performed in two wavelength ranges and with 1,4, 10 mm pathlength. Different spectral data pretreatment and chemometrics methods were used. We applied SIMCA classificator on spectral data of living and dead cells and obtained good accuracy when identifying each class. Bigger variation in the spectra of living cells with different concentrations was observed when compared to the same concentrations of dead cells. PLS was used to measure the number of cells in PBS. The best model for measurement of dead cells, as well as living cells, was developed when raw spectra in the 600-1098 nm region and 4 mm pathlength were used. Smoothing and second derivative spectral data pretreatment gave worst results. The analysis of PLS loading explained this result with the scatter effect found in the raw spectra and increased with the number of cells. Calibration for cell count in the 1100-2500 nm region showed to be very inaccurate.

Determination of Terazocin in Human Plasma by Liquid Chromatography and Bioequivalence Study of Teratonin® Tablets (액체크로마토그래프법에 의한 사람 혈장 중 테라조신의 정량 및 테라토닌® 정의 생물학적 동등성)

  • A rapid, selective and reproducible high-performance liquid chromatographic method has been developed for the determination of terazocin in human plasma. Terazocin plus the internal standard, prazocin hydrochloride, were extracted from alkalified plasma with tert-butylmethyl ether, back-extracted into 0.05% phosphoric acid. Fifty ${\mu}l-portions$ of extract were injected onto a octadecylsilane column and eluted with a mixture of acetonitrile, water and triethylamine (30 : 70 : 0.1 v/v, adjusted to pH 5.0 with dilute phosphoric acid) at a flow rate of 1.0 ml/min. The fluorescence intensity of column eluents was monitored at excitation wavelength of 250 nm and emission wavelength of 370 nm. No interference peaks were observed. The practical limit of quantitation was 5 ng/ml for terazocin. The average intraday and interday coefficients of variation were 4.15 and 3.54%, respectively. Also intraday and interday precisions over the range $5{\sim}60\;ng/ml$ were $0.49{\sim}2.92\;and\;0.38{\sim}5.12%$, respectively. The bioequivalence of two terazosin tablets, the $Hytrine^{\circledR}$ (Il Yang Pharmaceutical Co., Ltd.) and the $Teratonin^{\circledR}$ (Sam-A Pharmaceutical Co., Ltd.), was evaluated according to the guideline of Korea Food and Drug Administration (KFDA). Sixteen healthy male volunteers $(24.6{\pm}2.0\;years\;old)$ were divided into two groups and a randomized $2{\times}2$ cross-over study was employed. After one tablet containing 2 mg of terazosin was orally administered, blood was taken at predetermined time intervals and the concentration of terazosin in plasma was determined with a HPLC method using spectrofluorometric detector. AUC was calculated by the linear trapezoidal method. $C_{max}\;and\;T_{max}$ were compiled from the plasma drug concentration-time data. Analysis of variance (ANOVA) was utilized for the statistical analysis of the parameters. The results showed that the differences in $AUC_t,\;C_{max}\;and\;T_{max}$ between the two preparations were 0.21 %, 5.53% and 8.82%, respectively. The powers $(1-{\beta})\;for\;AUC_t,\;C_{max}\;and\;T_{max}$ were >99%, 97.49%, and 33.26%, respectively. Minimum detectable differences $({\Delta},\;%)\;at\;{\alpha}=0.1\;and\;1-{\beta}=0.8$ and the 90% confidence intervals were all less than ${\pm}20%$ except for $T_{max}.\;AUC_t\;and\;C_{max}$ met the criteria of KDFA for bioequivalence, indicating that $Teratonin^{circledR}$ tablets are bioequivalent to $Hytrine^{circledR}$ tablets.

Analysis of Voice and Swallowing Symptoms after Thyroidectomy in Patients without Recurrent Laryngeal Nerve Injury in Early Postoperative Period (반회후두신경 손상을 동반하지 않은 갑상선 절제술 환자에서 수술 초기의 음성 및 연하 기능의 변화에 대한 분석)

  • Background and Objectives : After thyroidectomy, many patients experience problems report such things as reduced voice range and vocal fatigue, swallowing problems without superior and recurrent laryngeal nerve injury. The purpose of this study was to evaluate voice and swallowing problems before and after thyroid surgery without laryngeal nerve injury. Materials and Methods : Ninety-three patients who underwent thyroidectomy without laryngeal nerve injury and completed the follow-up evaluations were studied between June 2013 and December 2015. Each evaluation was performed preoperatively, as well as 1 week, 1 month postoperatively. Analysis was performed including voice handicap index (VHI), dysphagia handicap index (DHI), and acoustic voice analysis. Results : Patients show significant variation of parameters in the fundamental frequency (F), maximal phonation time (MPT), shimmer, jitter and soft phonation index (SPI) early after operation, and most of them showed recovery of parameters after 1month of operation. Perceptive complaint of voice and swallowing also showed significant decreased after operation (p<0.005). After 1month of operation, MPT, highest frequency and frequency ranges still showed significant decreased parameters. Comparing acoustic and perceptive parameters of total thyroidectomy and lobectomy, there was no significant changes between them except highest frequency (p=0.042). Conclusion : The results from both subjective and objective evaluations show voice and swallowing disturbance after thyroidectomy even in the absence of laryngeal nerve and provide patients information about the recovery process after surgery. Highest frequency parameter showed most significant changes after operation.

  • Cytogenetic and hematological analysis was performed in peripheral blood obtained from pigs bred in the high background radiation areas (HBRA) (Cheongwon-gun and Boeun-gun) and a control area. The frequencies of gamma-ray induced micronuclei (MN) in the cytokinesis-blocked (CB) lymphocytes at several doses were measured in three pigs. An estimated dose of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the swine lymphocytes exposed in vitro to radiation over the range from 0 mGy to 1,969 mGy. The measurements performed after irradiation showed dose-related increases in the MN frequency in each donors. The results were analyzed using a linear-quadratic model with a line of best fit of $y=0.0005404D^2+0.04237D+0.00833$ [y = number of MN/cytokinesis-blocked (CB) cells and D = irradiation dose in Gy]. MN rates per 1,000 CB lymphocytes of pig from the HBRA (Cheongwon-gun, Boeun-gun) and the control area were $6.70{\pm}2.36$, $9.00{\pm}3.50$ and $11.00{\pm}2.98$, respectively. The MN frequencies of CB lymphocytes from pigs bred in three areas means that the values are within the background variation in this experiment. The MN frequencies and hematological values were similar regardless of whether the pigs were bred in the HBRA or the control area.

The Flow Behavior of Skin Collagen (피부조직 콜라겐의 유동 특성)

  • To obtain the basic information for the effective use of collagen, the flow behavior of collagen extracted from skin tissue was studied. The viscosity of collagen varied with sex, age and the kinds of collagen by extraction method. Regardless of the kinds of collagen, the viscosity of collagen extracted from $6{\sim}12$ week old rat was relatively high. In case of the same age, the viscosity showed higher in female than in male rat and in acid soluble collagen than in insoluble collagen. The solution of the collagen showed the characteristics of Bingham plastic and thixotropic fluid, and the viscosity varied distinctly with temperature, pH, ethanol concentration and collagen concentration. As collagen concentration increased to 6%, the consistency of acid soluble- and insoluble collagen showed a tendency to increase linearly(r = 0.972 for acid soluble collagen, r = 0.957 for insoluble collagen). In that range of collagen concentration, the increasing velocity of consistency was higher in acid soluble collagen than in insoluble collagen. The consistency of collagen solution was decreased according to temperature rising. In case of acid soluble collagen, the consistency is decreased abruptly between $30{\sim}40^{\circ}C$. According to pH variation, the consistency of acid soluble collagen showed biphasic phenomenon, though the consistency of insoluble collagen was found not to be influenced by pH. The consistency of acid soluble- and insoluble collagen according to ethanol concentration showed high between $40{\sim}60%$ of ethanol concentration.

Three-dimensional Simulation of Wave Reflection and Pressure Acting on Circular Perforated Caisson Breakwater by OLAFOAM (OLAFOAM에 기초한 원형유공케이슨 방파제의 반사율 및 작용파압에 관한 3차원시뮬레이션)

  • In this study, we proposed a new-type of circular perforated caisson breakwater consisting of a bundle of latticed blocks that can be applied to a small port such as a fishing port, and numerically investigated the hydraulic characteristics of the breakwater. The numerical method used in this study is OLAFOAM which newly added wave generation module, porous media analysis module and reflected wave control module based on OpenFOAM that is open source CFD software published under the GPL license. To investigate the applicability of OLAFOAM, the variations of wave pressure acting on the three-dimensional slit caisson were compared to the previous experimental results under the regular wave conditions, and then the performance for irregular waves was examined from the reproducibility of the target irregular waves and frequency spectrum analysis. As a result, a series of numerical simulations for the new-type of circular perforated caisson breakwaters, which is similar to slit caisson breakwater, was carried out under the irregular wave actions. The hydraulic characteristics of the breakwater such as wave overtopping, reflection, and wave pressure distribution were carefully investigated respect to the significant wave height and period, the wave chamber width, and the interconnectivity between them. The numerical results revealed that the wave pressure acting on the new-type of circular perforated caisson breakwaters was considerably smaller than the result of the impermeable vertical wall computed by the Goda equation. Also, the reflection of the new-type caisson breakwater was similar to the variation range of the reflection coefficient of the existing slit caisson breakwater.

An Analysis of Near-infrared Light Curves of δ Scuti Variable BO Lyn (δ Scuti형 변광성 BO Lyn의 근적외선 광도곡선 분석)

  • In order to investigate the light curve difference in visual and infrared wavelength of ${\delta}$ Scuti variable Bo Lyn, observations were performed using BOAO 1.8m reflecting telescope and an infrared detector, KASINICS, with J, H, and Ks filters. Infrared light curves of total 7 nights were obtained between March and April in 2011, and those were compared to the V-filter light curve to examine the differences in period, time of maximum light, amplitude, and shape. From the periodic analysis of infrared light curve, a single frequency of $f_1=10.712cycle/day$, $P=0.09335{\pm}0.00002days$ was obtained, and there was no difference in the period along different wavelengths. In the infrared light curve, a frequency of $2f_1$ was detected. This frequency well explains the asymmetric shape of light curve, one of the characteristics of high-amplitude ${\delta}$ Scuti variables. We compared the locations of the measured infrared maxima and the predicted maxima of V-filter, finding that the times of maxima were delayed about 0.3 phase at infrared wavelengths. Amplitude ratios were adopted to be ${\Delta}J/{\Delta}V=0.328$, ${\Delta}H/{\Delta}V=0.216$, and ${\Delta}Ks/{\Delta}V=0.211$, with the range of variation being smaller at longer wavelengths. It seems that the differences in the times of maxima and amplitude occurred because the changes in brightness of a pulsating variable star are mainly caused by the change in temperature.
