one key goal of software testing is to generate a 'good' test data set, which is consideres as the most difficult and time-consuming task. This paper discusses how genetic algorithns can be used for automatic generation of test data set for software testing. We employ mutation testing to show the effectiveness of genetic algorithms (GAs) in automatic test data generation. The approach presented in this paper is different from other in that test generation process requireas no lnowledge of implementation details of a program under test. In addition, we have conducted some experiments and compared our approach with random testing which is also regarded as a black-box test generation technique to show its effectiveness.
Twenty-eight clinical isolates of Escherichia coil, composed of thirteen norfloxacin resistant isolates (MIC of >16${\mu}$g/ml), one intermediately resistant isolate (MIC of 8${\mu}$g/ml), and fourteen susceptible isolates (MIC of <4${\mu}$g/ml), were randomly selected to study the norfloxacin resistance mechanism and phylogeny in clinical isolates in Korea. Eleven nofloxacin resistant isolates and one susceptible isolate were multi-drug resistant (MDR). Every norfloxacin resistant isolate with MIC higher than 32${\mu}$g/ml had the same three mutations: Ser83\longrightarrowLeu and Asp87\longrightarrowAsn or Tyr in GyrA and Ser80\longrightarrowIle in ParC. Whereas a resistant isolate with MIC of 16${\mu}$g/ml had three mutations but Asp87 in GyrA was replaced with Gly instead of Asn. The intermediately resistant isolate had the same two mutations in GyrA but a different mutation in ParC, Glu84\longrightarrowLys. Among the susceptible isolates, two isolates with MIC of 4${\mu}$g/ml had one mutation: Ser83\longrightarrowiLeu in GyrA, and no mutation was found in the susceptible isolates. Resistant isolates showed higher efflux activity than the susceptible ones, with random amplification of polymorphic DNA (RAPD), six susceptible isolates form a separate group from the rest of the isolates.

GAs, effective stochastic search algorithms based on the model of natural evolution and genetics, have been successfully applied to various optimization problems. When population size is not large, GAs often suffer from the phenomenon of premature convergence in which all chromosomes in the population lose the diversity of genes before they find the optimal solution. In this paper, we propose that a new heuristic that maintains the diversity of genes by adding some chromosomes with random mutation and selective mutation into population during evolution. And population size changes dynamically with supplement of new chromosomes. Experimental results for several test functions show that when population size is rather small and the length of chromosome is not long, this method is effective.
This article presents an adaptive directional differential evolution (ADDE) algorithm and its application in solving discrete sizing truss optimization problems. The algorithm is featured by a new self-adaptation approach and a simple directional strategy. In the adaptation approach, the mutation operator is adjusted in accordance with the change of population diversity, which can well balance between global exploration and local exploitation as well as locate the promising solutions. The directional strategy is based on the order relation between two difference solutions chosen for mutation and can bias the search direction for increasing the possibility of finding improved solutions. In addition, a new scaling factor is introduced as a vector of uniform random variables to maintain the diversity without crossover operation. Numerical results show that the optimal solutions of ADDE are as good as or better than those from some modern metaheuristics in the literature, while ADDE often uses fewer structural analyses.
The MTF1 gene of Saccharomyces cerevisiae encodes a 43 kDa MITOCHONDRIAL RNA polymerase specificity factor which recognizes mitochondrial promoters to initiate correct transcription. To better understand structure-function of the MTF1 gene as well as the transcription mechanism of mitochondrial RNA polymerase, two cold-sensitive alleles of the MTF1 mutation were isolated by plasmid shuffling method after PCR-based random mutagenesis of the MTF1 gene. The mutation sites were analyzed by nucleotide sequencing. These cs phenotype mtf1 mutants were respiration competent on the nonfermentible glycerol medium at the permissive temperature, but incompetent at 13.deg.C. The cs phenotype allele of the MTF1, yJH147, encoded an L146P replacement. The other cs allele, yJH148, contained K179E and K214M double replacements. Mutations in both alleles were in a region of Mtflp which is located between domains with amino acid sequence similarities to conserved regions 2 and 3 of bacterial s factors.
This paper proposes a DNA watermarking method for the privacy protection and the prevention of illegal copy. The proposed method allocates codons to random circular angles by using random mapping table and selects triplet codons for embedding target with the help of the Lipschitz regularity value of local modulus maxima of codon circular angles. Then the watermark is embedded into circular angles of triplet codons without changing the codes of amino acids in a DNA. The length and location of target triplet codons depend on the random mapping table for 64 codons that includes start and stop codons. This table is used as the watermark key and can be applied on any codon sequence regardless of the length of sequence. If this table is unknown, it is very difficult to detect the length and location of them for extracting the watermark. We evaluated our method and DNA-crypt watermarking of Heider method on the condition of similar capacity. From evaluation results, we verified that our method has lower base changing rate than DNA-crypt and has lower bit error rate on point mutation and insertions/deletions than DNA-crypt. Furthermore, we verified that the entropy of random mapping table and the locaton of triplet codons is high, meaning that the watermark security has high level.
Escherichia coli harboring pAC-LYCO4 and pDdxs was used for lycopene production. Three wild type strains of E. coli OW1, MG1655, and W3110 were compared with DH5${\alpha}$ used before for lycopene production. Lycopene productivity of E. coli MG1655 was similar to DH5${\alpha}$ and the highest among those wild type strain. Therefore, MG1655 strain was used for random transposon and NTG mutagenesis to increase lycopene productivity. Through transposon mutation, five transposon mutants with increased lycopene productivity were obtained. It was found that genes knocked out by transposon insertion were treB in Tn1 mutant, B2436 in Tn2 mutant, and rfaH in Tn3, 4, and 5 mutants. Lycopene productivity was the highest in Tn4 mutant among the Tn mutants, which was 6-fold and 8-fold higher in lycopene concentration and content, respectively, in comparison with those obtained with wild type strain. NTG4 mutant was acquired with NTG mutation. The highest lycopene productivity of 6 mg/L and 4 mg/g DCW was obtained from the NTG4 mutant when arabinose of 0.013 mM was added for induction of dxs, rate-limiting gene of MEP pathway. The lycopene productivity of NTG4 mutant was increased 18-fold and 12-fold in lycopene concentration and content, respectively when comparing with the wild type strain.
In multi-allelic model $X\;=\;(x_1,\;x_2,\;\cdots\;,\;x_d),\;M_f(t)\;=\;f(p(t))\;-\;{\int_0}^t\;Lf(p(t))ds$ is a P-martingale for diffusion operator L under the certain conditions. In this note, we examine the stochastic differential equation for model X and find the properties using stochastic differential equation.

Evolutionary Algorithm is the general method that can search the optimum value for the various problems. Evolutionary method consists of random selection, crossover, mutation, etc. Since the next generation is selected based on the fitness values, the crossover between chromosomes does not have any restrictions. Not only normal marriage but also consanguineous marriage will take place. In human world, consanguineous marriage was reported to cause various genetic defects, such as poor immunity about new diseases and new environment disaster, These problems translate into searching for the local optimum, not the global optimum. So, a new evolutionary algorithm is needed that prevents traps to...
