Ultrastructural studies on the Effect of Aqua-acupuncture to Liver Damage induced by Radia Aconiti (약침(藥鍼)이 부자(附子)로 유발(誘發)된 간손상(肝損傷)에 미치는 영향(影響)에 관한 전자현미경적(電子顯微鏡的) 연구(硏究))

  • In order to investigate experimentally that effect of Moschus, Bovis Calculus, Ursi Fel aqua-acupuncture on acutely damaged liver of rats induced by radix aconiti, the author gave Moschus, Bovis Calculus, Ursi Fel aqua-acupuncture according to method of manufacture stimulation to corresponding points, Kan-su (B18) and Ki-mun (Liv14), and carried out hematological, serological examination and electromicroscopical observation. The following results were obstained : 1. At 6 hours, cell organelles induced by radix aconiti toxicity prominently destructed in control and aqua-acupuncture treatment groups. 2. At 12 hours, cell organelles, destruction was some similarities between control group and aqua-acupuncture treatment group. 3. At 24 hours, rough endoplasmic reticulum and Golgi complex comparatively developed in aqua-acupuncture group treatment as compared with control group. 4. At 48 hours, cell organelles, destruction was recovered in aqua-acupuncture treatment group as compared with the control group and it was similiar normal findings. According to the above findings, it is considered that Moschus, Bovis Calculus, Ursi Fel aqua-acupuncture has effects of recovery of acutely damaged liver.

Literature Review on Safety of Pharmacopuncture according to Sasang Constitutional Medicine (동의수세보원(東醫壽世保元) 신정(新定) 처방(處方) 약재(藥材)의 약침 안전성 문헌 조사)

  • 1. Objectives: Pharmacopuncture according to Sasang Constitutional Medicine is new field of constitutional medicine. We investigated safety of pharmacopunctures to develop this treatment. 2. Methods: We searched for papers in fifteen Korean oriental medical journals and four online searching portal articles. Hepatotoxicity, nephrotoxicity and general toxicity tests were investigated about one hundred thirty kinds of pharmacopunctures. 3. Results: Toxicity test about 36 kinds of 130 pharmacopunctures were investigated. 1) Hepatotoxicity and nephrotoxicity test of pharmacopunctures were searched in 11 of 38 Soeumin herbs, 8 of 36 Soyangin herbs, 5 of 44 Taeeumin herbs and 1 of 12 Taeyangin herbs. From among these herbs, toxicity was detected in Radix aconiti lateralis preparata(附子), Fructus alpiniae oxyphyllae(益智仁), Semen crotonis(巴豆), Radix bupleuri(柴胡) and Semen armeniacae amarum(杏仁) pharmacopunctures. 2) General toxicity test of pharmacopunctures were searched in 8 of 38 Soeumin herbs, 9 of 36 Soyangin herbs, 6 of 44 Taeeumin herbs and 2 of 12 Taeyangin herbs. from among thes herbs, toxicity was detected in Radix aconiti lateralis preparata(附子), Radix ginseng(人蔘), Semen crotonis(巴豆), Rhizoma seu radix notopterygii(羌活), Radix aralia cordatae(獨活), Semen armeniacae amarum(杏仁) and Radix scutellariae(黃芩) and Cortex acanthopanacis(五加皮) pharmacopunctures. 4. Conclusions: From these results, we must decide extraction method, concentration and dose carefully to use the toxic pharmacopuncture. More toxicity test and further study is needed about pharmacopuncture treatment in Sasang Constitutional Medicine.

Recent Research Trends of Sayeok-tang Used in Ischemic Heart Disease (허혈성 심질환에 활용된 사역탕(四逆湯)의 최신 연구 동향)

  • Purpose : Sayeok-tang (Sini decoction, SND) is a cold-dispelling formula used for cold deficiency syndrome and is composed of Aconiti Lateralis Radix Preperata, Zingiberis Rhizoma, and Glycyrrhizae Radix et Rhizoma. It is used for diseases such as myocardial infarction, heart failure, acute and chronic gastroenteritis, and gastric effusion. This study proposes the possibility of expansion of basic research and clinical applications for ischemic heart disease (IHD) through systematic analysis of domestic and foreign studies on SND. Methods : We collected studies within the last 10 years on the use of SND in IHD and excluded those lacking relevance. Selected studies were classified by research method and the main themes of the studies were analyzed for each classification. Result: Out of 15 studies, there were 5 animal studies, 8 metabolite analyses in animals, 1 in vitro study, and 1 systematic review. Our review suggests that SND may be used as an adjuvant to nitroglycerin and percutaneous transluminal coronary angioplasty, and may improve symptoms and quality of life of patients with IHD. Myocardial protective effects through antioxidant, anti-inflammatory, anti-apoptotic, and anti-hypertensive actions were confirmed through these studies. Effects on carbohydrate, protein, and lipid metabolism were also reported. Conclusions : This study suggests that SND has potential as a treatment for IHD.

Effect of Radix Aconiti Extract on Cell Cycle Progression in HepG2 Human Hepatoma (HepG2 간암세포주기에 대한 부자 추출물의 효과)

  • This study was to investigate the cell cycle arrest effect and its mechanism of Radix Aconiti (RA) extract in HepG2 human hepatoma cells. We used the Flow Cytometer to investigate the effects on cell cycle arrest in RA extract-treated HepG2 cells. And protein levels involved in cell cycle progression such as p53, p21, and p21 are detected by Western blotting method. RA extract induced cell cycle arrest as confirmed by increase of G0/G1 cell population, and the mechanisms were related with up-regulation of p53, p21, p27 protein expressin in HepG2 cells. These results suggest that RA may be a valuable agent for the therapeutic intervention of human hepatomas.

Effect of Aconiti Radix on the Toxicity Induced by Glucose Oxidase in Cultured Myocardial Cells (배양심근세포에 있어서 Glucose Oxidase의 독성에 대한 천오두의 영향)

  • Cytotoxicity of glucose oxidase(GO) and neuroprotective effect of Aconiti Radix(AKR) against GO-induced neurotoxicity were measured for elucidating the mechanism of cardiotoxicity on cultured mouse myocardial cells by MTT assay after myocardial cells were cultured for 24 hours at various concentrations of GO. GO was toxic in a time-and dose-dependent manner on cultured myocardial cells after myocardial cells were grown for 24 hours in media containing 5~40mU/ml GO. While, cultures were pretreated with 50 μg/ml AKR for 2 hours increased remarkably cell viability. From the above results, it is suggested that GO is toxic on cultured mouse myocardial cells by the decrease of cell viability, and herb medicine such as AKR is very effective in the prevention of myocardial toxicity induced by GO.

Comparison of Immunomodualtory Effects of Water-extracted Aconiti lateralis Preparata Radix, Zingiberis Rhizoma, Cinnamomi Cortex and Evodiae Fructus (온리약인 부자, 건강, 육계, 오수유의 면역조절효과 비교)

  • This study was carried out to investigate the comparison of immunomodualtory effects of water-extracted Aconiti lateralis Preparata Radix(PR), Zingiberis Rhizoma(ZR), Cinnamomi Cortex(CC) and Evodiae Fructus(EF). The parameter examined to assess apparent immunomodulatory effect of the water-extracted PR, ZR, CC and EF included the regulation of Nitric oxide (NO). Also, ZR and EF represent the expression of Th1/Th2 type cytokine, the change of B cell phenotype. The water-extracted PR, ZR, CC and EF inhibited NO production and iNOS protein expression in LPS stimulated RAW 264.7 macrophage cells. In the Th1 and Th2 cytokine expression, the water-extracted ZR and EF induced IL-2, IFNr and IL-10 mRNA gene expression. Therefore, it seems that the water-extracted ZR and EF have a inducing effect of Th1 and Th2 type cytokines. In the Flow cytometry analysis, the water-extracted ZR and EF changed B cell phenotype (CD45R/B220), did NOT in PR and CC. The water-extracted PR, ZR, CC and EF have a reducing effect of immune suppression cause by Methotrexate (MTX), an agent of immune suppression. These results suggest that the immunomodulatory effects of the water-extracted ZR and EF may be, in part, associated with the inducing IL-2 and IFNr mRNA gene expression In and regulation of NO production in macrophage cells.

Apoptosis-inducing Effects of Radix Aconiti Extract in HL-60 Cells (혈액암 세포에서 부자(附子) 추출물의 Apoptosis 유도 효과)

  • The aim of this study was to investigate the apoptotic effect and its mechanism on Radix Aconiti (RA) extract in HL-60 human leukemia cell line. RA extract induced apoptosis as confirmed by discontinuous fragmentation of DNA. To clarify the mechanisms on RA extract-induced apoptosis, we examined the caspase-3, -8 enzyme activity and protein levels including Fas, FasL in HL-60 cells. Treatment with RA extracts resulted in the increase of caspase-3 enzyme activity in a time and dose-dependent manners, which was accompanied by the cleavage of poly-(ADP-ribose) polymerase (PARP). This activation of caspase-3 enzyme resulted from cleavage of procaspase-8, which was followed by increases of FasL, Fas protein expression in RA extracts-treated HL-60 cells. In conclusion, RA extract induced apoptosis of HL-60 human leukemia cell line. This results suggest that the apoptotic mechanisms of RA extract on HL-60 cells involved in FasL, Fas activation, procaspase-8 cleavage, activation of caspase-3 and cleavage of PARP. Collectively, these results suggest that RA may be a valuable agent as a anti-cancer drug.

Cytotoxic Effects of Radix Aconiti Extract in Lung Cancer Cell Lines (폐암세포에 대한 부자(附子) 추출물의 독성 효과)

  • The aim of this study was to investigate the cytotoxic effect and its mechanism on Radix Aconiti(RA) extract in lung cancer cell lines. RA extract treatment decreased the cell viability in a dose-dependent fashions in lung cancer cells including A549, H460, H23 and H157 cells. Many investigators reported that A549 and H460 cells expressed wild-type p53, but H23 and H157 cells preserved mutated p53. After treatment with RA extract in A549 and H460 cells, we measured the expression of p53 protein levels using Western blot. analysis. In both cells treated with RA extracts, p53 protein expressions were increased in a dose-dependent manner. In our experiments, RA extracts also have cytotoxic effects in H23 and H157, which have mutated p53. Treatment with RA extract decreased bcl-2 protein expressions in both cells. These results suggest that RA extracts have cytotoxic effects via p53 expression increase and bcl-2 inhibitable pathways in A549, H460 cells and H23, H157 cells, respectively.

Effects study of Aconiti Lateralis Radix Preparata extract on the regulation of heat and cold in PTU-induced hypothyroidism rats (포부자추출물의 갑상선기능저하증 흰쥐모델에서의 한열조절작용에 의한 개선효능 연구)

  • Objective : To suggest a scientific evidence of Aconitum carmichaeli Debx. (Aconiti Lateralis Radix Preparata: ALRP) as one of cooling and heating medicines on the regulation of body temperature, we investigated the effects of ALRP water extract on hypothyroidism. Methods : Hypothyroidism was induced by intradermal injection with PTU for 4 weeks in SD rats. ALRP extract or L-thyroxine as a control drug was orally administrated for 2 weeks with PTU injection in rats. The physiological and serological parameters were measured in rats. The histological change of thyroid tissues was observed by H&E staining, and also the expression of thermo-regulating proteins was determined by Western blot in dorsal root ganglia and brain tissues of rats. Results : The administration of ALRP extract in PTU-induced hypothyroidism rats was significantly increased body temperature, but did not changes on body weight, food and water intake. ALRP extract did not effect on the levels of TSH and T4 in the hypothyroidism rats. ALRP extract significantly decreased the levels of GPT, glucose, HDL-cholesterol, LDL-cholesterol, and total cholesterol in the hypothyroidism rats. In histological observation, the enlarged epithelium and atrophic follicles with higher concentration of follicular cells on hypothyroidism were improved by ALRP extract. In addition, ALRP extract increased the expression of TRPV1 and TRPM8 ion channel proteins in hypothyroidism rats. Conclusion : These results indicate that ALRP extract can improve PTU-induced hypothyroidism through regulation of body temperature and lipid accumulation. The action mechanism of ALRP extract is related with body temperature control by thermoregulation with TRP ion channels.
