Comparison of Skin Injury Induced by β- and γ-irradiation in the Minipig Model

  • Background: The effects of radiation on tissues vary depending on the radiation type. In this study, a minipig model was used to compare the effects of ${\beta}$-rays from $^{166}Ho$ and ${\gamma}$-rays from $^{60}Co$ on the skin. Materials and Methods: In this study, the detrimental effects of ${\beta}$- and ${\gamma}$-irradiation on the skin were assessed in minipigs. The histopathological changes in the skin from 1 to 12 weeks after exposure to 50 Gy of either ${\beta}$- (using $^{166}Ho$ patches) or ${\gamma}$- (using $^{60}Co$) irradiation were assessed. Results and Discussion: The skin irradiated by ${\beta}$-rays was shown to exhibit more severe skin injury than that irradiated by ${\gamma}$-rays at 1-3 weeks post-exposure; however, while the skin lesions caused by ${\beta}$-rays recovered after 8 weeks, the ${\gamma}$-irradiated skin lesions were not repaired after this time. The observed histopathological changes corresponded with gross appearance scores. Seven days post-irradiation, apoptotic cells in the basal layer were detected more frequently in ${\beta}$-irradiated skin than in ${\gamma}$-irradiated skin. The basal cell density and skin thickness gradually decreased until 4 weeks after ${\gamma}$- and ${\beta}$- irradiation. In ${\beta}$-irradiated skin lesions, and the density and thickness increased sharply back to control levels by 6-9 weeks. However, this was not the case in ${\gamma}$-irradiated skin lesions. In ${\gamma}$-irradiated skin, cyclooxygenase-2 (COX-2) was shown to be expressed in the epidermis, endothelial cells of vessels, and fibroblasts, while ${\beta}$-irradiated lesions exhibited COX-2 expression that was mostly limited to the epidermis. Conclusion: In this study, ${\beta}$-rays were shown to induce more severe skin injury than ${\gamma}$-rays; however, the ${\beta}$-rays-induced injury was largely repaired over time, while the ${\gamma}$-rays-induced injury was not repaired and instead progressed to necrosis. These findings reveal the differential effects of ${\gamma}$- and ${\beta}$-irradiation on skin and demonstrate the use of minipigs as a beneficial experimental model for studying irradiation-induced skin damage.

Radiation Dose Assessment Model for Terrestrial Flora and Fauna and Its Application to the Environment near Fukushima Accident

  • Background: To investigate radiological effects on biota, it is necessary to assess radiation dose for flora and fauna living in a terrestrial ecosystem. This paper presents a dynamic model to assess radioactivity concentration and radiation dose of terrestrial flora and fauna after a nuclear accident. Materials and Methods: Litter, organic soil, mineral soil, trees, wild crops, herbivores, omnivores, and carnivores are considered the major components of a terrestrial ecosystem. The model considers the physicochemical and biological processes of interception, weathering, decomposition of litter, percolation, root uptake, leaching, radioactive decay, and biological loss of animals. The predictive capability of the model was investigated by comparison of its predictions with field data for biota measured in the Fukushima forest area after the Fukushima nuclear accident. Results and Discussion: The predicted radioactive cesium inventories for trees agreed well with those for evergreens and deciduous trees sampled in the Fukushima area. The predicted temporal radioactivity concentrations for animals were within the range of the measured radioactivity concentrations of deer, wild boars, and black bears. The radiation dose for the animals were, for the whole simulation time, estimated to be much smaller than the lower limit (0.1 mGy·d-1) of the derived consideration reference level given by the International Commission on Radiological Protection for terrestrial flora and fauna. This suggested that the radiation effect of the accident on the biota in the Fukushima forest would be insignificant. Conclusion: The present dynamic model can be used effectively to investigate the radiological risk to terrestrial ecosystems following a nuclear accident.

방사선과 재학생의 수시출입자 방사선 피폭선량에 대한 고찰 (Consideration about Radiological Technology Student's Frequent Workers Exposure Dose Rate)

  • The Nuclear Safety Commission amended the Nuclear Safety Act by strengthening the safety management system for the frequent workers to the level of radiation workers. And students entering radiation management zones for testing and practical purposes are subject to frequent workers. It is inevitable that this will incur additional costs. In this paper, the validity of the amendment to the Nuclear Safety Act was to be assessed in terms of radiation protection. Study subjects are from 2014 to 2016, among university students in Seong-nam Korea and comparisons for analyses were made taking into account variables that are differences in annual, practical types, on-class and clinical practice students exposure dose. The analysis showed that exposures between on-class and clinical practice received were less than the annual dose limit of 1 mSv for the public. Then, some alternatives that excluding from frequent workers during on-class practice or mitigating the frequent workers' safety regulation for only on-class frequent workers can be considered. Optimization is how rational is the reduction in exposure dose to the costs required. Therefore, the results are hardly considered for optimization. If the data accumulated, it could be considered that the revision of the act could be evaluated and improved.

방사선비상시 비강스메어 시료의 전알파 분석 및 내부피폭선량평가 절차 (Gross Alpha Analysis of Nasal Smear Samples and Internal Dose Assessment Procedure in Radiation Emergency)

  • 방사선 사고시 피폭환자로부터 채취된 스메어 시료의 전알파 방사능 측정법을 수립하였으며 이를 바탕으로 피폭환자의 후속조치 절차를 확립하였다. 국내방사선사고 대응기관에서 사용중인 스메어용 면봉을 이용하여 계측 절차를 검증하였다. 액체섬광계수기를 이용하여 표준선원을 점적한 시료의 계측 결과 20% 이내에서 인증값과 잘 일치하였으며, 채취 조건은 세정제 등을 이용하는 것이 더 높은 스메어 효율을 보였다. 액체섬광계수법 특성상 소광현상의 영향을 배제하기 위해 건조 등의 최소한의 전처리가 필요 할 것으로 판단되었다. 계측결과를 바탕으로 방사선비상시 피폭환자에 대한 의료적 처치 기준 및 선량평가 절차를 수립하였다.

CHANGING OF RGS TRANSCRIPTS LEVELS BY LOW-DOSE-RATE IONIZING RADIATION IN MOUSE TESTIS

  • Deleterious effects of high dose radiation exposure with high-dose-rate are unarguable, but they are still controversial in low-dose-rate. The regulator of G-protein signaling (RGS) is a negative regulator of G protein-coupled receptor (GPCR) signaling. In addition, it is reported that irradiation stress led to GPCR-mediated mitogen-activated protein kinase (MAPK) and phosphotidylinositol 3-kinase (PI3-k) signaling. The RGS mRNA expression profiles by whole body radiation with low-dose-rate has not yet been explored. In the present study, we, therefore, examined which RGS was modulated by the whole body radiation with low-dose-rate ($3.49mGy{\cdot}h^{-1}$). Among 16 RGS expression tested, RGS6, RGS13 and RGS16 mRNA were down-regulated by low-dose-rate irradiation. This is the first report that whole body radiation with low-dose-rate can modulate the different RGS expression levels. These results are expected to reveal the potential target and/or the biomarker proteins associated with male testis toxicity induced by low-dose-rate irradiation, which might contribute to understanding the mechanism beyond the testis toxicity.

An Intercomparison of Counting Efficiency and the Performance of Two Whole-Body Counters According to the Type of Phantom

  • Background: Whole-body counters are widely used to evaluate internal contamination of the internal presence of gamma-emitting radionuclides. In internal dosimetry, it is a basic requirement that quality control procedures be applied to verify the reliability of the measured results. The implementation of intercomparison programs plays an important role in quality control, and the accuracy of the calibration and the reliability of the results should be verified through intercomparison. In this study, we evaluated the reliability of 2 whole-body counting systems using 2 calibration methods. Materials and Methods: In this study, 2 whole-body counters were calibrated using a reference male bottle manikin absorption (BOMAB) phantom and a Radiation Management Corporation (RMC-II) phantom. The reliability of the whole-body counting systems was evaluated by performing an intercomparison with International Atomic Energy Agencyto assess counting efficiency according to the type of the phantom. Results and Discussion: In the analysis of counting efficiency using the BOMAB phantom, the performance criteria of the counters were satisfied. The relative bias of activity for all radionuclides was -0.16 to 0.01 in the Fastscan and -0.01 to 0.03 in the Accuscan. However, when counting efficiency was analyzed using the RMC- II phantom, the relative bias of $^{241}Am$ activity was -0.49 in the Fastscan and 0.55 in the Accuscan, indicating that its performance criteria was not satisfactory. Conclusion: The intercomparison process demonstrated the reliability of whole-body counting systems calibrated with a BOMAB phantom. However, when the RMC-II phantom was used, the accuracy of measurements decreased for low-energy nuclides. Therefore, it appears that the RMC-II phantom should only be used for efficiency calibration for high-energy nuclides. Moreover, a novel phantom capable of matching the efficiency of the BOMAB phantom in low-energy nuclides should be developed.

Measurement of Neutron Production Double-differential Cross-sections on Carbon Bombarded with 430 MeV/Nucleon Carbon Ions

  • Background: Carbon ion therapy has achieved satisfactory results. However, patients have a risk to get a secondary cancer. In order to estimate the risk, it is essential to understand particle transportation and nuclear reactions in the patient's body. The particle transport Monte Carlo simulation code is a useful tool to understand them. Since the code validation for heavy ion incident reactions is not enough, the experimental data of the elementary reaction processes are needed. Materials and Methods: We measured neutron production double-differential cross-sections (DDXs) on a carbon bombarded with 430 MeV/nucleon carbon beam at PH2 beam line of HIMAC facility in NIRS. Neutrons produced in the target were measured with NE213 liquid organic scintillators located at six angles of 15, 30, 45, 60, 75, and $90^{\circ}$. Results and Discussion: Neutron production double-differential cross-sections for carbon bombarded with 430 MeV/nucleon carbon ions were measured by the time-of-flight method with NE213 liquid organic scintillators at six angles of 15, 30, 45, 60, 75, and $90^{\circ}$. The cross sections were obtained from 1 MeV to several hundred MeV. The experimental data were compared with calculated results obtained by Monte Carlo simulation codes PHITS, Geant4, and FLUKA. Conclusion: PHITS was able to reproduce neutron production for elementary processes of carbon-carbon reaction precisely the best of three codes.

Dose Estimation Model for Terminal Buds in Radioactively Contaminated Fir Trees

  • Background: After the Fukushima Daiichi Nuclear Power Plant (FDNPP) accident, biological alterations in the natural biota, including morphological changes of fir trees in forests surrounding the power plant, have been reported. Focusing on the terminal buds involved in the morphological formation of fir trees, this study developed a method for estimating the absorbed radiation dose rate using radionuclide distribution measurements from tree organs. Materials and Methods: A phantom composed of three-dimensional (3D) tree organs was constructed for the three upper whorls of the fir tree. A terminal bud was evaluated using Monte Carlo simulations for the absorbed dose rate of radionuclides in the tree organs of the whorls. Evaluation of the absorbed dose targeted 131I, 134Cs, and 137Cs, the main radionuclides subsequent to the FDNPP accident. The dose contribution from each tree organ was calculated separately using dose coefficients (DC), which express the ratio between the average activity concentration of a radionuclide in each tree organ and the dose rate at the terminal bud. Results and Discussion: The dose estimation indicated that the radionuclides in the terminal bud and bud scale contributed to the absorbed dose rate mainly by beta rays, whereas those in 1-year-old trunk/branches and leaves were contributed by gamma rays. However, the dose contribution from radionuclides in the lower trunk/branches and leaves was negligible. Conclusion: The fir tree model provides organ-specific DC values, which are satisfactory for the practical calculation of the absorbed dose rate of radiation from inside the tree. These calculations are based on the measurement of radionuclide concentrations in tree organs on the 1-year-old leader shoots of fir trees. With the addition of direct gamma ray measurements of the absorbed dose rate from the tree environment, the total absorbed dose rate was estimated in the terminal bud of fir trees in contaminated forests.

Evaluation of Radiological Effects on the Aptamers to Remove Ionic Radionuclides in the Liquid Radioactive Waste

  • Background: Aptamers are currently being used in various fields including medical treatments due to their characteristics of selectively binding to specific molecules. Due to their special characteristics, the aptamers are expected to be used to remove radionuclides from a large amount of liquid radioactive waste generated during the decommissioning of nuclear power plants. The radiological effects on the aptamers should be evaluated to ensure their integrity for the application of a radionuclide removal technique. Materials and Methods: In this study, Monte Carlo N-Particle transport code version 6 (MCNP6) and Monte Carlo damage simulation (MCDS) codes were employed to evaluate the radiological effects on the aptamers. MCNP6 was used to evaluate the secondary electron spectrum and the absorbed dose in a medium. MCDS was used to calculate the DNA damage by using the secondary electron spectrum and the absorbed dose. Binding experiments were conducted to indirectly verify the results derived by MCNP6 and MCDS calculations. Results and Discussion: Damage yields of about 5.00×10-4 were calculated for 100 bp aptamer due to the radiation dose of 1 Gy. In experiments with radioactive materials, the results that the removal rate of the radioactive 60Co by the aptamer is the same with the non-radioactive 59Co prove the accuracy of the previous DNA damage calculation. Conclusion: The evaluation results suggest that only very small fraction of significant number of the aptamers will be damaged by the radioactive materials in the liquid radioactive waste.

업종별 방사선작업종사자 피폭 기록 다각형 모델 분석 연구 (Polygonal Model Analysis on Occupational Exposure Record of Radiation Workers by Work Field)

  • 작업환경에 따라 방사선학적 위험성이 상이하므로 해당 업종 환경에 대한 분석을 통해 방호 수단과 정책을 개발해야 한다. 방사선을 이용하는 업종 분야에서의 특성을 평가하는 것은 해당 업종의 수치만을 보는 것이 아니라 다른 업종과의 비교 분석을 통해 수행되어야 한다. 본 연구는 업종별 방사선 피폭 기록 비교를 위해 부처별 종사자 피폭 기록으로부터 평가 인자를 도출하고 이를 상대적으로 평가할 수 있는 다각형 모델을 개발하여 8개 방사선 이용 업종에 대해 적용하였다. 2020년 방사선 피폭 기록을 바탕으로 다각형 모델을 적용하여 방사선 업종의 피폭 기록을 기반으로 특성을 비교 평가하였다. 이를 통해 다각형 모델의 유용성을 확인하였으며 업종에 대한 방호 정책 방안을 제안하였다.
