Investigation of 180W separation by transient single withdrawal cascade using Salp Swarm optimization algorithm

  • The 180W is the lightest isotope of Tungsten with small abundance ratio. It is slightly radioactive (α decay), with an extremely long half-life. Its separation is possible by non-conventional single withdrawal cascades. The 180W is used in radioisotopes production and study of metals through gamma-ray spectroscopy. In this paper, single withdrawal cascade model is developed to evaluate multicomponent separation in non-conventional transient cascades, and available experimental results are used for validation. Numerical studies for separation of 180W in a transient single withdrawal cascade are performed. Parameters affecting the separation and equilibrium time of cascade such as number of stages, cascade arrangements, feed location and flow rate for a fixed number of gas centrifuges (GC) are investigated. The Salp Swarm Algorithm (SSA) as a bio-inspired optimization algorithm is applied as a novel method to minimize the feed consumption to obtain desired concentration in the collection tank. Examining different cascade arrangements, it is observed in arrangements with more stages, the separation is further efficient. Based on the obtained results, with increasing feed flow rate, for fixed product concentration, the cascade equilibrium time decreases. Also, it is shown while the feed location is the farthest stage from the collection tank, the separation and cascade equilibrium time are well-organized. Finally, using SSA optimal parameters of the cascade is calculated, and optimal arrangement to produce 5 gr of 180W with 90% concentration in the tank, is proposed.

Positional correction of a 3D position-sensitive virtual Frisch-grid CZT detector for gamma spectroscopy and imaging based on a theoretical assumption

  • The virtual Frisch-grid method for room-temperature radiation detectors has been widely used because of its simplicity and high performance. Recently, side electrodes were separately attached to each surface of the detectors instead of covering the entire detector surface with a single electrode. The side-electrode structure enables the measurement of the three-dimensional (3D) gamma-ray interaction in the detector. The positional information of the interaction can then be utilized to precisely calibrate the response of the detector for gamma-ray spectroscopy and imaging. In this study, we developed a 3D position-sensitive 5 × 5 × 12 mm3 cadmium-zinc-telluride (CZT) detector and applied a flattening method to correct detector responses. Collimated gamma-rays incident on the surface of the detector were scanned to evaluate the positional accuracy of the detection system. Positional distributions of the radiation interactions with the detector were imaged for quantitative and qualitative evaluation. The energy spectra of various radioisotopes were measured and improved by the detector response calibration according to the calculated positional information. The energy spectra ranged from 59.5 keV (emitted by 241Am) to 1332 keV (emitted by 60Co). The best energy resolution was 1.06% at 662 keV when the CZT detector was voxelized to 20 × 20 × 10.

  • Research in the cytokine field has grown exponentially in recent years, and the validity of such studies relies heavily on the appropriate measurement of levels of cytokines in various biological samples. Transforming growth factor (TGF)-$\beta$, a hormonally active polypeptide found in normal and transformed tissue, is a potent regulator of cell growth and differentiation. The most widely used bioassay for TGF-$\beta$ is the inhibition of the proliferation of mink lung epithelial cells. Though detection of [$^3$H]thymidine incorporation is more sensitive than the MTT assay, it presents some disadvantages due to the safety and disposal problems associated with radioisotopes. In this study, we attempted to ascertain the experimental conditions which could be used for measuring the in vitro biological activity of TGF-$\beta$ in a safer and more sensitive way compared with the currently available methods. We compared the commonly used method, the MTT assay, to the XTT assay using different parameters including cell number, incubation time and the wave length used for detecting the product. We examined the anti-proliferative activities of TGF-$\beta$ in three different cell lines: Mv-1-Lu mink lung epithelial cells, MCF10A human breast epithelial cells and H-ras-transformed MCF10A cells. Herein, we present an experimental protocol which provides the most sensitive method of quantifying the biological activity of TGF-$\beta$, with a detection limit of as low as 10 pg/ml: Mv-1-Lu or H-ras MCF10A cells ($1{\times}10^5/well$) were incubated with TGF-$\beta$ at $37^{\circ}C$ in a humidified $CO_2$ incubator for 24 hr followed by XTT treatment and determination of absorbance at 450 or 490 nm. Our results may contribute to the establishment of an in vitro bioassay system, which could be used for the satisfactory quantitation of TGF-$\beta$.

Radiolabeling of nanoparticle for enhanced molecular imaging

  • The combination of nanoparticle with radioisotope could give the in vivo information with high sensitivity and specificity. However, radioisotope labeling of nanoparticle is very difficult and radioisotopes have different physicochemical properties, so the radioisotope selection of nanoparticle should be carefully considered. $^{18}F$ was first option to be considered for labeling of nanoparticle. For the labeling of $^{18}F$ with nanoparticle, Prosthetic group is widely used. Iodine, another radioactive halogen, is often used. Since radioiodine isotopes are various, they can be used for different imaging technique or therapy in the same labeling procedures. $^{99m}Tc$ can easily be obtained as pertechnatate ($^{99m}{TcO_4}^-$) by commercial generator. Ionic $^{68}Ga$ (III) in dilute HCl solution is also obtained by generator system, but $^{68}Ga$ can be substituted for $^{67}Ga$ because of the short half-life (67.8 min). $^{64}Cu$ emits not only positron but also ${\beta}-particle$. Therefore $^{64}Cu$ can be used for imaging and therapy at the same time. These radioactive metals can be labeled with nanoparticle using the bifunctional chelator. $^{89}Zr$ has longer half-life (78.4 h) and is used for the longer imaging time. Unlike different metals, $^{89}Zr$ should use the other chelate such as DFO, 3,4,3-(LI-1,2-HOPO) or DFOB.

  • Cellulose grafted with acrylic acid(AAc) was prepared by radiation grafting technique. The effects of reaction temperature. reaction time, monomer concentration, and the crosslinkers on the AAc grafting reaction on cellulose were examined. The amount of AAc grafted on the cellulose reached maximum at the concentration of 0.75vol% difunctional crosslinker and 1.0vol% trifunctional crosslinker, respectively. In the presence of acid, the amount of AAc grafted on the cellulose was decreased when reaction solution contains difunctional crosslinker, while that was increased when reaction solution contains trifunctional crosslinker. In the grafting reaction of cellulose with AAc and TMETA, mixture containing ferrous sulfate and acid enhanced further AAc grafting yield than mixture containing ferrous sulfate only.

Comparative study of 82Sr separation/purification methods used at Brookhaven National Laboratory and ARRONAX

  • Nuclear imaging is one of the most powerful measures for non-invasive diagnosis of myocardial vascular disease. Radionuclide such as 13N, 15O, 201Tl and 82Rb is used for the measurement of cardiac blood flow. 13N, 15O and 201Tl are produced in cyclotrons while 82Rb is obtained from generator. Rubidium (Rb), an alkali ion, behaves biologically like potassium, and accumulates in myocardial tissue. Rb has rapid blood clearance profile which allows the use of 82Rb with a short physical half-life of 75 s for non-invasive evaluation of regional myocardial perfusion. There are several advantages of 82Rb over other radioisotopes. An ultra-short half-life significantly reduces the exposure of patients to radiation and allows to repeat injections for studying the effects of medical intervention. As a positron emitter, 82Rb allows positron emission tomography (PET) imaging which have shown superior diagnostic performances. 82Rb can be produced from generator by decay of its parent 82Sr. However, the preparation of 82Sr is difficult, because appropriate purity is required to meet the specification of the product. Recently reported procedure from ARRONAX research institute showed that a Chelex-100 resin is sufficient for this purpose and additional column is not necessary. Whereas Brookhaven National Laboratory (BNL) procedure contains three ion exchange resin separation, including Chelex-100 resin. Currently, since 82Sr production site is non-existent in Korea, Korea Atomic Energy Research Institute (KAERI) has plan to produce 82Sr within specifications. We compared 82Sr purification procedures reported from ARRONAX and BNL to investigate the most suitable procedure for our conditions.

  • In this study, the sulfonated ion exchange fiber was synthesized by $Co^{60}\;{\gamma}-ray$ radiation-induced graft copolymerization. Degree of grafting (DG) increased with increasing the total dose and showed the highest value at 50 v/v% styrene monomer. And also, the degree of sulfonation (DS) increased with increasing the DG and reaction temperature. DS showed the maximum value at 20 min. Ion exchange capacity and swelling ratio of ion exchange fibers increased with increasing the DS and their maximum values were 4.76 meq/g and 23.5%, respectively. Ammonia adsorption increased as increasing the ammonia concentration and ion exchange capacity and remained constant over 10 cycles.

  • Because of a high degree of unsaturation of perilla seed oil vitamin E deficiency can be produced in chicks by using this particular oil diet. It is therefore convenient to use this oil for elucidating more detailed function (s) of vitamin E. The present study was undertaken to investigate the nutritional and pathological aspects both of high PUFA and of low vitamin E in the diet eventually to elucidate the function(s) of vitamin E. The present study examines the relationships between PUFA, vitamin E and Se in the experimental diets by measuring vitamin E levels both in serum and in liver and activities of SGPT and SGOT and by examining electron micrographs of the chick hepatocytes. Vitamin E concentrations in serum and liver responded to dietary treatments. Serum levels of vitamin E were more affected than those in liver, the values of groups IV(15% fat,- Vit.E) and V(15% fat,-Vit E,-Se) were significantly lower than those of groups I (5% fat) and 111 (15% fat) with P/S ratio of 1. The activities of SGOT and SGPT were found not to be different significantly among different diet groups. Electron microscopic observations of the chick hepatocyte revealed degeneration of mitochondria and appearance of vesicles in the cytoplasm of groups fed diets high in PUFA and deficient in vitamin E alone or deficient in both vitamin E and Se. In the same group pyknotic nuclei and deterioration of the sinusoidal border, showing diminution in microvilli were also observed. More detailed studies concerning biochemical aspects should be carried out by using radioisotopes both in the in vitro and in vivo systems and morphological study should have a focus on the changes in nucleus which seems already suggestive in the present observation.

  • Perfusion magnetic resonance imaging (pMRI) is a special technique for evaluation of blood flow. Exogenous pMRI methods which are dynamic susceptibility contrast (DSC) and dynamic contrast-enhanced (DCE) use an intravenous bolus injection of paramagnetic contrast agent. In contrast, an endogenous pMRM method which is arterial spin labeling (ASL) use diffusible blood in body. In order to scan pMRI in human, technical optimizations are very important according to disease conditions. For examples, DSC is popularly used in patients with acute stroke due to its short scan time, while DSC or DCE provides the various perfusion indices for patients with tumor. ASL is useful for children, women who are expected to be pregnant, and in patients with kidney diseases which are problematic in nephrogenic systemic fibrosis (NSF). Perfusion MRI does not require any injection of radioisotopes. We expect that demand for perfusion MRI will be higher in evaluating drug efficacy and other treatment effects.

How Computed Tomography Contrast Media and Magnetic Resonance Imaging Contrast Media Affect the Changes of Uptake Counts of 201Tl

  • The purpose of the study is to investigate how uptake counts of $^{201}Tl$ of radioisotopes in the human body could change, when taking computed tomography and magnetic resonance imaging right after injecting contrast media. $^{201}Tl$ radioisotope substances of iodine contrast medium, which is a computed tomography contrast medium, and paramagnetic contrast medium, which is an magnetic resonance imaging contrast medium, were used as study materials. First, $^{201}Tl$ was put into 4 cc of normal saline in test tube, and then a computed tomography contrast medium of Iopamidol$^{(R)}$ or Dotarem$^{(R)}$, was put into 2 cc of normal saline in test tube. An magnetic resonance imaging contrast medium of Primovist$^{(R)}$ or Gadovist$^{(R)}$ was also put into 2 cc of normal saline in test tube. Each contrast medium was distributed to make $^{201}Tl$ as 3 mCi, with a total of 4 cc. Gamma camera, low energy high resolution collimator, and pinhole collimator were used to obtain images. The uptake count of $^{201}Tl$ was measured with 1000 frames of images, and obtained after 10 times of repetition. This study revealed that the use of Gadovist$^{(R)}$, which is an magnetic resonance imaging contrast medium, showed the smallest number of uptake count, after measuring $^{201}Tl$ uptake count by low energy high resolution collimator. On the other hand, the use of Iopamidol$^{(R)}$, which is a computed tomography contrast medium, showed the biggest difference in uptake count, when measuring $^{99m}Tc$ uptake count by Pinhole collimator. When examining with gamma camera, using contrast medium and $^{201}Tl$, identifying the changes of uptake count is very important for improving the value of diagnosis.
