Effect of Schizandra chinensis Extracts on Oxidative Damage

  • In this study, we evaluated the protective effects of supercritical extracts and two step ethanol extracts after supercritical extraction from Schizandra chinensis on antioxidant activities and oxidative DNA and cell damages. Supercritical extracts removed DPPH (1,1-diphenyl-2-picryldrazyl) radical by 85.5% at 200 ${\mu}g$/ml, but showed low activities of scavenging and chelating the hydroxyl radical and ferrous iron. However, two step ethanol extracts showed low activities of scavenging the DPPH radical, but removed the hydroxyl radical by 86% at 200 ${\mu}g$/ml. In addition, we tested the activities of extracts for reducing hydroxyl radical-induced DNA and cell damage. Two step ethanol extracts showed protective effect against the oxidative DNA damage by reducing DNA segmentation, inhibiting DNA migration and decreasing the expression of phospho-H2AX. Also, two step ethanol extracts showed protective effect against the oxidative cell damage by inhibiting lipid peroxidation and increasing the expression of p21 protein. Taken together, we suggest that two step ethanol extracts from S. chinensis have a role as useful inhibitors against oxidative damages.

해면으로부터 분리된 Bromotopsentin의 항산화활성 (Antioxidant Activities of Bromotopsentin from the Marine Sponge Spongosorites sp.)

  • Bromotopsentin (BSM) is a bisindole alkaloid compound, which is recognized as a metabolite of the marine sponge Spongosorites sp. In this study, the antioxidant activity of BSM was investigated. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, the trolox equivalent antioxidant capacity (TEAC) assay, the superoxide radical scavenging (NBT) assay, the lipid peroxidation and hydroxyl radical-induced DNA damage assays were carried out to evaluate the antioxidant activity of BSM. It was found that BSM had stronger scavenging activity on the stable free radical DPPH and superoxide radical than L-ascorbic acid with an $IC_{50}$ value of 62 and 64 ${\mu}M$, respectively. The TEAC value which indicated the total antioxidant capacity of BSM was about 0.8, which was also stronger than L-ascorbic acid. About 1.3 ${\mu}M$ of BSM induced 50% inhibition of lipid peroxidation. 60 nM of BSM exhibited a significant protective activity against DNA strand scission by hydroxyl radical on pBR322 DNA. Taken together, we suggest that BSM possesses strong antioxidant activity, and could be a valuable new addition to the list of anti-aging chemotherapeutic agents.

녹차첨가 서리태청국장의 이화학적 특성 및 유리기 소거능 (Radical Scavenging Effects and Physicochemical Properties of Seolitae Chungkukjang Added with Green Tea)

  • 본 연구에서는 청국장의 기능성과 관능적 품질개선을 위한 연구의 일환으로 서리태청국장에 녹차를 0.5%, 2.0%, 5.0% 첨가(w/w)하여 청국장을 제조한 후 특성을 비교하고 항산화능을 검토하였다. 녹차 5% 첨가한 서리태청국장이 in vitro상에서 DPPH와 ${\cdot}OH$ 소거능이 우수하였고 관능검사 결과는 향, 맛, 종합적 기호도가 일반청국장과 유사하게 높게 나타났다. 이상의 결과를 종합하여 볼 때 녹차 5% 첨가 서리태청국장이 청국장의 관능적 품질이 뛰어날 뿐만 아니라 항산화활성이 가장 높은 것으로 평가되었다.

Antioxidant and Antidiabetic Activities of Aralia elata Seeds

  • Aralia elata seeds were successively extracted with water, methanol, ethanol, acetone and chloroform. The crude extracts were investigated for antioxidant and antidiabetic activities. The antioxidant properties of various extracts were evaluated by antioxidant tests, such as DPPH free radical-scavenging activity, hydroxyl radical-scavenging assay, metal-chelating activity, lipid peroxidation inhibition activity and reducing power assay. The 70% methanol extract exhibited the highest activity in the in vitro models of DPPH free radical-scavenging activity, metal-chelating activity, and reducing power assay. Acetone extract showed good effects on lipid peroxidation inhibition and hydroxyl radical-scavenging assay at a low concentration. In addition, the $\alpha$-glucosidase inhibition assay showed that 70% methanol extract had the highest activity. These results indicate the high possibility of using A. elata seeds for medical application due to their efficient antioxidant properties.

갓김치 Chlorophylls 및 Carotenoids의 항산화 효과 (Antioxidative Effect of Cholorophylls and Carotenoids in Mustard Leaf Kimchi Activity)

  • Antioxidative effects of crude chlorophylls and carotenoids extracts from mustard leaf kimchi on the lipid peroxidation in rat liver homogenate, egg phosphatidyl choline (EPC) liposome and superoxide anion radical were examined. The extracts were found to inhibit the increase of the thiobarbituric acid (TBA) value and show the effect of antioxidative activity on the liposomal phospholipid membrane. The oxidation index of EPC liposome was markedly decreased in the prescence of the extracts. The antioxidative activity of the extracts from mustard leaf kimchi was not related with fermentation period of the kimchi. The extracts from mustard leaf showed the similar antioxidative activity of $\alpha$-tocopherol within in the given level of addition. However, the oxidation index. When the effect of the extracts from mustard leaf kimchi on free radical scavenging was observed by the determination of the superoxide anion radical scavenging activity, it had similar value to that of $\alpha$-tocopherol.

Toxicological Studies on the Essential Oil of Eugenia caryophyllata Buds

  • The essential oil (EC-oil) obtained from the buds of Eugenia caryophyllata (Myrtaceae) was examined for its free radical-scavenging activity, cytotoxicity, and in vivo toxicity. To find the xenobiotic properties of EC-oil, serum thiobarbituric acid reactive substances (TBARS) level and hepatic drug-metabolizing enzyme activities were measured. It was found that EC-oil displayed xenobiotic properties like bromobenzene. The cytotoxicities of eugenol and of the EC-oil were greatly attenuated by the sulfhydryl-containing N-acetyl-L-cysteine (NAC), suggesting that eugenol was susceptible to nucleophilic sulfhydryl. In addition, eugenol also showed potent free radical-scavenging activity in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Moreover, methyleugenol considerably exhibited less cytotoxicity and less potent free radical-scavenging activity than eugenol, and the cell viability of the methyleugenol was more increased with NAC treatment than the eugenol. These results indicate that the phenolic OH in eugenol may play a crucial role in both cytotoxicity and free radical-scavenging activity. The fashion on oxidative stress and hepatic drug-metabolizing enzyme activities of eugenol resembled those of bromobenznene.

CRDS Study of Tropospheric Ozone Production Kinetics : Isoprene Oxidation by Hydroxyl Radical

  • The tropospheric ozone production mechanism for the gas phase additive oxidation reaction of hydroxyl radical (OH) with isoprene (2-methyl-1,3-butadiene) has been studied using cavity ring-down spectroscopy (CRDS) at total pressure of 50 Torr and 298 K. The applicability of CRDS was confirmed by monitoring the shorter (~4%) ringdown time in the presence of hydroxyl radical than the ring-down time without the photolysis of hydrogen peroxide. The reaction rate constant, $(9.8{\pm}0.1){\times}10^{-11}molecule^{-1}cm^3s^{-1}$, for the addition of OH to isoprene is in good agreement with previous studies. In the presence of $O_2$ and NO, hydroxyl radical cycling has been monitored and the simulation using the recommended elementary reaction rate constants as the basis to OH cycling curve gives reasonable fit to the data.

  • Reactivity and reaction mechanism for the reactions of 1,1'-(azodicarbonyl) dipiperidine with triphenylphosphines are investigated using kinetic method. The cation radical, Ph3P and the anion radical, -N-N - are produced during the course of the reaction. The cation radical is formed by the transfer of an electron from phosphorus to the nitrogen atom. The anion radical is formed by the addition of the one electron to the azo rad-ical. The rate constants are decreased by electron withdrawing groups while they are increased by electron donating groups present in triphenylphosphine. The electron density increases on nitrogen, while positive charge is developed on phosphorus in the transition state.

도토리분말을 첨가한 현미설기떡의 제조 및 품질평가 (Quality Evaluation of Brown Rice Sulgidduk Containing Acorn Powder)

  • This study examined the quality characteristics of brown rice Sulgidduk containing acorn powder and the optimal mixing rate (0%, 5%, 10%, and 15%). The moisture contents of brown rice Sulgidduk increased with increasing amount of acorn powder. The DPPH free radical scavenging activities (16.15%~28.06%) and ABTS free radical scavenging activities (22.98%~42.81%) of the brown rice Sulgidduk increased with increasing acorn powder content. The L value of the Hunter's color value decreased with increasing amount of acorn powder. The hardness and chewiness of brown rice Sulgidduk increased with increasing amount of acorn powder. The brown rice Sulgidduk containing 10% acorn powder showed the highest score with regard to the sensory characteristics. These results suggest that the most desirable amount of acorn powder is 10% and the addition of acorn powder could contribute positively toward the quality characteristics of brown rice Sulgidduk.

숙성된 3종 인삼추출물의 항산화 및 주름개선 효과 (Antioxidant and Anti-wrinkle Effects of Steamed Three Ginseng Extracts)

  • 숙성된 인삼은 인간의 건강을 회복시키고 향상시키는 강장제로서 알려져 있으며, 백삼보다 더 많은 약리적인 활성을 가진다고 알려져 왔다. 전이 가능한 종양, 림프구 증식과 쥐간의 지질과산화에 대한 숙성된 인삼의 효과도 연구되었다. 본 연구는 3가지 인삼 추출물의 항산화와 항노화 효과를 평가하기 위해서 실험하게 되었다. 시장에서 이미 만들어진 홍삼엑기스를 사용하였고, 그 외 수삼, 건삼을 구매하여 홍삼제조기로 만들어진 추출물로 실험에 사용하였다. 숙성된 인삼의 3가지 추출물들은 superoxide radical 소거 활성, hydroxyl radical 소거 활성, 리놀산 자동산화 억제, 섬유아세포에서 collagenase 억제와 collagen 합성의 효과를 연구했다. 숙성시켜 만든 수삼은 100 ug/mL의 농도에서 가장 높은 superoxide radical 소거 활성뿐만 아니라 vitamin C보다 더 높은 hydroxyl radical 소거 활성을 나타냈다. 또한 리놀산 자동산화 억제, collagen 합성과 collagenase 억제에서 가장 높은 활성을 보였다.
