Gene Expression Profiles Following High-Dose Exposure to Gamma Radiation in Salmonella enterica serovar Typhimurium

  • Microarrays can measure the expression of thousands of genes to identify the changes in expression between different biological states. To survey the change of whole Salmonella genes after a relatively high dose of gamma radiation (1 kGy), transcriptome dynamics were examined in the cells by using DNA microarrays. At least 75 genes were induced and 89 genes were reduced two-fold or more after irradiation. Several genes located in pSLT plasmid, cyo operon, and Gifsy prophage were induced along with many genes encoding uncharacterized proteins.While, the expression of genes involved in the virulence of Salmonella as well as metabolic functions were decreased. Although the radiation response as a whole could not be illustrated by using DNA microarrays, the data suggest that the response to high dose of irradiation might be more complex than the SOS response.

자궁경부암세포에서 방사선조사시 차등 발현되는 유전자 동정 (Identification of Differentially Expressed Radiation-induced Genes in Cervix Carcinoma Cells Using Suppression Subtractive Hybridization)

  • 목적 : 자경경부암세포에서 polymeric chain reaction (PCR)원리를 이용한 suppression subtractive hybridization (SSH) 방법으로 방사선조사 시 차등 발현되는 유전자를 동정하고자 하였다. 대상 및 방법 : 자궁경부암세포주인 HeLa세포주에 방사선조사 전과 후 총 RNA와 poly $(A)^+$ mRNA를 분리였다. SSH방법으로 forward 및 reverse-subtracted cDNA libraries를 만들었다. 차등 발현된 유전자를 screening하기 위해 reverse Northern blotting (dot blot analysis)을 이용하여 각각의 library에서 88개의 클론을 선택하였고 Nothern blotting으로 확인 후 sequencing하였다. 결과 : screening상 176개 클론 중 forward-subtracted library에서 10개의 유전자가 reverse-subtracted library에서 9개의 유전자가 동정되었다. forward-subtracted library로부터 3개의 유전자가 Northern blotting에 의하여 확인되었고 이중 telomerase catalytic subunit and sodium channel-like protein 유전자와 1개의 ESTs (expressed sequence tags) 유전자가 방사선선량에 따라 증가하쳐다. 결론 : 본 연구를 통해 자궁경부암세포주에서 방사선에 의해 유도되는 유전자를 SSH 방법을 통해 동정할 수 있었다. 그러나 이러한 유전자가 어떤 생물학적인 기능을 갖고 있는지에 대한 계속적인 연구가 필요하다

Bacillus lentimorbus WJ5의 감마선유도 돌연변이체들에서 공통으로 발현되는 방사선 관련 유전자의 microarray 분석 (Microarray Analysis of Radiation Related Gene Expression in Mutants of Bacillus lentimorbus WJ5 Induced by Gamma Radiation)

  • 본 연구에서는 감마선으로 유도된 돌연변이체들에서 공통으로 발현되는 방사선 관련 유전자들의 발현을 연구하기 위하여, B. lentimorbus WJ5 의 방사선 유도 돌연변이체에서 발현되는 유전자를 DNA microarray로 동시에 탐색하였다. DNA microarray는 B. lentimorbus WJ5 genome을 무작위로 절단하여 2,000 단편으로 구성하였으며, 감마선 $(^{60}/Co)$으로 유도된 7 돌연변이체의 발현을 정량적으로 관찰하였다. 클러스터 분석결과 발현된 408 유전자 중 27개가 감마선 유도 돌연변이체 모두에서 유의하게 발현이 증가되었다. 특히, 복구(mutL, mutM) 에너지 대사 (acsA, sdhB, pgk, yhjB, citB), protease (npr), 산화자극에 대한 환원 (HMM)관련 유전자들이 동시에 증가되었다. 이는 감마선 유도 돌연변이체들에서 자발적인 직/간접 복구 관련 유전자의 발현 증가는 방사선 노출 직후 보이는 stress response와는 다른 현상임을 나타내는 것으로 생각된다.

Gamma Radiation Induced Mutational Spectrum of Laccase Gene in Pleurotus ostreatus

  • To investigate the mutational spectrum of laccase (Lac) genes (lac -A and lac -B) involved in degrading lignin which is the recalcitrant cell wall polymer, the genes of the Pleurotus ostreatus mutants induced by gamma ray radiation were amplified by PCR and were cloned. All partial lac-A genes of 4 mutants (PO-6, -7, -14 and -15) consisted of 1763 base pairs due to the deletion of two bases (491-nt and 492-nt) and addition of one base (875-nt) in 1764 base pairs of lac -A gene of PO-1. Totally 36 mutational hot spots were detected and 32 positions were mutated in all of those 4 mutants simultaneously. These mutations were predominantly A : T -> G : C transitions (40%). Putative amino acid sequences of lac -A genes of mutants have one simultaneous mutated residue (from Thr-44 to Ala-44). The 1764 bp of partial lac -B gene was cloned only in PO -5 mutant and contained 19 mutated bases. These mutations were predominantly G : C->A : T transitions (45%). Lac-B protein of PO-5 has two mutated residues of Glu-290 and His-363 from Ala-290 and Phe-363, respectively. The hyper-mutational positions were concentrated in specific regions of between 50-nt and 900-nt in lac genes. These results suggested that the mutational hotspots responded to gamma radiation could be in some genes, at least lac -A and lac -B of p. ostreatus.

인체세포주 A431에서 방사선 조사 후 DNA수선 유전자 발현과 세포고사와의 관계에 관한 연구 (Relationship between Radiation Induced Activation of DNA Repair Genes and Radiation Induced Apoptosis in Human Cell Line A431)

  • 목적: 피부세포인 A431세포주에서 방사선 조사에 의한 세포고사가 방사선량과 방사선 조사 후 경과시간에 따라서 어떻게 변하는지를 밝혀보고, 방사선에 의해 유도된 수선유전자의 발현을 방사선량별, 조사 후 경과시간별로 분석하여 세포고사와 어떤 관계가 있는지 알아보고자 하였다. 대상 및 방법: 한국 세포주은행으로부터 분양받은 피부상피암 세포의 일종인 A431을 Cs-137 세포조사기를 이용하여 5 Gy, 25 Gy씩 조사하고 4, 12, 48시간이 지난 다음 세포를 모아 유세포계측법을 이용하여 고사세포를 계수하였다. 또한 이 세포들을 Northern blot analysis, Western blot analysis를 시행하여 방사선량별, 경과시간별로 유전자의 변화를 분석하였다. 각 실험군간의 통계적 유의성은 SPSS 통계프로그램을 사용하여 MANOVA test에 의해 검정하였으며, p값 0.05 미만을 유의한 수준으로 판정하였다. 결과: 유세포 계측기로 측정한 고사세포의 비율은 방사선 조사 후 12시간째에 가장 유의하게 증가하였다 (p<0.01). DNA수선유전자의 발현은 5 Gy 조사 후 p53, p21, hRAD 유전자가 12시간째에 증가하였고, 25 Gy 조사 후에는 hRAD50과 p21이 12시간에 증가하였으며, p53과 GADD45는 12시간까지 별 변화가 없었으나 이후 증가하여 48시간에 가장 높은 발현을 보였다. 결론: 피부상피암세포에서 방사선에 의해 유도되는 세포고사는 방사선 조사 후 12시간에 가장 현저해지는 것을 알 수 있었으며, 이 세포고사에 DNA수선 유전자가 밀접한 관련이 있을 것으로 보이는데, 특히 최근에 발견된 hRAD50 유전자도 세포고사와 밀접한 관련이 있을 것으로 사료되었다.

Exploiting cDNA Microarray-Based Approach Combined with RT-PCR Analysis to Monitor the Radiation Effect: Antioxidant Gene Response of ex vivo Irradiated Human Peripheral Blood Lymphocyte

  • Although ionizing radiation (IR) has been used to treat the various human cancers, IR is cytotoxic not only to cancer cells but to the adjacent normal tissue. Since normal tissue complications are the limiting factor of cancer radiotherapy, one of the major concerns of IR therapy is to maximize the cancer cell killing and to minimize the toxic side effects on the adjacent normal tissue. As an attempt to develop a method to monitor the degree of radiation exposure to normal tissues during radiotherapy, we investigated the transcriptional responses of human peripheral blood lymphocytes (PBL) following IR using cDNA microarray chip containing 1,221 (1.2 K) known genes. Since conventional radiotherapy is delivered at about 24 h intervals at 180 to 300 cGy/day, we analyzed the transcriptional responses ex-vivo irradiated human PBL at 200 cGy for 24 h-period. We observed and report on 1) a group of genes transiently induced early after IR at 2 h, 2) of genes induced after IR at 6 h, 3) of genes induced after IR at 24 h and on 4) a group of genes whose expression patters were not changed after IR. Since Biological consequences of IR involve generation of various reactive oxygen species (ROS) and thus oxidative stress induced by the ROS is known to damage normal tissues during radiotherapy, we further tested the temporal expression profiles of genes involved in ROS modulation by RT-PCR. Specific changes of 6 antioxidant genes were identified in irradiated PBL among 9 genes tested. Our results suggest the potential of monitoring post-radiotherapy changes in temporal expression profiles of a specific set of genes as a measure of radiation effects on normal tissues. This type of approach should yield more useful information when validated in in vivo irradiated PBL from the cancer patients.

Salt-Responsive Genes in Salt Tolerant Rice Mutants Revealed through Microarray Analysis

  • Transcriptional regulation in response to salt in mutant lines was investigated using oligonucleotide microarrays. In order to characterize the salt-responsive genes in rice, the expression profiles of transcripts that responded to salt-treatment were monitored using the microarrays. In the microarray analysis, among 37,299 reliable genes, 5,101, 2,758 and 2,277 genes were up-regulated by more than 2-fold using the salt treatment, while the numbers of down-regulated genes were 4,619, 3,234, and 1,878 in the WT, ST-495, and ST-532, respectively. From genotype changes induced by gamma ray mutagenesis, 3,345 and 2,397 genes were up-regulated, while 2,745 and 2,075 genes were down-regulated more than 2-fold in the two untreated mutants lines compared with untreated WT, respectively. A total of 3,108 and 2,731 genes were up-regulated more than 2-fold, while 3,987 and 3,660 genes were down-regulated by more than 2-fold in the salt treatment of the two mutants lines compared with the salt treated WT, respectively. The expressions of membrane transporter genes such as OsAKT1, OsKUP, and OsNAC increased more severely in ST-495 and ST-532 than in the WT. The expressions of the proline accumulation related genes such as OsP5CS and OsP5CR were also markedly increased in the salt tolerant mutants when compared to the WT plant.

Expression of Antioxidant Isoenzyme Genes in Rice under Salt Stress and Effects of Jasmonic Acid and ${\gamma}$-Radiation

  • Analysis of chlorophyll (Chl) fluorescence implicated treatment of 40 mM NaCl decreased maximal photochemical efficiency of photosystem II (PSII) (Fv/Fm), actual quantum yield of PSII (${\Phi}_{PSII}$), and photochemical quenching (qP) in rice, but increased non-photochemical quenching (NPQ). Decreases in Fv/Fm, ${\Phi}_{PSII}$, and qP were significantly alleviated by $30\;{\mu}M$ jasmonic acid (JA), while NPQ increase was enhanced. Transcription levels of antioxidant isoenzyme genes were differentially modulated by NaCl treatment. Expression of cCuZn-SOD2 gene increased, while those of cAPXb, CATb, and CATc genes decreased. JA prevented salt-induced decrease of pCuZn-SOD gene expression, but caused greater decrease in mRNA levels of cAPXa and Chl_tAPX genes. Investigation of vacuolar $Na^+/H^+$ exchanger (NHX2) and 1-pyrroline-5-carboxylate synthetase (P5CS) gene expressions revealed transcription level of NHX2 gene was increased by JA, regardless of NaCl presence, while that of P5CS gene slightly increased only in co-presence of JA and NaCl. Unlike JA, ${\gamma}$-radiation rarely affected expressions of antioxidant isoenzyme, NHX2, and P5CS genes, except for increase in mRNA level of Chl_tAPX and decrease in that of pCuZn-SOD. These results demonstrate enhanced salt-tolerance in JA-treated rice seedlings may be partly due to high transcription levels of pCuZn-SOD, NHX2, and P5CS genes under salt stress.

MCF-7 세포주의 γ선에 의한 DNA 손상 반응 유전자 발현 양상의 분석 (A DNA-Damage Response Gene Expression Analysis in MCF-7 followed by γ-Radiation)

  • Cell response to genotoxic agents is complex and involves the participation of different classes of genes including cell cycle control, DNA repair and apoptosis. In this report, we presented a approach to characterize the cellular functions associated with the altered transcript profiles of MCF-7 exposed to low-dose in vitro gamma-irradiation. We used the method of human 2.4 k cDNA microarrays containing apoptosis, cell cycle, chromatin, repair, stress and chromosome genes to analyze the differential gene expression characterization that were displayed by radiation-exposed cell, human breast carcinoma MCF-7 cell line, such as 4 Gy 4 hr, 8 Gy 4 hr, and 8 Gy 12 hr. Among these genes, 66 were up-regulated and 49 were down-regulated. Specific genes were concomitantly induced in the results. Cyclin dependent kinase 4 (Cdk4) is induced for starting the cell cycle. This regulation is required for a DNA damage­induced G1 arrest. In addition to, an apoptotic pathways gene Bcl-w was concomitantly induced. Mismatch repair protein homologue-l (hMLH1), a necessary component of DNA mismatch protein repair (MMR), in G2-M cell cycle checkpoint arrest. The present study provides new information on the molecular mechanism underlying the cell response to genotoxic stress, with relevance to basic and clinical research.

Radiation-induced Tumorigenesis

  • During the past 2 decades, radiation tumorigenesis researchers have focused on cellular and molecular mechanisms. We reviewed some of these research fields, since they may specifically relate to the induction of cancer by ionizing radiation. First, radiation-mediated mutation was discussed. Then the initiating event in radiation carcinogenesis, as well as other genetic events that may by involved, is discussed in terms of the possible role of the activation of genes and the loss of cell-cycle checkpoints.
