Effects of Ultraviolet-B Radiation on Growth and Photosynthesis in Cucumber Primary Leaves

  • In the present study we studied the growth, photosynthetic traits and protective mechanisms against oxidative stress in the primary loaves of cucumber (Cucumis sativus L.) seedlings with or without UV-B treatment. Cucumber seedings were irradiated with UV-B for 10 days in environment-controlled growth chambers. The primary leaves irradiated with UV-B showed reduction in leaf length and decreased biomass production. The reduced biomass production seemed to be due to a negative effect of UV-B radiation on the photosynthetic process. Changes in chemical properties of leaf, such as chi a/b ratio affected photosynthesis. UV-B significantly affected chl b content compared with chi a in the light harvesting complex resulting reduced photosynthetic activity Fv/Fm decreased with an UV-B stress, suggesting that the photosynthetic apparatus, and particularly, PS II was damaged under UV-B stress. Malondialdehyde(MDA) concentration which represents the state of membrane lipid peroxidation Increased significantly under UV-B stress confirming an oxidative stress. UV-B exposure with SA solution(0.1-1.0 mM) can partially ameliorated some of the detrimental effects of UV-B stress. Leaf injuries including loss of chlorophyll and decreased ratio of Fv/Fm were reduced with combined application of UV-B and SA. ABA and JA showed similar mode of action in physiological effects on photosynthetic activities though the levels were lower than those from SA treated plants. Chloroplast ultrastructure was also affected by UV-B exposure. The thickness of leaf tissue components decreased and the number of grana and thylakoids was reduced in chloroplast applied UV-B or SA alone. At combined stress granal and stromal thylakoids were less affected. The leaves under combined stress acquired a significant tolerance to oxidative stress. From these results, it can be suggested that SA may have involved a protective role against UV-B induced oxidative damage.

라디에이션 응력의 재평가 (Re-estimation of Radiation Stress)

  • 일반적으로 선형파동이론에 의해 산정된 라디에이션 응력은 쇄파대내 및 부근에서 과대 평가되는데, 이것이 평균수위변동의 계산치와 실험치와의 불일치의 원인이 된. 오(1995)는 Svendsen (1984)이 제안한 라디에이션 응력을 이용하여 평균수위변동을 산정하고 실험치와 비교한 결과, 파형경사가 작은 경우에는 잘 일치하고 있으나 파형경사가 큰 경우에는 실험치와 잘 맞지 않는 것을 보였다. 본 연구에서는 Svendsen(1984)이 제안한 무차원 라디에이션 상수를 수정하여 쇄파점에서의 상대수심 및 심해파 파형경사의 함수로 표현하여 계산하고, 그 결과를 선형이론, Stive(1984)모델, 쇄파의 성분파고 스펙트럼에 기초한 Sawaragi 등(1984)의 모델 및 기왕의 실험 데이터와 비교하였다. 수정, 제안된 Svendsen 모델의 결과는 실험치와 매우 양호한 일치를 보여 주었다.

방사선으로 인한 산화적 손상에서 phloroglucinol의 모낭 보호 효과 (Protective effect of phloroglucinol against gamma radiation-induced oxidative stress in hair follicles)

  • When exposed to gamma-rays, hair follicular cells immediately go through apoptosis, which hampers their rapid differentiation essential for the regeneration of hair. Phloroglucinol (PG) is a phenolic compound of Ecklonia cava, brown algae abundant in Jeju island, Korea. Containing plentiful polyphenols, PG is known for its instructive effects by inhibiting apoptosis, scavenging oxygen radicals, and protecting cells against oxidative stress. In this study, we demonstrate that PG rescues radiosensitive hair follicular cells from gamma radiation-induced apoptosis and DNA damage. To identify protective capacity of PG on hair follicles, we irradiated with 8.5 Gy (1.5 Gy/min) of gamma-rays to the whole body of C57BL/6 mice at day 6 after depilation with or without PG. In mice exposed to radiation, the expression of proapoptotic molecule p53 was downregulated in the skin of PG treated group. On immunohistochemical observation of the skin, PG inhibited the immunoreactivity of p53 and cleaved caspase-3. PG treatment protected hair follicular cells from cell death due to gamma-radiation. Our results suggest that PG presents radioprotective effects by inhibiting apoptosis of radiosensitive hair follicular cells and can protect hair follicular cells from gamma-ray induced damage.

약용식물 추출물의 산화적 스트레스에 대한 방어효과 (Protective Effect against Oxidative Stress in Medicinal Plant Extracts)

  • 본 연구에서는 약용식물 추출물울 대상으로 하여 산화적 스트얘스에 대한 방어효과를 검색하였다. 항산화 또는 항염증 효과가 보고된 총 48 종의 약용식물에서 메탄올 추출물 시료를 제조하고 1차적으로 이들의 과산화수소수에 의해 유도된 산화적 스트레스에 대한 방어활성을 검색하였다. 이들 중 33 시료에서 다양한 정도의 활성이 검색되었으며, 그 중 감초의 메탄올 추출물 시료가 $100{\mu}g/m{\ell}$의 농도에서 25.9% 의 가장 높은 활성을 보였다. 그 외 $100{\mu}g/m{\ell}$의 농도에서 20% 이상의 방어 활성이 나타난 시료는 감인, 단삼, 두충, 목단피, 부평초, 연자육 등이었다. 우수한 활성이 검색된 시료에 대하여 감마선에 의해 유도된 산화적 스트레스에 대한 방어활성을 검색한 결과 양강의 메탄올 추출물에서 17.8% (at $20{\mu}gm{\ell}$)의 가장 높은 활성이 검색되었다. 방어 활성이 우수한 10개의 시료에 대하여 각각 5개의 분획시료률 제조하여 방어활성을 검색하였다. 이 들 분획시료 중 빈랑자 및 부평초의 butanol 분획이 각각 78.8% 및 77.2% 로 가장 우수한 방어 활성이 검색되었다.

Protective Activity against Ionizing Radiation of Antioxidative Plants Indigenous to Korea

  • We have screened the cytoprotective effect on ${\gamma}-ray$ radiation induced oxidative stress from forty one Korean plant extracts. Carpinus laxiflora (caulis), Quercus salicina (caulis), and Castanopsis cuspidata (caulis) were found to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and intracellular reactive oxygen species (ROS). As a result, extracts of three plants reduced cell death of Chinese hamster lung fibroblast (V79-4) cells induced by $H_2O_2$ treatment. In addition, these extracts protected cell death of V79-4 cells damaged by ${\gamma}-ray$ radiation. In addition, these extracts scavenged ROS generated by radiation. Taken together, the results suggest that Carpinus laxiflora, Quercus salicina, and Castanopsis cuspidata protect V79-4 cells against oxidative damage by radiation through scavenging ROS.

  • The immune system is specifically sensitive to oxidative stress induced by ionizing radiation because of its rapid proliferative activity. For this reason, an instructive immune system is one of the best ways to minimize side effects, such immunodeficiency, of gamma radiation. Over the past few decades, several natural plants with antioxidant and immunomodulatory properties have been identified as adjuncts for nontoxic and successful radiotherapy. Hizikia fusiforme extract (HFE) containing plentiful dietary fiber and fucoidan is known for its instructive antioxidant capacity, immunomodulation abilities, and immune activation. In this study, we determined whether HFE protects radiosensitive immune cells from gamma radiation-induced damage. C57BL/6 mice were irradiated with gamma-ray. The effect of HFE on the ionizing radiation damage of immune cells was then evaluated with an MTT assay, 3H-thymidine incorporation assay, and PI staining. We found that HFE stimulated the proliferation of gamma-ray irradiated immune cells without cytotoxic effects. We also observed that HFE not only decreased DNA damage but also reduced gamma radiation-induced apoptosis of the immune cells. Our results suggest that HFE can protect immune cells from gamma-ray damage and may serve as an effective, non-toxic radioprotective agent.

Effects of Water Deficit and UV-B Radiation on Accumulation of Functional Metabolites in Crops: A Review

  • With increasing social concerns for healthy food, the studies on the cultivation of crops to increase accumulation of functional metabolites in crops have been investigated. Accumulation of the metabolites in crops is highly affected by various types of stress, such as nutrient deficiency, water deficit (WD), extreme temperature and UV-B radiation as well as their own life cycle. This review summarizes the previous studies on the effects of environmental stresses, especially WD and UV-B radiation, on accumulation of functional metabolites in crops. UV-B radiation and WD during specific period (mainly at maturation stage) activates the adaptation and/or defense system in crops, thereby increasing biosynthesis of the metabolites. Although WD and UV-B radiation tend to decrease in crop yield, the decrease can be compensated by the production of high value crops having high content of functional metabolites.

Purpurogallin Protects Keratinocytes from Damage and Apoptosis Induced by Ultraviolet B Radiation and Particulate Matter 2.5

  • Purpurogallin, a natural phenol obtained from oak nutgalls, has been shown to possess antioxidant, anticancer, and anti-inflammatory effects. Recently, in addition to ultraviolet B (UVB) radiation that induces cell apoptosis via oxidative stress, particulate matter 2.5 ($PM_{2.5}$) was shown to trigger excessive production of reactive oxygen species. In this study, we observed that UVB radiation and $PM_{2.5}$ severely damaged human HaCaT keratinocytes, disrupting cellular DNA, lipids, and proteins and causing mitochondrial depolarization. Purpurogallin protected HaCaT cells from apoptosis induced by UVB radiation and/or $PM_{2.5}$. Furthermore, purpurogallin effectively modulates the pro-apoptotic and anti-apoptotic proteins under UVB irradiation via caspase signaling pathways. Additionally, purpurogallin reduced apoptosis via MAPK signaling pathways, as demonstrated using MAPK-p38, ERK, and JNK inhibitors. These results indicate that purpurogallin possesses antioxidant effects and protects cells from damage and apoptosis induced by UVB radiation and $PM_{2.5}$.

양산형 일광차단모의 개발(제1보) -고안 및 마네킹 착용실험- (Development of the sunshade hat with a large brim(Part I) -Development and test with manikins-)

  • This study was to develop the sunshade hat which reduced stress from solar radiation and ultraviolet radiation (UV), in order to keep the farmer's health and to promote their work efficiency. The new sunshade hat with a large brim, special structure for ventilation, stability and portability was designed and tested with manikin heads outdoors. Two newly designed sunshade hats(A, B) and three existing hat were tested Sunshade hat A made of double fabric with aluminum coating-nylon and black cotton cloth with a polyester mix(T/C) was the most protective from solar radiation. Sunshade hat B with larger brim was the most protective from ultraviolet radiation, even though it was made of aluminum coating-nylon single fabric.

Retrospective dosimetry using fingernail electron paramagnetic resonance response

  • Human fingernails were used to estimate the radiation dose via electron paramagnetic resonance measurements of radiation-induced radicals. The limiting factors in this research were mechanically induced electron paramagnetic resonance signals due to the mechanical stress during the preparation of the samples. Therefore, different treatment methods of fingernails were used to reduce the mechanically induced signals. The results demonstrate that the mechanically induced and radiation-induced signals have apparently different microwave power saturation behaviors. In addition, the mechanically induced signal shows a fading evolution over time and reaches a constant value. Chemical treatment using the different reagents showed that the minimum mechanically induced signal was obtained using the dithiothreitol reagent. The dose-response curves of the samples treated with dithiothreitol for 30 minutes demonstrated a greater linearity than those of samples treated for 5 minutes. Therefore, to find an unknown absorbed dose in a fingernail sample using a calibration curve, we recommend adopting the mentioned chemical treatment procedure to reduce the uncertainty.
