Development and Evaluation of a SYBR Green Real-time PCR Assay for Canine Cytokine Gene Expression (SYBR Green 실시간 역전사 중합효소연쇄반응을 이용한 개 싸이토카인 유전자 발현의 정량)

  • Cytokines are important mediators of the immune response, and quantitating cytokine mRNA is a highly sensitive and attractive method for measuring cytokine production. The objective of the current study was to develop and validate a SYBR green quantitative real-time reverse transcriptase PCR (qRT-PCR) assay for measuring canine cytokine mRNA. The optimal annealing temperatures ($T_a$) of the designed primers were $62^{\circ}C$ for interleukin (IL)-$1{\beta}$, IL-6 and IL-10; $60^{\circ}C$ for glyceraldehyde 3-phosphate dehydrogenase (GAPDH) and tumor necrosis factor (TNF)-${\alpha}$; and $58^{\circ}C$ for high mobility group box 1 (HMGB1). Primer efficiencies of all primers calculated for standard curve samples were between 97.1% and 102.6%. No evidence of secondary structure or primer-dimer formation was seen via melt-curve analysis or gel electrophoresis. The developed qRT-PCR assays are highly specific and sensitive and can be used to quantify gene expression levels of canine cytokines.

Derivation of External Exposure Characteristics of Industrial Radiography Based on Empirical Evidence

  • Background: This study aims to derive the characteristics of each work type for industrial radiography based on empirical evidence through expert advice and a survey of radiation workers of various types of industrial radiography. Materials and Methods: According to a Korean report, work types of industrial radiography are classified into indoor tests, underground pipe tests, tests in a shielded room (radiographic testing [RT] room test), outdoor field tests, and outdoor large structure tests. For each work type, exposure geometry and radiation sources were mainly identified through the expert advice and workers' survey as reliable empirical evidence. Results and Discussion: The expert advice and survey results were consistent as the proportion of the work types were high in the order of RT room test, outdoor large structure test, underground pipe test, outdoor field test, and indoor test. The outdoor large structure test is the highest exposure risk work type in the industrial radiography. In most types of industrial radiography, radiation workers generally used 192Ir as the main source. In the results of the survey, the portion of sources was high in the order of 192Ir, X-ray generator, 60Co, and 75Se. As the exposure geometry, the antero-posterior geometry is dominant, and the rotational and isotropic geometry should be also considered with the work type. Conclusion: In this study, through expert advice and a survey, the external exposure characteristics for each work type of industrial radiography workers were derived. This information will be used in the reconstruction of organ dose for health effects assessment of Korean radiation workers.

A Study on Optimum Mass of TMD for Improving Seismic Response Control Performance of Retractable-Roof Spatial Structure (개폐식 대공간 구조물의 지진 응답 제어 성능 향상을 위한 TMD의 최적 질량에 관한 연구)

  • In this study, the retractable-roof spatial structure was chosen as the analytical model and a tuned mass damper (TMD) was installed in the analytical model in order to control the seismic response. The analysis model is mainly consisted of runway trusses (RT) and transverse trusses (TT), and the displacement response was analyzed by installing TMD on those trusses. The mass of the single TMD which is installed in the analytical model was set to 1% of the total structure mass and the total TMD mass ratio was set to be 8% or 6%. In addition, the mass of a single TMD was varied depending on the number of installations. As a result of analyzing the optimal number of installations of TMD, the displacement response was reduced in all cases compared to the case without TMD. Above all, the case with 8 TMDs was the most effective in reducing he displacement response. However, in this case, as the load on the upper structure of the retractable-roof spatial structure increases, the total mass ratio of TMD was maintained and the number of TMDs was increased to reduce the mass ratio of one TMD.

  • Precedent researches on lyocell are mostly on lyocell structure, characteristics, physical properties by finishing and dyeability, and the like; there are little cases of researches on interlining for lyocell clothes production. To serve as a base information helpful to select fusible interlining and improve the efficiency of sewing process considering the characteristics of lyocell, this study performed t-est, F-test toward to the difference in the appearance-related properties and the mechanical properties of lyocell-interlining bonded fabric in accordance with the characteristics of fusible interlining and lyocell face fabric. This study reached following conclusions. Drapability and Crease recovery of lyocell-interlining bonded fabric are influenced by the structure of fusible interlining; stiffness of lyocell-interlining bonded fabric, by the structure of fusible interlining and the yam number of lyocell face fabric. WT is influenced by the mixture rate and yam number of lyocell face fabric. RT is influenced by the yam number of lyocell face fabric; G, 2HG, 2HG5, by the structure(woven and non-woven) of fusible interlining; B, HB, by the weight, structure(woven and non-woven) of fusible interlining, and yam number of lyocell face fabric; WC, by the weight, structure of fusible interlining; RC, by the structure of fusible interlining; thickness, by the weight of fusible interlining and the yarn number of lyocell face fabric; weight, by mixture rate and yam number of lyocell face fabric.

  • We have studied on the SLDs utilizing InAs chirped QD structure. The output power and spectral bandwidth are obtained as CW 40 mW at RT and about 100 nm, respectively. More high performance of SLD can be possible with optimized design for the chirped QD structures.

Cloning, Expression and Hormonal Regulation of Steroidogenic Acute Regulatory Protein Gene in Buffalo Ovary

  • In mammalian ovary, steroidogenic acute regulatory (StAR) protein mediates the true rate-limiting step of transport of cholesterol from outer to inner mitochondrial membrane. Appropriate expression of StAR gene represents an indispensable component of steroidogenesis and its regulation has been found to be species specific. However, limited information is available regarding StAR gene expression during estrous cycle in buffalo ovary. In the present study, expression, localization and hormonal regulation of StAR mRNA were analyzed by semi-quantitative RT-PCR in buffalo ovary and partial cDNA was cloned. Total RNA was isolated from whole follicles of different sizes, granulosa cells from different size follicles and postovulatory structures like corpus luteum and Corpus albicans. Semi-quantitative RT-PCR analyses showed StAR mRNA expression in the postovulatory structure, corpus luteum. No StAR mRNA was detected in total RNA isolated from whole follicles of different size including the preovulatory follicle (>9 mm in diameter). However, granulosa cells isolated from preovulatory follicles showed the moderate expression of StAR mRNA. To assess the hormonal regulation of StAR mRNA, primary culture of buffalo granulosa cells were treated with FSH (100 ng/ml) alone or along with IGF-I (100 ng/ml) for 12 to 18 h. The abundance of StAR mRNA increased in cells treated with FSH alone or FSH with IGF-I. However, effect of FSH with IGF-I on mRNA expression was found highly significant (p<0.01). In conclusion, differential expression of StAR messages was observed during estrous cycle in buffalo ovary. Also, there was a synergistic action of IGF-I on FSH stimulation of StAR gene.

Plasticity and Fracture Behaviors of Marine Structural Steel, Part V: Effects of Strain Rate and Temperature (조선 해양 구조물용 강재의 소성 및 파단 특성 V: 온도 의존성을 고려한 변형률 속도에 관한 실험적 연구)

  • This is the fifth in a series of companion papers dealing with the dynamic hardening properties of various marine structural steels at intermediate strain rates. Five steps of strain rate levels (0.001, 1, 10, 100, 200/s) and three steps of temperature levels (LT ($-40^{\circ}C$), RT, and HT ($200^{\circ}C$)) were taken into account for the dynamic tensile tests of three types of marine structural steels: API 2W50 and Classifications EH36 and DH36. The total number of specimens was 180 pieces. It was seen that the effects of dynamic hardening became clearer at LT than at RT. Dynamic strain aging accompanying serrated flow stress curves was also observed from high temperature tests for all kinds of steels. The dynamic hardening factors (DHFs) at the two temperature levels of LT and RT were derived at the three plastic strain levels of 0.05, 0.10, 0.15 from dynamic tensile tests. Meanwhile, no DHFs were found for the high temperature tests because a slight negative strain rate dependency due to dynamic strain aging had occurred. A new formulation to determine material constant D in a Cowper-Symonds constitutive equation is provided as a function of the plastic strain rate, as well as the plastic strain level. The proposed formula is verified by comparing with test flow stress curves, not only at intermediate strain rate ranges but also at high strain rate ranges.

Biological, Physical and Cytological Properties of Pepper mottle virus-SNU1 and Its RT-PCR Detection

  • A strain of Pepper mottle virus (PepMov) was isolated from chili pepper plants in Korea. In host range study, this virus, designated PepMoV-SNU1, shared most characteristics with PepMoV isolates reported previously. Thermal inactivation point ($45^{\circ}C\;to\;75^{\circ}C$) and dilution end point ($10^{-1}\;to\;10^{-4}$) of PepMoV-SNU1 showed differences depending on the propagation hosts. Cylindrical and pinwheel-shaped inclusions were always observed in pepper leaf tissues infected with the virus alone. Unexpectedly, a special structure of pinwheel shaped inclusion surrounded with unknown small spots was also observed in the leaf section when co-infected with a strain of pepper mild mottle virus. The partial sequence of coat protein gene and 3' untranslated region of PepMoV-SNU1 showed 98% identity with those of other PepMoV isolates. A primer pair derived from 3' end of the coat protein gene and poly A tail regions were designed. Optimal detection condition of PepMoV-SNU1 by RT-PCR was tested to determine appropriate annealing temperature and additional volumes of oligo-dT (18-mer), dNTP, and Taq polymerase. Under the optimized condition, an expected 500 Up PCR-product was detected in pepper leaves infected with PepMoV-SNU1 but not in healthy plants.

Effects of Boshimgeonbi-tang on Gene Expression in Hypothalamus of Immobilization-stressed Mouse (보심건비탕(補心健脾湯) 투여가 Stress 유발 Mouse의 Hypothalamus 유전자 발현에 미치는 영향)

  • The genetic effects of restraint stress challenge on HPA axis and the therapeutic effect of Boshimgeonbi-tang on the stress were studied with cDNA microarray analyses, RT-PCR on hypothalamus using an immobilization-stress mice as an animal model. Male CD-1 mice were restrained in a tightly fitted and ventilated vinyl holder for 2hrs once a day, and this challenge was repeated for seven· consecutive days. In the change of body weight it showed that the Boshimgeonbi-tang is effected recovery on weight loss caused by the immobilization-stress. Seven days later, total RNA was extracted from the organs of the mouse, body-labeled with $CyDye^{TM}$ fluorescence dyes and then hybridized to CDNA microarray chip. Scanning and analyzing the array slides were carried out using GenePix4000 series scanner and GenePix $Pro^{TM}$ analyzing program, respectively. The expression profiles of 109 genes out of 6000 genes on the chip were significantly modulated in hypothalamus by the immobilization stress. Energy metabolism-, lipid metabolism-, apoptosis-, stress protein, transcriptional factor, and signal transduction-related genes were transcriptionally activated whereas DNA repair-, protein biosysthesis-, and structure integrity-related genes were down-regulated in hypothalamus. The 58 genes were up-regulated by the mRNA expression folds of 1.5 to 7.9. and the 51 genes were down-regulated by 1.5 - 5.5 fold. The 11 genes among them were selected to confirm the expression profiles by RT-PCR. The mRNA expression levels of Tnfrsf1a (apoptosis), Calm2 (cell cycle), Bag3 (apoptosis), Ogg1 (DNA repair), Aatk (apoptosis), Dffa (apoptosis), Fkbp5 (protein folding) were restored to the normal one by the treatment of Boshimgeonbi-tang.

Photoemission study on the reactivity of organic molecules on chemically modified TiO2(001) surfaces

  • Adsorption and subsequent catalytic reactions of ethanol and acetaldehyde on chemically modified rutile TiO2(001) surfaces are probed by x-ray photoemission spectroscopy (XPS) using synchrotron radiation. TiO2 is a well-known photocatalyst for various catalytic reactions including oxidation of organic molecules. In this respect, the surface atomic structure has been found to play a vital role in determining the catalytic reactivity and selectivity of TiO2. In this study, we employ an atomically well-ordered reduced TiO2(001) surface which is prepared in a UHV chamber by repeated Ar+-sputtering and annealing (900 K) cycles. We systematically modify the surface by treating the surface with H2O or O2 at room temperature (RT). The catalytic reactivity of the surface-modified TiO2(001) is evaluated by dosing ethanol/acetaldehyde onto the surface at RT and by subsequent annealing to higher temperatures (400~600 K). XPS spectra of C 1s core level are intensively used to probe any change in the oxidation state of carbon atoms. We find that the reactivity as well as the saturation coverage are significantly affected by the RT-treatment of the TiO2 surface with H2O or O2. For both reactant molecules (ethanol/acetaldehyde), oxidation reactions are found to be enhanced on the O2-treated surface compared with the reduced or H2O-treated surfaces. Possibly reaction pathways are discussed based on the observed XPS spectra.
