Reverse Transcription Recombinase Polymerase Amplification Assay for Rapid and Sensitive Detection of Barley Yellow Dwarf Virus in Oat

  • Barley yellow dwarf virus (BYDV) is an economically important plant pathogen that causes stunted growth, delayed heading, leaf yellowing, and purple leaf tip, thereby reducing the yields of cereal crops worldwide. In the present study, a reverse transcription recombinase polymerase amplification (RT-RPA) assay was developed for the detection of BYDV in oat leaf samples. The RT-RPA assay involved incubation at an isothermal temperature (42℃) and could be performed rapidly in 5 min. In addition, no cross-reactivity was observed to occur with other cereal-infecting viruses, and the method was 100 times more sensitive than conventional reverse transcription polymerase chain reaction. Furthermore, the assay was validated for the detection of BYDV in both field-collected oat leaves and viruliferous aphids. Thus, the RT-RPA assay developed in the present study represents a simple, rapid, sensitive, and reliable method for detecting BYDV in oats.

  • The purpose of this study was to evaluate the stress distribution developed in supporting structures by distal extension removable partial denture with 4-types of direct retainer. The direct retainers examined were Akers clasp, RPI clasp, RPA clasp and RPL clasp in bilateral & unilateral free end case. 3-dimensional photoelastic stress analysis was used to record the isochromatic fringe patterns and to calculate the compressive stress at measuring points. The results were as follows. 1. In bilateral free end case, RPI clasp exhibited the similar stress distribution on distal and mesial alveolar crest but Akers clasp exhibited higher stress concentration on distal alveolar crest than mesial alveolar crest. 2. In bilateral free end case, RPA clasp and RPL clasp exhibited the similar stress distribution on distal and mesial alveolar crest and RPL clasp exhibited higher stress concentration on buccal alveolar crest than lingual alveolar crest. 3. Akers clasp produced high stress concentration on residual alveolar ridge distally, but RPI clasp, RPA clasp and RPL clasp produced even stress distribution on residual alveolar ridge. 4. Removable partial denture in unilateral free end case exerted higher stress on abutment tooth root apex than bilateral distal extension removable partial denture.

  • Many studies have focused on the influences and outcomes of online game addiction. However, few studies have investigated how the online game addiction prevention behavior differs depending on online game user. In order to fill this gap, this study attempts to classify online game users based on risk perception attitude (RPA) framework. More specifically, this research tries to show online game user can be classified into four groups based on perceived risk of online game addiction and efficacy beliefs of online game addiction protection, and to identify how the groups differ in terms of motivation, information seeking, and behaviors for online game addiction prevention. For this purpose, analysis on survey data from 240 Korean college students who use online game reveals that the users can be classified into responsive, avoidance, proactive, indifference groups. Furthermore, there are differences between groups in terms of motivation, information seeking, and behaviors for online game addiction prevention. This study contributes to expand existing literature by providing tailored guidelines for implementation of online game addiction prevention strategies and policy.

  • Seismic analysis of local site conditions is fundamental for a reliable site seismic hazard assessment. It plays a major role in mitigation of seismic damage potential through the prediction of surface ground motion in terms of amplitude, frequency content and duration. Such analysis requires the determination of the transfer function, which is a simple tool for characterizing a soil profile by estimating its vibration frequencies and its amplification potential. In this study, numerical simulations are carried out and are then combined with a statistical study to allow the characterization of design sites classified by the Algerian Building Seismic Code (RPA99, ver 2003), by average transfer functions. The mean transfer functions are thereafter used to compute RPA99 average site factors. In this regard, coming up seismic fields are simulated based on Power Spectral Density Functions (PSDF) defined at the rock basement. Results are also used to compute average site factor where, actual and synthetic time histories are introduced. In absence of measurement data, it is found that the proposed approach can be used for a better soil characterization.

A Theory on Phase Behaviors of Diblock Copolymer/Homopolymer Blends

  • The local structural and thermodynamical properties of blends A-B/H of a diblock copolymer A-B and a homopolymer H are studied using the polymer reference interaction site model (RISM) integral equation theory with the mean-spherical approximation closure. The random phase approximation (RPA)-like static scattering function is derived and the interaction parameter is obtained to investigate the phase transition behaviors in A-B/H blends effectively. The dependences of the microscopic interaction parameter and the macrophase-microphase separation on temperature, molecular weight, block composition and segment size ratio of the diblock copolymer, density, and concentration of the added homopolymer, are investigated numerically within the framework of Gaussian chain statistics. The numerical calculations of site-site interchain pair correlation functions are performed to see the local structures for the model blends. The calculated phase diagrams for A-B/H blends from the polymer RISM theory are compared with results by the RPA model and transmission electron microscopy (TEM). Our extended formal version shows the different feature from RPA in the microscopic phase separation behavior, but shows the consistency with TEM qualitatively. Scaling relationships of scattering peak, interaction parameter, and temperature at the microphase separation are obtained for the molecular weight of diblock copolymer. They are compared with the recent data by small-angle neutron scattering measurements.

Detection of Apple Scar Skin Viroid by Reverse Transcription Recombinase Polymerase Amplification Assay

  • The aim of the present study was to develop a sensitive and specific detection method for the rapid detection of apple scar skin viroid (ASSVd) in apple leaves. The resulting reverse transcription recombinase polymerase amplification (RT-RPA) assay can be completed in 10 min at 42℃, is 10 times more sensitive than conventional reverse transcription polymerase chain reaction, and can specifically amplify ASSVd without any cross-reactivity with other common apple viruses, including apple stem grooving virus, apple stem pitting virus, and apple chlorotic leaf spot virus. The reliability of the RT-RPA assay was assessed, and the findings suggested that it can be successfully utilized to detect ASSVd in field-collected samples. The RT-RPA assay developed in the present study provides a potentially valuable means for improving the detection of ASSVd in viroid-free certification programs, especially in resource-limited conditions.

Analysis of Essential Oils from the Peel of Mandarine (Citrus unshiu Marc. Var. Okitsu) (온주 밀감의 껍질로부터 정유의 분리 및 분석)

  • Essential oils were isolated from the peel of mandarin (Citrus unshiu Marc. Var. Okitsu) cultivated in Cheju Island. Among three different isolation methods investigated, simultaneous distillation gave higher yield 1.14%(w/w) than solvent extraction or cold pressing. The densities of essential oils were between 0.8409 and 0.8530, which showed no significant effect of the isolation methods. Some variations in the number of constituents detected and the content of d-limonene in the peel oils were observed depending upon the isolation methods. The citrus fruit was collected seasonally from the field and used as a sample for analysis. The peel thickness and the peel content of citrus fruit were around 2 mm and about 17% on wet weight basis, respectively, in harvest season. The maximum yield of essential oils was obtained in September from citrus peels just prior to ripeness and thereafter the yield had a tendency of decrease but remained over 1%(w/w). The relative content of d-limonene, the major constituent of citrus peel oils. increased gradually with ripening up to 68.69%(relative peak area, RPA). The other minor constituents were ${\gamma}-terpinene,\;{\beta}-elemene$, farnesene, hexadecanoic acid, ${\alpha}-pinene,\; {\beta}-myrcene$ and linalool in the decreasing order of their own contents of 7.75, 2.96, 2.29, 1.76, 1.63, 1.56 and 1.46%(RPA).

  • The PBA of ship weapon system should be installed and operated under harsh environmental conditions and so it should be highly reliable to endure the mission profiles during its entire lifetime. In the case of PBA failure during operation, rapid maintenance is highly likely to be difficult due to problems such as supply of parts, which can have a devastating effect on the mission. In order to validate the reliability of PBA, a series of tests are performed with PBA samples, but they require time, testing facilities, samples, expenses and failure analysis if failed. The reliability of PBA is predicted on the basis of specifications such as MIL-HDBK-217F, but this specification does not take into account failure mechanisms for specific design details, environment and usage, interconnects and its characteristics that drive many failures of PBA in the field. Therefore, this study predicts the reliability of PBA using an RPA tool and proposes the RPA methodology as a validation process at the design stage. With RPA, it is now possible to achieve design validation including inherent failure mechanism, identification of weakest link, alternative design options, and test plan development.
