  • The COVID-19 pandemic is not over despite the emergency use authorization as can see recent COVID-19 daily confirmed cases. The viruses are not only difficult to diagnose and treat due to random mutations, but also pose threat human being because they have the potential to be exploited as biochemical weapons by genetic manipulation. Therefore, it is inevitable to the rapid antibody-based therapeutic platform to quickly respond to future pandemics by new/re-emerging viruses. Although numerous researches have been conducted for the fast development of antibody-based therapeutics, it is sometimes hard to respond rapidly to new viruses because of complicated expression or purification processes for antibody production. In this study, a novel rapid antibody-based therapeutic platform using single B cell sorting method and mRNA-antibody. High immunogenicity was caused to produce antibodies in vivo through mRNA-antigen inoculation. Subsequently, antigen-specific antibody candidates were selected and obtained using isolation of B cells containing antibody at the single cell level. Using the antibody-based therapeutic platform system in this study, it was confirmed that novel antigen-specific antibodies could be obtained in about 40 days, and suggested that the possibility of rapid response to new variant viruses.

A Subpopulation of RNA3 of Cucumber mosaic virus Quasispecies

  • This study examined the existence of genetically diverse population of Cucumber mosaic virus (CMV), known as quasispecies, from lily, Nicotiana benthamiana and from purified virions. Based on the conserved sequences of CMV lily isolates in intergenic region (IR) on RNA3, the genetic variation of IR from three different sources was investigated by a specific restriction endonuclease hydrolysis of amplified reverse transcription-polymerase chain reaction (RT-PCR) products using virus-specific primers, and was compared with IR sequences. The IR nucleotide sequences of CMV lily isolates were highly conserved, however, quasispecies was detected from all three sources in low level, containing sub-populations of RNA3. These subpopulations of RNA3 were inoculated onto zucchini squash by in vitro transcripts from corresponding full-length cDNA clones together with Eny RNA1 and 2 transcripts. The systemic symptom of zucchini plants infected by these quasispecies was chlorotic spotting, which was milder than severe mosaic and stunt symptom caused by Eny-CMV. The severity of symptom was correlated with RNA accumulation of viruses. These results suggest that the genome of CMV lily isolates consists of quasispecies populations.

Hepatitis E Virus Papain-Like Cysteine Protease Inhibits Type I Interferon Induction by Down-Regulating Melanoma Differentiation-Associated Gene 5

  • Upon viral infection, the host cell recognizes the invasion through a number of pattern recognition receptors. Melanoma differentiation associated gene 5 (MDA5) and retinoic acid-inducible gene-I (RIG-I) recognize RNA molecules derived from invading viruses, activating down-stream signaling cascades, culminating in the induction of the type I interferon. On the other hand, viruses have evolved to evade type I interferon-mediated inhibition. Hepatitis E virus has been shown to encode a few antagonists of type I interferon and it is not surprising that viruses encode multiple mechanisms of viral evasion. In the present study, we demonstrated that HEV PCP strongly down-regulates MDA5-mediated activation of interferon ${\beta}$ induction in a dose-dependent manner. Interestingly, MDA5 protein expression was almost completely abolished. In addition, polyinosinic polycytidylic acid (poly(I:C))- and Sendai virus-mediated activation of type I interferon responses were similarly abrogated in the presence of HEV PCP. Furthermore, HEV PCP down-regulates several molecules that play critical roles in the induction of type I IFN expression. Taken together, these data collectively suggest that HEV-encoded PCP is a strong antagonist of type I interferon.

Detection of Puumala and Hantaan Viruses among Bats in Korea by Nested RT-PCR

  • Hantavirus is a genus of the Bunyaviridae family consisting following serotype groups: Hantaan, Seoul, Puumala, Prospect Hill, Thailand, Belgrade, Thotta palayam, Sin Nombre. Most of Hantavirus group have been associated with many clinically similar disease known collectively as hemorrhagic fever with renal syndrome (HFRS). Hantaan virus is the prototype of the genus hantavirus, originally isolated from Apodemus agrarius. Bat was found as a natural host for Hantaan virus in Lee's lab for the first time. Then, Hantaan-like virus was isolated Hantaan-like virus from bat. To identify hantaviruses that are present in Korea among bats, bats were collected from Jeong-Sun, Won-Joo, Chung-Ju and Hwa-Cheon area, RNA was isolated from lung and serum. RT-PCR was performed with a universal primer from M segment. Nested RT-PCR was carried out to differentiate Hantaan, Seoul and Puumala virus using serotype specific primers. As we expected, Hantaan viruses were detected in bats and Seoul virus was not detected. Interestingly, Puumala viruses were also detected in bats from Won-Ju, but not in other areas. Puumala virus is originally isolated from Clethrinomys glareolus, and cause light HFRS. Recently, Paradoxomis webbiana, a wild bird turn out to be a reservoir for Puumala virus in Korea. These data indicate that bat is a new natural reservoir of Puumala virus.

The Innate Immune Viral Sensors and Their Functional Crosstalk

  • The precise and elaborate regulation of signaling cascades by diverse cytoplasmic and endosomal antiviral sensors is crucial for maintaining immune homeostasis and defending against viral pathogens. Receptors and enzymes that recognize foreign nucleic acids play a pivotal role in inducing antiviral interferon programs, serving as the first line of defense against various DNA and RNA viruses. Recent research has increasingly highlighted the crosstalk between nucleic acid sensors in detecting multiple virus invasions, resulting in amplified antiviral signals and compensating for any missing roles. This review provides an update on recent findings regarding the interplay of RNA sensors for DNA virus recognition.

Inhibitory Effects of Antisense RNA on Expression of Cholesteryl Ester Transfer Protein in Vaccinia Virus Expression System

  • Cholesteryl ester transfer protein (CETP), a hydrophobic glycoprotein promoting transfer of cholesteryl esters (CE) from high-density lipoproteins (HDL) to lower-density lipoproteins in the plasma, has been recognized a potent atherogenic factor during the development of coronary artery diseases. This study demonstrated a possible utilization of antisense RNA to inhibit expression of the CETP gene using vaccinia virus as an expression system. The CETP cDNA was inserted into a transfer vector (pSC11) in sense and antisense orientations and used to generate recombinant viruses. Recombinants containing sense or antisense orientations of the CETP cDNA were isolated by $TK^-$ selection and X-gal test. The inserted CETP cDNAs in the recombinants were identified by Southern blot analysis and allowed to transcribe in host cells (CV-1). Expressions of the exogenous CETP mRNA, extracted from the CV-1 cells coinfected with viruses containing sense and antisense DNAs, were monitored by Northern blot analysis using the CETP cDNA probe, by Western blot analysis using monoclonal antibody against the C-terminal active region of the CETP and by the CETP assay. Decreased expressions of the exogenous CETP cDNA were clearly evident in the Northern and Western blot analyses as the dose of antisense expression increased. In the CETP assay, the CETP activities decreased compared to the activity obtained from the cell extracts infected with sense construct only.

In vitro Evaluation of AraC Prodrugs for Their Antiviral Activity

  • The araC prodrugs (1~5) carrying a special acyl group at 5'-Ο-or $N^4$-position were evaluated for in vitro antiviral activity against various human viruses. When tested against HSV-1 and HSV-2 cultured in the verso cells, the prodrugs exhibited slightly higher $ED_{50}$ values compared with one of the parent araC but showed more increased $CC_{50}$ values in all cases. Consequently the overall selectivity indexes of prodrugs were higher than that of arab. The prodrugs, except compound 5, exhibited very potent activity similar to that of araC ($ED_{50}$ about $0.12{\mu}g/mι$) when evaluated against another human DNA virus, cytomegarovirus. However, theses araC prodrugs were completely inactive against RNA viruses i.e. poliovirus and coxackie B3 virus at the concentration of 4250{\mu}g/mι.$

Identification of Chinese Yam Necrotic Mosaic Virus(ChYNMV) infecting Chinese yam(Dioscorea opposita Thunb. cv. Jang-Ma) in Korea.

  • Chinese yam(D. opposita Thunb. cv. Jang-Ma) plants showing necrotic mosaic symptom were collected from their growing fields in Andong, Korea. Electron microscoptic examination of negatively stained preparations showed filamentous particles of 660nm in length. The virues purified partially were used to isolate Viral RNA as template for RT-PCR to amplify the CP gene with ChYNMV specific and oligo dT primers. Amino acids sequeces revealed that the viruses shared 99.3% similarity with ChYNMV(AB044386) which was known as the member of macluravirus. So the viruses from Chinese yam(D. opposita Thunb. cv. Jang-Ma) plants were identified as ChYNMV. Comparing the amino aced sequences of ChYNMV strains with other macluraviruses such as CdMV, NLV and MacMV revealed that N-terminal was the most variable region and conserved regions were present within the genus Macluravirus.

Original Antigenic Sin Response to RNA Viruses and Antiviral Immunity

  • The human immune system has evolved to fight against foreign pathogens. It plays a central role in the body's defense mechanism. However, the immune memory geared to fight off a previously recognized pathogen, tends to remember an original form of the pathogen when a variant form subsequently invades. This has been termed 'original antigenic sin'. This adverse immunological effect can alter vaccine effectiveness and sometimes cause enhanced pathogenicity or additional inflammatory responses, according to the type of pathogen and the circumstances of infection. Here we aim to give a simplified conceptual understanding of virus infection and original antigenic sin by comparing and contrasting the two examples of recurring infections such as influenza and dengue viruses in humans.

Identification of Tomato Aspermy Virus (TAV) and Chrysanthemum Virus B (CVB) from Dendranthema indicum in Korea

  • Chrysanthemums showing leaf mottling were collected from three southern locations in Korea in 1998. Two kinds of viruses were isolated from the leaves and were identified as tomato aspermy virus ch-TAV) and chrysanthemum virus B (ch-CVB), according to their host range, morphology, intracellular location, agar gel double diffusion test, and double-stranded RNA (dsRNA) analysis. The purified ch-TAV was spherical particles of approximately 29 nm in diameter and ch-CVB was filamentous particles of 685 nm long. Inclusion bodies were not observed in ch-TAV and/or ch-CVB infected chrysanthemum. ch-TAV showed positive serological reaction with TAV antiserum (ATCC-127) but not with CMV-pepper antiserum. In dsRNA analysis, four kinds of viral dsRNA were observed on ch-TAV and one viral dsRNA was shown on ch-CVB. Rate of co-infection with TAV and CVB in commercial chrysanthemums was 20.9%. On the other hand, infection with CVB alone was 97.2%. However, chrysanthemums naturally infected with TAV alone were not found.
