Study on the Mechanism of Vascular Relaxation of Methanol Extract of Rose multiflora Radix (장미근(薔薇根) 메탄올 추출물의 혈관이완 기전에 대한 연구)

  • Vascular tone plays an important role in the regulation of blood pressure. In the present study, the methanol extract of Rosae multiflora Radix (MRM) induced dose-dependent relaxation of phenylephrine-precontracted aorta, which was abolished by removal of functional endothelium. Pretreatment of the endothelium-intact aortic tissues with $N^G$-nitro-L-arginine methly ester (L-NAME) or 1H-[1,2,4]-oxadiazole-[4,3-${\alpha}$]-quinoxalin-1-one (ODQ) inhibited the relaxation induced by MRM, respectively. But, the relaxation effect of MRM was not blocked by indomethacine, glibenclamide, tetraethylammonium (TEA), verapamil, diltiazem, atropine, and propranolol, respectively. Moreover, incubation of endothelium-intact aortic rings with MRM increased the production of cGMP. Taken together, the present results suggest that MRM relaxes vascular smooth muscle via endothelium-dependent nitric oxide/cGMP signaling. These results would be useful for further study to MRM on animal models with cardiovascular diseases.

  • A variety of experiments concerning the development of ideal prosthetic grafts for correcting circumferential tracheal defects have been performed. The requirements for an ideal tracheal prosthesis are impermeability to air, consistency to prevent collapse, and acceptance by the host tissue causing a minimum inflammatory reaction, allowing fibroblastic infiltration and epithelialization. The synthetic material, polyurethane(PU), is known as a biocompatible polymer with an inert component. In this study, the tracheal prosthesis was made from microporous PU(30 micrometer in diameter) coated with gelatin and reinforced with isoplastic rings. This procedure provides the prosthesis with a compression strength. The out side diame er of the prosthesis was 20 mm with a length of 30 mm. The gelatin used in the study was obtained from pig skin and immobilized and cross-linked by irradiation(60 Co gamma ray) to promote host tissue incorporation and render the prosthesis epithelization after implantation. Animal experiments using 10 mongrel dogs were performed to compare three kinds of prosthesis; gelatin coated polyurethane graft, uncoated polyurethane graft, and prosthesisf pericadium complex graft. After 6 weeks of implantation, the epithelialization of implants was seen on the gelatin-coated and prosthesisfpericadium complex grafts. Implanted prosthesis were complicated by airway obstruction due to anastomosis granuloma. Early tracheal stenosis was found in the uncoated graft group. Two kind of anastomosis techniques were tested on the gelatin-coated prosthesis. Everted anastomosis resulted severe granuloma than the inverted anastomosis. In the prosthesislpericadium complex graft, bacteria and inflammation at a anastomotic site was found. Based on these results, gelatin coated porous polyurethane trachea prosthesis is biocompatible and may be useful in clinical application with further investigation.

High Ferrihydrite Turbidity in Groundwater of Samdong-Myeon (Ulsan) by Carbonate-Water Inflow of Deep Origin (심부 탄산수의 유업에 의한 울산시 삼동면 지하수의 높은 페리하이드라이트 탁도)

  • The turbidity in several wells of Samdong-myeon, Ulsan, exceeded potable groundwater standard (1 NTU). Mineralogical analysis showed that the fine suspended particles are ferrihydrite spheres with a size of less than $0.5\;{\mu}m$ and helical iron-oxidizing bacterial filaments, and their aggregates. Ferrihydrite was almost amorphous only showing two electron diffraction rings, and contained Si and P. Helical bacterial filaments were almost replaced by ferrihydrite. The helical bacteria have played an important role in the ferrihydrite formation by becoming the loci for ferrihydrite precipitation as well as oxidizing ferrous iron. The physicochemical conditions of low pH, low redox potential, high Ca concentration, and high alkalinity are consistent with the hydrogeochemical characteristics of carbonate groundwater, implicating that the inflow of deep ferriferous carbonate groundwater and its oxidation have caused the ferrihydrite turbidity in several wells of the study area.

AN EXPERIMENTAL STUDY ON THE FRACTURE STRENGTH OF CLASS II POSTERIOR RESTORATIONS (II급 와동에서 각종 구치용 수복물의 파절강도에 관한 실험적 연구)

  • The purpose of this study was to evaluate the fracture strength of class II restored premolars with amalgam, posterior composite, amalgam - Ketac silver, resin - Ketac silver restorations at marginal ridge. Fifty extacted maxillary and mandibular premolar teeth that were caries free, fracture free, and restoration free were selected and randomly divided into five groups : Group 1 : 10 intact teeth, Group 2 : 10 teeth with class II cavities and restored with, amalgam, Group 3: 10 teeth with class II cavities and restored with posterior resin, Group 4 : 10 teeth with class II cavities and restored with amalgam - ketac silver, Group 5 : 10 teeth with class II cavities and restored with resin - Ketac silver. All teeth were mounted in base of dental stone within metal rings of 2cm diameter, exposing only the crown portion. Class II mesio - occlusal or disto - occlusal cavities were prepared into specimens of Group 2 through 5 by using a No. 710 fissure bur. The occlusal portion was prepared to a faciolingual width of 1.5mm and a pulpal depth of 1.5mm. The proximal protion was prepared to a faciolingual width of 4mm, a occlusogingival height of 4mm, and a gingival floor of 1.5mm. The teeth in Group 2 and 3 were resotored with silver amalgam apd posterior resin respectively. In Group 4 and 5, proximal portions were first filled with Ketac silver 1.5mm gingivally and remaining cavities were restored with amalgam and posterior resin respectively. All specimens were stored in 100 % relative humidity at $37^{\circ}C$ for 48 hours before testing. All teeth were subjected to a compressive load in a Universal Instron Testing Machine at marginal ridges. The loads required to fracture the restorations were recorded in killograms and the data obtained were subjected to statisticall analysis. The results were all follows : 1. The fracture strength of Group 1 which were unprepared were $100{\pm}10.1\;kg$ and the higher values than Group 2, 3, 4, 5 which were prepared and resotred. 2. In restored groups, Group 2 had the higher fracture strength($81.8{\pm}12.4\;kg$) than other groups and Group 4 had the lowest fracture strength($66.8{\pm}9.2kg$). 3. There were significant differences between fracture strength of between Group 1 and Group 3, 4, 5(P<0.05), but not significant difference between fracture strength of Group 2, 3, 4, 5(P>0.05).

  • When $[U-^{14}C]$ 3,3', 4,4'-tetrachloroazobenzene$([U-^{14}C]\;TCAB)$ was added to the $MM_2$ medium as a sole carbon source for the isolated microorganisms and incubated, some radioactive metabolites were detected by autoradiography. No $^{14}CO_2$ was evolved from $[U-^{14}C]\;TCAB$ which was added as a sole carbon source to an organic matter-free soil inoculated by the isolates, wetted with the $MM_2$ salt medium, and incubated at $30^{\circ}C$. One of the metabolites in pure culture of Achromobacter group VD, which was isolated and identified, was tentatively identified as a compound of m/z 250 by means of GC/MS. The possible pathways for its formation are thought to include dechlorination from the TCAB structure, hydroxylation, ortho fission of the two benzene rings, and reduction of the resulting carboxyl group.

Biodegradation of Recalcitrant Chlorinated Aromatic Compounds via Microbial Dechlorination (미생물의 탈염소화 작용에 의한 난분해성 염화방향족 오염물질의 분해)

  • Chlorinated aromatic compounds are one of the largest groups of environmental pollutants as a result of world-wide distribution by using them as herbicides, insecticides, fungicides, solvents, hydraulic and heat transfer fluids, plasticizers, and intermediates for chemical synthesis. Because of their toxicity, persistence, and bioaccumulation, the compounds contaminated ubiquitously in the biosphere has attracted public concerns in terms of serious influences to wild lives and a human being, such as carcinogenicity, mutagenicity, and disturbance in endocrine systems. The biological recalcitrance of the compounds is caused by the number, type, and position of the chlorine substituents as well as by their aromatic structures. In general, the carbon-halogen bonds increase the recalcitrance by increasing electronegativity of the substituent, so that the dechlorination of the compounds is focused as an important mechanism for biodegradation of chlorinated aromatics, along with the cleavage of aromatic rings. The removal of the chlorine substituents has been known as a key step for degradation of chlorinated aromatic compounds under aerobic condition. This can occur as an initial step via oxygenolytic, reductive, and hydrolytic mechanisms. The studies on the biochemistry and genetics about microbial dechlorination give us the potential informations for microbial degradation of xenobiotics contaminated in natural microcosms. Such investigations might provide biotechnological approaches to solve the environmental contamination, such as designing effective bioremediation systems using genetically engineered microorganisms.

  • The spermatozoa of bagrid catfish, Pseudobagrus fulvidraco are approximately $76{\mu}m$ in length, and a relatively simple and elongated cell composed of a spherical head, a short middle piece and a tail. The ultrastructure of spermatozoa of P. fulvidraco is characterized by the following features. The acrosome is absent as in most teleost. The round nucleus measuring about $1.67{\mu}m$ in length and diameter is depressed with a deep nuclear fossa. The nuclear fossa, the length of which is about three-fifths of the nuclear diameter, contains the proximal and distal contrioles. The two centrioles are oriented approximately $160^{\circ}$ to each other. The filamentous materials give rise to satellite appendages arranged tangentially from the triplets of the distal centriole and the doublets of the anterior end of the axoneme toward the nuclear envelope. The mitochondria are not fused and their number is 20 or more. They are arranged in two or three layers and two rings within the cytoplasmic collar and surround the axoneme. They are separated from the axoneme by the cytoplasmic canal. The axoneme is of the 9+2 microtubular pattern and has inner but no outer dynein arms. The two lateral fins are in the same plane with the two central microtubules, the doublets 3 and 8, which are ultrastructural characteristics of the sperm tail unlike other siluroids lacking the lateral fins.

  • This study is the subsequent full-scale research of a TV drama writer who has been out of scholarly pursuits as it explores Su-Hyeon Kim's underlying consciousness with focusing on her heroines, the and . The author Su-Hyeon Kim clearly distinguishes a TV melodrama from a TV home-drama by her own self-control, which is a rare case in TV drama genres, therefore, her consciousness lights up at her melodrama. This study applies Lacanian theory to the author's melodramas for examining the author's under-lying thought. For Lacan the subject is an 'alienated', 'privative', 'fractured' 'being' as an imperfect language, the symbolic order, forms the subject through its signification. The subject desires the other's desire, and wants to become an object for the other's desire. The desire constantly demands an integral world, a perfect love, the wholly harmonious imaginary order. And it lasts up as it refuses the symbolic order's imperfection while it works its unconscious fantasy. Lacan states that only the 'traversing the fantasy', 'separation' would give birth to the real, liberated subject. Despite a 20-years of rift within two works, the and have an identical conflict core, that is a subject's constitutive, fundamental privation and desire of a human being. Su-Hyeon Kim's underlying consciousness complies with her continued theses of an inquiry into the subject's real liberation and freedom when desire of the rings produces the subject's alienation, privation, and the pursuit of a impossible perfect love.

The Growth of the Cultured Scallop, Patinopecten yessoensis (양식 가리비의 성장)

  • The seedlings of the scallop, Patinopecten yessoensis, which were collected in April 1980, April 1981 and May 1981, and the grown-up scallops collected in the sea bottom in November 1979, were reared by hanging method up to the July 1981. The growth of the seedlings collected in April 1980 was greatly affected by the time of the seedling collection, manipulation of the net cage, density in the net cage and the annual water temperature fluctuation. The scallops reared from the spat collected in April 1980 reached the sizes of 0.33 mm, 1.23 mm, 29.34 mm and 59.59 mm in shell length in 40,75, 285 and 450 days respectively. Since then, growth rate was determined as follows based on the age estimated by the year rings on the shell:84.96mm in 19 months, 99.3mm in 31, 112.3mm in 37 and 113.64mm in 43 months. The meat and the adductor muscle weight increased with the shell length. The meat weight roached about 15g when the shell length was 60-70mm and about 94.13 g when 130-140mm, and the adductor muscle weight reached about 4.89 g when the shell length was 60-70 mm and about 39.59g when 120-130 mm. But the growths of the meat and the adductor muscle weight were in stagnancy after scallops reached 125 mm in shell length.

  • Samples of Potamocorbula ustulata ustulata were collected monthly from October 2004 to November 2005 in the Jujin estuary of Gochang, Chollabuk-do, west coast of Korean peninsula. Age of P. ustulata ustulata was determined by the rings on the shell. The relationship between the shell length and the ring diameter in each ring group was expressed as a regression line. Therefore, there is a correspondence in each ring formation. Based on the monthly variation of the marginal index (MI') of the shell, it is assumed that the ring of this species was formed once a year during October to December. The relationship between the shell length (SL) and the shell height (SH; mm) was highly correlated with shell height as the following equation: SH = 0.6438 SL + 0.5642 ($r^2\;=\;0.978$). The shell length (SL) - shell width (SW) relation was also expressed by the following equation: SW = 0.4352 SL - 0.5675 ($r^2\;=\;0.957$). Shell length (SL; mm) and the total weight (TW; g) followed: $TW\;=\;6.999\;{\times}\;10^{-5}\;SL^{3.2542}(r^2\;=\;0.975)$. Growth curves for the shell length and the total weight fitted to the von Bertalanffy's growth curve were expressed respectively as: $$SL_t=30.77[1-e^{-0.4572(t+0.7371)}],\;TW_t=4.87[1-e^{-0.4572(t+0.7371)}]^{3.2542}.$$
