Antioxidant Activity and Ginsenoside Pattern of Fermented White Ginseng

  • Ethanol and water extracts of white and fermented ginseng were prepared and their ginsenoside composition and antioxidant effects were assessed. The main ginsenosides in white ginseng were $Rb_1$ > Re > $Rg_1$, and those in fermented ginseng were $Rb_2+Rb_3$ > Rd > $Rg_1$. Ginsenosides Rd and $Rg_3$ in fermented ginseng were enriched 11 and 58 times, respectively, over that in white ginseng through fermentation with five Bacillus spp. The greatest levels of 2-deoxyribose and superoxide anion dismutase-like activities were found in 50% ethanol extracts of fermented ginseng. Thus, these data suggest that white ginseng has the greatest free radical scavenging activity and that fermented ginseng has the highest antioxidant activity.

Component analysis of cultivated ginseng, cultivated wild ginseng, and wild ginseng and the change of ginsenoside components in the process of red ginseng (인삼.산양삼.자연산 산삼의 ginsenoside 함량 분석 및 홍삼화 후의 변화 관찰)

  • Objectives: The aim of this experiment is to provide an objective differentiation of cultivated ginseng, cultivated wild ginseng, and wild ginseng through component analysis, and to know the change of ginsenoside components in the process for making red ginseng. Methods: Comparative analysis of ginsenoside $Rb_1,\;Rb_2$, Rc, Rd, Re, Rf, $Rg_1,\;Rg_3,\;Rh_1$ and $Rh_2$ from the cultivated ginseng 4 and 6 years, cultivated wild ginseng, and wild ginseng were conducted using High Performance Liquid Chromatography(hereafter HPLC). And the same analyses were conducted in the process of red ginseng. Results: 1. For content comparison of ginsenoside $Rb_1$, Rc, Rd, Rf, $Rg_1$ and $Rh_1$, wild ginseng showed high content, followed cultivated ginseng 4 and 6 years, cultivated wild ginseng showed low content than any other samples. 2. For content comparison of ginsenoside $Rb_2$ and Re, cultivated ginseng 4 years showed high content, followed wild ginseng and cultivated ginseng 6 years, cultivated wild ginseng showed low content than any other samples. 3. For content comparison of ginsenoside $Rg_3$, wild ginseng and cultivated wild ginseng were only showed low content. 4. For content comparison of ginsenoside $Rh_2$, cultivated wild ginseng was only showed low content. 5. In the process of red ginseng, ginsenoside $Rb_1,\;Rb_2$, Rc, Rd, $Rg_3$ and $Rh_1$ were increased, and ginsenoside Re and $Rg_1$ were decreased in cultivated wild ginseng. 6. In the process of red ginseng, ginsenoside $Rg_3$ and $Rh_1$ were increased, and ginsenoside $Rb_2$, Rc, and Re were decreased in cultivated ginseng 4 years. 7. In the process of red ginseng, ginsenoside $Rb_1,\;Rb_2$, Rf and $Rh_1$ were increased, and ginsenoside Rc and Rd were decreased in cultivated ginseng 6 years. Conclusions: Distribution of ginsenoside contents to the cultivated ginseng, cultivated wild ginseng, and wild ginseng was similar and was not showed special characteristics between samples. And the change of ginsenoside to the process of red ginseng, cultivated ginseng and cultivated wild ginseng were showed different aspect.

Biological Activities and Chemical Characteristics of Monascus-Fermented Korean Red Ginseng (홍국발효홍삼의 이화학적 특성 및 생리활성)

  • This study investigated the biological activities and chemical characteristics of Monascus-fermented Korean red ginseng (MFRG). The comparative activities of water, ethanol, and methanol extracts from MFRGE and Korean red ginseng (RG) were tested in vitro of anti-oxidative models of linoleic acid peroxidation by thiocyanate and thiobarbituric acid (TBA) methods and DPPH ($\alpha,\alpha'$-diphenyl-$\beta$-picrylhydrazyl)radical scavenging activities. In addition, measurements of their bioactive total phenolic compounds and minerals, and extract yield, were obtained. The extract yield of each solvent extracted from MFRG and RG was aqueous by 6.58% and 5.83%, ethanol by 0.62% and 0.98%, and methanol by 1.27% and 3.04%, respectively. Total phenolic compounds were higher in all solvents extracted from MFRG than those from RG. Major mineral contents (ppm) of MFRG and RG were K by 16,936 and 22,386, Ca by 2,310 and 3,693, Mg by 2,703 and 2,647, respectively. The DPPH radical scavenging activities were higher in all solvents extracted from MFRG than those from RG, however, all these extracts exhibited a relatively low level of radical-scavenging activity compared to the butylated hydroxytoluene (BHT). In antioxidative activities determined by TBA method using linoleic acid peroxidation, 70% methanol extract from MFRG and RG showed the highest antioxidative activity at a concentration of 0.1%. These results may provide the basic data to understand the biological activities of bio-active materials derived from MFRG.

Bioconversion of Ginsenosides in Red Ginseng Extract by Saccharomycescerevisiae and Saccharomyces carlsbergensis (홍삼농축액에서 Saccharomyces cerevisiae와 Saccharomyces carlsbergensis에 의한 Ginsenosides의 bioconversion)

  • Ginseng(Panax ginseng C.A. Meyer) is reported to have many pharmaceutical activities. The minor ginsenosides(Rd, Rg3, Rh2 and compound K) display pharmaceutical properties superior to those of the major ginsenosides. These minor ginsenosides, which contribute a very small percentage, are produced by hydrolysis of the sugar moieties of the major ginsenosides. The pH of red ginseng extracts fermented with S. cerevisiae and S. carlsbergensis decreased rapidly during 3 days of fermentation, with no further significant change thereafter. After 20 days of fermentation, a relatively small difference remained in the acidity of extracts fermented with S. cerevisiae (0.54%) and S. carlsbergensis (0.58%). Reducing sugar in the S. cerevisiae and S. carlsbergensis extracts decreased from 25.86 to 4.54 mg/ml and 4.32 mg/ml glucose equivalents, respectively; and ethanol contents increased from 1.5% at day 0 to 16.0 and 15.0%, respectively, at 20 days. Ginsenosides Rb1, Rb2, Rc, Re, Rf, and Rg1 decreased during the fermentation with S. cerevisiae, but Rd and Rg3 increased by 12 days. Ginsenosides Rb1, Rb2, Rc, Re and Rg1 decreased gradually in the extract with S. carlsbergensis, but Rd and Rg3 were increased at 6 days and 9 days.

Evaluation of ginsenoside bioconversion of lactic acid bacteria isolated from kimchi

  • Background: Panax ginseng is a physiologically active plant widely used in traditional medicine that is characterized by the presence of ginsenosides. Rb1, a major ginsenoside, is used as the starting material for producing ginsenoside derivatives with enhanced pharmaceutical potentials through chemical, enzymatic, or microbial transformation. Methods: To investigate the bioconversion of ginsenoside Rb1, we prepared kimchi originated bacterial strains Leuconostoc mensenteroides WiKim19, Pediococcus pentosaceus WiKim20, Lactobacillus brevis WiKim47, Leuconostoc lactis WiKim48, and Lactobacillus sakei WiKim49 and analyzed bioconversion products using LC-MS/MS mass spectrometer. Results: L. mesenteroides WiKim19 and Pediococcus pentosaceus WiKim20 converted ginsenoside Rb1 into the ginsenoside Rg3 approximately five times more than Lactobacillus brevis WiKim47, Leuconostoc lactis WiKim48, and Lactobacillus sakei WiKim49. L mesenteroides WIKim19 showed positive correlation with b-glucosidase activity and higher transformation ability of ginsenoside Rb1 into Rg3 than the other strains whereas, P. pentosaceus WiKim20 showed an elevated production of Rb3 even with lack of b-glucosidase activity but have the highest acidity among the five lactic acid bacteria (LAB). Conclusion: Ginsenoside Rg5 concentration of five LABs have ranged from ${\sim}2.6{\mu}g/mL$ to $6.5{\mu}g/mL$ and increased in accordance with the incubation periods. Our results indicate that the enzymatic activity along with acidic condition contribute to the production of minor ginsenoside from lactic acid bacteria.

Antimutagenic Effects of Ginsenoside Rb$_1$, Rg$_1$ in the CHO-K1 Cells by Benzo[a]pyrene with Chromosomal Aberration Test and Comet Assay

  • The usage and types of chemicals are advancing, specializing, large-scaled increasing, and new chemical exposed workers are concerning to occupational disease. The generation of reactive oxygen in the body from carcinogen, mutation and DNA damage in cancer is protected by natural antioxidants (phytochemicals) with antimutagenic effect. There were many reports of ginsenoside Rb$_1$, Rg$_1$ grievances of the genetic mutation to suppress the effect confirm the genetic toxicity test with chromosomal aberration test and the Comet (SCGE) assay confirmed the suppression effect occurring chromosomal DNA damage. We had wanted to evaluate the compatibility and sensitivity between the chromosomal aberration (CA) test and the Comet assay. We used the CA test and Comet assay to evaluate the anti-genotoxicity of ginsenoside Rb$_1$ and Rg$_1$, in CHO-K1 (Chinese hamster ovary fibroblast) cell in vitro, composed negative control (solvent), positive control (benzo[a]pyrene), test group (carcinogen+variety concentration of ginsenoside) group. The positive control was benzo[a]pyrene (50 $\mu$M), well-known carcinogen, and the negative control was the 1 % DMSO solvent. The test group was a variety concentration of ginsenoside Rb$_1$, Rg$_1$ with 10$^{-8}$%, 10$^{-6}$%, 10$^{-4}$%, 10$^{-2}$%, 1%, 10%. In chromo-somal aberration test, we measured the number of cells with abnormally structured chromosome. In Comet assay, the Olive tail moment (OTM) and Tail length (TL) values were measured. The ratio of cell proliferation was increased 8.3% in 10$^{-8}$%, 10$^{-6}$%, 10$^{-4}$%, 10$^{-2}$%, 1%, 10% Rb$_1$ treated groups, and increased 10.4% in 10$^{-10}$%, 10$^{-8}$%, 10$^{-6}$%, 10$^{-4}$%, 10$^{-2}$%, 1% Rg$_1$ treated groups. In the CA test, the number of chromosomal aberration was decreased all the Rb$_1$ and Rg$_1$ treated groups. In the Comet assay, the OTM values were decreased in all the Rb$_1$ and Rg$_1$ treated groups. To evaluate the compatibility between CA and Comet assay, we compared the reducing ratio of chromosomal abnormalities with its OTM values, it was identified the antimutagenicity of ginsenoside, but it was more sensitive the CA test than the Comet assay. Ginsenoside Rb$_1$ and Rg$_1$ significantly decrease the number of cells with chromosomal aberration, and decrease the extent of DNA migration. Therefore, ginsenoside Rb$_1$, Rg$_1$ are thought as an antioxidant phytochemicals to protect mutagenicity. The in vitro Comet assay seems to be less sensitive than the in vitro chromosomal aberration test.

Quantitative Analysis of Ginsenosides in Red Ginseng Extracted under Various Temperature and Time (홍삼의 추출 시간 및 온도에 따른 Ginsenosides 함량 비교분석)

  • This study compared the contents of ginsenoside according to the extract conditions of red ginseng to provide basic information for developing functional food using red ginseng. According to the result, the content of crude saponin was highest in 72 hours of extraction at $82^{\circ}C$ (RG-823). The content of prosapogenin (ginsenoside $Rh_1,\;Rh_2,\;Rg_2,\;Rg_3$) was highest in 48 hours of extraction, and followed by 72 and 24 hours at $82^{\circ}C$. And at $93^{\circ}C$ the prosapogenin contents were highest in the order of 48 hours, and next in 24 and 72 hours. In addition, ginsenoside $Rb_1,\;Rb_2$ Rc and Re were not detected in 72 hours of extraction at $93^{\circ}C$ (RG-933) presumedly due to hydrolysis, but ginsenoside Rd, Rf and $Rg_1$ were detected as long as 72 hours of extraction. These results show that protopanaxatriol group is relatively more resistant to heat than protopanaxadiol group.

Comparison of Physicochemical Properties of Extruded Ginseng Samples

  • This study compared the physicochemical properties of root hair of white ginseng (WG), root hair of tissue cultured mountain ginseng (MG), root hair of red ginseng (RG) and extruded ginseng samples. The comparison of crude ash and total sugar resulted insignificant differences between extruded and raw samples. MG had a higher content of crude ash, crude protein, amino acids and polyphenolic compound than WG and RG; the total sugar and reducing sugar were highest in RG. Crude fat and acidic polysaccharide in RG and WG were similar to and higher than MG. Crude saponin of treated samples WG1 (moisture content 25%, barrel temperature $110^{\circ}C$) and WG3 (moisture content 35%, barrel temperature $110^{\circ}C$) were 9.80% and 9.73%, respectively, which were the highest among ginseng samples. In conclusion, the extrusion process can be applied to red ginseng manufacturing, and some characteristics of MG were higher than in RG and WG.

Change of Ginsenosides and Free Sugars in Seeds During Stratification and Seedling During Early Growth Stage of Panax ginseng (인삼의 종자개갑시와 묘생육초기의 Ginsenosides 및 유리당의 변화)

  • For the elucidation of saponin synthesis during ontogeny changes of ginsenosides and free sugars in seeds during stratification and seedlings in early growth stage were investigated with high performance liquid chrom-atography. Embryo plus endosperm at 40-day stratification showed 80% decrease of total saponin, disappear-ance of Rc, Rb$_2$ and Rb$_1$ and appearance of Rg$_3$ (probable) and 20-Glc-Rf (probable). Leaf ginsenoside F$_3$ was found not in fruit plup but seed and decreased during stratification. Both decomposition and synthesis of saponin seemed to occure during stratification. Ginsenosides in endosperm and embryo might be originated from fruit pulp by penetration. In seedling saponin appeared first in shoot and in root about one month later. Ginsenoside Rc, Rb$_2$, Rb$_1$ appeared in root at the last investigation (June 30) indicating normal saponin synthetic capacity of root. Saponin synthetic rate was twice in leaf than in root. Leaf ginsenoside F$_3$ was found in seedling root. Root saponin Rg$_3$ and 20-Glc-Rf were found in leaf and stem in seedling and decreased with growth suggesting that rate saponin is not such in certain growth stage. Total saponin content was negatively correlated with PT/PD in seeds and arial parts of seedling due to greater change of PD. than PT. Seed at 70days stratification showed high sucrose content. In seedling glucose was main sugar in stem all the while and sucrose in root at early stage while glucose, fructose and sucrose were found in leaf.

A Comparative Study of Image Classification Method to Detect Water Body Based on UAS (UAS 기반의 수체탐지를 위한 영상분류기법 비교연구)

  • Recently, there has been a growing interest in UAS(Unmanned Aerial System), and it is required to develop techniques to effectively detect water body from the recorded images in order to implement flood monitoring using UAS. This study used a UAS with RGB and NIR+RG bands to achieve images, and applied supervised classification method to evaluate the accuracy of water body detection. Firstly, the result for accuracy in water body image classification by RGB images showed high Kappa coefficients of 0.791 and 0.783 for the artificial neural network and minimum distance method respectively, and the maximum likelihood method showed the lowest, 0.561. Moreover, in the evaluation of accuracy in water body image classification by NIR+RG images, the magalanobis and minimum distance method showed high values of 0.869 and 0.830 respectively, and in the artificial neural network method, it was very low as 0.779. Especially, RGB band revealed errors to classify trees or grasslands of Songsan amusement park as water body, but NIR+RG presented noticeable improvement in this matter. Therefore, it was concluded that images with NIR+RG band, compared those with RGB band, are more effective for detection of water body when the mahalanobis and minimum distance method were applied.
