Effect of Light Intensity on Growth Characteristic and Flower Color Change of New Guinea Impatiens 'Fishlimp 149' (광도 차이에 따라 나타나는 뉴기니아 봉선화 생육 특성 및 화색 변화)

  • New Guinea impatiens (Impatiens hawkeri) shows very sensitive responses to different light conditions. Due to these phenomena, testers of DUS (distinctness, uniformity and stability) for granting plant variety protection right often have problems distinguishing genetic or physiological differences. New Guinea impatiens 'Fishlimp 149' was grown under several light intensities in a rain-sheltered vinyl house to observe differences of plant growth and flowering. As compared with the control (avg. $1,010{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$), treatments of shade-1 (avg. $599{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and shade-2 (avg. $88{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) showed increased plant height, plant width, leaf size and pedicel length. On the contrary, these growth parameters decreased in shade-3 (avg. $30{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) with the lowest light intensity. Shade-1 treatment enhanced flower characteristics such as flower diameter, upper petal width, side petal width and lower petal length. However, these characteristics were suppressed by lower light intensities than that in shade-1. Anthocyanin contents of shoot, leaf and pedicel decreased with increasing shading, but that of flower petal was the greatest of the shade-1 treatment. Shade-1 treatment showed the greatest Hunter a value analyzed by a colorimeter, and L and b values increased with increasing shading. Shade-1 treatment seemed to be provided the most proper light condition for DUS test of New Guinea impatiens. Additionally, anthocyanin accumulation on New Guinea impatiens during DUS test was due to not genetic differences but physiological phenomenon.

In vivo Antinociceptive Antiinflamatory and Antioxidative Effects of the Leaf and Stem Bark of Kalopanax pictus in Rats (음나무 잎 및 수피의 진통소염효과 및 아주반트로 유발된 산화적 스트레스에 대한 효과)

  • The leaves (KPL) of Kalopanax pictus (KP) are used as a vegetable or a functional food in Korean society. The stem bark (Kalopanacis Cortex, KPS) has been traditionally used to treat neurotic pain, rheumatoid arthritis and diabetic disease. This research was undertaken to demonstrate that the leaf extract of KP (KPL) has also the antinociceptive and antiinflammatory effects like the extract (KPS) of Kalopanacis Cortex and to compare the activity levels of several extracts obtained from KP. Antinociceptive and antiinflammatory effects were measured against the extracts described as followings; KPL-1 (the MeOH extract obtained from the leaf shoot of KP collected on May), KPL-2 (the MeOH extract from KP collected on June), KPL-3 (the MeOH ectract from KP with no thorns), KPS-1 (MeOH extract from KPS of a Korean habitat), KPS-2 (MeOH extract from KPS of a Chinese habitat). The antimociceptive test undertaken by acetic acid-induced writhing, hot plate-, and tail-flick methods using mice. The anti-inflammatory test was also undertaken by measuring the edema in the carrageenan-induced test. The order of activity potency in the antinociceptive and antiinflammatory assays was commonly shown as followings: KPL-3>KPS>1>KPS-2>KPL-1>KPL-2. This order was also observed in acetic acid-induced vascular permeability test. The antiinflammatory activity in carrageenan-induced assay was also observed as the following order: KPL-3>KPS- 1>PS-2>KPL-1>KPL-2. In addition, adjuvant-induced rats were used for a model to assess the oxidative stress. Treatment of the rat with the extracts reduced serum thiobarbituric acid-reactive substances (TBARS), hydroxy radical(OH) and superoxide dismutase(SOD) activity caused by FCA together together with the inhibition of hepatic TBARS level and lipofuscin content. The above finding suggests that the leaf extract has the antinociceptive and antinflammatory activity. It is also suggested that KPL-3 with more potent activity than other tested extracts could be developed for a new available biomaterial.

Single Oral Dose Toxicity Test of Coix lacryma-jobi var. ma-yuen Stapf Sprout in Sprague-Dawley Rats (의이엽 (薏苡葉)의 Sprague-Dawley 랫드를 이용한 단회경구투여 독성시험)

  • Objectives : 'Johyun' yulmoo which is a new variety of Coix lacryma-jobi var. ma-yuen Stapf sprout was developed and registered by Rural development administration in 2004. This variety was derived from the cross between single cross of Suwon-6 and Okayama and UCN300-25 as F1. It is characterized by early maturity, short plant height, a strong resistance, and a superior yield and is suitable for the central and northern regions. Accordingly, we were performed and evaluated single oral dose toxicity test of 'Johyun' yulmoo sprout (JYS) in Sprague-Dawley (SD) rats. Methods : Single oral dose toxicity test was performed using with male and female rats. Rats were divided into two groups: Group 1, vehicle-treated rats (Control); Group 2, JYS 5000 mg/kg-treated rats. JYS was orally administered to male and female rats at dose levels of 5000 mg/kg. Animals were monitored on the mortality, clinical signs, body weight changes, and necropsy findings for 14 days. groups : Group 1, vehicle-treated rats (Control); Group 2, JYS 5000 mg/kg-treated rats. Results : After oral treatment of JYS, we could not find any mortality at 5000 mg/kg. Compared with the control group, there were also no significant differences in clinical sign, weight changes, weight gain, and gross abnormalities in JYS 5000 mg/kg-treated group. Conclusions : Taken together, these results suggest that approximate lethal dose of JYS was considered as over 5000 mg/kg. Results from this study provide scientific evidence for the safety of JYS. Moreover, this study could be used as a basis for dose-setting data of the repeated dose 13-week oral toxicity test of JYS.

  • This study was carried out in order to reduce the amount of sawdust for recycling the generated manure from livestock farms, and to investigate the effects on the reducing usage of sawdust and quality of produced compost. To do this, a cylindrical horizontal composting device were used in the experiments and compost was analyzed for judging produce compost quality. The experiment was carried out separately under different cases of operational control conditions. The first case was produced by using sawdust and pig manure mixture(Test-1); the second case was produced by using sawdust, pig manure and the previously produced compost(Test-2). In the second case, Except for some heavy metal content, The water content and C/N ratio were found to be suitable for fertilizer process specification of the RDA(Rural Development Administration) and it was found to reduce the sawdust 1.25tons usage.

  • Maize is one of the world's three largest crops and has a long cultivation history, and is an important crop used for various purposes such as food, feed, and industrial raw materials. Recently, the agricultural environment is changing, in which the limit of cultivation of crops is shifted to the north due to the rise in temperature due to climate change. This study was conducted in experimental field of Suwon in 2022 by setting a seeding period earlier than the sowing time to establish the North Korean agricultural climatic zone and meteorological conditions. The test cultivars were silage cultivars, Kwangpyeongok and Dacheongok. As a priming test method, it was used to directly plant seeds in the field through immersion using 4mM zinc (Zn) and 2.5mM manganese (Mn), which are trace elements for seeds. The planting season was early on March 15th, April 1st, and April 15th. The number of days from sowing to silk stage of the two cultivars sown on March 15, April 1, and April 15 was 107, 93, and 85 days for Kwangpyeongok and 109, 95, and 87 days for Dacheongok, respectively. The seed priming test did not show any difference from the control group in the growth survey up to the middle stage of growth. In another test, low-temperature recovery was confirmed through nitrogen (2-5%) foliar fertilization after 3 days, 5 days, and 7 days in refrigeration (0 degrees), a selective low temperature treatment for com in the third leaf stage. As a result of this study, it was confirmed that the low-temperature damaged com treated at 0℃ showed the same growth as that of the untreated com through nitrogen foliar fertilization. These results suggest that urea foliar fertilization for low-temperature damage reduction of corn for silage in high-latitude climates will be helpful. In addition, through the results of the study, additional studies are needed on the recovery mechanism and field application through urea foliar fertilization.

Development of Wax-typed Pheromone Dispenser for Mating Discruption of the Oriental Fruit Moth, Grapholita molesta, and Its Application Technique (복숭아순나방(Grapholita molesta) 교미교란용 왁스형 페로몬방출기와 그 적용 기술 개발)

  • A wax-typed pheromone dispenser has been developed and applied to control outbreak of the oriental fruit moth, Grapholita molesta, in apple orchard. To optimize its application technique, this study analyzed effect of different amounts of the pheromone dispenser on mating disruption('MD') of G. molesta. Different pheromone dispenser amounts significantly influenced the MD effect assessed by cumulative male adult catches monitored respectively by sticky delta trap and food trap, and resulted in differential damage on host plants. In a field test during entire growing season, a standard amount(120 g per 0.117 ha) of wax-typed pheromone dispenser was proved to be effective to suppress outbreak of G. molesta adults and to prevent host plant damage as much as a current commercial MD product($Isomate^{(R)}$). This study also demonstrated an effectiveness of deployment of food trap barriers around MD-treated area to prevent immigration of mated females from outside untreated areas. These results indicate that the wax-typed pheromone dispenser can be applied to control field G. molesta populations and its co-application with fuod trap barriers would be optimal to maximize MD efficacy.

The Comparison in Daily Intake of Nutrients, Quality of Diets and Dietary Habits between Male and Female College Students in Daejeon (대전지역 일부 대학생의 영양소 섭취상태 식사의 질과 식습관에 있어서 남녀의 차이에 관한 연구)

  • This study was conducted to investigate the differences in daily nutrient intakes, dietary habits and nutrition knowledge between male and female college students. Male undergraduate students 004 subjects) and female undergraduate students (229 subjects) , enrolled at H University in Daejeon, were recruited for this study. The mean age of the subjects was 22.9 year in male and 20.4 year in female students. Daily intakes of energy and nutrients were calculated from the records of one day of dietary food intakes by 24-hour recall method, and general information, dietary habits and attitudes, food intake frequency and food preferences and knowledge for food and nutrition were surveyed through a questionnaire. About $70.1\%$ of male and $66.1\%$ of female students were in the normal range of BMI (18.5 - 23), and $25.2\%$ of male students were over-weighted in contrast to $27.7\%$ of female students were underweight. Males and females were taking $77.2\%$ and $77.9\%$ of RDA for energy, respectively, and $54.8\%$ of male and $48.0\%$ of female students were taking energy under $75\%$ of RDA. Many of them showed deficient intakes of calcium, iron, vitamin A and riboflavin. Average of MAR was 0.75 in male and 0.72 in female students. NAR for calcium, iron, vitamin A and riboflavin in male students were 0.55, 0.69,0.75 and 0.61, respectively, compared to 0.53, 0.51, 0.70 and 0.67 in female students. The nutrients, which have INQ less than 1, were calcium and riboflavin in male, compared to calcium, iron, and riboflavin in female students. There were no gender differences in meal regularity and meal skipping rates, but female students showed higher rates of skipping dinner than males (p < 0.001) About $50.8\%$ of female students were ingesting snacks 1 - 2 time/day, compared to $27.1\%$ of male students. For the food intake frequency, fruit group was significantly eating more for female than male students (p< 0.001) Though female students got higher scores for nutritional knowledge test (p < 0.001) than male students, they did not show better dietary habits or dietary attitudes than male students actually. Therefore, a more active and actual education program accustomed to the different genders and ages with focus on real changing of dietary behaviors needs to be developed and run in schools and local departments.

Genetic Diversity and Pathotypes of Xanthomonas orzyae pv. oryzae Isolated in Korea (국내 수집 벼흰잎마름병균의 유전적 다양성 및 병원형)

  • Xanthomonas oryzae pv. oryzae, causal agent of bacterial leaf blight (BLB) of rice, had been collected and identified using Biolog and fatty acid analysis. Epidemics of BLB had been occurred all the times at several rice cultivating areas in Korea in 1999-2004. Most X. oryzae pv. oryzae isolated in 1999 and 2002 belonged to Korean race K1, but more than 50% of the pathogen isolated in 2003 belonged to Korean race K3. Especially, most pathogens isolated in Jeonnam and Joenbuk provinces belonged to Korean race K3. Inoculation test of near isogenic lines (NIL) of rice carrying single resistance genes against BLB showed that many isolates belonging to Korean race 1 reacted differently to diverse resistant monogenic lines of rice. Southern blot analysis also showed that the bacterial pathogens belonged to the same race had different numbers of avirulence genes. This results suggested that each Korean race type may respond to many resistance genes of rice. All the K3 races isolated in Jeonnam and Joenbuk provinces were able to cause disease on Xa3 monogenic lines of rice. Since most rice cultivars cultivated in Jeonnam and Jeonbuk were carrying Xa3 resistance genes, the bacterial pathogens isolated in Jeonnam and Jeonbuk were likely to develop to adapt to Xa3 resistance gene. Together with avirulence gene patterns of the bacterial isolates and the results of disease reaction of monogenic lines of rice to them, Korean X. oryzae pv. oryzae was classified into 19 pathotypes. This newly classified pathotypes should help the breeding of new resistance rice cultivars in Korea.

A Survey on the Perception of Consumers to Develop Processing Products of Mushroom Processed Foods (버섯가공식품 제품 개발을 위한 소비자 인식조사)

  • This study was carried out to investigate the recognition, preference of mushrooms and mushroom processed foods among general consumers in order to develop new mushroom processed foods. The questionnaires are consists of general questions, health status, intake frequency of mushrooms, reason for prefer mushrooms, have purchased experience or not of mushroom processed foods and prefer mushroom processed foods to want development by age. A total 1173 questionnaires were analyzed for statistical analysis. The statistical analysis was completes using SPSS Win program (Version 14.0) for descriptive analysis and $X^2$-test. Main results of this study were as follow: Most of respondents prefer mushrooms and 28.5% of the respondents like Pleurotus eryngii most. The most reasons for the prefer mushrooms are freshness. The frequency of intake mushrooms was 6-8 times per months. The most consideration factor in purchasing mushrooms was freshness/hygiene. 55.6% of the respondents had purchasing experience of mushroom processed foods to eat. The most purchased mushroom processed foods were soup and hor cereal. The most consideration factor in purchasing mushroom processed foods was improvement of taste.

Profiles of Toxin genes and Antibiotic Susceptibility of Staphylococcus aureus Isolated from Perilla Leaf Cultivation Area (들깻잎 재배단지에서 분리한 Staphylococcus aureus의 독소 유전자와 항생제 감수성 분석)

  • Thirty one of Staphylococcus aureus isolated from perilla leaf cultivation areas in Miryang were investigated on the characteristics, such as enterotoxin genes and antibiotic susceptibility. Five toxin genes (sea, seb, sec, sed, and see) were examined by PCR method. Disc diffusion method was used to examine the antibiotic susceptibility of S. aureus by using 18 types of antibiotic discs with different concentrations. Among enterotoxin-encoding genes, sea and sed genes were co-detected from 4 isolates (12.9%), sed gene was founded in 9 isolates (29.0%), and see gene was founded in 1 isolate (3.2%). However seb and sec and tsst were not detected in any isolates. As a result of antibiotic susceptibility test, 7 isolates (22.6%) were resistant to 12 antibiotics (penicillin, ampicillin, oxacillin, amoxicillin-clavulanic acid, cefazolin, cephalothin, imipenem, gentamicin, tetracycline, ofloxacin, norfloxacin, and erythromycin). 2 isolates (6.5%) were resistant to 5 antibiotics (penicillin, ampicillin, amoxicillin-clavulanic acid, gentamycin, and telithromycin). MRSA (Methicilline Resistant Staphylococcus aureus) was founded in packing vinyl, hands, and perilla leaves.
