Radiographic diagnosis of canine heartworm disease without clinical signs (임상증세를 보이지 않는 개심장사상충증의 방사선학적 소견)

  • Dirofilaria immitis, the causative agent of canine heartworm disease, inhabits mainly in the pulmonary arteries and the heart of dogs and cats, causing circulatory and respiratory disorders. Although diagnosis of the disease is based on the presence of microfilaria in the peripheral blood or of specific antigens released from the adult worms into the peripheral blood, the severity of the infection and the assessment of disease progression are based on the clinical signs and radiographic image analysis. We analysed 12 mixed-bred Jindo dogs naturally infected with Dirofilaria immitis without any clinical signs and compared the radiographic images of the heart and the lung with the number of adult worms at necropsy. The dorsoventral radiographs of 12 infected dogs revealed that the right caudal lobar pulmonary arteries (RCaLPA) were dialated in 66.7% of dogs, whereas the main pulmonary artery segment was enlarged in 50.3% of dogs. The cranial lobar pulmonary arteries (RCrLPA) were dilated or pruned in 33.3% of dogs. All dogs displayed the interstitial lung pattern, while the vertebral heart size (VHS) was of normal range. Although a range of 9 to 166 adult worms (av. 45.6) was found in the pulmonary arteries, in the heart or in the vena cava, no correlation with the radiographic findings and/or with the clinical signs was observed.

Application of 16S rDNA PCR-RFLP Analysis for the Rapid Identification of Weissella Species (Weissella 속 유산균의 빠른 동정을 위한 16S rDNA PCR-RFLP 분석법의 적용)

  • A polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP) analysis was applied to detect and identify ten Weissella spp. frequently found in kimchi. The previously reported genus-specific primers designed from 16S rDNA sequences of Weissella spp. were adopted but PCR was performed at the increased annealing temperature by $4^{\circ}C$. The sizes of amplified PCR products and restricted fragments produced by AluI, MseI, and BceAI endonucleases were well correspond with the expected sizes. W. kandleri, W. koreensis, W. confusa, W. minor, W. viridescens, W. cibaria, and W. soli were distinguished by AluI and MseI and W. hellenica and W. paramesenteroides were identified by BceAI. W. thailandensis was distinguished when restriction pattern of other species was compared but identified by the single use of MspI.

Implementation of Object Identifier, Mobile RFID and QR Code Exploiting End-of-Life Treatment Information of Internet of Things Devices (사물인터넷 디바이스의 폐기 처리 정보를 활용한 객체 식별자, 모바일 RFID 및 QR 코드 구현)

  • In a situation in which around 50 million metric tons of electrical and electronic products is generated globally per year, the total installed base of Internet of Things (IoT) devices is projected to amount to around 75 billion worldwide by 2025. However, there is very little research on identification schemes for end-of-life treatment (EoLT) of IoT devices. To address this issue, this paper proposes new identifiers including EoLT information such as recyclability rate (Rcyc) and recoverability rate (Rcov) of an IoT device, recycling rate (RCR) and recovery rate (RVR) of each part in the IoT device, etc. and implements them by using object identifier (OID), mobile radio frequency identification (RFID) and quick response (QR) code. The implemented OID and mobile RFID can be used with cooperation of a remote server via communication networks and the implemented QR code can be used simply with a smartphone app.

Rotator cuff retear after repair surgery: comparison between experienced and inexperienced surgeons

  • Background: We hypothesized in this study that the characteristics of retear cases vary according to surgeon volume and that surgical outcomes differ between primary and revision arthroscopic rotator cuff repair (revisional ARCR). Methods: Surgeons performing more than 12 rotator cuff repairs (RCRs) per year were defined as high-volume surgeons, and those performing fewer than 12 RCRs were considered low-volume surgeons. Of the 47 patients who underwent revisional ARCR at our clinic enrolled in this study, 21 cases were treated by high-volume surgeons and 26 cases by low-volume surgeons. In all cases, the interval between primary surgery and revisional ARCR, degree of "acromial scuffing," number of anchors, RCR technique, retear pattern, fatty infiltration, retear size, operating time, and clinical outcome were recorded. Results: During primary surgery, significantly more lateral anchors (p=0.004) were used, and the rate of use of the double-row repair technique was significantly higher (p<0.001) in the high- versus low-volume surgeon group. Moreover, the "cut-through pattern" was observed significantly more frequently among the cases treated by high- versus low-volume surgeons (p=0.008). The clinical outcomes after revisional ARCR were not different between the two groups. Conclusions: Double-row repair during primary surgery and the cut-through pattern during revisional ARCR were more frequent in the high- versus low-volume surgeon groups. However, no differences in retear site or size, fatty infiltration grade, or outcomes were observed between the groups.

Evaluation of Lead Exposure Characteristics by Process Category and Activity (작업공정 및 활동에 따른 국내 작업장 납 노출특성 평가)

  • Objectives: The purpose of this study is to systematically identify situations where exposure levels are expected to be high by structuring domestic lead measurement data according to exposure processes and activities. Methods: Occupational exposure data on lead was collected from the results of the Evaluation of Reliability of Working Environment Measurement conducted by the government from 2019 to 2020. Lead exposure characteristics were analyzed by PROC (process category) and activity. The Risk Characterization Ratios (RCRs) of five PROCs according to ventilation type and lead content were evaluated using the MEASE (Metal's EASE) model. Results: The exposure data on lead (n=250) was classified into 12 PROCs and 12 activities, with an average concentration of 0.040 mg/m3 and about 14% exceeding the occupational exposure limit of 0.05 mg/m3. Processes with high exposure levels were PROC 7 (industrial spraying), 23 (open processing and transfer operations of molten metal), 24 (mechanical treatment), 25 (welding), and 26 (handling of powder containing lead). The results of evaluating RCR for the five PROCs were greater than 1 or close to 1 even if local exhaust ventilation was used. Conclusions: There is a possibility that the concentration of exposure is high in the casting and tapping of molten metal containing lead, mechanical treatment such as fracturing and abrasion, handling of powder, spraying, battery manufacturing, and waste battery recycling processes. It is necessary to implement chemical management policies for workplaces with such processes.

Effect of Chemotaxis on Nodulation in Bradyrhizobium-Soybean Symbiosis (근류균의 화학주성이 근류형성에 미치는 영향)

  • To research the effect of chemotaxis of Rhizobia toward the root exudate on nitrogen fixing ability in soybean Rhizobia symbiosis system. Root exudate from seedlings of Glycine max. L was collected aseptic conditions. B. japonicum KCTC 2422 induced the formation of symbiotic nitrogen fixing nodules on the root of soybean plant and possessed motility and chemotaxis toward the 2mM proline. LPN-100 mutant was $Nod^-$, $Che^+$, and LPN-101 was $Che^-$, $Nod^+$ strains. Physiological properties of mutants were similar to parent strain. The crude root exudate was tested for its chemotactic ability using the capillary tube method. Chemotactic responses of RCR 3407 toward crude root exudate were 2.2, 2.6, 2.9, those of KCTC 2422 were 2.3, 2.9, 3.0, respectively. The crude root exudate was fractionated into neutral, cationic and anionic fractions. Chemotactic responses of KCTC 2422 was least with anionic fraction, most with neutral and intermediate with cationic fraction. B. japonicum KCTC 2422 was attracted by carbohydrates, amino acids and carboxylic acid. Carbohydrates and amino acids were good chemoattractants and carboxylic acids were intermediate chemoattractants. The peak concentration was $10^{-3}M$ for ribose, glucose, glutamine, aspartic acid and carboxylic acids, with exception of xylose, arabinose, tryptophan, which elicited maximum responses at $10^{-4}M$. The formation of nodules and nitrogenase activity of soybean inoculated with KCTC 2422 was determined in 7days after inoculation, and those of LPN-101 was detected in 15days after inoculation, but LPN-100 didn't form of nodules in soybean plants.

  • Rotavirus is the main cause of severe diarrhea in infants and young children of the world. However, the frequency of genetic alterations makes it hard to control the prophylaxis. Therefore, continuous monitoring of the rotavirus's genetic change is inevitable to prevent disease prevalence and is useful in inventing an efficient vaccine. From January 2005 to December 2010, we investigated 11,607 stool samples of acute gastroenteritis patients in the Incheon metropolitan area. About 13.18% (1,530 stool samples) of all samples had a positive reaction against rotavirus using an antigen capture enzyme-linked immunosorbent assay (ELISA). Then, the 160 stool samples were searched for subtypes of group A rotavirus by using a reverse transcription polymerase chain reaction (RT-PCR) and a nested multiplex RCR. In P sub-typing, P8 (56.3%) was an extremely prevalent genotype, followed by P6 (21.3%), and P1A (10.0%). G1 (39.4%) was most widespread in the G subtype, followed by G4 (25.0%) and G3 (18.8%). G1P8 (35.5%) was the most common G and P subtype combination, followed by G4P6 (19.3%) and G3P8 (13.1%). These results might be useful data for understanding the epidemiological status of group A-rotavirus dispersion in the Incheon metropolitan area.

Isolation and Identifieation of Entomopathogenic Nematodes from Soil and Insect (토양과 곤충 사체로부터 곤충병원성 선충의 분리 및 동정)

  • Nematodes were isolated using silkwom trap through the investigation of 100 soil samples from various biotopes in Korea. The 30 nematode strains from soil and dead insects by the pathogenicity aganinst silkworms (Bombyx mori mori) and insect pests of Calliphora vomitoria, Pseufazetia separata, Palomena angulosa, and Melolontha incana. Mortailty of the silkworm larvae and pupae were as high as 100% by nematode infection, those of insect of pests were varied from 20 to 100%. The 30 strains of entemopathogenic nematodes were classified into five groups of Rhabditidae, Diplogatroidae, Heterorhabitidae, Steinernematidae, and Tylenchida by morphological criteria. The genetic relationships among the 30 nematode strains were analyzed by various RAPD bands with twenty primers. The 30 nematode strains were classified into six major subgroups on the basis of the genetic similarity coefficient of 0.853. The grouping by RAPD was agree with those of morphological taxa in discrimination of the higher group, however, was not completely agree in the subgroup. The family Steinernematidae belong to Rhabditida was clarified as closer to the Tylenchida, rather than the other Rhabditida of Heterorhabitidae, Rhabditidae, and Diplogatroidae in genetic distance valule. From the result of the morphological classification and RAPD of the genomic DNA showed that genetic relationship analysis furnish infurmation on phylogenetic classification and relationships of entomopathogenic nematodes. The application of genetic similarity will overcome the limitation of taxonomy and classification of morphologically simple nematode. Several primers were confirmed those utility of identification for individual nematode strains, the methods of molecular genetics secured the simplicity, rapidity and accuracy on the selection of entomopathogenic nematodes.

IDENTIFICATION OF PORPHYROMONAS ENDODONTALIS USING POLYMERASE CHAIN REACTION(RCR) (중합효소연쇄반응(Polymerase Chain Reaction)을 이용한 Porphyromonas endodontalis의 동정에 대한 연구)

  • Porphyromonas endodontalis, an anaerobic Gram negative cocobacillus which was known to be associated with the infected root canals and periapical lesions, is very difficult to culture and to detect by the traditional method in that it requires much time to induce the specific black pigmentation, and it is very sensitive to oxygen and the antibiotics added in the culture medium. In this study, the nucleotide sequences of the 'probe h' (0.73kb), one of the specific DNA probes top. endodontalis (ATCC 35406) which had been developed by our department, was determined and then a pair of primers for PCR amplification was fabricated to identify P. endodontalis. The plasmids containing 'probe h' were purified by $Wizard^{TM}$ Midipreps DNA Purification System (Promega Corp.), and the nucleotide sequences of the 'probe h' were determined by the dideoxy chain termination method using TaqTrack Sequencing System (Promega Corp.) and detected by fluorescent labelling method. The sense/antisense PCR primers were designed with computer software (Lasergene, DNASTAR Ind. PCR was done with a programmable GeneAmp PCR System 2400 (Perkin Elmer-Cetus Co.). Each sample containing the whole genomic DNA of P. endodontalis and other black-pigmented Bacteroides was itailly denatured at $94^{\circ}C$ for 5 min and then subjected to 30 cycles, each of them consisting of 60s at $94^{\circ}C$, 60s at $60^{\circ}C$, and 90s. at $72^{\circ}C$. The amplified DNA was resolved electrophoretically in a 1.0 % agarose gel in 1X TAE buffer, stained with EtBr, and photographed on a UV transilluminator. The results were as follows : 1. The nucleotide sequences of 'probe h' (743 base pairs) were obtained by dideoxy chain termination method, and from that results the specific primers to P. endodontalis (ATCC 35406), 'Primer H1/ Primer H2', were designed. 2. It has been found that 'Primer H1/H2' could detect P. endodontalis (ATCC 35406) using PCR. 3. The PCR system with this primers may be a powerful technique to amplify the specific sequences of 'probe h' of P. endodontalis (ATCC 35406) that produce distinct identification of it from other black-pigmented Bacteroides, and this could help us to determine the nature of periapical disease.

Comparative Study of Korean Workers' Exposure to Dichloromethane by Process Category between Work Environment Monitoring Program and ECETOC TRA (국내 디클로로메탄 제조·사용 사업장 근로자의 공정별 노출수준에 대한 작업환경측정값과 ECETOC TRA 모델값 비교연구)

  • Objectives: By law, companies in Korea must periodically measure workers' exposure to harmful chemicals (the system is called the Work Environment Monitoring Program (WMP)[a]) and report the results to the government. The government also measures exposure to monitor the WMP's reliability (called Reliability Assessment (RA) for WMP[b]). The issue is that measured data from these two sources are so different that the objectivity of WMP needs to be confirmed by comparing the results using the European Centre for Ecotoxicology and Toxicology of Chemicals' Targeted Risk Assessment (ECETOC TRA). Methods: Step 1: Data collection from WMP reports submitted by companies (n=586) and RA for WMP written by the government (n=33). Step 2: Data Standardization by key information included. Step 3: Data conversion to input-variables required to run the ECETOC TRA model, and run the model with specific data (n=514) which meet the predetermined exposure scenario. Step 4: Statistical data analysis by process category (PROC) and ventilation type from each source ([A] and [B]). Step 5: Additional analysis of any unexpected results. Results: The process categories of the production and handling of Dichloromethane were classified into 12 PROCs, and ten of them were selected to run ECETOC TRA. Modeled values tended to be higher than measured values from both sources. For the measured values from WMP, RCR distribution by PROC was narrow (0.197-0.267, 95% CI) and did not have a relationship with ventilation type, which differs from the tendency of the modeling result. Meanwhile, the measured values from RA for WMP were relatively widely distributed (0.301-1.177, 95% CI) by PROC. In particular PROCs (13,19) were high enough to exceed 1. Also, they become low with better ventilation types and appear differently depending on the ventilation type, similar to the model result. Conclusions: This study revealed that ECETOC TRA might have the potential to serve as a screening tool for exposure assessment and to be used as assistive method for WMP to estimate exposure. Further empirical study is required to confirm its availability as a screening tool.
